An Improved Method for Multiresidue Analysis of Pesticides in Lettuce, Chinese Cabbage and Green Pepper by Gas Chromatography

  • For the improvement of gas chromatographic analysis of multiple pesticide residues in green pepper, lettuce and Chinese cabbage, multiresidue test mixtures (MRTMs) of 10 groups (ECD 5 groups and NPD 5 groups) and a recovery test mixture (RTM) of 18 compounds (11 compounds for ECD and 7 compounds for NPD) were established based on retention time and response to relevant detectors. A new extraction solvent (acetone: acetonitrile=1 : 9) and a clean up eluent (hexane: dichloromethane : acetonitile = 50 : 48.5 : 1.5) for solid-phase extraction (SPE) cartridge were selected to test two types of multiresidue methods (MRM I and MRM II). MRM II provided high recovery better than MRM I when RTM was tested Recovery experiment with MRTMs which was conducted using MRM II resulted in that more than seventy percents of compounds were recovered in the range of $50{\sim}140%$, while 9% of compounds were over 140% of recovery and only $7{\sim}8$ compounds failed to detect. MRM II, an improved method, could be employed for screening residues of 190 pesticides in those vegetables.

GC/NPD를 이용한 다성분 잔류농약의 동시분석 (Simultaneous Analysis of Multi-residual pesticides using GC/NPD)

  • Pesticides were extracted from samples with 70% acetone and methylene chloride in order, and then cleaned up via open-column chromatography apparatus packed with florisil, and finally analyzed simultaneously the organophosphorus pesticides using GC/NPD. Ultra-2 and Ultra-1 fused silica capillary columns were used to separate and identify the products. Recovery of most analytes from soybean sample, taken from pesticide residues well, was greater than(80%) for all except(6) analytes. This method can simultaneously determine multiple pesticides with a high degree of accuracy and precision.

GC/ECD와 NPD를 이용한 건강기능식품 주요 원료 중 다성분 잔류농약 분석 (Analysis of Multiple Pesticide Residues in Raw Materials Used in Dietary Supplements by GC/ECD and NPD)

  • 본 연구는 건강식품섭취 후 부작용 발생 현황과 건강기능식품 주원료 및 생산 및 출하액, 국내 생산 및 수입을 바탕으로 원료를 선정하고 공전시험법(2002)에 의하여 GC-ECD와 GC-NPD로 분석 가능한 농약(36종)에 대하여 분석한 후 GC-MS로 확인하여 분석하였다. 16종 139품목에 대하여 분석한 결과 19품목에서 농약이 검출되었으며, 국내산 원료와 수입 원료의 검출 현황은, 국내산 원료의 경우 61품목 중 12품목이 검출되었고, 수입 원료의 경우 78품목 중 7품목이 검출되었다. 국내산 원료인 인삼의 경우 13품목 중 3품목이 검출되었고, 운지버섯은 4품목 중 2품목, 구기자는 4품목 중 3품목, 오미자는 5품목 중 2품목, 천궁 4품목 중 2품목이 검출되었다. 수입 원료의 경우 인삼은 8품목 중 3품목이 검출되었으며, 홍삼은 4품목 중 1품목, 운지버섯 6품목 중 1품목, 쑥은 5품목 중 1품목, 천궁 5품목 중 1품목이 검출 되었다. 농약별 분포현황을 보면 총 16종 139품목에 대하여 불검출은 120건이었으며, chlorpyrifos는 13건으로 가장 높게 나타났다. 본 연구는 한정된 시료를 분석한 것이나, 건강기능식품에 사용되는 원료의 잔류농약 존재확인은 지속적인 모니터링의 필요성을 보여주고 있다. 또한 건강기능식품시장이 성장세를 이루고 있는 추세에 건강기능식품 섭취 후 부작용이 발생하였다는 원료에 대한 위해분석을 하는데 있어서의 기초 자료와 국내에서 기준 규격이 설정되어 있지 않은 식품 원료에 대한 기준 규격의 검토 자료로 활용 가능성이 기대된다.

High Performance Liquid Chromatography-Diode Array Detector(HPLC-DAD)에 의한 가미홍화탕 (KH-19)의 지문 분석 (Fingerprint of Marker Substances in Gami-Honghwa-Tang(KH-19) by HPLC-DAD)

  • Objectives : This study was aimed to evaluate marker substances in Gami-Honghwa-Tang (KH-19) by high performance liquid chromatography-photodiode array detector (HPLC-DAD). Gami-Honghwa-Tang is composed of nine crude herbs, Rehmanniae Radix Preparata, Angelicae Gigantis Radix, Cnidii Rhizoma, Paeoniae Radix, Corni Fructus, Moutan Cortex Radicis, Lycii Fructus, Carthami Flos and Glycyrrhizae Radix. Methods : The separation was performed on an Aquasil C18 (4.6×250mm) column by gradient elution with 0.05% TFA in H2O - 0.05% TFA in acetonitrile (0 min 100:0, 20 min 90:10, 40 min 70:30, 60 min 50:50, 80 min 0:100, 90 min 100:0) as the mobile phase at a flow-rate of 1.0 ml/min with detection at 190-800nm. Also we examined the contents for bacteria, pesticide residue and harmful heavy metals. Results : HPLC-DAD was employed to determine the quantities and the qualities of several marker substances such as 5­hydroxymethyl-2-furaldehyde (5-HMF), paeonol, loganin, paeoniflorin, glycyrrhizin, and decursin in the KH-19. There were no bacterial contents, pesticide residues, or harmful heavy metals. Conclusions : We suggest these results could be a useful evidence for quality control of KH-19. This method permits fingerprints of selected individual marker substances from herbal prescriptions without derivatization, multiple purification steps, or lengthy separation times.

배추에 살포된 몇 가지 농약제제에 따른 성분의 휘산과 비산 (Drift and Volatilization of Some Pesticides Sprayed on Chinese Cabbages)

  • 배추 재배시 살포된 농약의 비산 휘산정도를 알아보기 위해 유기인계인 chlorpyrifos, EPN, pirimiphos-methyl, phenthoate, 유기염소계인 tetradifon, dicofol, 피레스로이드계인 bifenthrin 농약을 이용하여 포장실험을 수행하였다. 이들의 잔류량 분석은 다성분 분석법을 이용하였는데 농약성분에 따라 다르나 회수율은 78${\sim}$92%로 우수하였으며 표준편차도 ${\pm}0.68{\sim}3.52%$로서 재현성이 있었다. 농약의 살포량은 농약사용지침서에서 권하는 량으로 하였으며, 1차실험시에는 바람이 불어오는 첫 구간에 농약을 살포하였고, 2차 실험시에는 풍향에 관계없이 정중앙에 살포하고 방사형으로 시료를 채취하였다. 실험결과 살포농약 대부분이 비산이나 휘산으로 인해 인접 작물에 전이 잔류하는 것을 확인하였다. 살포 중 비산되는 양은 농약성분과 풍향, 거리에 따라 달리 나타났으며 살포구간의 농작물 중 농약잔류량에 비하여 비살포구에서의 농작물 중 농약잔류량 비율은 3m거리에서 평균 0.66%, 6m 거리에서 0.10%, 9m 거리에서 0.05%이었고 근접지역에서는 최고 1.83%에 이르렀다.

심리측정 패러다임을 이용한 대구 지역 주부들의 식품 위해요소에 대한 위험 지각 수준 및 우려도에 영향을 미치는 요인 분석 (Analysis of Levels of Risk Perception using Psychometric Paradigm and Factors Affecting Concerns about Food Risk Elements of Housewives in Daegu)

  • This study examined the levels of risk perception of food risk elements by adopting a psychometric paradigm and analyzed factors affecting concerns about food risk elements to obtain basic materials for food safety policy. The data were collected from 296 housewives in Daegu, Korea, by a self-administered questionnaire. Frequency distributions, Pearson's correlation coefficient analysis, factor analysis, Cronbach's ${alpha}$, and multiple regression analyses were conducted by SPSS 21.0. The mean level of concern for food safety was 3.75/5.00 points, and the mean percentage of correct answers about heavy metal contamination was highest among food risk elements. The respondents perceived radioactive contaminated foods, GM foods, and endocrine disruptors as a new, delayed, scientifically unknown, involuntary, serious, and uncontrollable risk in risk perception. According to the result of factor analysis for risk perception, two factors such as non-controllability and dread were categorized. In the risk perception map, radioactive contaminated foods and GM foods were considered as an uncontrollable and dreaded risk, heavy metal contamination, endocrine disruptors, and pesticide residues as a controllable and dreaded risk, and foodborne illness and food additives as a controllable and less dreaded risk. On the other hand, the levels of concerns about food risk elements were higher in order of radioactive contaminated foods, GM foods, and endocrine disruptors. The results of multiple regression analysis showed that age, concern for food safety, percentage of correct answers about food risk elements, non-controllability, and dread influenced the concerns about food risk elements. These results imply that food safety policies should consider differences in consumer's risk perception of food risk elements.
