Effect of GM-CSF on the Embryonic Development and the Expression of Implantation Related Genes of Mouse Embryos (Granulocyte-Macrophage Colony Stimulating Factor (GM-CSF)의 첨가가 생쥐 수정란의 발생과 착상관련 유전자 발현에 미치는 영향)

  • Objective : The purpose of the current series of experiments were to assess the effect of GM-CSF, as a medium supplement, on the development of mouse embryos and the expression of LIF and IL-1? mRNA. Materials and Methods: Mouse 2-cell embryos were collected from the oviducts of 6 weeks old ICR mice at 48 hours after hCG injection. Embryos were cultured in P-1 medium supplemented with mouse GM-CSF (0, 1, 5, 10 ng/ml). The embryo development to blastocysts and hatching blastocysts was assessed and the cell number in blastocyst was also examined. Using RT-PCR, the expressions of LIF and IL-1? mRNA in blastocyst were evaluated in the GM-CSF supplemented group and control group. Results: In mouse, the addition of GM-CSF increased the percentage of blastocysts (65.5%, 68.6%, 73.0% and 76.1% for control and 1, 5 and 10 ng/ml, respectively), and increased the proportion of hatching blastocysts (35.2%, 36.4%, 43.2% and 53.0% for control and 1, 5 and 10 ng/ml, respectively). The mean cell numbers in blastocyst were significantly increased in GM-CSF supplemented groups compared to control group. LIF and IL-1? expression in blastocyst were significantly higher in GM-CSF supplemented group than in control group. Conclusion: The results of experiment by mouse embryos showed beneficial effects of GM-CSF as a medium supplement. Furthermore, the addition of GM-CSF significantly increased the expression of LIF and IL-1? in mouse embryos. These results suggest that GM-CSF might be a important molecule in embryo implantation.

In Vitro/In Vivo Development of Vitrified Immature Mouse Oocytes (초자화 동결된 생쥐 미성숙란의 체외/체내 발달)

  • This study was carried out to investigate in vitro/in vivo development of vitrified-thawed immature mouse oocytes. Immature mouse oocytes were vitrified with EFS40 (40% ethylene glycol, 18% ficoll and 0.5 M sucrose). Thawed oocytes were matured for 16 hr in vitro. Matured oocytes with the first polar body were fertilized with the concentration of 1~2$\times$10$^{6}$ $m\ell$ of epididymal sperm. After fertilization, cleavage ($\geq$ 2-cell) and in vitro/in vivo development rates were examined. $\pi$ Ie results were summarized as follows: in vitro maturation rate of immature mouse oocytes in vitrified-thawed group was similar to that in exposed group (67.5%) and control (66.3%), but cleavage rate of vitrified-thawed oocytes (64.9 %) and blastocyst formation rate (59.0%) were significantly different compared to those of exposed group (83.7 and 74.7%) and control (90.7 and 83.7%) (p<0.05). However, when the blastocysts derived from immature mouse oocytes vitrified-thawed were transferred to pseudopregnant mouse, total implantation (31.3%) was slightly lower than that in control (40.8%), but live fetus formation rate (66.7%)was slightly higher than that in control (58.1%), there was not significantly different. Therefore, when the blastocyts produced in vitro were transferred into recipients, although the development in vitro of oocytes vitrified-thawed was decreased, live fetus formation rate was similar to that of control group. The present results indicate that immature mouse oocytes can be frozen successfully by vitrification with EFS40.

Production of Identical Multiplets from Deep-Frozen Embryos (동결보존배를 이용한 일란성 다태생산에 관한 연구)

  • Although plenty of experiments for production of genetically identical twins have been reported, most of these reports were focused on microsurgical bisection of morula stage embryos, and little research has been performed for the production with frozen emb교os. The objective of these experiments were to assess the in vitro and in vitro developmental potential of blastomeres isolated from frozen-thawed 4-cell and 8-cell mouse embryos and to produce the identical multiplets from those embryos. The percentages of isolated 1/4, 2/4, 4/4, (control), 1/8, 2/8, 3/8, 4/8 and 8/8(control) blastomeres of 4-cell and 8-cell mouse emb교os which developed to normal blastocyst were 38.7%, 73.6%, 96.2(control), 15.1%, 40.1%, 65.9%, 88.3%, and 98.5%(control), respectively. The percentages of isolated 1/4, 2/4, 4/4(control), 1/8, 2/8, 3/8, 4/8 and 8/8(control) blastomeres of frozen-thawed 4-cell and 8-cell mouse embryos which developed to normal blastocyst were 35.6%, 68.5%, 100.0%(control), 16.1%, 50.6%, 71.7%, 90.9% and 100.0%(control), respectively. Normal 26 pups were obtained by transfer of 240 blastocyts developed 1, on 1/4 to 8/8 blastomeres. Those 26 pups obtained, 4 identical twins were produced from 2/4, 2/8, 3/8 and 4/8 blastomeres.

  • Genetically engineered mouse models are commonly preferred for studying the human disease due to genetic and pathophysiological similarities between mice and humans. In particular, Cre-loxP system is widely used as an integral experimental tool for generating the conditional. This system has enabled researchers to investigate genes of interest in a tissue/cell (spatial control) and/or time (temporal control) specific manner. A various tissue-specific Cre-driver mouse lines have been generated to date, and new Cre lines are still being developed. This review provides a brief overview of Cre-loxP system and a few commonly used promoters for expression of tissue-specific Cre recombinase. Also, we finally introduce some available links to the Web sites that provides detailed information about Cre mouse lines including their characterization.

Effects of Water Extracts of Camelia sinensis L on Blood Alcohol Concentration and Activities of Sub-acute Alcohol Metabolic Enzymes in ICR Mouse (ICR Mouse의 아급성 알코올 대사에 보이차(Camelia sinensis L) 추출물이 미치는 효과)

  • An eight-week-old male ICR mouse, which was induced with acute alcohol and sub-acute alcohol poisoning condition, was administered with bohee tea(Camelia sinensis L) extract. Under the inducement of the sub-acute alcohol poisoning condition, no considerable differences could be found in the blood alcohol concentration of the positive control group and the bohee tea group(p<0.05). The GOT activity of the three groups: bohee tea, Drink, and Alcodex decreased than that of the normal control group(9.064±4.687 unit)(p<0.05). In addition, the blood GOT activity of the dark green tea group dropped by 81.44% compared with that of the positive control group. On the other hand, the blood GTP activity of the bohee tea group decreased by 5.2% as opposed to that of the positive control and the Drink that decreased by 7.5% as opposed to that of the positive control. The hepatic ADH activity of the bohee tea increased by 22.7%, as opposed to that of the positive control group. The Drink, however, had an increase rate of 33.6%. In the case of the hepatic ALDH activity of the liver, no significant differences were ever recorded among all groups, except for the positive control group. Due to an intake of bohee tea extract, the hepatic ALDH activity decreased by 77.27% which could not be seen in the positive control group. However, Drink and A1codex had a decrease could be seen(p<0.05).

  • An eight-week-old male ICR mouse, which was induced with acute alcohol and sub-acute alcohol poisoning condition, was administered with bohee tea(Camelia sinensis L) extract. After oral administration of bohee tea and inducement of acute alcohol poisoning condition, the mouses blood alcohol concentration became as low as that of the normal control group. Its decrease rate was 87.26%, in comparison with that of the positive control group. Moreover, its blood GOT activity decreased with a rate of 93.1 % until it reached the normal level, as opposed to that of the positive control group. In addition, the GOT activity, despite rising after the alcohol intake, decreased(p<0.05) significantly after administration of each sample and reached the normal level. The bohee tea group experienced a significant decrease in the GOT activity, compared with the A1codex group and the Drink group. The GPT activity of the Alcodex group decreased by 11 % compared with that of the positive control group. The CTP activity of the bohee tea group decreased by 8.2%, while that of the Drink group decreased by 6.5%(P<0.05). However, there were no significant differences between the results in the control group and those of the test group. The bohee tea group's hepatic ADH activity increased by 22.7% compared with that of the positive control group. On the other hand, the hepatic ADH activity of the Drink group increased by 33.6% while that of the A1codex group increased by 20.4%. On the contrary, the bohee tea extract, the hepatic ALDH did not manifest any significant difference as compared with the normal control group. However, its decrease rate was about 16.67% as compared with that of the positive control group. The Drink group, meanwhile, obtained a decrease rate of about 21.59%.

  • Effect of the egg yolky from laying hens intubated, p.o., astaxanthin (designated AEY) on the oxidation of mouse liver microtome was investigated using female ICR mouse (6~7 weeks of age). Mice were adapted in a temperature- and humidity-controlled house for one week and randomly divided into 7 treatment groups (10 mice/cage/treatment). Mice were intubated p.o., AEY (50, 100 and 150 mg) or control egg yolk (CEY, 150 mg) every week for 3 weeks. BHT (5 mg) and e -tocopherol (50 mg) were fed to mice as positive control. At week 4, the liver microsome was prepared from sacrificed mice. Protein content of mouse liver microsome with AEY treatment was relatively higher than that with CEY treatment. AEY treatment remarkably lowered the content of unsaturated fatty acids including oleic acid and linoleic acid, but raised that of the saturated fatty acids including stearic acid. AEY group showed relatively higher antioxidative activity than CEY, when used Asc/F $e^{+2}$ or NADPH/F $e^{+2}$ as oxidant. Antioxidative activity of AEY was more effective than $\alpha$-tocopherol, but less effective than BHT.

Head Mouse System Based on A Gyro and Opto Sensors (각속도 및 광센서를 이용한 헤드 마우스)

  • We proposed the device to control a computer mouse with only head movements and eye blinks so that disabilities by car or other accidents can use a computer. The mouse position were estimated from a gyro-sensor which can measure head movements, and the mouse events such as click/double click were from opto sensors which can detect the eyes flicker, respectively. The sensor was mounted on the goggle in order not to disturb the visual field. There was no difference in movement speed between ours and a general mouse, but it required 3$\sim$4 more times in the result of the experiment to evaluate spatial movements and events detection of the proposed mouse because of the low accuracy. We could eliminate cumbersome work to periodically remove the accumulated error and intuitively control the mouse using non-linear relative point method with dead zones. Optical sensors are used in the event detection circuitry designed to remove the influence of the ambient light changes, therefore it was not affected in the change of external light source.

Hand Mouse System Using a Pre-defined Gesture for the Elimination of a TV Remote Controller

  • Many hand gesture recognition systems using advanced computer vision techniques to eliminate the need for a TV remote controller have been proposed. Nevertheless, some issues still remain, such as high computational complexity and insufficient information on the target object and background. Moreover, none of the proposed techniques consider how to enter the control mode of the system. This means that they may need a TV remote controller to enter the control mode. This paper proposes a hand mouse system using a pre-defined gesture with high background adaptability. By doing so, a remote controller to enter the control mode of the IPTV system can be eliminated.
