DNA chip 통합분석 프로그램을 이용한 효모의 세포주기 유전자 발현 통합 데이터의 분석 (Analysis of Combined Yeast Cell Cycle Data by Using the Integrated Analysis Program for DNA chip)

  • 효모의 세포주기 관련 유전자 발현 통합 데이터를 사용하여 본 연구실에서 개발한 유전자 발현 통합 분석프로그램을 사용하여, 클러스터링 알고리즘의 성능을 비교하고 데이터내에 존재하는 클러스터 개수를 추정하기 위해 FOM 분석을 적용하였으며, 이 분석방법을 통하여 K-means, SOM, Fuzzy c-means 클러스터링 방법의 성능을 서로 비교하였다. 클러스터 개수를 추정한 다음 3가지 클러스터링 방법에 대한 클러스터링 결과 비교, 클러스터의 기능할당 및 모티프 분석을 시도하였다. 본 논문에서 제시하는 분석 방법은 DNA chip 발현 데이터의 일반적인 분석방법을 유전자 발현 패턴의 유사성을 토대로 한 클러스터링 방법에 근간을 두고 있다. 본 논문에서는 클러스터링한 후 각 클러스터의 기능할당 및 모티프 분석에 대한 일반적인 분석방법을 제시하였으며, 본 연구실에서 개발한 유전자 발현분석 통합 프로그램이 효율적으로 사용될 수 있음을 보여주고 있다.

해양 생물 모티브를 응용한 크루즈 룩 디자인 (Cruise Look Design Inspired by the Ocean Life Motif)

  • The purpose of this study was to develop cruise look design inspired by the ocean life motif using by computer CAD system. Domestic and international brands were selected for this study researching cruise line collection. A variety of literature and online site for concept of cruise look and sustainable design was studied. Adobe Illustrator CS2 and Adobe Photoshop CS2 program were used for cruise look design through analysis of 07/08 S/S trend. The diffusion of cruise look reflects a societal trend to be changed life pattern. The results of this study are as below. First, as this society has become modernized with advancement of technology, people have been increasingly interested in the pursuit of happiness and the concept of 'quality of life', thereby creating a new trend of consumption culture. As a result, fashion market place changed targeting sensible and self-enrichment customers. Second, as combining rapid technological innovation and mass production, industrial design caused environmental pollution and the problems are being worsen. Fashion design has same problems and led to eco-friendly design by the large. Third, as the change of style influenced by economic and social condition, cruise look re-defined and became more common. The change of various fields led to fashion market place should be changed. The cruise look design using computer program will give more benefits to many designers.

조각보의 조형성을 응용한 패턴의 감성 이미지 연구 -면 구성과 색채를 중심으로- (Research on Sensibility Image of Pattern Applying the Formative Elements of a Traditional Jokakbo -The Surface Composition and Colors-)

  • Through the formative analysis of the traditional jokakbo, we have composed surface composition that was made by a perfect square, triangle, a weather vane type, vertical type, cintamani type, and a mixed rectangular. And we have composed 18 kinds of motif stimulants that was made by vivid tone, pale tone, and so on, and finally analyzed sensibility image, preference rate of them. The cause of composition for the sensibility image of stimulant was composed by the cause of simplicity characteristics, interesting characteristics, rigid-flexibility characteristics, and modern characteristics. And from these things, the cause of simplicity characteristics and the cause of interesting characteristics were revealed as important dimensional factors of sensibility image. The sensibility image of motif was revealed as a simple image of perfect square, a feminine image of cintamani type, a modern image of mixed rectangular. And black-white colored mixed rectangular and chromatic colored cintamani type were also revealed as an interesting image. The preference rate of motif is related with the cause of interesting characteristics and simplicity characteristics, it also has been revealed to prefer the pale tone than others. And it also has been revealed that achromatic colored mixed rectangular, cintamani type of vivid tone, perfect square and rectangular of pale tone were mostly preferred.

The Construction of Regulatory Network for Insulin-Mediated Genes by Integrating Methods Based on Transcription Factor Binding Motifs and Gene Expression Variations

  • Type 2 diabetes mellitus is a complex metabolic disorder associated with multiple genetic, developmental and environmental factors. The recent advances in gene expression microarray technologies as well as network-based analysis methodologies provide groundbreaking opportunities to study type 2 diabetes mellitus. In the present study, we used previously published gene expression microarray datasets of human skeletal muscle samples collected from 20 insulin sensitive individuals before and after insulin treatment in order to construct insulin-mediated regulatory network. Based on a motif discovery method implemented by iRegulon, a Cytoscape app, we identified 25 candidate regulons, motifs of which were enriched among the promoters of 478 up-regulated genes and 82 down-regulated genes. We then looked for a hierarchical network of the candidate regulators, in such a way that the conditional combination of their expression changes may explain those of their target genes. Using Genomica, a software tool for regulatory network construction, we obtained a hierarchical network of eight regulons that were used to map insulin downstream signaling network. Taken together, the results illustrate the benefits of combining completely different methods such as motif-based regulatory factor discovery and expression level-based construction of regulatory network of their target genes in understanding insulin induced biological processes and signaling pathways.

지각자의 내적 특질에 따른 문양의 감성이미지 및 선호도 (Effect of Perceiver's Internal Traits on Sensory Image of Pattern Categories and Colors)

  • The purpose of the study was to examine the effect of internal traits of perceivers on sensory images of fashion materials and dress wearers. Perceiver's internal traits included aspects of Gender-body Ideology (GBI), as well as fashion lifestyles and fashion interest. The research was conducted on a quasi experimental basis, with subjects numbering 187 male and 207 female college students. Data were collected in the period from March 19th to March 31st, 2007. Two sets of stimuli and three measuring tools (to assess GBI, fashion life styles and fashion interest) were used. The stimuli were thirteen fabric specimens and thirteen photographs of dress styles, manipulating by two variables: (a) motif colors (white, grey, pinks and blue); (b) motif categories (plain, paisley, floral, stripes, and zebra effect). The data were analysed by factor analysis, ANOVA, Duncan's multiple range test. The major findings were as follows: Gender-body ideology was separated into agonic and hedonic dimensions. Fashion life style was separated into 3 dimensions(shopping life-style pursuit, social activity pursuits and brand life style pursuits). The internal traits of perceivers influenced preferences and buying intentions. Shopping life-style pursuits had more impact on preferences and buying intentions than other internal traits.

효모 ABF1 단백질의 DNA Binding 부위에 대한 구조 기능 연구 (Structure-Function Analysis of DNA Binding Domain of the Yeast ABF1 Protein)

  • ABF1(Autonomously replicating sequence Binding Factor 1)은 효모 genome에서 $RTCRYN_5ACG$의 염기 서열을 가지고 있는 promoter, mating-type silencer, ARS에 결합하는 DNA binding 단백질이다. E. coli 에서 ABF1 유전자를 발현하기 위하여, ABF1 유전자를 pMAL-c2 벡터에 cloning하였다.(pMAHW). pMAHW를 E. coli에 형질전환하여, ABF1 융합단백질을 발현시키고, amylose resin affinity chromatography에 의하여 분리하였다. Factor Xa protease를 이용하여 분리된 융합단백질로부터 maltose binding protein을 잘라낸 후에 gel retardation analysis 방법으로 분리된 ABF1이 ARS1에 결합하는 능력을 지니고 있음을 확인하였다. DNA 결합에 관련된 부위를 찾기 위하여, 비전형적인 zinc finger motif가 위치하는 자리에서 pMAHW의 ABF1 유전자에 His-61을 다른 아미노산으로 치환하였다. DNA binding 부위로 추정되는 ABF1 단백질의 중간지역에 Leu-353, Leu-360를 다른 아미노산으로 치환하였다. Site-specific mutagenesis 를 통해 만들어진 mutant를 gel retardation analysis와 complementation test를 통해서 비전형적인 zinc finger motif이외에 다른 DNA binding motif가 있는 것을 알 수 있었다.

인상주의 회화의 화면등가의 법칙에 기반 한 패션디자인 연구 (An Analysis of Fashion Designs Based on the Laws of the Screen Equivalent of Impressionist Paintings)

  • This study reviews the principles for the techniques of Impressionist paintings as well as analyzed contemporary fashion designs with a focus on a motif-building technique based on the laws of a screen equivalent as a visual formative approach. We provide design principles based on fashion design painting techniques. Previous research on the laws of the screen equivalent of Impressionist paintings were studied and a qualitative analysis was conducted on fashion design cases from 2011, 2012 S/S and F/W collections. The analysis resulted in the following outcomes. First, the development of new motifs were found directly correlated to the creativity of design if it was a motif-building design. Second, in the selected fashion design cases, cutting lines and details were covered by motifs and their shapes collapsed in regards to overall visual uniformity so that specific details were hard to identify. Third, clothing shapes are recognized the changing colors of motifs and not through construction pattern lines; therefore, the expressions of diverse visual forms were available without being disturbed by construction pattern lines. This is deemed equivalent to an Impressionist painting style that depicts shapes with colors instead of lines. Lastly, the cases covered in this study have created new visual aspects that replace the stereoscopic spatial depth of clothes with a 'sensuous surface'. The pleasures derived from the sensuous surface are deemed equivalent to the visual pleasures created by Impressionist paintings.

텍스타일 디자이너의 인지 모형에 대한 탐색적 접근 -모티브 개발 단계를 중심으로- (An Exploratory Approach to Textile Designer's Cognition Model -focused on the Stage of Motif Development-)

  • 본 연구는 비디오/오디오 프로토콜 분석 방식을 이용해서 텍스타일 디자인 프로세스 중 모티브 개발 단계에서의 디자이너의 인지 모형을 분석하기 위한 탐색적 연구이다. 텍스타일 디자인 프로세스에 대한 프로토콜 분석을 위해 선행 연구를 고찰하여 디자인 행동 범주의 틀을 하향식(top-down)방식으로 설정하고 실제 행동 프로토콜(action protocol) 분석을 통해 상향식(bottom-up)방식으로 세부 디자인 행동을 분석하였다. 본 연구에서는 모델 휴먼 프로세서(Model Human Processor)이론에 기초하여 텍스타일 디자인 행동을 모터 행동(motor action), 지각 행동(perceptual action), 인지 행동(cognitive action)의 세 가지 범주로 나누었으며 이를 토대로 하여 텍스타일 디자인 프로세스에 적합한 코딩 스킴(coding scheme)을 개발하였다. 본 연구에서는 이 새로운 코딩 스킴을 토대로 하여 모티브 개발 단계에서의 텍스타일 디자이너의 인지 모형을 제시하였다.

Structural resemblance of the DNAJA-family protein, Tid1, to the DNAJB-family Hsp40

  • The specific pair of heat shock protein 70 (Hsp70) and Hsp40 constitutes an essential molecular chaperone system involved in numerous cellular processes, including the proper folding/refolding and transport of proteins. Hsp40 family members are characterized by the presence of a conserved J-domain (JD) that functions as a co-chaperone of Hsp70. Tumorous imaginal disc 1 (Tid1) is a tumor suppressor protein belonging to the DNAJA3 subfamily of Hsp40 and functions as a co-chaperone of the mitochondrial Hsp70, mortalin. In this work, we performed nuclear magnetic resonance spectroscopy to determine the solution structure of JD and its interaction with the glycine/phenylalanine-rich region (GF-motif) of human Tid1. Notably, Tid1-JD, whose conformation was consistent with that of the DNAJB1 JD, appeared to stably interact with its subsequent GF-motif region. Collectively with our sequence analysis, the present results demonstrate that the functional and regulatory mode of Tid1 resembles that of the DNAJB1 subfamily members rather than DNAJA1 or DNAJA2 subfamily proteins. Therefore, it is suggested that an allosteric interaction between mortalin and Tid1 is involved in the mitochondrial Hsp70/Hsp40 chaperone system.
