Mechanisms of Macromolecular Interactions Mediated by Protein Intrinsic Disorder

  • Intrinsically disordered proteins or regions (IDPs or IDRs) are widespread in the eukaryotic proteome. Although lacking stable three-dimensional structures in the free forms, IDRs perform critical functions in various cellular processes. Accordingly, mutations and altered expression of IDRs are associated with many pathological conditions. Hence, it is of great importance to understand at the molecular level how IDRs interact with their binding partners. In particular, discovering the unique interaction features of IDRs originating from their dynamic nature may reveal uncharted regulatory mechanisms of specific biological processes. Here we discuss the mechanisms of the macromolecular interactions mediated by IDRs and present the relevant cellular processes including transcription, cell cycle progression, signaling, and nucleocytoplasmic transport. Of special interest is the multivalent binding nature of IDRs driving assembly of multicomponent macromolecular complexes. Integrating the previous theoretical and experimental investigations, we suggest that such IDR-driven multiprotein complexes can function as versatile allosteric switches to process diverse cellular signals. Finally, we discuss the future challenges and potential medical applications of the IDR research.

  • For a given p-valent analytic function g with positive coefficients in the open unit disk $\Delta$, we study a class of functions $f(z) = z^p - \sum\limits{_{n=m}}{^\infty} a_nz^n(a_n{\geq}0)$ satisfying $$\frac 1 {p}{\Re}\;(\frac {z(f*g)'(z)} {(f*g)(z)})\;>\;\alpha\;(0{\leq}\;\alpha\;<\;1;z{\in}{\Delta})$$ Coefficient inequalities, distortion and covering theorems, as well as closure theorems are determined. The results obtained extend several known results as special cases.

  • Let A(p) be the class of functions $f\;:\;z^p\;+\;\sum\limits_{j=1}^{\infty}a_jz^{p+j}$ analytic in the open unit disc E. Let, for any integer n > -p, $f_{n+p-1}(z)\;=\;z^p+\sum\limits_{j=1}^{\infty}(p+j)^{n+p-1}z^{p+j}$. We define $f_{n+p-1}^{(-1)}(z)$ by using convolution * as $f_{n+p-1}\;*\;f_{n+p-1}^{-1}=\frac{z^p}{(1-z)^{n+p}$. A function p, analytic in E with p(0) = 1, is in the class $P_k(\rho)$ if ${\int}_0^{2\pi}\|\frac{Re\;p(z)-\rho}{p-\rho}\|\;d\theta\;\leq\;k{\pi}$, where $z=re^{i\theta}$, $k\;\geq\;2$ and $0\;{\leq}\;\rho\;{\leq}\;p$. We use the class $P_k(\rho)$ to introduce a new class of multivalent analytic functions and define an integral operator $L_{n+p-1}(f)\;\;=\;f_{n+p-1}^{-1}\;*\;f$ for f(z) belonging to this class. We derive some interesting properties of this generalized integral operator which include inclusion results and radius problems.

  • Let $Q_{n+p-1}(\alpha)$ denote the- dass of functions $$f(z)=z^{P}-\sum_{n=0}^\infty{a_{(p+k)}z^{p+k}$$ ($a_{p+k}{\geq}0$, $p{\in}N=\left{1,2,{\cdots}\right}$) which are analytic and p-valent in the unit disc $U=\left{z:{\mid}z:{\mid}<1\right}$ and satisfying $Re\left{\frac{D^{n+p-1}f(\approx))^{\prime}}{pz^{p-a}\right}>{\alpha},0{\leq}{\alpha}<1,n>-p,z{\in}U.$ In this paper we obtain sharp results concerning coefficient estimates, distortion theorem, closure theorems and radii of p-valent close-to- convexity, starlikeness and convexity for the class $Q_{n+p-1}$ ($\alpha$). We also obtain class preserving integral operators of the form $F(z)=\frac{c+p}{z^{c}}\int_{o}^{z}t^{c-1}f(t)dt.$ c>-p $F\left(z\right)=\frac{c+p}{z^{c}}\int_{0}^{z} t^{c-1}f\left(t \right)dt. \qquad c>-p$ for the class $Q_{n+p-1}$ ($\alpha$). Conversely when $F(z){\in}Q_{n+p-1}(\alpha)$, radius of p-valence of f(z) has been determined.

  • Let $T^{\alpha}_{\lambda}$(p, A, B) denote the class of functions $$f(z)=z^p-{\sum\limits^{\infty}_{k=1}}{\mid}a_{p+k}{\mid}z^{p+k}$$ which are regular and p valent in the unit disc U = {z: |z| <1} and satisfying the condition $\left|{\frac{{e^{ia}}\{{\frac{f^{\prime}(z)}{z^{p-1}}-p}\}}{(A-B){\lambda}p{\cos}{\alpha}-Be^{i{\alpha}}\{\frac{f^{\prime}(z)}{z^{p-1}}-p\}}}\right|$<1, $z{\in}U$, where 0<${\lambda}{\leq}1$, $-\frac{\pi}{2}$<${\alpha}$<$\frac{\pi}{2}$, $-1{\leq}A$<$B{\leq}1$, 0<$B{\leq}1$ and $p{\in}N=\{1,2,3,{\cdots}\}$. In this paper, we obtain sharp results concerning coefficient estimates, distortion theorem and radius of convexity for the class $T^{\alpha}_{\lambda}$(p, A, B). It is further shown that the class $T^{\alpha}_{\lambda}$(p, A, B) is closed under "arithmetic mean" and "convex linear combinations". We also obtain class preserving integral operators of the form $F(z)=\frac{p+c}{z^c}{\int^z_0t^{c-1}}f(t)dt$, c>-p, for the class $T^{\alpha}_{\lambda}$(p, A, B). Conversely when $F(z){\in}T^{\alpha}_{\lambda}$(p, A, B), radius of p valence of f(z) has also determined.

MHC Multimer: A Molecular Toolbox for Immunologists

  • The advent of the major histocompatibility complex (MHC) multimer technology has led to a breakthrough in the quantification and analysis of antigen-specific T cells. In particular, this technology has dramatically advanced the measurement and analysis of CD8 T cells and is being applied more widely. In addition, the scope of application of MHC multimer technology is gradually expanding to other T cells such as CD4 T cells, natural killer T cells, and mucosal-associated invariant T cells. MHC multimer technology acts by complementing the T-cell receptor-MHC/peptide complex affinity, which is relatively low compared to antigen-antibody affinity, through a multivalent interaction. The application of MHC multimer technology has expanded to include various functions such as quantification and analysis of antigen-specific T cells, cell sorting, depletion, stimulation to replace antigen-presenting cells, and single-cell classification through DNA barcodes. This review aims to provide the latest knowledge of MHC multimer technology, which is constantly evolving, broaden understanding of this technology, and promote its widespread use.
