Stock Characterization of the Fleshy Prawn (Penaeus chinensis) in the Yellow Sea by Intraspecific Sequence Variation of the Cytochrome c Oxidase Subunit I Gene

  • To determine the amount of genetic variation among populations of Penaeus chinensis (Osbeck) in the Yellow Sea, 342 bp region of the mitochondrial cytochrome c oxidase subunit I gene was amplified and sequenced. Six haplotypes, which differ by from one to four nucleotide sustitutions, were detected from 34 individuals of 4 populations examined. Mean sequence divergence between pairs of haplotypes was $0.68\%$. Most individuals from 4 populations were shared by the most common genotype. This genotype was distributed evenly in the Korean and Chinese populations. This result is in accordance with findings observed using RFLPs analysis of mtDNA (Hwang et al., 1997). Therefore, it is suggested that P. chinensis should be treated as one unit stock in the Yellow Sea.

Phylogenetic Analysis of Korean Native Goats Based on the Mitochondrial Cytochrome b Gene (mtDNA Cytochrome b 유전자에 기초한 한국재래염소의 계통유전학적 분석)

  • The goal of this study was to verify the phylogenetic status of the Korean native goats (KNG). We determined the complete sequence of the mitochondrial cytochrome b gene in 48 goats among four populations. We also analyzed genetic variability within goats, and a phylogenetic analysis was performed by comparison with other country's goats. Three nucleotide substitutions were detected, and two of these were missense mutations that occurred due to a substitution of amino acid. Four haplotypes were defined from KNG. Three of these haplotypes were only found in the Chinese goat. However, the other haplotype was KNG-specific. In the phylogenetic analysis, four clades (A~D) were classified among domestic goats, and the KNG was classified into clade 1 that estimated as lineage A based on the D-loop sequence. Each haplotype from the KNG was clustered closely with that of the Chinese goat. The results of haplotype distribution and phylogenetic location suggest that strong gene flow occurred from China to the Korean Peninsula.

Intraspecific variation of gene structure in the mitochondrial large subunit ribosomal RNA and cytochrome c oxidase subunit 1 of Pyropia yezoensis (Bangiales, Rhodophyta)

  • Red algal mitochondrial genomes (mtDNAs) can provide useful information on species identification. mtDNAs of Pyropia / Porphyra (Bangiales, Rhodophyta) have shown diverse variation in their size and gene structure. In particular, the introns and intronic open reading frames found in the ribosomal RNA large subunit gene (rnl) and cytochrome c oxidase subunit 1 gene (cox1) significantly vary the mitochondrial genome size in Pyropia / Porphyra species. In this study, we examined the exon / intron structure of rnl and cox1 genes of Pyropia yezoensis at the intraspecific level. The combined data of rnl and cox1 genes exhibited 12 genotypes for 40 P. yezoensis strains, based on the existence of introns. These genotypes were more effective to identify P. yezoensis strains in comparison to the traditional DNA barcode cox1 marker (5 haplotypes). Therefore, the variation in gene structure of rnl and cox1 can be a novel molecular marker to discriminate the strains of Pyropia species.

Expression of Metallothionein mRNA in Cadmium Treated A549 Cell Line Derived from Human Lung Epithelial Cell (인간 폐포세포 유래 A549세포주에서의 Cadmium 처리에 의한 메탈로치오닌 유전자 발현)

  • Metallothionein gene expression activity of cadmium was investigated in a human lung epithelial cell line. Cells, grown to near confluence, were exposed to 0∼10 ${\mu}$M Cd metal for 6 hours. Cadmium did not cause morphological alteration in lung epithelial cells that are characteristic of cell damages such as cell shrinkage, detachment of the cell from its neighbors, cytoplasmic and chromatic condensation. However, metallothionein genes of MT-1 and MT-2 were rapidly induced in the treated cell measured by RT-PCR. Regarding the induction pattern of motallothionein mRNA, MT-1 mRNA was induced in a dependent manner. MT-2 mRNA induction, which was measured using oligo primers based on cDNA of human reticulocytes, seemed to be slightly increased in low doses but decreased at high concentration used in the experiment.

  • Sperm from mud loach (Misgurnus mizolepis) were electroporated in the presence of plasmid DNA, pRSV/luc or pMT/hGH over a range of field strength of 0-1,625 V/cm with capacitance from 0 to 1,000 ${\mu}F$, and the effects of electroporation on fertilization, hatching, early survival, and efficiency of gene transfer were investigated. Average fertilization rate, hatching rate and early survival rate up to yolk sac absorption of all experimental groups were not significuntly different (P>0.05). The proportion of fish carrying pRSV/luc based on the polymerase chain reaction (PCR) analysis was ranged from 0 to $20\%$, however, the values of gene transfer efficiency from the different eledctroporation conditions were not significantly different. PCR analysis of pMT/hGH transferred groups revealed that screening of pMT/hGH transferred fish by PCR was difficult because of significant nonspecific amplifications resulted from the homologous sequences in the genome of mud loach.

  • NAD4 as a gene encoding NADH dehydrogenase subunit 4 in the micodhondrion from maize has been cloned using RT-PCR and sequenced for examining RNA edited sites. Analysis of mt cDNA sequences showed the typical RNA editing patterns and unusual base changes as well;RNA editing from cDNA sequences occured base change from c to U in most cases, however transitions from t to g and G to A were also observed. Even though those editings appared to be occurred randomly, RNA edited sites showed mostly in exon 1 and exon 4 regions, when compared with NAD4 cDNA from wheat, locations of edited sites did not consistent with each other suggesting that the phenomenon of RNA editing occured randomly not site-specific manner.

  • MtDNA cyt b gene was used to investigate the geographic variation of 11 populations (106 individuals) of the planktonic developing, periwinkle Littorina brevicula, throughout Eastern, Western, and Southern coastal regions in Korea. The sequence of 500 base pairs and 13 different haplotypes were determined. Haplotype LbA was predominated through the populations studied with frequence of 0.877. Haplotypes were shown different frequencies in each coastal region (0.82, 0.90, and 1.00, respectively). enetic analysis of the 61 individuals of L. brevicula from the polluted and unpolluted sites yielded 8 distinct haplotypes. Haplotype LbA also was most common, and it was shared by 0.872 of frequency among specimens.

Low Genetic Diversity and Shallow Population Structure of the Japanese Halfbeak Hyporhamphus sajori Revealed from Mitochondrial DNA in the Northeast Asia (Mitochondrial DNA를 이용한 동북아시아 학꽁치 Hyporhamphus sajori의 유전적 다양성과 집단 구조)

  • This study was conducted to know the genetic diversity and population structure of Japanese halfbeak (Hyporhamphus sajori) in the Northeast Asia, using mitochondrial DNA control region. In the present study, a total of 70 individuals were collected from three locations of China (Liaoning), Korea (Tongyeong) and Japan (Wakasa Bay), and 47 individuals sequences from three locations of Japan (Wakasa Bay, Toyama Bay and Mikawa Bay) were downloaded from genbank. A total of 7 haplotypes were identified with 7 polymorphic sites from 358 bp length sequences. Haplotype and nucleotide diversity were very low and ranged from 0 to 0.295±0.156 and 0 to 0.0009±0.0011, respectively. Ancestral haplotype was shared by 94% individuals. An extremely low haplotype and nucleotide diversity, and starlike minimum spanning tree indicated that the species have undergone a recent population expansion after bottleneck. Pairwise FST values were low and there was no significant differences among populations suggesting a gene flow among the populations. Dispersal of the eggs with the aid of drifting seaweed and currents might be the major responsible factor for the genetic homogeneity.
