Background: CD30 is a member of TNF receptor family and expressed on lymphocytes and other hematopoietic cells following activation as well as Hodgkin and Reed-Sternberg cells in Hodgkin's lymphoma. In this study, CD30-mediated regulation of cell adhesion molecule expression on normal activated mouse T cells was investigated. Methods: Mouse T cells were activated with anti-CD3 antibody for induction of CD30, which was cross-linked by immobilized anti-CD30 antibody. Results: High level of CD30 expression on T cells was observed on day 5, but only little on day 3 even under culture condition resulting in an identical T cell proliferation, indicating that CD30 expression requires a prolonged stimulation up to 5 days. Cross-linking of CD30 alone altered neither proliferation nor apoptosis of normal activated T cells. Instead, CD30 appeared to promote cell adherence to culture substrate, and considerably upregulated ICAM-1 and, to a lesser extent, ICAM-2 expression on activated T cells, whereas CD2 and CD18 (LFA-1) expression was not affected. None of cytokines known as main regulators of ICAM-1 expression on tissue cells (IL 4, $IFN{\gamma}$ and $IFN{\alpha}$) enhanced ICAM-1 expression in the absence of CD30 signals. On the other hand, addition of $NF-{\kappa}B$ inhibitor, PDTC (0.1 mM) completely abrogated the CD30-mediated upregulation of ICAM-1 expression, but not CD2 and ICAM-2 expression. Conclusion: This results support that CD30 upregulates ICAM-1 expression of T cell and such regulation is not mediated by higher cytokine production but $NF-{\kappa}B$ activation. Therefore, CD30 may play important roles in T-T or T-B cell interaction through regulation of ICAM-1, and -2 expression.
생체 내 (in vivo) 실험에서 톳 추출물을 2주 동안 격일로 0, 50, 500 mg/kg B.W.의 농도로 마우스에 경구투여한 후 비장세포 증식능 및 복강 대식세포에서 분비하는 cytokine ($IL-1{\beta}$, IL-6, $TNF-{\alpha})$ 생성량을 검색한 결과 50 mg/kg B.W.과 500 mg/kg B.W.의 농도에서 Con A나 LPS로 자극 시 대조군에 비해 높은 비장세포 증식능을 보였고 특히 50 mg/kg b.w. 농도로 투여한 군에서 비장세포 증식능이 최대를 나타내는 것을 알 수 있었다. 복강 대식세포에 의한 cytokine 생성량을 측정한 결과 LPS 첨가시 $IL-1{\beta}$의 분비량은 2주 경구 투여 시에 50 mg/kg B.W.의 농도군에서 가장 높은 생성량을 보였다. 이는 LPS를 투여한 군과 투여하지 않은 군에서 모두 동일한 경향을 보였다. IL-6 분비량은 LPS 첨가시 50 mg/kg B.W.의 농도 투여군에서 가장 높은 생성을 나타냈다. $TNF-{\alpha}$의 경우는 유의적인 차이를 보이지 않았다. 이상의 결과로 톳 열수 추출물은 대식세포의 활성화에 작용하여 사이토카인 생성을 증가시키는데 영향을 미칠 수 있는 것으로 예측된다. 따라서 톳 추출물은 마우스 면역계의 조절기전에 작용하여 비장세포와 대식세포의 활성화를 유도함으로써 면역세포 활성을 직접적으로 촉진시키거나 또는 이와 관련된 다른 면역반응에 영향을 미침으로서 면역 활성에 효과적으로 작용할 가능성이 있으리라 사료된다.
Increasing evidences suggest that ischemia-induced vascular damage is an integral step in the cascade of the cellular and molecular events initiated by cerebral ischemia. In the present study, employing a mouse brain endothelioma-derived cell line, bEnd.3, and oxygen-glucose deprivation (OGD) as an in vitro stroke model, the role of nuclear factor kappa B (NF-${\kappa}B$) activation during ischemic injury was investigated. OGD was found to activate NF-${\kappa}B$ and to induce bEnd.3 cell death in a time-dependent manner. OGD phosphorylated neither 32 Ser nor 42 Tyr of $I{\kappa}B{\alpha}$. OGD did not change the amount of $I{\kappa}B{\alpha}$. The extents of OGD-induced cell death after 8 h, 10 h, 12 h and 14 h of OGD were 10%, 35%, 60% and 85%, respectively. Reperfusion following OGD did not cause additional cell death, indicating no reperfusion injury after ischemic insult in cerebral endothelial cells. Three known as NF-${\kappa}B$ inhibitors, including pyrrolidine dithiocarbamate (PDTC) plus zinc, aspirin and caffeic acid phenethyl ester (CAPE), inhibited OGD-induced NF-${\kappa}B$ activation and increased OGD-induced bEnd.3 cell death in a dose dependent manner. There were no changes in the protein levels of bcl-2, bax and p53 which are modulated by NF-${\kappa}B$ activity. These results suggest that NF-${\kappa}B$ activation might be a protective mechanism for OGD-induced cell death in bEnd.3.
Baicalein, a natural flavonoid obtained from the rhizome of Scutellaria baicalensis Georgi, has been reported to have anticancer activities in several human cancer cell lines. However, its antimetastatic effects and associated mechanisms in melanoma cells have not been extensively studied. The current study examined the effects of baicalein on cell motility and anti-invasive activity using mouse melanoma B16F10 cells. Within the noncytotoxic concentration range, baicalein significantly inhibited the cell motility and invasiveness of B16F10 cells in a concentration-dependent manner. Baicalein also reduced the activity and expression of matrix metalloproteinase (MMP)-2 and -9; however, the levels of tissue inhibitor of metalloproteinase-1 and -2 were concomitantly increased. The inhibitory effects of baicalein on cell motility and invasiveness were found to be associated with its tightening of tight junction (TJ), which was demonstrated by an increase in transepithelial electrical resistance and downregulation of the claudin family of proteins. Additionally, treatment with baicalein markedly reduced the expression levels of lipopolysaccharide-induced phosphorylated Akt and the invasive activity in B16F10 cells. Taken together, these results suggest that baicalein inhibits B16F10 melanoma cell migration and invasion by reducing the expression of MMPs and tightening TJ through the suppression of claudin expression, possibly in association with a suppression of the phosphoinositide 3-kinase/Akt signaling pathway.
천연물로부터 치은에 생성되는 멜라닌의 양을 효과적으로 조절하여 치은 미백을 목표로 하는 물질을 분리하고자 문헌에 소개되고 있는 미백 효과를 가진 전통한약을 비롯하여, 민간에서 사용되고 있는 약 100여 종의 고등식물을 대상으로 B-16 mouse melanoma cell lines에서 melanin 생성 억제 효과를 screening하였다. 그 결과 천궁의 methylene chloride 분획에서 활성을 나타내어 실험재료로 선택하여 2개의 화합물을 분리하였으며 각종 spectral data를 검토하여 linoleic acid methyl ester(1), 1,3-dilinoleoyl-2-stearoyl glycerol(2)로 구조를 규명하였다. 이들 화합물은 B-16 mouse melanoma cell lines에서의 melanin 생성억제 활성을 Kojic acid를 비교 물질로하여 측정하였다. 2종의 화합물 중 1,3-dilinoleoyl-2-stearoyl glycerol은 높은 활성을 보이지 않았으나 linoleic acid methyl ester는 Kojic acid에 비하여 강한 활성을 나타내었다.
Objective: Mammalian embryos undergo changes of energy environment for transfer from oviduct to uterus. Also, the human reproductive organ (oviduct, uterus) contains energy sources of different concentration (oviduct - glucose: 0.5 mM, pyruvate: 0.32 mM, lactate: 10.5 mM; uterus - goucose: 3.15 mM, pyruvate: 0.1mM, lactate: 5.87 mM, respectively). This study was conducted to examine the effect of these energy sources added in DMEM with glutamine on the mouse embryo development. Methods: There was used ICR female mouse. Two cell embryos of mouse are collected by method of 'flushing'. Flushing fluid was used Ham's F-10 added to 20% FBS. The collected 2 cell embryos were cultured in media such as Control (only DMEM), group A and B (DMEM supplemented with 0.5 mM and 3.15 mM glucose), and group C and D (DMEM supplemented with 0.1 mM and 0.32 mM pyruvate), and group E and F (DMEM supplemented with 5.87 mM and 10.5 mM lactate). All experimental media supplemented with 20% hFF, respectively. Pattern of embryo development was observed to interval at 24hr during 96hr. Results : The media with glutamine added glucose (group A: 51.0%; group B: 48.4%) was significantly (p<0.05) higher than other experimental group in development into the morula stage after 24 hr in culture, but not significantly different compared with control and the rate of development into the blastocyst was significantly (p<0.05) low in the both of pyruvate (group C: 7.9% group D: 6.8%) and lactate (group E: 7.1%, group F: 7.1%) treatment group after 48 hr in culture. Development into the blastocyst and hatched balstocyst after 72 hr in culture revealed similarly in control (81.9%) and glucose treatment group (group A: 83.3%, group B: 82.8%). However, development into the hatched and attached blastocyst after 96hr in culture revealed significantly (p<0.05) development in the glucose treatment group (group A: 82.3%, group B: 78.5%) than control (63.2%), and its of pyruvate (group C: 34.1%, group D: 34.1%) and lactate (group E: 25.9%, group F: 33.3%) treatment group were significantly (p<0.05) lower than control similar to previous observations. Conclusion : The glucose added to the DMEM with only glutamine, as energy source, was highly to the rate of development compared with control, but the other energy sources were not, synthetically. Above refer to, the human reproductive organ (oviduct, uterus) contains energy sources of different concentration. Thus, further studies are will examine continuously to effects by interaction of different energy sources in the mouse embryo development, and these results will provide to foundation on the human embryo culture.
Ultraviolet (UV) radiation of the sunlight, especially UVA and UVB, is the primary environmental cause of skin damage, including topical inflammation, premature skin aging, and skin cancer. Previous reports show that activation of nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) in human skin fibroblasts and keratinocytes after UV exposure induces the expression and release of proinflammatory cytokines, such as interleukin-1 (IL-1) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), and subsequently leads to the production of matrix metalloproteases (MMPs) and growth factor basic fibroblast growth factor (bFGF). Here, we demonstrated that TNFR2-SKEE and TNFR2-SKE, oligopeptides from TNF receptor-associated factor 2 (TRAF2)-binding site of TNF receptor 2 (TNFR2), strongly inhibited the interaction of TNFR1 as well as TNFR2 with TRAF2. In particular, TNFR2-SKE suppressed UVB- or $TNF-{\alpha}$-induced nuclear translocalization of activated $NF-{\kappa}B$ in mouse fibroblasts. It decreased the expression of bFGF, MMPs, and COX2, which were upregulated by $TNF-{\alpha}$, and increased procollagen production, which was reduced by $TNF-{\alpha}$. Furthermore, TNFR2-SKE inhibited the UVB-induced proliferation of keratinocytes and melanocytes in the mouse skin and the infiltration of immune cells into inflamed tissues. These results suggest that TNFR2-SKE may possess the clinical potency to alleviate UV-induced photoaging in human skin.
Objectives The purpose of this study is to investigate the effect of Li Zhong Tang(LZH-T) on atopic dermatitis by using NC/Nga atopic dermatitis mouse. Methods First, in vitro, we isolated B cells from NC/Nga mouse which induced atopic dermatitis-like skin for 18 weeks. We analyzed FACS by intracellular staining of $IFN-{\gamma}$, GATA-3+ and also analyzed cytokines by using real-time PCR. Secondly, in vivo, after administration of LZH-T to the 12 weeks old mouse with atopic dermatitis. We analyzed serum IgE, $IFN-{\gamma}$ level and observed the changes of activated cell. Results In vitro, LZH-T decreased the levels of CD4+/$IFN-{\gamma}$ and increased the levels of CD4+/GATA3+. In vivo, serum IgE levels were decreased and $IFN-{\gamma}$ levels were increased in LZH-T group compared to the control group. In PBMCs, the percentage of activated cell - granulocytes, CD3+, CD3+CD4+, B220+CD23+, and CCR3+ were decreased and CD19+, CD3+CD8+ were increased in LZH-T group compared to the control group. Conclusions This study demonstrates immunological activity of GPJST on atopic dermatitis-like model mice.
The immune system and neuroendocrine systems are the two key components that maintain bodily homeostasis. Peripheral blood specimens can indicate abnormalities in a body, which often cause various threats to human health, including devastating autoimmune or metabolic diseases. To develop a treatment regimen for such diseases, experimental animal models are indispensable to researchers in academic fields. In this study, we examined the peripheral blood of 3 representative mouse strains (ICR, Balb/c, and C57Bl/6), which are widely used, to investigate whether there is a difference in reference range according to animal model. We performed hematological and chemistry analysis on individuals of both genders. The results of hematology analysis showed that the number of most types of blood cells was lower in ICR than in the other two strains. The results of chemical analysis revealed no specific pattern, but different patterns according to the individual indicator. Although the distinction between ICR and B6 was prominent, differences between Balb/c and B6 were also observed for several indicators. For some indicators, totally different patterns existed between females and males. Conclusively, this study provides the information that 3 experimentally representative mouse models have their own basal levels of blood components, suggesting the importance of a careful choice of a proper mouse model in research into immune or metabolic diseases, to exclude any biases.
본 연구는 인간 난관액 또는 자궁액 내에 존재하는 에너지원이 생쥐 2-세포기 배의 체외 발달에 미치는 영향을 조사하기 위하여 실시하였다. ICR 암 생쥐에 5 IU hCG 주사 후 $46{\sim}50$ 시간에 2-세포기 배를 회수하였다. 회수된 배는 3가지 배양 조건 [대조군: 0 mM Group A: glucose(G) 0.5 mM + pyruvate(P) 0.32 mM + lactate(L) 10.5 mM, Group B: G 3.15 mM + P 0.1 mM + L 5.83 mM]에서 72시간 배양하였다. 배양 24 시간에 상실배 출현율은 group A (72.3%)와 group B (56.6%)가 대조군(34.9%)보다 유의하게 높았다.(p<0.05). 그러나 48시간에 배반포기 배 출현율은 대조군(51.8%)이 group A (39.8%)와 group B (25.9%)보다 유의하게 (p<0.05) 높았다. 72시간에 투명대 부착 ($ZiB,\;41.0{\sim}51.8%$), 투명대 탈출 ($ZeB,\;18.1{\sim}32.5%$) 및 총 배반포기 배 출현율 ($68.7{\sim}73.5%$)은 실험군 간에 통계적인 차이가 없었다. 배반포기 배의 평균 세포수와 ICM 세포수는 group A (70.8, 13.4)와 group B (64.4, 11.8)가 대조군 (53.1, 5.7)보다 유의하게(p<0.05) 많았고, 통계적인 유의차는 없었으나 group A가 group B보다 많은 경향이었다. 총 세포수에 대한 ICM 비율은 group A(22.9%)와 group B(23.7%)가 대조군(14.2%)보다 유의하게(p<0.05) 높았다. 영양배엽(TE) 세포수($34.1{\sim}45.1$)는 실험군 간에 통계적인 차이가 없었다. ICM에 대한 TE 비율(ICM:TE ratio)은 대조군(1:6.0)이 group A(1:3.4)나 group B(1:3.4)보다 유의하게(p<0.05) 높았다. 생쥐 2-세포기 배를 배양하여 72시간까지의 배 발달율을 살펴보면 배양액에 에너지원을 첨가하는 것이 효과적이었으며, 자궁액 농도보다는 난관액 농도로 에너지원을 조절했을 때 배 발생 능력이 높은 경향을 보였다.
