터너 증후군에서 신기형의 발생에 미치는 레닌-안지오텐신계 유전자 다형성의 영향 (Impact and Prevalence of Renin-angiotensin System Gene Polymorphism of Renal Anomalies in Turner Syndrome)

  • 목적 : 레닌-안지오텐신계는 신장의 성장과 발달에 중요한 역할을 하는 것으로 알려져 있으며, 안지오텐신 II는 신장형성기 동안 중요한 역할을 담당한다. 저자들은 터너 증후군 환자에서 신기형의 발생빈도를 조사하고, 신기형이 동반된 군과 동반되지 않은 군으로 분류하여 양군 사이에 레닌-안지오텐신계 유전자의 분포에 차이가 있는지를 조사하였다. 방법 : 말초혈액의 임파구를 사용하여 세포분 석학적 방법으로 진단된 33명의 터너 증후군 환자를 대상으로 신요로계의 기형여부를 초음파검사와 경정맥 요로 조영 검사, DMSA scan으로 진단하고, 더불어 안지오텐신전환효소의 유전자형, 안지오텐시노겐 유전자형, 안지오텐신 수용체유전자형의 분포를 핵산증폭법을 사용해 조사하였다. 결과 : 33명의 터너 증후군 환자 중 12명에서 신기형이 동반되어 전체적으로 36.4%의 빈도를 나타내었다. 핵형에 따른 신기형의 빈도를 살펴보면 전형적인 45,X형의 경우는 18명 중 8명에서 관찰되어 44.4%의 빈도를 나타내었고, 모자이시즘과 X 염색체구조상의 이상인 경우에는 15명 중 4명에서 관찰되어 26.7%의 빈도를 나타내어, 전형적인 45,X형의 경우에서 발생빈도가 높았으나 통계적으로 유의하지는 않았다(P>0.05). ACE 유전자형의 분포는 신기형 동반군에서 DD형이 0%, ID형이 73%, 그리고 II형이 27%이었으며, 신기형이 동반되지 않은 군에서는 DD형이 11%, ID형이 56%, II형이 33%로 양군 사이에는 ACE 유전자형의 분포는 유의한 차이가 없었다. AGT M235T 유전자형의 분포는 신기형 동반군에서 MM형이 82%, MT형이 8%, 그리고 TT형이 0%이었으며, 신기형이 동반되지 않은 군에서는 MM형이 56%, MT형이 33%, TT형이 11%로 양군 사이에는 AGT M235T 유전자형의 분포는 유의한 차이가 없었다. ATR 1 유전자형의 분포는 신기형 동반군에서 AA형이 91%, AC형이 9%, 그리고 CC형이 0%이었으며, 신기형이 동반되지 않은 군에서는 AA형이 94%, AC형이 6%, CC형이 0%로 양군 사이에는 ATR 1 유전자형의 분포도 유의한 차이가 없었다. 결론 : 터너증후군 환자에서 신기형의 동반율은 36.4%였으며, 45,X형에서 발생빈도가 높았으나 통계적으로 유의하지는 않았다. 신기형이 동반된 군과 동반되지 않은 군 사이에는 레닌-안지오텐신계 유전자의 분포는 모두 유의한 차이가 관찰되지 알았다.

형질 전환된 포플러에 대한 nos-NPT II 유전자의 기관별 발현 특성 (Organ Specific Expression of the nos-NPT II Gene in Transgenic Hybrid Poplar)

  • 임목을 대상으로 삽입된 외래 유전자의 공간적, 시기별 발현 특성을 이해하기 위한 기초연구로서 온실에서 생육 중인 형질전환된 2년생 잡종 포플러 (Populus alba X P. grandidentata) Hansen 클론을 대상으로 삽입된 외래 유전자의 발현정도를 각 기관별로 조사하였다. Agrobacterium binary vector pRT45, pRT102 및 pRT104에 의해서 형질전환된 3계통의 형질전환체 Tr15, Tr345, Tr665 모두는 선발가능한 표식 유전자로서 nos promoter-NPT II 유전자가 대상 식물체의 genome에 삽입되어 있으며, 그외에, pin2 promoter-CAT 유전자(pRT45), nos promoter-PIN2 유전자(pRT102), Cauliflower Mosaic Virus 35s promoter-PIN2 유전자(pRT104)가 3계통의 형질전환체에 제각각 삽입되어 있는 잡종 포플러이다. 이들 3계통의 형질전환 포플러 식물체의 DNA를 PCR 검정 기법을 이용하여 분석해 본 결과 선발 가능한 표식 유전자인 NPT-II가 삽입되어 있음이 입증되었다 발현 정도를 비교 분석하기 위해서 NPT-ELISA 검정을 실시하였다. 삽입된 NPT II 유전자는 형질전환된 포플러의 잎, 엽병, 형성층 조직, 줄기의 목질부, 뿌리에서 발현되었으며, 발현 정도는 형질전환된 식물체의 계통에 따라서, 그리고 형진전환된 식물체의 부위에 따라서 다양하게 나타났다. pRT45에 의해서 형질전환된 Tr15 형질전환체의 경우, 늙은 잎과 엽병에서 NPT II 유전자가 가장 높은 수준으로 발현되었으며, 어린 잎과 뿌리 조직에서 가장 낮게 발현되었다. 삽입된 외래 유전자가 각 식물체간에, 각 기관에 따라서 각각 상이한 발현 정도를 나타내는 이와 같은 결과는 형질전환된 식물체에 대한 효과적인 선발과정이 요구됨을 의미함은 물론이고, 형질전환 식물체의 발달 과정에 따라서 삽입된 외래 유전자가 공간적, 시기적으로 각각 다르게 발현할 수 있다는 것을 나타낸다.

Development of Species-Specific PCR Primers for the Rapid and Simultaneous Identification of the Six Species of Genus Takifugu

  • Pufferfish (Takifugu spp.) are economically important edible marine fish. Mistakes in pufferfish classification can lead to poisoning; therefore, accurate species identification is critical. In this study, we used the mtDNA cytochrome c oxidase subunit I gene (COI) to design specific primers for six Takifugu species among the 21 domestic or imported pufferfish species legally sold for consumption in Korea. We rapidly and simultaneously identified these pufferfish species using a highly efficient, multiplex polymerase chain reaction (PCR) system with the six species-specific primers. The results showed that species-specific multiplex PCR (multiplex species-specific polymerase chain reaction; MSS-PCR) either specifically amplified PCR products of a unique size or failed. MSS-PCR yielded amplification fragment lengths of 897 bp for Takifugu pardalis, 822 bp for T. porphyreus, 667 bp for T. niphobles, 454 bp for T. poecilonotus, 366 bp for T. rubripes, and 230 bp for T. xanthpterus using the species-specific primers and a control primer (ca. 1,200 bp). We visualized the results using agarose gel electrophoresis to obtain accurate contrasts of the six Takifugu species. MSS-PCR analysis is easily performed and provides identification results within 6 h. This technique is a powerful tool for the discrimination of Takifugu species and will help prevent falsified labeling, protect consumer rights, and reduce the risk of pufferfish poisoning..

Mitochondrial DNA Sequence Variation of the Oriental Mole Cricket, Gryllotalpa orientalis (Orthoptera: Gryllotalpidae) in Korea

  • The mole crickets, Gryllotalpa, are insect pest distributed in the world. In Korea, G. orientalis was reported to occur, but previous ecological studies suggested the presence of two ecological types. To test this hypothesis, we sequenced a portion of mitochondrial (mt) genome from 48 G. orientali individuals collected over five Korean localities: Busan, Suwon, Okchon, Wonju, and Gangneung. From the sequence analysis, only two haplotypes were obtained, but the sequence divergence between the two haplotypes was 11 %, suggesting the presence of two distinct genetic groups in Korea. Although the population of Busan, Okchon, Wonju, and Gangneung was identified as a single haplotype, but that of Suwon was occupied by both hapotypes. Considering sequence divergence of other insect species occurring in Korea, the divergence estimate found between the two haplotypes seems to be too large to be considered as identical species. This result may suggest that the two differentiated haplotypes found in this study may reflect the previously reported two ecological types found in Suwon, Korea. To further understand the genetic divergence of the two phylogenetic groups, analysis of more variable regions of G. orientalis genome is required.

우리나라 연안해역 난 및 자치어의 분포특성 (Distribution of Eggs and Larvae in Coastal Waters of Korea)

  • The distribution and abundance of fish eggs and larvae were investigated from February to December 2020 along the coastal waters of Korea. The eggs and larvae were identified using the mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) and 16s rRNA gene. During the study period, eggs of overall 45 taxa belonging to 26 families were collected and larvae of overall 39 taxa belonging to 23 families were collected. In Yeongil Bay, eggs of Engraulis japonicus, which accounted for 83.9% of the total population, was the most dominant species, followed by Sardinops sagax (4.0%), Repomucenus valenciennei (3.8%) and E. japonicus larvae, which accounted for 34.9% of the total population. These were followed by Sebastiscus marmoratus (31.0%). In Gomso Bay, E. japonicus eggs accounted for 61.7% of the total population, followed by Sillago japonica (14.0%), Johnius grypotus (8.8%) and Pholis fangi larvae, which accounted for 53.5% of the total population, followed by Ammodytes personatus (34.1%). In Jinhae Bay, E. japonicus eggs accounted for 86.0% of the total population, followed by Leiognathus nuchalis (4.1%), Konosirus punctatus (3.7%) and E. japonicus larvae, which accounted for 48.7% of the total population, followed by Parablennius yatabei (21.6%).

Molecular Identification of Anisakis Larvae Extracted by Gastrointestinal Endoscopy from Health Check-up Patients in Korea

  • Anisakiasis is a zoonotic disease induced by anisakid nematodes, and endoscopic inspection is used for a diagnosis or remedy for it. Anisakis simplex, Anisakis physeteris, and Pseudoterranova decipiens had been reported to be the major species causing human infections, particularly, in Japan. However, in Korea, recent studies strongly suggested that Anisakis pegreffii is the major species of human infections. To support this suggestion, we collected anisakid larvae (n=20) from 20 human patients who were undergone gastrointestinal endoscopy at a health check-up center in Korea, and molecular identification was performed on the larvae using PCR-RFLP analysis and gene sequencing of rDNA ITS regions and mtDNA cox2. In addition, anisakid larvae (n=53) collected from the sea eel (Astroconger myriaster) were also examined for comparison with those extracted from humans. The results showed that all human samples (100%) were identified as A. pegreffii, whereas 90.7% of the samples from the sea eel were A. pegreffii with the remaining 9.3% being Hysterothylacium aduncum. Our study confirmed that A. pegreffii is the predominant species causing human anisakiasis in Korea, and this seems to be due to the predominance of this larval type in the fish (sea eels) popularly consumed by the Korean people. The possibility of human infection with H. aduncum in Korea is also suggested.

Molecular Systematics of Korean Cobitids Based on Mitochondrial Cytochrome b Sequence

  • We compared the complete mitochondrial cytochrome b gene sequences of Korean and European cobitids to provide independent evidence for assessment of systematic and biogeographic relationships of species in the genus Cobitis. The data suggested monophyly of the genus Cobitis and the inclusion of Korean Cobitis species within the group having one lamina circularis, a primitive condition. Also, all the phylogenetic analyses using maximum parsimony, maximum likelihood, and neighbor joining methods showed a monophyletic relationship among Cobitis. The basal position of the Caspian C. cf. sibirica reported here reflects the eastern Asiatic origin cf. the European Cobitis and establishes C. cf. sibirica as an independent lineage. The Korean C. pacifica diverged next to C. cf. sibirica in basal group from the genus Cobitis. This result is in agreement with the hypothesized Asiatic origin of some European freshwater fish lineages. The phylogenetic relationships in this study showed a close affinity between C. zanadreai and C. sinensis. Two new species, C. tetralineata and C. pacifica in Korea also are closely related to monophyletic group clustering the type species of the Acanestrinia subgenus (C. elongata) with all the endemic Italian species (C. bilineata and C. zanandreai). This may suggest that the affinity between the Korean and Danubian-Italian imply genetic convergence or genetic plesiomorphic state between allopatric species that are separated for the Miocene. The mtDNA-based phylogeny for the species of the genus Cobitis from Kores and Europe permits phylogenetic assessment of the morphological transitions of Iamina circularis.

The Expression Patterns of AtBSMT1 and AtSAGT1 Encoding a Salicylic Acid (SA) Methyltransferase and a SA Glucosyltransferase, Respectively, in Arabidopsis Plants with Altered Defense Responses

  • We reported previously that overexpression of a salicylic acid (SA) methyltransferase1 gene from rice (OsBSMT1) or a SA glucosyltransferase1 gene from Arabidopsis thaliana (AtSAGT1) leads to increased susceptibility to Pseudomonas syringae due to reduced SA levels. To further examine their roles in the defense responses, we assayed the transcript levels of AtBSMT1 or AtSAGT1 in plants with altered levels of SA and/or other defense components. These data showed that AtSAGT1 expression is regulated partially by SA, or nonexpressor of pathogenesis related protein1, whereas AtBSMT1 expression was induced in SA-deficient mutant plants. In addition, we produced the transgenic Arabidopsis plants with RNAi-mediated inhibition of AtSAGT1 and isolated a null mutant of AtBSMT1, and then analyzed their phenotypes. A T-DNA insertion mutation in the AtBSMT1 resulted in reduced methyl salicylate (MeSA) levels upon P. syringae infection. However, accumulation of SA and glucosyl SA was similar in both the atbsmt1 and wild-type plants, indicating the presence of another SA methyltransferase or an alternative pathway for MeSA production. The AtSAGT1-RNAi line exhibited no altered phenotypes upon pathogen infection, compared to wild-type plants, suggesting that (an)other SA glucosyltransferase(s) in Arabidopsis plants may be important for the pathogenesis of P. syringae.

Development of Near-Isogenic Lines (NILs) Conferring Xa4, xa5 and Xa21 Genes Resistant to Bacterial Blight (Xanthomonas oryzae pv. oryzae) in japonica rice Genetic Background

  • Near-isogenic lines (NILs) carrying bacterial blight resistance genes (Xa4, xa5 and Xa21) were developed in japonica rice using Suweon345 as genetic background. NILs were selected by gene specific DNA markers and inoculation of K1 or K3a race. NILs conferring Xa4 were resistant to K1, K2, K3, and moderately resistant to K3a. NILs conferring xa5 were resistant to K1, K2, K3, and K3a. NILs having Xa21 were susceptible to K1, while resistant to K2, K3 and K3a. Target genes of NILs with the genetic background of Suweon345 were also confirmed by using eleven Philippines races and International Rice Bacterial Blight (IRBB) NILs carrying Xa4, xa5 and Xa21. All NILs had no significant difference from their recurrent parents in the major agronomic traits except for panicle length and brown rice 1,000 grain weight. Heading date of NILs ranged from Aug. 10 to Aug. 11, which was similar to that of recurrent parent, Suweon345. Culm length, number of grains per panicle and ratio of ripened grain of NILs were similar to those of Suweon345. Milled rice of NILs was ranged from 4.82 to 4.93MT/ha. These NILs will be useful for improving resistance to K3a race of bacterial blight pathogens in Korean japonica cultivars.

Mitochondrial Genetic Diversity and Phylogenetic Relationships of Siberian Flying Squirrel(Pteromys volans) Populations

  • Siberian flying squirrel, an endangered species in South Korea, is distributed through major mountain regions of South Korea. The number of Siberian flying squirrel(Pteromys volans) in South Korea has decreased and their habitats are fragmented and isolated because of anthropogenic activities. So far no molecular genetic data has, however, been available for their conservation and management. To obtain better information concerning genetic diversity and phylogenetic relationships of the Siberian flying squirrel in South Korea, we examined 14 individuals from South Korea, 7 individuals from Russia, and 5 individuals from northeastern China along with previously published 29 haplotypes for 1,140 bp of the mtDNA cytochrome b gene. The 14 new individuals from South Korea had 7 haplotypes which were not observed in the regions of Russia and Hokkaido. The level of genetic diversity(0.616%) in the South Korean population was lower than that in eastern Russia(0.950%). The geographical distribution of mtDNA haplotypes and reduced median network confirmed that there are three major lineages of Siberian flying squirrel, occupying; Far Eastern, northern Eurasia, and the island of Hokkaido. The South Korean population only slightly distinct from the Eurasia, and eastern Russian population, and is part of the lineage Far Eastern. Based on these, we suggest that the South Korean population could be considered to belong to one partial ESU(Far Eastern) of three partial ESUs but a different management unit. However, the conservation priorities should be reconfirmed by nuclear genetic marker and ecological data.
