High-Frequency Interchange Network for Multispectral Object Detection (다중 스펙트럼 객체 감지를 위한 고주파 교환 네트워크)

  • Object recognition is carried out using RGB images in various object recognition studies. However, RGB images in dark illumination environments or environments where target objects are occluded other objects cause poor object recognition performance. On the other hand, IR images provide strong object recognition performance in these environments because it detects infrared waves rather than visible illumination. In this paper, we propose an RGB-IR fusion model, high-frequency interchange network (HINet), which improves object recognition performance by combining only the strengths of RGB-IR image pairs. HINet connected two object detection models using a mutual high-frequency transfer (MHT) to interchange advantages between RGB-IR images. MHT converts each pair of RGB-IR images into a discrete cosine transform (DCT) spectrum domain to extract high-frequency information. The extracted high-frequency information is transmitted to each other's networks and utilized to improve object recognition performance. Experimental results show the superiority of the proposed network and present performance improvement of the multispectral object recognition task.

A $G_{4}$ Sequence within PHR1 Promoter Acts as a Gate for Cross-Talks between Damage-Signaling Pathway and Multi-Stress Response

  • Rph1 and Gisl are damage-responsive repressors involved in PHR1 expression. They have two $C_{2}$H/ sub 2/ zinc finger motifs as putative DNA binding domains and N-terminal conserved domain with unknown function. They are also found in the human retinoblastoma binding protein 2 and the mouse jumonji- encoded protein. The repressors are able to bind to A $G_{4}$ sequence within a 39-bp sequence called upstream repressing sequence of PHR1 promoter (UR $S_{PHR1}$) responsible for the damage-response of PHR1. We report here that Rph1 is predominantly localized in the nucleus as examined by fluorescence microscopic analysis with GFP-Rph1 fusion protein. On the basis of the fact that the A $G_{4}$ sequence that is recognized by Rph1 and Gisl is also recognized by Msn2 and Msn4 in a process of stress response, we a1so tried to examine the in vivo function of A $G_{4}$ and the role of Msn2 and Msn4 in PHR1 expression. Our results demonstrate that Msn2 and Msn4 are actually required for the basal transcription of PHR1 expression but not for its damage induction. When A $G_{4}$ sequence was inserted into the minimal promoter of the cyc1-LacZ reporter, the increased LacZ expression was observed indicating its involvement in transcriptional activation. The data suggest that the A $G_{4}$ is primarily required for basal transcriptional activation of PHR1 or CYC1 promoter through the possible involvement of Msn2 and Msn4. However, since the A $G_{4}$ is also involved in the repression of PHR1 via Rphl and Gisl, it is proposed that A $G_{4}$ functions as either URS or upstream activating sequence (UAS) depending on the promoter context.t.

Hetero-core Spliced Fiber Optical Sensing System for an Environment Monitoring (구조물 모니터링을 위한 헤테로 코어형 광센싱 시스템)

  • A multi-purpose environmental monitoring system has been developed as a commercially available standard using the technique of hetero-core spliced fiber optic sensors, for the purposes of monitoring large-scale structures and preserving natural environments. The monitoring system has been tested and evaluated in a possible outdoor condition, in view of the full-scale operation at actual sites to be monitored. Additionally, the developed system in this work conveniently provides us with various options of sensor modules intended for monitoring such physical quantities as displacement, distortion, pressure, binary states, and liquid adhesion. Two channels of optical fiber line were monitored in each channel, three displacement sensor modules were connected in series, in order to examine the performance to a pseudo-cracking experiment in the outdoor situation and to clarify temperature influences an the system, in terms of the coupling of optical connectors and the OTDR stability. The results from the pseudo-cracking experiment agreed with the actual cracks, by means of calculation, based an the detected displacement values and their geometrical arrangement of the used sensor modules. The temperature change, ranging from 10 to $20^{\circ}C$ resulting from the 10-days free running operation, was found to influence the system stability of ${\pm}10{\mu}m$, primarily due to the coupling instability of the used optical connectors. It was found that fusion splicing, rather than the use of connectors, reduced the fluctuation dawn to ${\pm}2{\mu}m$. The specification and performance of various option modules have been demonstrated to show the capability of inspecting various physical quantities by use of the single system, which would be suitable for multi-purpose environmental monitoring.

Bactericidal Effect of Cecropin A Fused Endolysin on Drug-Resistant Gram-Negative Pathogens

  • The rapid spread of superbugs leads to the escalation of infectious diseases, which threatens public health. Endolysins derived from bacteriophages are spotlighted as promising alternative antibiotics against multi-drug resistant bacteria. In this study, we isolated and characterized the novel Salmonella typhimurium phage PBST08. Bioinformatics analysis of the PBST08 genome revealed putative endolysin ST01 with a lysozyme-like domain. Since the lytic activity of the purified ST01 was minor, probably owing to the outer membrane, which blocks accessibility to peptidoglycan, antimicrobial peptide cecropin A (CecA) was fused to the N-terminus of ST01 to disrupt the outer membrane. The resulting CecA::ST01 has been shown to have increased bactericidal activity against gram-negative pathogens including Pseudomonas aeruginosa, Klebsiella pneumoniae, Acinetobacter baumannii, Escherichia coli, and Enterobacter cloacae and the most affected target was A. baumannii. In the presence of 0.25 µM CecA::ST01, A. baumannii ATCC 17978 strain was completely killed and CCARM 12026 strain was wiped out by 0.5 µM CecA::ST01, which is a clinical isolate of A. baumannii and resistant to multiple drugs including carbapenem. Moreover, the larvae of Galleria mellonella could be rescued up to 58% or 49% by the administration of CecA::ST01 upon infection by A. baumannii 17978 or CCARM 12026 strain. Finally, the antibacterial activity of CecA::ST01 was verified using 31 strains of five gram-negative pathogens by evaluation of minimal inhibitory concentration. Thus, the results indicate that a fusion of antimicrobial peptide to endolysin can enhance antibacterial activity and the spectrum of endolysin where multi-drug resistant gram-negative pathogens can be efficiently controlled.

Use of Human Serum Albumin Fusion Tags for Recombinant Protein Secretory Expression in the Methylotrophic Yeast Hansenula polymorpha (메탄올 자화효모 Hansenula polymorpha에서의 재조합 단백질 분비발현을 위한 인체 혈청 알부민 융합단편의 활용)

  • The thermotolerant methylotrophic yeast Hansenula polymorpha is an attractive model organism for various fundamental studies, such as the genetic control of enzymes involved in methanol metabolism, peroxisome biogenesis, nitrate assimilation, and resistance to heavy metals and oxidative stresses. In addition, H. polymorpha has been highlighted as a promising recombinant protein expression host, especially due to the availability of strong and tightly regulatable promoters. In this study, we investigated the possibility of employing human serum albumin (HSA) as the fusion tag for the secretory expression of heterologous proteins in H. polymorpha. A set of four expression cassettes, which contained the methanol oxidase (MOX) promoter, translational HSA fusion tag, and the terminator of MOX, were constructed. The expression cassettes were also designed to contain sequences for accessory elements including His8-tag, $2{\times}(Gly_4Ser_1)$ linkers, tobacco etch virus protease recognition sites (Tev), multi-cloning sites, and strep-tags. To determine the effects of the size of the HSA fusion tag on the secretory expression of the target protein, each cassette contained the HSA gene fragment truncated at a specific position based on its domain structure. By using the Green fluorescence protein gene as the reporter, the properties of each expression cassette were compared in various conditions. Our results suggest that the translational HSA fusion tag is an efficient tool for the secretory expression of recombinant proteins in H. polymorpha.

3D Line Segment Detection using a New Hybrid Stereo Matching Technique (새로운 하이브리드 스테레오 정합기법에 의한 3차원 선소추출)

  • We present a new hybrid stereo matching technique in terms of the co-operation of area-based stereo and feature-based stereo. The core of our technique is that feature matching is carried out by the reference of the disparity evaluated by area-based stereo. Since the reference of the disparity can significantly reduce the number of feature matching combinations, feature matching error can be drastically minimized. One requirement of the disparity to be referenced is that it should be reliable to be used in feature matching. To measure the reliability of the disparity, in this paper, we employ the self-consistency of the disunity Our suggested technique is applied to the detection of 3D line segments by 2D line matching using our hybrid stereo matching, which can be efficiently utilized in the generation of the rooftop model from urban imagery. We carry out the experiments on our hybrid stereo matching scheme. We generate synthetic images by photo-realistic simulation on Avenches data set of Ascona aerial images. Experimental results indicate that the extracted 3D line segments have an average error of 0.5m and verify our proposed scheme. In order to apply our method to the generation of 3D model in urban imagery, we carry out Preliminary experiments for rooftop generation. Since occlusions are occurred around the outlines of buildings, we experimentally suggested multi-image hybrid stereo system, based on the fusion of 3D line segments. In terms of the simple domain-specific 3D grouping scheme, we notice that an accurate 3D rooftop model can be generated. In this context, we expect that an extended 3D grouping scheme using our hybrid technique can be efficiently applied to the construction of 3D models with more general types of building rooftops.

A Design of Development Process Model of Product Lines for Developing Embedded Software (임베디드 소프트웨어 개발을 위한 제품계열 중심의 개발프로세스 모델 설계)

  • Recently, the requirements of the embedded software are getting diverse as the diversity of embedded software application fields increases. The systematic development methods are issued to deal with the dependency between hardware and software. However, the existing development methods have not considered the software's close connection to hardware and the high-level reusability for common requirements of several similar domains. In this paper, we propose a design method of development process model of product lines to support an efficient development method for embedded software. For this, we firstly suggest a domain scoping method and an IDEF0(Integration DEFinition)-based business model for extracting the efficient requirements. Next, we present a component deriving method based on the service architecture and an architecture design method after considering the hardware dependency. And we explain the artifacts of MSDFS(Multi Sensor Data Fusion System) at each design step in order to show how the proposed model can be applied to the embedded software development.

A Study on the Measurement of the Relative Nuclear Reaction Cross-Section of the natW(p,xn)176Re Reaction using 100 MeV Proton (100 MeV 양성자를 이용한 natW(p,xn)176Re 핵반응의 상대 핵반응단면적 측정에 대한 연구)

  • This study derives the relative cross-section for the natW(p,xn)176Re nuclear reaction by measuring the gamma rays generated from the nuclear reaction with natural tungsten using a 100 MeV linear accelerator of the Korea Multi-purpose Accelerator Complex in the Korea Atomic Energy Research Institute. In general, research on isotopes with a short half-life always shows a tendency that the intensity of radioactivity decreases rapidly within a short period of time, making it very difficult to measure itself. In particular, 176Re is one of the relatively short radionuclides with a half-life of 5.3 minutes. In this study, 109.08 keV gamma rays generated from the 176Re isotope having such a short half-life were measured using a high-purity Ge detector(HPGe detector). The obtained relative measurements were the results in the 8 to 14 MeV proton energy domain published by Richard G. in 1967, and the TENDL-2019 value, which was the result of A. J. Koning in 2019, which evaluated the nuclear reaction cross-section by calculation based on this comparative analysis was performed. The results of this study are expected to be usefully applied to the design of nuclear fusion reactor which is known as future energy sources, elements ratio for the nuclear synthesis of astrophysics.
