  • Regional distribution and relative frequency of endocrine cells in ten portions of the gastrointestinal tract of the Korean native cattle were observed by immunocytochemical methods using specific antisera against chromogranin, serotonin, somatostatin, glucagon, bovine pancreatic polypeptide(BPP), motilin, gastric inhibitory polypeptide(GIP), neurotensin, secretin, gastrin and substance P. The results observed are summarized as follows: In the abomasum, chromogranin-, serotonin-, somatostatin-, motilin-, glucagon-, gastrin-, and substance P-immunoreactive cells were found. Chromogranin-and serotonin-immunoreactive cells were more numerous in the fundic region than pyloric region. Somatostatin- and gastrinimmunoreactive cells were numerous in the pyloric region than in the fundic region. In the small intestine, chromogranin-, serotonin-, somatostatin-, glucagon-, BPP-, motilin-, gastrin-, GIP-, neurotensin-, secretin-, and substance P-immunoreactive cells were detected. Chromogranin-, somatostatin-, GIP- and secretin-immunoreactive cells were most numerous in the duodenum, while BPP-, motilin-, glucagon-, neurotensin- and substance P-immunoreactive cells were rarely seen in the small intestine. In the large intestine, chromogranin-, serotonin- and BPP-immunoreactive cells were widely distributed and most numerous in the rectum. Somatostatin-, glucagon- and substance P-immunoreactive cells were rarely seen in the large intestine.

  • Interstitial cells of Cajal (ICCs) are pacemaker cells that exhibit periodic spontaneous depolarization in the gastrointestinal (GI) tract and generate pacemaker potentials. In this study, we investigated the effects of ghrelin and motilin on the pacemaker potentials of ICCs isolated from the mouse small intestine. Using the whole-cell patch-clamp configuration, we demonstrated that ghrelin depolarized pacemaker potentials of cultured ICCs in a dose-dependent manner. The ghrelin receptor antagonist [D-Lys] GHRP-6 completely inhibited this ghrelin-induced depolarization. Intracellular guanosine 5'-diphosphate-${\beta}$-S and pre-treatment with $Ca^{2+}$-free solution or thapsigargin also blocked the ghrelin-induced depolarization. To investigate the involvement of inositol triphosphate ($IP_3$), Rho kinase, and protein kinase C (PKC) in ghrelin-mediated pacemaker potential depolarization of ICCs, we used the $IP_3$ receptor inhibitors 2-aminoethoxydiphenyl borate and xestospongin C, the Rho kinase inhibitor Y-27632, and the PKC inhibitors staurosporine, Go6976, and rottlerin. All inhibitors except rottlerin blocked the ghrelin-induced pacemaker potential depolarization of ICCs. In addition, motilin depolarized the pacemaker potentials of ICCs in a similar dose-dependent manner as ghrelin, and this was also completely inhibited by [D-Lys] GHRP-6. These results suggest that ghrelin induced the pacemaker potential depolarization through the ghrelin receptor in a G protein-, $IP_3$-, Rho kinase-, and PKC-dependent manner via intracellular and extracellular $Ca^{2+}$ regulation. In addition, motilin was able to depolarize the pacemaker potentials of ICCs through the ghrelin receptor. Therefore, ghrelin and its receptor may modulate GI motility by acting on ICCs in the murine small intestine.

  • In order to investigate the regional distribution and relative frequency of the serotonin-, somatostatin- and motilin-immunoreactive cells in the gastrointestinal tract GIT) of the bullfrog (Rana catesbeiana) with developmental stages, group of bullfrogs subdivided into the tadpole with hindlegs, metamorphosed bullfrog with tail, 2 wk after metamorphosed bullfrog and adult bullfrog, were stained by immunohistochemical methods (PAP methods). Multifarious changes of the regional distribution and relative frequency of the gastrointestinal endocrine cells were observed as developmental stages but motilin-immunoreactive cell ere not detected in this study. Serotonin-immunoreactive cells were detected from 2 wk after metamorphosed bullfrog in the whole GIT. The positive cells were predominant in the pylorus regions. In the adult bullfrog, these cells increased in fundus. On the other hand, somatostatin-immunoreactive cells were observed from the tadpole with hindlegs in the stomach regions. In the metamorphosed bullfrog with tail, they were found in the whole GIT except for the ileum, and increased with developmental stages except for rectum. In the rectum, these cells were not detected after that period. Our results show that most of the immunoreactive cells appear from completed metamorphosed periods when the histological structure of GIT is completely differentiated.

  • The relative frequency and distribution of occurrence of immunoreactive cells in the proventriculus, diverticulum, cardia, fundus and pylorus of the stomach of pigs were investigated by PAP method using specific antisera against BCG, Gas/CCK, 5-HT, somatostatin, glucagon, BPP, motilin and insulin. In the diverticulum and cardia, BCG-, 5-HT-, somatostatin- and glucagon-immunoreactive cells were detected. In the fundus, BCG-, 5-HT- and somatostatin-immunoreactive cells were also found. In the pylorus, BCG-, Gas/CCK-, 5-HT-, somatostatin- and glucagon-immunoreactive cells were observed. However, no BPP-, motilin- and insulin-immunoreactive cells were found in the stomach epithelium of the pigs. These results showed that the occurrence of the endocrine cells confirmed in the diverticulum as the cardia and suggest that the function of diverticulum may be similar to that of cardia in the pigs.

  • We previously identified quantitative trait loci (QTL) for body weight and average daily gain in a common region between ADL0198 (chr 1: 171.7 Mb) and ABR0287 (chr 1: 173.4 Mb) on chicken chromosome 1 in an $F_2$ resource population produced by crossing low- and high-growth lines of the Hinai-dori breed. Motilin receptor (MLNR) is a candidate gene affecting growth traits in the region. In this study, we genotyped polymorphisms of the MLNR gene and investigated its association with growth traits in a Hinai-dori $F_2$ intercross population. All the exons of the MLNR gene in the parental population were subjected to PCR amplification, nucleotide sequenced and haplotypes identified. To distinguish resultant diplotype individuals in the $F_2$ population, a mismatch amplification mutation assay was performed. Three haplotypes (Haplotypes 1-3) were accordingly identified. Six genotypes produced by the combination of three haplotypes (Haplotype 1, 2, and 3) were examined in order to identify associations between MLNR haplotypes and growth traits. The data showed that Haplotype 1 was superior to Haplotype 2 and 3 in body weight at 10 and 14 weeks of age, average daily gain between 4 and 10 weeks, 10 and 14 weeks, and 0 and 14 weeks of age in female in $F_2$ females. It was concluded that MLNR is a useful marker of growth traits and could be used to develop strategies for improving growth traits in the Hinai-dori breed.

The results obtained are summarized as follows: 1. Two kinds of endocrine cells were demonstrated with two specific staining methods. Argyrophil cells(reactive cells for Grimelius method) were found most frequently in the intestinal region, and were infrequent in the rectum, whereas argentaffin cells (reactive cells for Masson-Hamperl method) were found most frequently in the rectum and in the other legions were infrequent. These reacting cells were mainly found in the intestinal glands, whereas a small number in the mucosa. 2. Twelve kinds of endocrine cells, gastrin(Gas)-, somatostatin (Som)-, serotonin(5-HT)-, glucagon(Glu)-, bovine pancreatic polypeptide(Bpp)-, cholecystokinin(Cck)-, secretin(Sec)-, motilin(Mot)-, glicentin(Gli)-, gastric inhibitor polypeptide(GIp)-, substance P(Sp)-, and neurotensin(Neu)-immunoreactive cells, were identified by immunohistochemical method. Gas-, Som-, 5-HT-, Glu-, Cck-, Sec-, Mot-, Gli-, Sp-, and Neu-reactive cells were observed in the duodenum, and among these Gas- and 5-HT-reactive cells were moderately found while the others were infrequent. In the jejunoileum Gas-, Som-, 5-HT-, Glu-, Cck-, Mot-, Gli-, GIp-, SP-, and Neu-reactive cells were found, and among these 5-HT- and GIp-reactive cells were moderately found while the others were infrequent. In the colon Sec-reactive cell was not detected. 5-HT-reactive cells were found most frequently and the others were infrequent in this region. 5-HT-, Bpp-, GIi- and Neu-reactive cells were found in the rectum. Among these 5-HT-reactive cells were found most frequently. 3. Electron microscopically, five types of endocrine cells, EC, ECL, D, G, A-like cell, were identified in the intestinal region. • The regional distribution and relative frequency of gastrointestinal endocrine cells were studied immunohistochemically in the gastrointestinal mucosa and pancreas of the fresh water turtle. Ten kinds of endocrine cells were identified in the gastrointestinal tract. Cholecystokinin-8-, bovine pancreatic polypetide-and glucagon-immunoreactive cells were seen throughout the gastrointestinal tract, also among them cholecystokinin-8-immunoreactive cells were most predominant in segment III. Although gastrin- and gastrin/cholecystokinin-immunoreactive cells were found from segment III to VI and X, respectively, they were numerous in segment III. Somatostatin-immunoreactive cells were observed from segment I to VII. 5-hydroxytryptamine- immunoreactive cells were detected in segment I, III, VIII, IX and X. Human pancreatic polypeptide-immunoreactive cells were demonstrated in segment V, VI, VIII, IX and X. Insulin-immunoreactive cells were found from segment III to X except for segment VIII, but rare in segment VII. Neurotensin-immunoreactive cells were found to be restricted to segment VIII, IX and X. No porcine chromogranin-, substance P- and bombesin-immunoreactive cells were detected throughout the gastrointestinal tract of the fresh water turtle. Although typical mammalian pancreatic islets encapsulated by connective tissue were not present in this species, five kinds of endocrine cells-glucagon, insulin, somatostatin, bovine pancreatic polypeptide and 5-hydroxytryptamine-were found in forming small or large groups and scattered in the exocrine gland region. However porcine chromogranin- and motilin-immunoreactive cells could not be demonstrated in the pancreas.
