상엽, 상지 및 잠분 에탄올 추출물의 품종별 세포독성 효과 (Cytotoxicity of the ethanol extracts of mulberry leaves, branches and silkworm feces)

  • The cytotoxicity of the ethanol extracts of varietal samples of mulberry leaves and branches and silkworm feces was measured using CT-26 cells originated from murine metastatic colon cancer, using dye uptake assay in order to find potential anticancer agents. Two ethanol extracts (varietal mulberry leaves and mulberry branches) were prepared from 16 varietal mulberries and used as partial extract materials for the activity assay. Among these, the ethanol extracts from Shinkwangppong leaves showed a little anticancer activity, and those from Sugaeppong, Cheongunppong and Gumsulppong branches showed some anticancer activity as well as cytotoxicity. In contrast, ethanol extracts from freeze-dried, the 3rd day of 5th instar feces showed more potent anticancer activity than that of other mulberry leaves, mulberry branches and other 5th silkworm instar larva feces on the basis of high UV absorbance at 665 nm.

지각과 상엽 혼합추출물이 지질 개선에 미치는 영향 (The Improvement of Mixed Extracts of Fructus Aurantii and Mulberry Leaves on Lipid Metabolism)

  • Background : Diabetes is a chronic disease characterized by deficiency of insulin. It has been reported that Fructus aurantii and mulberry leaves have the effect of anti-diabetes. Methods : To investigate the improvement of mixed extracts of Fructus aurantii and mulberry leaves on lipid metabolism, we performed double-blinded, randomized and placebo-controlled multi-center clinical trial. Results : Mixed extracts of Fructus aurantii and mulberry leaves significantly reduced total cholesterol. Conclusion : These results suggest that mixed extracts of Fructus aurantii and mulberry leaves improve lipid metabolism.

뽕잎과 민들레 추출물을 급여한 계육의 저장성에 미치는 영향 (Effects of Dietary Supplementation of Mulberry Leaves and Dandelion Extracts on Storage of Chicken Meat)

  • 본 시험은 육계에 뽕잎과 민들레 추출물을 각각 1%와 2%를 급여하여 5주간 사육한 계육의 pH, 육색, TBARS 및 VBN을 조사하였다. 실험구는 뽕잎과 민들레 추출물을 급여하지 않은 대조구, 뽕잎 추출물 1% 급여구는 T1, 뽕잎 추출물 2% 급여구는 T2, 민들레 추출물 1% 급여구는 T3 그리고 민들레 추출물 2% 급여구를 T4 등 5개 처리구로 나누어 사육한 후 도계하여 4주간 냉장 온도($4{\pm}1^{\circ}C$)에서 저장하면서 실험하였다. 계육의 pH는 저장 기간이 경과하면서 감소하였고(P<0.05), 대조구에 비해 뽕잎과 민들레 추출물 급여구에서 낮았으며, 특히 민들레 2% 급여구인 T4에서 가장 낮았다(P<0.05). 가슴살의 육색 중 $L^*$값과 $a^*$값은 저장 기간이 지남에 따라 일정한 변화가 없었고 $b^*$값은 증가하였으며, 다리살의 $L^*$값은 감소하였고, $a^*$값은 증가하였다. TBARS는 저장 기간이 지나면서 모든 처리구에서 증가하였고(P<0.05), 가슴살과 다리살에서 대조구보다 뽕잎과 민들레 추출물 급여구에서 낮았으며, 뽕잎과 민들레 추출물 급여에 의한 차이는 없었다. VBN 함량은 계육을 저장하는 동안 증가하였고, 대조구에 비해 뽕잎과 민들레 추출물 급여구에서 낮았으며(P<0.05), 뽕잎과 민들레 추출물 급여에 의한 차이는 없었다. 이상의 결과를 종합적으로 고찰해 보면 뽕잎과 민들레 추출물을 계육에 급여하면 pH, TBARS 및 VBN 함량이 낮아져 계육의 저장성 개선에 도움이 되리라 사료된다.

고탄수화물 식이 섭취 마우스에서 상엽 및 누에 추출물의 혈당강하 효과 (Blood Glucose Lowering Effects of Mulberry Leaves and Silkworm Extracts on Mice Fed with High-Carbohydrate Diet)

  • Mulberry leaves(Mori folium) and silkworm(Bombyx mori) are potnet inhibiters of intestinal $\alpha$-glycosidase, and inhibit the digestion of starch and sucrose in the small intestine. They are able to prevent postprandial hyperglycemia and decrease blood insulin levels. In this study , a high-carbohydrate diet(CHO ; 67.5%, protein ; 20.8%, fat : 11.7%) was received by the control group. In contrast, the experimental groups received a high-carbohydrate diet with extracts of mulberry leaves and silkwork(50mg.100g diet), and acarbose(6.7mg/100g diet). after a 10 week study period , the experimental groups had lower blood glucose and triglyceride levels. The experimental groups tended to have lwer Hb Alc levels. Also, blood insulin levels were lower than the control groups in accordance with blood glucose levels. The activities of intestinal $\alpha$-glucosidase in the middle and distal parts of small intestine were induced by the extracts of mulberry leaves and silkworm in the experimental groups. However, the activities of liver lysosomal glucosidase and the contents of glycogen in the liver were not affected by the mulberry leave and silkworm extracts nor by acarbose. Mulberry leaves and silkworm were able to prevent sudden postprandial peaks in blood glucose as a result of $\alpha$-glycosidase, inhibition, there by decreasing unnecessary insulin secretion.

뽕잎의 유산발효에 의한 Piperidine Alkaloid 함량 증진 (Enhancement of Piperidine Alkaloid Contents by Lactic Acid Fermentation of Mulberry Leaves (Morus alba L.))

  • This study was carried out to investigate solid-state fermentation method using cellulolytic lactic acid bacteria Lactobacillus plantarum TO-2100 in order to increase piperidine alkaloid contents in mulberry leaves. Piperidine alkaloid, one type of which include 1-deoxynojirimycin (1-DNJ), is known to inhibit ${\alpha}$-glycosidase activities. Using this strain, the optimal solid-state fermentation conditions on mulberry leaves powder were found as the following: initial moisture content, temperature and relative humidity were 20%, $30{\sim}35^{\circ}C$ and 60 ~ 70%, respectively, and the fermentation time was 72 hrs. The piperidine alkaloid contents in the fermented mulberry leaves were 2.86% on dry powder, which is 7-fold increase from that of non-fermented mulberry leaves. The 1-deoxynojirimycin contents after applying preparative thin layer chromatography were 2.02% on dry powder, which is 8 times higher than that of non-fermented mulberry leaves. ${\alpha}$-Glycosidase activities was inhibited by 65.7 ~ 84.7% with 3 ~ 5% treatments of hot-water extracts of the fermented mulberry leaves, compared to 16.2 ~ 40.2% with 3 ~ 5% treatments of hot-water extracts of non-fermented mulberry leaves. Therefore, the results suggest that solid-state fermentation method does indeed increase of piperidine alkaloid contents on mulberry leaves.

Antifungal activities of extracts from different parts of mulberry plant against Alternaria alternata and Fusarium sp.

  • In the present study, we investigated the antifungal activity of methanol and ethanol extracts of different parts (leaves, twigs, and root bark) of mulberry plant against Alternaria alternata and Fusarium sp. Among them, the methanol and ethanol extracts of mulberry root bark exerted the highest inhibitory activity against the mycelial growth of A. alternata ($70.6{\pm}1.6$ to $80.8{\pm}6.7%$ and $58.7{\pm}0.0$ to $80.8{\pm}6.7%$, respectively) and Fusarium sp. ($15.5{\pm}2.7$ to $39.3{\pm}3.4%$ and $26.4{\pm}2.7$ to $47.6{\pm}4.8%$, respectively). In contrast, the methanol and ethanol extracts from mulberry leaves and twigs did not suppress the mycelial growth of these fungal species. Importantly, the methanol and ethanol extracts of mulberry leaves tended to even accelerate the mycelial growth of A. alternata and Fusarium sp. Therefore, the results of this study indicate that methanol and ethanol extracts of mulberry root bark can be used as control agents against A. alternata and Fusarium sp.

Comparison with various mulberry leaves' and fruit's extract in lipid accumulation inhibitory effect at adipocyte model

  • In relation to the global increase trend of obesity population, there is a demand for the development of foods having high functional activity by mass-extracting anti-obesity active substances using mulberry product such as leaf and fruit. Therefore, we evaluated the antiobesity efficacy according to varieties by using the mulberry leaves and fruit extracts. At the same time, high active varieties were selected. For this purpose, the effects of the extracts of the mulberry leaf and fruit on 3T3-L1 adipocyte differentiation were examined. As a result, in the case of mulberry leaves, the lipid accumulation inhibitory rate of 'Cheongolppong' was higher than that of the control at $500{\mu}g/ml$ treatment. And in the case of the extract of mulberry fruit, 'Daesim' showed the highest lipid accumulation inhibitory rate compared with the control at 50 times of diluted extract.

품종이 다른 뽕잎추출물의 Helicobacter Pylori에 대한 항균활성 탐색과 항산화 효과 (The Antimicrobial Activity against Helicobacter pylori and Antioxidant effect from the extracts of Mulberry leaves (Morus Alba L.))

  • 108종의 표본묘목으로부터 채취한 뽕나무잎을 이용하여 항산화효과와 Helicobacter pylort에 대한 억제효과를 측정하였다. 항산화효과 중 DPPH에 대한 전자공여능은 물 추출물에서는 양면상 등 13종의 뽕잎에서 80% 이상의 높은 전자공여능이 확인되었으며, 80% ethanol 추출물에서는 고수9 등 59종의 뽕잎에서 80% 이상의 높은 전자공여능이 확인되어, 물 추출물보다 상대적으로 높은 효과를 나타내었다. ABTS radical decolorlization을 측정한 결과 물 추출물과 80% ethanol 추출물 모두에서 대부분의 뽕잎 추출물들이 90% 이상의 높은 항산화 효과를 나타내었다. TBARS 값은 control값이 $0.493{\times}10^{-2}{\mu}M$로 나타났으며, 물 추출물에서는 청운뽕, 도내, 축무, 사천금, 수원상 1호, 불전 2호, 오노다마구와, 야마나까다까스께를 제외한 모든 품종에서 대조구보다 낮게 나타났으며, 80% ethanol 추출물에서도 대축면을 제외한 107종의 품종에서 대조구보다 낮은 TBARS 값을 나타내어 산화촉진인자를 binding하는 능력이 뛰어난 것으로 판단되었다. TBARS와 같이 지용성 물질의 항산화력을 나타내는 antioxidant protection factor(PF)에서는 많은 종류의 뽕잎 80% ethanol 추출물에서 2이상의 높은 PF를 나타내었으며, TBARS값과 PF값 모두 80% ethanol 추출물이 물 추출물보다 효과가 우수한 것으로 나타났다. 뽕잎 품종별 추출물로 Helicobacter pylori균을 저해하는 물질을 탐색한 결과 108종의 뽕잎 중에서 감락상, 다호조생, 신일뽕, 국상 27호, 개량십문자, 국부, 수중상, 백광 6호, 이상, 사천금, 영변추우, 사175, 당상 2호, 청목노상 및 수중상 등 15 품종의 뽕잎에서 저해환을 나타냈으며, 그 중 물 추출물에서는 다호조생 품종이 지름 13mm의 저해환을, 알코올 추출물에서는 청목노상 품종이 지름 15mm의 저해환으로 효과가 가장 우수하여 천연 항균제 및 생리활성 물질로의 이용가능성을 추측할 수 있었다.

식용대두유에 대한 뽕잎추출물의 항산화 작용 (The Antioxidative Activities of Mullberry leaves Extracts on Edible Soybean Oil)

  • Recently, the concern about safety and functional substances of foods are increased therefore antioxidant substances and plants which had pharmacological effect have been studied actively. It has been known that the mulberry leaf is effective in curing many diseases. Nowaday, the pharmachological effects of mulberry leaves on diabetes mellitus and their usage for many aspects were confirmed. Mulberry leaves are known for effective in prevention of diabetes mellitus, because of specific amino acids and fibers. In this study, methanol, hexane, chlorform, ethyl acetate, and butanol extracts obtained from mulberry leaves were added to soybean oil and they were stored for 30 days at $60{\pm}2^{\circ}C$ and peroxide value(POV) and conjugated diene value(CDV) were measured periodically. Results of this study were obtained as follows; 1. The POV of soybean oil after the addition of each mulberry leaves powder(MLP) extracts generally enhanced as the storage time was prolonged, so the POV of all samples was reached higher than 100meq./kg.oil after 10 days storage without the addition of butanol, methanol, ethylacetate, hexane extracts at 0.1% level. Especially, the POV of soybean oils including butanol extract was 87.35meq/kg.oil after 10 days storage and antioxidant activity of butanol extract was shown to be superior to that of BHT. The pattern of the changes of the CDV of soybean oil after the addition of MLP extracts at 0.02%, 0.05% and 0.1%, respectively, were almost constant during 10 days of storage and then rapidly increased during the rest of experimental periods. During 10 days of storage in case of 0.1% adding level, the antioxidant activities of the butanol extract was superior to that of the each MLP extracts.

뽕잎추출액코팅농도에 따른 뽕잎쌀밥의 품질에 관한 연구 (The Study on the Characteristic of Cooked Rice According to the Different Coating Ratio of Mulberry Leaves Extracts)

  • The purpose of this study was to assess the optimum coating ratio for rice, using various ratios of mulberry leaves extract, 1.0, 1.5, and 2.0%, and to determine the optimum ratio of added water, in proportion to the total weight of mulberry rice. The moisture content of the soaked rice, and the optimum water uptake rate, moisture content of the cooked rice, as well as its blue and color values, mechanical characteristics, internal structure and sensory evaluation, were analyzed. The statistical data analyses were completed using the SAS program. The results are summarized as follows: The moisture content of mulberry rice was less than that of raw rice. The average optimum water uptake of the soaked mulberry rice at the different water temperatures, 10, 20 and 30, was 20% of the total weight of the raw mulberry rice. As for the results of the sensory evaluation,; 140% water, in proportion to the total weight of raw mulberry rice, was judged to be the optimum. The average moisture content of the cooked mulberry rice was 45∼50%, but there was no significant difference in the various coating ratios. The blue value of the cooked mulberry rice awas highest on the first day of cooking. The L- and a-values decreased with increasing coating ratio, but the b-value increased under the same conditions. As for the mechanical characteristics,; the adhesiveness, hardness and springiness decreased during 2 days of storage. The internal structure of the mulberry rice, observed by SEM, showed a close structure on increasing the coating ratios of mulberry leaves extracts. It was concluded that the optimum coating ratio of mulberry rice and ratio of added water for cooking wereas 1.5 and 140%, respectively, in proportion to the total weight of raw mulberry rice.
