Consumption of a high-fat-high-sucrose diet partly diminishes mechanical and structural adaptations of cardiac muscle following resistance training

  • [Purpose] The purpose of this study was to investigate the effects of a high-fat high-sucrose (HFHS) diet on previously reported adaptations of cardiac morphological and contractile properties to resistance training. [Methods] Twelve-week-old rats participated in 12-weeks of resistance exercise training and consumed an HFHS diet. Echocardiography and skinned cardiac muscle fiber bundle testing were performed to determine the structural and mechanical adaptations. [Results] Compared to chow-fed sedentary animals, both HFHS- and chow-fed resistance-trained animals had thicker left ventricular walls. Isolated trabecular fiber bundles from chow-fed resistance-trained animals had greater force output, shortening velocities, and calcium sensitivities than those of chow-fed sedentary controls. However, trabeculae from the HFHS resistance-trained animals had greater force output but no change in unloaded shortening velocity or calcium sensitivity than those of the chow-fed sedentary group animals. [Conclusion] Resistance exercise training led to positive structural and mechanical adaptations of the heart, which were partly offset by the HFHS diet.

Regulatory Effects of Exercise and Dietary Intervention in Mitogen Activated Protein Kinase Signaling Pathways in Rats

  • As a central component of a novel protein kinase cascade, the activation of the mitogen-activated protein (MAP) kinase cascade has attracted considerable attention. We sought to determine the effect of exercise and diet on the activation of the extracellular-signal regulated protein kinase (ERK) 1/2 and the p38 MAP kinase pathways in rat soleus muscle. Forty-eight Sprague-Dawley rats were assigned to one of two dietary conditions: high-carbohydrate (CHO) or high-fat (FAT). Animals having each dietary condition were further divided into one of three subgroups: a sedentary control group that did not exercise (NT), a group that performed 8 weeks of treadmill running and was sacrificed 48 h after their final treadmill run (CE), and a group that was sacrificed immediately after their final routine exercise training (AE). A high-fat diet did not have any significant effect on phosphorylated and total forms of ERK 1/2 or p38 MAP kinase. In chronically trained muscle that was taken 48 h after the last training, phosphorylated ERK 1/2 significantly increased only in the FAT but not in the CHO groups. In the case of total ERK 1/2, it increased significantly for both groups. In contrast, both phosphorylated and total forms of p38 MAP kinase decreased markedly compared to sedentary muscle. In muscle that was taken immediately after a last bout of exercise, phosphorylated ERK 1/2 increased in both groups but statistical significance was seen only in the CHO group. Total ERK 1/2 in acutely stimulated muscle increased only in the CHO-AE group even though the degree was much lower than the phosphorylated status. Muscle that was taken immediately after the routine training increased in phosphorylation status of p38 MAP kinase for both dietary conditions. However, statistical significance was seen only in the CHO group owing to a large variation with FAT. In conclusion, a high-fat diet per se did not have any notable effect versus a high-carbohydrate diet on MAP kinase pathways. However, when diet (either CHO or FAT) was combined with exercise and/or training, there was differentiated protein expression in MAP kinase pathways. This indicates MAP kinase pathways have diverse control mechanisms in slow-twitch fibers.

Muscle Fiber, Connective Tissue and Meat Quality Characteristics of Pork from Low Birth Weight Pigs as Affected by Diet-Induced Increased Fat Absorption and Preferential Muscle Marbling

  • This study investigated how birth weight differences in piglets affected carcass and muscle fiber properties as well as meat quality at slaughter. Within litters, piglets were grouped according to their birth weight as either normal (NBW; 1.62-1.73 kg) or low (LBW; 1.18-1.29 kg). At 5 weeks of age, NBW piglets were randomly transitioned to control (C) or isocaloric high fat diets derived from non-dairy (HF), while LBW piglets were randomly transitioned to high fat diets derived from non-dairy (HF) or dairy sources (HFHD). Piglets were reared in individual pens under standardized housing and feeding conditions. Live weight was recorded weekly, and pigs were slaughtered at 12 weeks of age. Hot carcass weights, dressing percentages, lean meat yield, and primal cut proportions were determined. The m. longissimus thoracis was collected from the right side of the carcass for measurement of physical and chemical properties of meat and muscle fiber characteristics. Results indicated that LBW pigs compensated for their live weight compared to NBW pigs at 6 weeks of age. The mean muscle fiber diameter of LBW-HFHD group is significantly higher than NBW-C and NBW-HF group, and the type I muscle fiber diameter is significantly higher than NBW-C group. Dairy fat inclusion in LBW pig diet reduced carcass back fat thickness. This increased the calculated lean meat yield to be comparable to that of NBW pigs fed a commercial diet. Incorporating dairy-sourced high-fat into LBW pigs' diets appears to be an effective strategy for producing carcasses equivalent to NBW pigs.

흰쥐 골격근에서 안정화 이산화염소(Stabilized $ClO_2$)의 부패억제에 관한 현미경적 연구 (Microscopic Study of Decomposition-Inhibition in Stabilized $ClO_2$ Gas in Skeletal Muscle of Rat)

  • 강한 산화력과 소독효과로 인해 광범위하게 사용되고 있는 안정화 이산화염소(S-$ClO_2$)의 근육조직에 대한 부패억제 효과를 알아보고자 본 연구를 수행하였다. 본 실험을 위해 8주령 SD계 흰쥐 골격근(skeletal muscle)을 사용하였고, 안정화 이산화염소를 처리하지 않은 대조군과, 안정화 이산화염소의 분말과 수용액을 처리한 실험군으로 분류하여 광학 및 투과전자현미경으로 관찰하였다. 광학현미경 관찰 결과, 대조군 1일부터 근섬유막이 분리되기 시작하였고 핵의 붕괴가 관찰되었다. 2일군에서는 근섬유막이 붕괴되어 근섬유의 형태만 남아 있었고, 근원섬유도 붕괴되는 양상을 나타내었다. 3일군 이후 대조군의 경우는 대부분의 근섬유막과 결합조직의 붕괴로 근섬유의 구분이 불분명하였다. 실험군의 경우, 3일군에서 근섬유 사이 결합조직의 부분적인 붕괴가 관찰되었다. 그러나 4, 5일군에서도 3일군과 유사한 양상을 나타내었다. 한편, 전자현미경적 소견은 대조군의 경우 1일군부터 근섬유막의 붕괴로 근섬유의 경계가 불분명하였고, 미토콘드리아막도 부분적인 파괴가 관찰되었다. 2일째부터 근섬유막이 모두 붕괴되었고 근섬유 사이에서 세균이 관찰되었다. 3일군 이후는 근육의 형태를 구별할 수 없었다. 그러나 실험군의 경우, 1일과 2일군은 전형적인 근육의 미세구조를 관찰할 수 있었고, 3일군부터 부분적인 미토콘드리아막의 붕괴가 관찰되었다. 이후 시간경과에 따라 결합조직이 붕괴되어 동심원상의 구조물이 일부 관찰되었다. 따라서 본 연구결과, 대조군은 1일군부터 조직의 붕괴가 시작되어 3일군 이후는 대부분 붕괴되어 형태를 구분할 수 없었으나, 실험군의 경우는 3일군부터 부분적인 붕괴가 시작되었고 이후 시간이 경과되어도 전체적인 근육의 형태가 유지되어 안정화 이산화염소가 근육에 부패억제 효과가 있는 것으로 확인되었다.

Effects of Water-misting Sprays with Forced Ventilation after Transport during Summer on Meat Quality, Stress Parameters, Glycolytic Potential and Microstructures of Muscle in Broilers

  • Effects of water-misting sprays with forced ventilation after transport during summer on meat quality, stress parameters, glycolytic potential and microstructures of muscle in broilers were investigated. A total of 105 mixed-sex Arbor Acres broilers were divided into three treatment groups: i) 45-min transport without rest (T group), ii) 45-min transport with 1-h rest (TR group), iii) 45-min transport with 15-min water-misting sprays with forced ventilation and 45-min rest (TWFR group). The results showed the TWFR group significantly increased (p<0.05) initial muscle pH ($pH_i$) and ultimate pH ($pH_u$) and significantly reduced $L^*$ (p<0.05), drip loss, cook loss, creatine kinase, lactate dehydrogenase activity, plasma glucose content, lactate and glycolytic potential when compared with other groups. Microstructure of the muscle from TWFR group broilers under light microscopy showed smaller intercellular spaces among muscle fibers and bundles compared with T group. In conclusion this study indicated water-misting sprays with forced ventilation after transport could relieve the stress caused by transport under high temperature, which was favorable for the broilers' welfare. Furthermore, water-misting sprays with forced ventilation after transport slowed down the postmortem glycolysis rate and inhibited the occurrence of PSE-like meat in broilers. Although rest after transport could also improve the meat quality, the effect was not as significant as water-misting sprays with forced ventilation after transport.

말초신경 손상 백서모델에서 초음파 치료의 신경 및 근육 재생효과 (The Effect of Ultrasound Treatment on Regeneration Nerve and Muscle on Injured Peripheral Nerve of Rats Model)

  • The purposes of this study is to discuss and analyze the effect on the recovery from cut in sciatic nerve. This study used 9 weeks male rats of Sprague-Dawley family. Rat in groups 4 were treated with pulsed therapeutic ultrasound for 3 minutes. 3 times weekly at 3MHz respectively (intensity; $0.2W/cm^2,\;0.5W/Cm^2,\;10W/cm^2$); rat in group 1 received placebo ultrasound. In addition, changes of serum aspartate amino-transferase(AST) and creatine phosphokinase(CPK) levels were also demonstrated with diameter of individual muscle fasciculate and number of muscle fiber in each of three types of muscles located in gastrocnemius, soles. The results of comparing the changes in groups are as follows; 1. We found out that hypertrophic epineurium was present in sciatic nerve injured ultrasound treatment of groups. 2. In the gastrocnemius morphological investigation of the group I (control group), severe muscle atrophy were observed at the 7th days of the sciatic nerve injury. however, muscle atrophy of the group IV ($1.0W/cm^2$) were slightly recovered at the 14th days after treatment ultrasound. At the 28th days, muscular fibers were formed in polygon and were significantly recovered. 3. C-fos immunoreactive of the group II ($0.2W/cm^2$), III ($0.5W/cm^2$) were remarkably increased at the 1th day after treatment of ultrasound. Group IV were markedly deceased. 4. Brain-Derived Neurotrophic Factor(BDNF) immunoreactive of the group II, III were increased after 7 days of the sciatic nerve injury. Group IV were markedly increased from 14th days to 28th days after treatment of ultrasound. 5. A significant increase of serum AST levels were demonstrated in control group. However, serum AST levels of massage groups were significantly decreased compared to that of control group in followed order ; ($0.2W/cm^2<0.5W/cm^2<1.0W/cm^2$). 6. A significant increase of serum CK levels were demonstrated in control of group. However, serum CK levels of massage groups were significantly decreased compared to that of control group in followed order ; ($0.2W/cm^2<0.5W/cm^2<1.0W/cm^2$). The above results suggest that ultrasound treatment after peripheral nerve injury might reduce noxious stimuli, facilitate nerve recovery and effective in the functional improvement delaying muscle atrophy or degeneration.

신경근육 접합부의 종판 폭과 분포에 따른 운동단위 수의 추정에 관한 연구 (A Study on Estimation of Numbers of Motor Unit related to the Widths and Distribution of Endplate in Neuromuscular Junction)

  • 본 논문에서는 표면 근전도(surface electromyogram : SEMG)와 근육모델링을 이용하여 신경근육 접합부(neuromuscular junction, NMJ)의 종판(end plate) 폭(widths)과 분포(distribution)에 따른 운동단위(motor unit, MU)수를 추정하는 새로운 방법을 제안하였다. 이를 위하여 MU-시뮬레이터(motor unit simulator)와 EPZ-시뮬레이터(end plate zone simulator)를 설계하고, 본 연구에서 제안된 방법과 기존방법들을 비교하였다. 제안된 MU-시뮬레이터로 시뮬레이션 된 SMUAP(single motor unit action potential : 단일운동단위활동전위)와 CMAP(compound muscle action potential : 복합근활동전위)은 검출된 근신호와 유사하였다. EPZ-시뮬레이터는 신경근육 접합부의 종판 폭과 분포를 바꾸어 가면서 운동단위수를 추정하기 위하여 설계하였다. 실험결과 운동단위 수는 약 450 개, 근섬유수 약 340 개, 종판 폭은 약 6 mm이고, 종판분포는 불규칙하게 분포된 것 (randomly distributed)으로 추정되었다. 본 연구에서 제안된 방법은 인간 근육의 생체조직검사로 측정한 운동단위의 수와 비교 가능한 결과가 나왔다.

흰쥐에서 허혈시간에 따라 재관류후 나타나는 근조직의 미세구조 변화에 allopurinol이 미치는 영향 (Effect of Allopurinol on Ultrastructural Changes in Ischemia Reperfusion Injury to Skeletal Muscle of Rats After Graded Periods of Complete Ischemia)

  • It has been well known that ischemia and reperfusion injury to skeletal muscle following an acute arterial occlusion causes significant morbidity and mortality. The skeletal muscle, which contains high energy phosphate compounds, has ischemic tolerance. During the ischemia, the ATP is catalyzed to hypoxanthine anaerobically and hypoxanthine dehydrogenase is converted to xanthine oxidase. During reperfusion, the hypoxanthine is catalyzed to xanthine by xanthine oxidase under $O_2$, presence and that results in production of cytotoxic oxygen free radicals. These cytotoxic free radicals, $O_2^-,\;H_{2}O_2,\;OH^-$, are toxic and make lesions in skeletal muscle during reperfusion. The authors perform the present study to investigate the effects of allopurinol, the inhibitor of xanthine oxidase, on reperfused ischemic skeletal muscles by observing the ultrastructural changes of the muscle fibers. A total of 48 healthy Sprague-Dawley rats weighing from 200 g to 250 g were used as experimental animals. Under urethane(3.0mg/kg., IP) anesthesia, lower abdominal incision was done and the left common iliac artery were ligated by using vascular clamp for 1, 2 and 6 hours. The left rectus femoris muscles were obtained at 6 hours after the removal of vascular clamp. In the allopurinol pretreated group, 50mg/kg of allopurinol was administered once a day for 2 days and before 2 hours of ischemia. The specimens were sliced into $1mm^3$ and prepared by routine methods for electron microscopic observations. All preparations were stained with uranyl acetate and lead citrate, and then observed with Hitachi -600 transmission electron microscope. The results were as follows: 1. In 1 hour ischemia/6 hours reperfused rectus femoris muscles of rats, decreased glycogen particles and electron density of mitochondrial matrix and dilated terminal cisternae are seen. In 2 hours ischemia/6 hours repersed rectus femoris muscles of rats, mitochondria with electron lucent matrix, irregularly dilated triad and spheromembranous bodies are observed. In 6 hours ischemia/6 hours reperfused rectus femoris muscles of rats, irregularly arranged myofibrils, and many spheromembranous bodies, fat droplets and lysosome are seen. 2. In 1 hour ischemia/6 hours reperfused rectus femoris muscles of rats pretreated with allopurinol, decreased glycogen particle and dilated cisternae of sarcoplasmic reticulum and triad are observed. In 2 hours ischemia/6 hours reperfused rectus femoris muscles of rats pretreated with allopurinol decreased electron density of mitochondrial matrix and spheromembranous bodies are seen. In 6 hours ischemia/6 hours reperfused rectus femoris muscles of rats pretreated with allopurinol, mitochondria with electron lucent matrix, spheromembranous bodies and dilated cisternae of sarcoplasmic reticulum and terminal cistern are observed. The results suggest that the allopurinol attenuates the damages of the skeletal muscles of rats during ischemia and reperfusion.

성장 단계에 따른 한국 재래 오골계 근육의 조직학 및 생리학적 특성 (Histochemical and Physiological Characteristics during Korean Native Ogol Chicken Development)

  • 본 연구는 한국 재래 오골계의 성장에 따른 조직학적 및 생리학적 특성을 분석하기 위해 수행하였다. 계군들의 체중, 흉근과 가자미근의 무게 측정 및 시료의 채취는 부화 직후 및 3, 5, 15주령에 이루어졌다. 근섬유 특성은 부화 직후에서 15주령까지의 오골계 흉근과 가자미근을 이용하여 분석하였고, DNA, RNA 및 단백질 함량은 왼쪽 흉근에서 측정하였다. 체중의 상대적인 증가율은 부화 직후에서 3주령까지의 기간에 가장 크게 나타났다. 가슴의 무게는 부화 직후에서 3주령까지의 기간에 약 24.46배가 증가했으며, 가슴 중 흉근도 동일한 기간 동안 약 31.14배 증가하였다. 소퇴부 및 가자미근의 무게 역시 부화 직후에서 3주령까지의 시기에 뚜렷한 증가가 나타났다. 모든 근섬유의 단면적은 성장 기간 동안 증가하는데, 15주의 실험 기간 동안 흉근의 근섬유 단면적은 65배 증가($34.06\;{\pm}\;3.08$에서 $2238\;{\pm}\;177\;{\mu}m^2$)하였다. 흉근의 근섬유 단면적은 부화 직후에서 3주령까지의 기간에는 약 18배, 3주령에서 5주령까지의 기간 동안에는 약 1.6배 증가를 보였다. 따라서 흉근의 근섬유 단면적과 무게 역시 3주령 때까지 가장 큰 증가를 보임을 알 수 있다. 핵산 및 단백질의 함량을 분석한 실험에서도 부화 직후에서 3주령 사이에 가장 큰 증가가 나타났는데, 부화부터 3주령까지의 기간 동안 각각 DNA 총량은 13배, RNA 총량은 21배, 단백질 총량은 30배 증가한 것으로 나타났다.

익지인(益智仁), 두충(杜沖), 백강잠(白殭蠶) 혼합추출물이 남성갱년기 증상 개선에 미치는 영향 (Effects of Fructus Amomi Amari, Eucommiae Cortex, Bombyx Batryticatus Extract on Improving Symptoms of Late-onset Hypogonadism)

  • In recent times, the number of men with late-onset hypogonadism has increased, and interest on this topic has also increased. This study was conducted to investigate effects of the mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus on improve late-onset hypogonadism. The experimental subjects consisted of three groups: a control group consisting of 8-week-old male ICR mice that had undergone no treatment, an aging-elicited group (AE group) consisting of 50-week-old ICR male mice that had undergone no treatment, and a Mixed herbal extract treatment group (MT group) consisting of 50-week-old ICR male mice that had undergone the mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus treatment (0.1 g/kg/day) for 6 months. After the experiment, the mice from all the experimental groups were dissected, and they were analyzed through histochemical and immunohistochemical methods. The mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus reduces aging-induced cell damage and oxidative stress and increases the secretion of serotonin and B-endorphin in aged mice, and promotes spermatogenesis in seminiferous tubules and reduces apoptosis and oxidative stress, and increases androgen receptor, $17{\beta}-HSD$ and GnRH, increases the ratio of smooth muscle to collagen fibers in the corpus cavernosum, increases eNOS, decreases PDE-5 and oxidative stress in aged mice, so it improves depression, reproductive, sexual problems caused by Late-onset hypogonadism. the mixture extract of Fructus amomi Amari, Eucommiae cortex, Bombyx batryticatus inhibits the induction of osteoporosis by increasing decreased bone matrix distribution due to aging, increasing the activities of OPC and OPN, which are produced in osteoblasts, and decreasing RANKL, MMP-3 activity, increasing OPG activity. It also reduces muscle damage, oxidative stress, inflammation and apoptosis of muscle tissue, and increases Myo-D in the sartorius muscle of aged mice for improving muscle atrophy caused by by Late-onset hypogonadism.
