Supplement of tauroursodeoxycholic acid in vitrification solution improves the development of mouse embryos

  • This study was performed to determine whether supplementation of tauroursodeoxycholic acid (TUDCA), an endoplasmic reticulum (ER) stress inhibitor, during vitrified cryopreservation enhances the development of frozen mouse embryos. Mouse 8-cell stage embryos were collected and exposed to a cryoprotectant solution containing TUDCA or TM (tunicamycin, an ER stress inhibitor) at room temperature and stored in liquid nitrogen following vitrification. The final concentration of TUDCA or TM was $50{\mu}M$. The survival and development rates of mouse 8-cell stage embryos exposed to TUDCA- or TM-containing solutions at room temperature or stored in liquid nitrogen following vitrification were measured. There were no significant differences in survival rate and blastocyst formation rate among control, TUDCA, and TM groups after embryos were exposed to vitrification solutions at RT. When mouse 8-cell stage embryos were treated with TUDCA or TM and then stored in liquid nitrogen, the survival rates of control and TUDCA groups were significantly higher than for the TM group. Blastocyst formation rate of the TUDCA group following in vitro culture was significantly higher than that in control or TM groups. The TM group showed a lower (p < 0.05) blastocyst formation rate than the other two groups. Our results indicate that TUDCA supplementation during cryopreservation of mouse embryos could enhance their development capacity.

Addition of interleukin-6 to mouse embryo culture increases blastocyst cell number and influences the inner cell mass to trophectoderm ratio

  • Objective: In vitro culture of preimplantation embryos is improved by grouping embryos together in a drop of media. Individually cultured embryos are deprived of paracrine factors; with this in mind, we investigated whether the addition of a single embryo-secreted factor, interleukin-6 (IL-6), could improve the development of individually cultured embryos. Methods: Mouse embryos were cultured individually in $2{\mu}L$ of G1/G2 media in 5% oxygen and supplemented with a range of doses of recombinant mouse or human IL-6. Results: Mouse IL-6 increased hatching at doses of 0.01 and 10 ng/mL compared to the control (93% and 93% vs. 78%, p< 0.05) and increased the total number of cells at a dose of 0.1 ng/mL compared to the control ($101.95{\pm}3.36$ vs. $91.31{\pm}3.33$, p< 0.05). In contrast, the highest dose of 100 ng/mL reduced the total number of cells ($79.86{\pm}3.29$, p< 0.05). Supplementation with human IL-6 had a different effect, with no change in hatching or total cell numbers, but an increase in the percentage of inner cell mass per embryo at doses of 0.1, 1, and 100 ng/mL compared to the control ($22.9%{\pm}1.1%$, $23.3%{\pm}1.1%$, and $23.1%{\pm}1.1%$ vs. $19.5%{\pm}1.0%$, p< 0.05). Conclusion: These data show that IL-6 improved mouse embryo development when cultured individually in complex media; however, an excess of IL-6 may be detrimental. Additionally, these data indicate that there is some cross-species benefit of human IL-6 for mouse embryos, but possibly through a different mechanism than for mouse IL-6.

Effect of DOGO phreatic water containing sulphur on Atopic Dermatitis in NC/Nga mouse (아토피 피부염 유발 NC/Nga mouse에서 도고 온천수 유황 성분의 효과)

  • Objectives : The purpose of this study is to investigate the effects of DOGO phreatic water containing sulphur on Atopic Dermatitis in NC/Nga mouse. Methods : We made DOGO phreatic water removed sulphur using Twin Alternating Sulfate Eater. After making atopic dermatitis caused by sensitizing NC/Nga mouse to DNCB(dinitrochlorobenzene), we made mouse swim in tanks each filled with distilled water, tap water, DOGO phreatic water(contain sulphur), DOGO phreatic water(remove sulphur) for 30minutes everyday. 3weeks later, we analyzed skin clinical score, total IgE levels(by ELISA), WBC differential counting(Neutrophils, Monocytes), absolute cell number of $Neutrophil^+Gr-1^+$, CCR3 mRNA expressions(by Real-time PCR), IL-4, IFN-${\gamma}$ production levels(by ELISA), histologic test(by H&E staining, toluidine blue staining). Results : The results of making NC/Nga mouse induced atopic dermatitis swim in tanks filled with DOGO phreatic water(contain sulphur) are as follows. 1. Skin clinical scores were decreased significantly in comparison to control group. 2. Total IgG levels were decreased significantly in comparison to control group. 3. WBC differential counting(Neutrophils, Monocytes) were decreased significantly in c.mparison to control group. 4. Absolute cell number of $Neutrophil^+Gr-1^+$ were decreased significantly in comparison to control group. 5. CCR3 mRNA expressions were decreased significantly in comparison to control group. 6. IL-4, IFN-${\gamma}$ production levels were decreased significantly in comparison to control group. 7. The epithelial tissue thickness, leucocytes infiltration, erythema, edema, excoriation, scaling, mast cells infiltrations in dorsal skin were decreased in comparison to control group. Conclusions : These results indicate that DOGO phreatic water(contain sulphur) can be used for helping treat atopic dermatitis.

Effect of Bopheyangyoungjeon on the Lung Tumor of A/J Mouse Induced with Urethane (보폐양영전(保肺養營煎)이 Urethane으로 유발(誘發)된 A/J mouse의 폐암에 미치는 영향(影響))

  • A/J mice injected with urethane(1mg/ g of body weight) develop tumors with distinct histological patterns, which are classified as solid and papillary. I divided the mice into 3 groups; control group treatment with saline, BYA group treatment with herbal formula, Bopheyangyoungjeon 0.3g/kg and BYB groups treatment with herbal formula 1.5g/kg. The administration of herbal medicine was done every day for 56 days. The experimental results from herbal medicine treatment were compared to those from a saline-treated control group. Serial sections of the whole lung(150 to 200 sections per mouse) showed solid and papillary tumors arose from the pulmonary acinus, invading the bronchioles only as the tumors grew. The number of tumor, in the 8-weeks groups, are decreased in the experimental groups compared to control group. COX-2 protein and IGF- I protein expression was more increased in lung tumors of the control group compared to BYA and BYB groups. These results suggest that Bopheyangyoungjeon extract suppress the carcinogenesis of lung in the A/J mouse.

The Effects of Sasammaickmoondong-tang against Duodenal Mucosal Lesions (Mouse의 십이지장 점막 염증에 대한 사삼맥문동탕의 면역반응 연구)

  • Objectives : This study was carried out to investigate the effects of Sasammaickmoondong-tang(SME) on duodenal mucosal lesions induced by indomethacin in mouse. Methods : The normal group is that no inflammation elicitated mouse. Control group is that gastro-inflammation elicitated mouse. Sample group is that SME administered mouseice after gastro-inflammation elicitation. Results : In the common morphology and histochemical change, control group was observed various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell-by hemorrhagic erosion, while sample group was as same as normal group. In the immunohistochemical change, the distributions of COX-1, Bcl-2, and BrdU treated with SME noticeably increased than control group(p<0.05). The distributions of TUNEL, $NF-{\kappa}B$, COX-2, $IL-2R-{\alpha}$, NK-1.1, ICAM-1, and CD11b/18 treated with SME noticeably decreased than control group(p<0.05). And the distribution of SBA was as same as normal group. Conclusion : According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to duodenal mucosal lesions.

Coordinate Estimation of Mobile Robot Using Optical Mouse Sensors (광 마우스 센서를 이용한 이동로봇 좌표추정)

  • Coordinate estimation is an essential function for autonomous navigation of a mobile robot. The optical mouse sensor is convenient and cost-effective for the coordinate estimation problem. It is possible to overcome the position estimation error caused by the slip and the model mismatch of robot's motion equation using the optical mouse sensor. One of the simple methods for the position estimation using the optical mouse sensor is integration of the velocity data from the sensor with time. However, the unavoidable noise in the sensor data may deteriorate the position estimation in case of the simple integration method. In general, a mobile robot has ready-to-use motion information from the encoder sensors of driving motors. By combining the velocity data from the optical mouse sensor and the motion information of a mobile robot, it is possible to improve the coordinate estimation performance. In this paper, a coordinate estimation algorithm for an autonomous mobile robot is presented based on the well-known Kalman filter that is useful to combine the different types of sensors. Computer simulation results show the performance of the proposed localization algorithm for several types of trajectories in comparison with the simple integration method.

Subacute Toxicity Test of Guh Sung Y.L.S.-95 (Guh Sung Y.L.S.-95의 아급성 독성시험)

  • Guh Sung Y.L.S.-95 is one of the polyacidic solution of which main component is acetic acid. We investigated the subchronic toxicity of the Guh Sung Y.L.S.-95 using SPF ICR mouse for 4 weeks. The Guh Sung Y.L.S.-95 was administered by gastric intubation, 1.0, 2.5, 5.0 g/kg body weight. The results are as follows: 1. There are no adverse effects on the clinical obserbation and body weight changes. Also, there are some significant changes in organ weight, but it was meaningless because of the absence of dose-response relationships. 2. In the hematological patterns of administered mouse, there are no significant changes between the treated groups. Also, there are no serological enzymatic changes in the treated mouse. In the 1.0 g/kg treated group, ASP activity was increased significnatly compared with control group. But, this level of activity was fall under the normal physiological range of control mouse. 3. Histopathological findings of the brain, liver, heart, spleen, kidneys, stomach, lung, testis, ovary, uterus and thymus were not observed in the treated mouse. From the above results, the Guh Sung Y.L.S.-95 has no toxicity upto the 5.0 g/kg/day of oral dose for 4 weeks.

  • In recent years, the use of touch screens has expanded, and devices such as remote controllers have been developed in various ways to control and access contents at long range. The wireless-based touch screen is used in classroom, seminar room, and remote video conversation in addition to the TV remote control. The purpose of the study is to design a smart phone-based intuitive interface that can perform the role of a wireless mouse and a wireless keyboard at range using Bluetooth and to develop an application that integrates functions of a mouse and a keyboard. Firstly, touch interaction model for controlling software such as PowerPoint by connecting to a general PC on a smart phone has been studied. The most simple touch operation interface is used to reproduce the function of existing devices and design more simply. The studies of the extension of interfaces with various functions are important, but development of optimized interfaces for users will become more important in the future. In this sense, this study is valuable.
