감마선 조사가 Staphylococcal Enterotoxin B의 비장세포 증식률 및 Interleukin-2 분비능에 미치는 영향 (Effect of Gamma-Irradiation on the Cell Proliferating and Interleukin-2 Producing Activity of Mouse Splenocytes of Staphylococcal Enterotoxin B)

  • The purpose of this study was to investigate the cell proliferating and interleukin-2 producing activity of staphylococcal enterotoxin B by gamma-irradiation. Staphylococcal enterotoxin B was gamma-irradiated with the various doses of 0, 2, 20 and 50 kGy. SDS-PAGE analysis showed that gamma-irradiation caused the sharp decrease of the content of staphylococcal enterotoxin B and the effect was irradiating dose-dependent. Non-irradiated staphylococcal enterotoxin B increased the cell proliferation of splenocytes isolated from female Balb/c mouse, whereas 2 kGy-irradiated toxin significantly decreased the activity. 20 and 50 kGy-irradiated staphylococcal enterotoxin B was no effect. A similar effect on the interleukin-2 production of mouse splenocytes was observed with non-irradiated and irradiated staphylococcal enterotoxin B. It was considered to be due to the decrease of the antigenicity of staphylococcal enterotoxin B by gamma-irradiation. Therefore, these results suggest that gamma-irradiation can be effective for the decrease of the antigenicity of staphylococcal enterotoxin B as superantigen.

Mouse의 십이지장 점막 염증에 대한 사삼맥문동탕의 면역반응 연구 (The Effects of Sasammaickmoondong-tang against Duodenal Mucosal Lesions)

  • Objectives : This study was carried out to investigate the effects of Sasammaickmoondong-tang(SME) on duodenal mucosal lesions induced by indomethacin in mouse. Methods : The normal group is that no inflammation elicitated mouse. Control group is that gastro-inflammation elicitated mouse. Sample group is that SME administered mouseice after gastro-inflammation elicitation. Results : In the common morphology and histochemical change, control group was observed various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell-by hemorrhagic erosion, while sample group was as same as normal group. In the immunohistochemical change, the distributions of COX-1, Bcl-2, and BrdU treated with SME noticeably increased than control group(p<0.05). The distributions of TUNEL, $NF-{\kappa}B$, COX-2, $IL-2R-{\alpha}$, NK-1.1, ICAM-1, and CD11b/18 treated with SME noticeably decreased than control group(p<0.05). And the distribution of SBA was as same as normal group. Conclusion : According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to duodenal mucosal lesions.

절단마우스 이분배의 동결보존실험 1. 마우스 절단이분배의 체외 발육능에 대하여 (Effects of Freezing on Bisected Mouse Embryos 1. Developmental Potentials of Bisected Mouse Embryos in vitro)

  • Mouse embryos of 8-cell stage and compacted morulae(approximately 16 cells) containing different number of blastomeres were bisected and cultured in vitro to determine the developmental potentials of the divided embryos compared with those of unmanipulated control embryos. The results were as follows. 1. Micromanipulation was performed successfully by means of a simple manipulator which holds a fine glass, needle, without the use of any micro-instruments for support. 2. The percentage of bisected morulae with 7-9 blastomeres that developed to eu-blastocyst was 94.1% while only 64.8% of the bisected 8-cell embryos with 4 blastomeres developed to eublastocysts (p<0.05). 3. The percentage of eu-blastocysts decreased, while that of pseudoblastocysts and trophectodermal vesicle increased as the number of blastomeres decreased in the bisected embryos of the two stages. 4. The time of the blastocoele re-formation of the bisected and control embryos was not significantly different in morulae stage embryos, but it was significantly delayed in the 8-cell stage embryos (Eu-B, Pseudo-B) compared with control embryos (P<0.01, P<0.05 respectively).

Differential Signaling via Tumor Necrosis Factor-Associated Factors (TRAFs) by CD27 and CD40 in Mouse B Cells

  • Background: CD27 is recently known as a memory B cell marker and is mainly expressed in activated T cells, some B cell population and NK cells. CD27 is a member of tumor necrosis factor receptor family. Like CD40 molecule, CD27 has (P/S/T/A) X(Q/E)E motif for interacting with TNF receptor-associated factors (TRAFs), and TRAF2 and TRAF5 bindings to CD27 in 293T cells were reported. Methods: To investigate the CD27 signaling effect in B cells, human CD40 extracellular domain containing mouse CD27 cytoplamic domain construct (hCD40-mCD27) was transfected into mouse B cell line CH12.LX and M12.4.1. Results: Through the stimulation of hCD40-mCD27 molecule via anti-human CD40 antibody or CD154 ligation, expression of CD11a, CD23, CD54, CD70 and CD80 were increased and secretion of IgM was induced, which were comparable to the effect of CD40 stimulation. TRAF2 and TRAF3 were recruited into lipid-enriched membrane raft and were bound to CD27 in M12.4.1 cells. CD27 stimulation, however, did not increase TRAF2 or TRAF3 degradation. Conclusion: In contrast to CD40 signaling pathway, TRAF2 and TRAF3 degradation was not observed after CD27 stimulation and it might contribute to prolonged B cell activation through CD27 signaling.

CD72 is a Negative Regulator of B Cell Responses to Nuclear Lupus Self-antigens and Development of Systemic Lupus Erythematosus

  • Systemic lupus erythematosus (SLE) is the prototypic systemic autoimmune disease characterized by production of autoantibodies to various nuclear antigens and overexpression of genes regulated by IFN-I called IFN signature. Genetic studies on SLE patients and mutational analyses of mouse models demonstrate crucial roles of nucleic acid (NA) sensors in development of SLE. Although NA sensors are involved in induction of antimicrobial immune responses by recognizing microbial NAs, recognition of self NAs by NA sensors induces production of autoantibodies to NAs in B cells and production of IFN-I in plasmacytoid dendritic cells. Among various NA sensors, the endosomal RNA sensor TLR7 plays an essential role in development of SLE at least in mouse models. CD72 is an inhibitory B cell co-receptor containing an immunoreceptor tyrosine-based inhibition motif (ITIM) in the cytoplasmic region and a C-type lectin like-domain (CTLD) in the extracellular region. CD72 is known to regulate development of SLE because CD72 polymorphisms associate with SLE in both human and mice and CD72-/- mice develop relatively severe lupus-like disease. CD72 specifically recognizes the RNA-containing endogenous TLR7 ligand Sm/RNP by its extracellular CTLD, and inhibits B cell responses to Sm/RNP by ITIM-mediated signal inhibition. These findings indicate that CD72 inhibits development of SLE by suppressing TLR7-dependent B cell response to self NAs. CD72 is thus involved in discrimination of self-NAs from microbial NAs by specifically suppressing autoimmune responses to self-NAs.

Mouse cell에서 탁리소독음(托裏消毒飮)의 항산화작용과 항염증 효과 (The Effects of Taglisodog-eum Extract on Antioxidant and Antiinflammatory ability in mouse cell)

  • Background and Objectives : The aim of this study was to investigate the anti-oxidant and anti-inflammatory effects of the Taglisodog-eum(TSE) extract on the RAW264.7 cell Methods : The RAW264.7 cell was cultured using Dulbecco's modified Eagle's medium(DMEM, USA), including the 10% fetal-bovine serum(FBS; Sigma, USA) in a $37^{\circ}C$, 5% CO2 incubator. Results : The anti-oxidant ability of TSE were dose-dependantly increased. The LPS-induced IKK, iNOS and COX-2 mRNA expression were dose-dependantly decreased in the RAW264.7 cells treated with TSE. $NF-kB$ activation was suppressed. Conclusion : The findings in this study show that TSE has anti-oxidant and anti-inflammatory effects, such as the inhibition of $NF-kB$ activity.

자외선 B 조사에 의한 마우스 피부 ATPase 양성 가지세포의 변화 및 녹차투여의 효과 (The Change of ATPase-positive Dendritic Cell and the Effect of Green Tea in Mouse Skin by Ultraviolet B Irradiation)

  • In this study we assessed the influences of ultraviolet (UV) light B radiation on epidermal ATPase-positive dendritic cell (DC) and the effect of green tea treatment in ICR mouse. The extent of changes following 200 mJ/$cm^2$ (0.5 mW/sec) was studied at 0, 6, 12, 18, 24, 30 or 36 hours after exposure. SBCs were decreased by 6 hours after irradiation. There was tendency to decrease from 6 hours to 24 hours and had little further change from then to 36 hours after irradiation. The mice that received 0, 50, 100, 200, 300 or 400 mJ/$cm^2$ of UVB were examined 24 hours after irradiation. The DCs were decreased as the radiation dose increases from 100 to 400 mJ/$cm^2$. The frequency of UVB (200 mJ/$cm^2$)-induced DC decrease was reduced by treatment of green tea (i.p. and topical application, p<0.01).

NC/Nga 생쥐에서 가미청심연자탕(加味淸心蓮子湯)의 GATA-3 조절에 의한 항아토피 피부염 효과 (Anti-dermatitis Effects of KamiCheongsimyeonjatang on GATA-3 Regulation in NC/Nga Mouse)

  • Objectives : The purpose of this study is to investigate effectiveness of KamiCheongsimyeonjatang(KCSYJT) medicines to suppress atopic dermatitis in mouse model experimentally. Methods : First, in vitro, we isolated B cells from 18 weeks of atopicdermatitis-like skin NC/Nga mouse. Then we analyzed FACS(Fluorescence Activated Cell Sorter) by intracellular staining of IFN-$\gamma$, GATA-3+ analyzed cytokines by using real-time PCR. Secondly, in vivo, after administration of KCSYJT to atopic dermatitis NC/Nga mouse at 12 weeks of age, we analyzed serum IgE and the change of activated cell in PBMCs(Peripheral Blood Mononuclear Cells). Results : In vitro, KCSYJT medicines supressed IL-1$\beta$, IL-6, TNF-$\alpha$, and TGF-$\beta$ mRNA and increased IL-10 mRNA in B cells. Also, KCSYJT medicines decreased the levels of GATA-3$^+$CD4$^+$ and increased the levels of IFN-$\gamma^+$CD4$^+$T Cell. In vivo, serum IgE levels dreased in KCSYJT group than control group and In PBMCs, the activated cell percentage of granulocytes, CD3+, CD3+/CD4+, B220+/CD23+, and CCR3+ decreased and CD19+, CD3+/CD8+ increased in KCSYJT group than control group. Conclusions : This study demonstrates immunological activity of KCSYJT on atopic dermatitis-like model mice.

수수 물 추출물이 마우스 면역세포와 항체 생성능에 미치는 영향 (Effect of Sorghum bicolor L. Moench(Sorghum, su-su) Water Extracts on Mouse Immune Cell Activation)

  • This study was performed to evaluate the effect of Sorghum bicolor L. Moench(Sorghum, su-su) extracts on mouse immune cell activation. As ex vivo experiment, different concentrations(0, 50, 500mg/kg B.W.) of Sorghum bicolor L. Moench water extracts were orally administrated into mouse every other day for four weeks. The proliferation of mouse splenocytes, the number of plaque forming cells(PFC) and the cytokine IL-1β production by activated macrophage were used as indices for immunocompetence. Splenocyte proliferation was enhanced in mouse orally administrated with 50mg/kg B.W./day concentration compared to that of control group. Especially, the highest proliferation of spleoncyte was seen in the mouse orally administrated at the concentration of 50mg/kg B.W./day. The number of plaque forming cells(PFC) to SRBC were significantly enhanced when compared with control group. Also, the mouse of Sorghum bicolor L. Moench water extracts 50mg/kg B.W./day supplementation group with LPS stimulation enhanced level of IL-1$\beta$ cytokine production. This study suggest that supplementation of Sorghum bicolor L. Moench water extracts may enhance the immune function by regulating the splenocytes proliferation, increasing the number of PFC and enhancing the cytokine production by activated macrophage.

가미청폐화담탕이 천식 유발 병태 모델에서 천식 관련 활성 면역세포에 미치는 영향 (Effects of Gamicheungpyehwadam-tang on Immune-cell Regulation in Association with Bronchial Asthma in OVA-induced Mouse Model)

  • These studies were investigated the effects of Gamicheungpyehwadam-tang (CPHDT) on immune-cell regulation in association with bronchial asthma in OVA-induced mouse model. The administration of 400 mg/kg CPHDT significantly reduced the number of total cells in lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model. The administration of 400 mg/kg CPHDT significantly reduced $CD3^+,{\;}CD19^+$and $CD3^+,{\;}CD69^+$ cell numbers separated from lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model. CPHDT significantly reduced $CD3^+/CCR3^+,{\;}CD4^+,{\;}B220^+/IgE^+$, and $CD3^+/DX5^+$ cell numbers separated from lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model in a dose dependent manner, However, CPHDT significantly reduced $CD8^+$ cell numbers from only lung and spleen. The administration of CPHDT significantly reduced $NK^+$ cell numbers separated from lung of OVA-induced bronchial asthma mouse model in all concentrations, but 200 mg/kg CPHDT reduced $NK^+$ cell numbers separated from peripheral lymph node. These results suggest that CPHDT has anti-asthma and anti-allergy effects. In addition to, CPHDT may be useful treatment of asthma based on the further studies about the individual efficacy search of the components of CPHDT and the adding of variety drugs to CPHDT.
