Morphological and rheological properties of culture broth of Cephalosporium acremonium M25

  • Cephalosporium acremonium is a filamentous microorganism producing cephalosporin C. The morphological differentiation of C. acremonium in submerged culture is closely related with the rheological properties of culture broth and production of cephalosporin C. In this study, the rheological and morphological properties of culture broth of C. acremonium were investigated. In the seed broths of shake-flask and fermenter culture, the Herschel-Berkley equation was in excellent agreement with experimental results in the whole range of shear rate. In the seed broths of shake-flask culture, morphological differentiation into arthrospores affected to changes of apparent viscosity. But results in the fermenter culture, morphological factors such as mean hyphal thickness and the number of tips gave more effect on changes of apparent vitacosity than differentiation into arthrospores. Overall, it suggested that the morphological parameters measured by image analysis can be used as a good parameter to indicate the rheological properties of culture broth of C. acremonium M25.

β-Carotene, Cucumisin Content and Fruit Morphology of Melon (Cucumis melo L.) Germplasm Collections

  • Melon fruits are a year-old plant in the family Cucurbitae and are the most cultivated fruit in tropical countries. Melon flesh is an important source of anti-cancer, antioxidant effects, attracting attention as a functional food. We investigated the morphological properties, β-carotene content, and cucumisin activity of seeds of 58 melon genetic resources. Melon resources have shown various morphological properties. Melons grouped by morphological properties were the five groups. β-carotene content varied between 82.34 mg/kg, 86.75 mg/kg, 25.56 mg/kg, 86.25 mg/kg, and 54.65 mg/kg. Between β-carotene, cucumisin activity and other quantitative fruit morphological properties, the color of the pulp and the firmness of the pulp had a significant amount of correlation between the β-carotene content. However, cucumisin activity and β-carotene content had significant negative correlation, and the color of the fruit and shape of the fruit had significant negative correlation with β-carotene content and activity of cucumisin. Our study vindicated that high diversity in melon morphological characters of genetic sources that provide beneficial baseline data for the future and in the activity of β-carotene and cucumisin, and these results could predict indicators of β-carotene content by the length of leaf, the width of fruit, the length of joint, and the length of seed, and collectively the morphological properties of melons could help predict indicators of β-carotene content and help develop functional sarcoma and farmhouse cultivation.

Characterization of Biometry and Chemical and Morphological Properties of Fibers from Bagasse, Corn, Sunflower, Rice, and Rapeseed Residues in Iran

  • The biometry, morphological properties and chemical composition of bagasse, corn, sunflower, rice, and rapeseed residues plants were analyzed. The results revealed differences in biometry properties and chemical composition of the different types of agricultural resides investigated. The greatest proportion of fiber length (1.32 mm) and cellulose (55.56%) was found in residues of bagasse plants, with a low ash (1.78%) and lignin (20.5%). The lignin of all types of agricultural resides was less than hardwood and softwood. In addition, the rice and rapeseed residues plants had highest amount of ash and extractive component. The slenderness and flexibility ratios of the all types of agricultural resides fibers were similar to some of hardwood and softwood species.

  • Corn stalk, one of the most abundant agricultural residue in the world, was examined in this study to use it as an alternative fiber source of wood fiber. In order to find the proper way to utilize corn stalk more efficiently, the morphological properties and the papermaking properties of the, bast fibers and the corn pith, were evaluated respectively. Although the bast fiber fraction showed comparable properties to those for hardwoods in the fiber properties and the papermaking properties, the pith resulted in low brightness and low drainage rate. But the short and flexible fibers in pith fraction led to dense and compact handsheet structure, correspondingly the higher sheet strength. There big differences in properties between bast fiber and pith should be considered for the fully utilization of corn stalk.

  • The morphological properties of the traditional handmade papers made in Korea, China and Japan were identified using CLSM(Confocal Laser Scanning Microscope). It was shown that the primary and secondary wall of the Hanji fibers were separated. The lumen of the fibers was not observed. Dochim, a traditional way of Hanji calendering, was turned out to accelerate the separation of the two walls, by which the unique effects of Dochim could be obtained. There were no separation of the two walls in Chinese handmade paper and Japanese handmade paper. Lumen was not observed in Senji fibers but it was observed in Whasi fibers. It was postulated that the morphological properties of the fibers are closely related with the calligraphic writings or drawings properties of handmade papers.

Effects of Kneading Treatment on the Properties of Various Pulp Fibers (Kneading 처리가 다양한 펄프 섬유들의 특성에 미치는 영향)

  • In this study, effects of kneading treatment on the properties of hardwood bleached kraft pulp (HwBKP), softwood bleached kraft pulp (SwBKP) and hardwood bleached chemi-thermo-mechanical pulp (HwBCTMP) were elucidated with a laboratory two-shaft kneader. Kneading treatment was performed at 30% (w/w) of pulp concentration and the number of passes through the kneader was adjusted from 0 to 10 passes. Then, changes in properties of pulp fibers were evaluated. It was found that fiber characteristics were influenced by kneading treatment. Fiber length was decreased with kneading while other morphological properties such as fiber width, curl and kink became increased as the number of passes through the kneader increased from 0 to 5 passes. The magnitude of changes in the morphological properties of softwood chemical pulp was the largest, followed by hardwood chemical pulp. The morphological properties of HwBCTMP were little influenced by kneading treatment. Swelling of fiber measured by WRV was increased with kneading except of HwBCTMP.

Morphological Properties and Target Strength Characteristics for dark banded rockfish (Sebastes inermis) (볼락의 형태학적 특징과 음향반사강도 특성)

  • Morphological properties of dark banded rockfish (Sebastes inermis) were analysed to investigate its acoustic scattering characteristics. Total of 18 live samples was prepared for X-ray photos and collected morphological coordinates of their body and swim bladder shapes. Kirchhoff-ray mode model was used to calculate acoustic scattering pattern for broad-band frequency range. Inclination of swim bladder ranged from 17 to 30 and the averaged value was about $25.2^{\circ}$ (S.D.(standard deviation)=3.15). There were no any tendency of increase or decrease in volume and area ratio of swim bladder to fish body and ranged from 2.2 % to 4.43 % and 14.85 % to 21.31 %, respectively. The averaged value of volume and area ratio was 3.13 % (S.D.=0.52) and 17.6 % (S.D.=1.5). $b_{20}$ values were -69.01 for 38 kHz, -69.83 for 70 kHz, -70.17 for 120 kHz and -70.93 for 200 kHz, recpectively. Broadband acoustic patterns of dark banded rockfish for 20 ~ 200 kHz were similar among samples and they reflected size and morphological properties of fish species.

Analysis of Reconstituted Tobacco Products by Characterizing Morphological Properties of Major Structure Materials (국내외산 판상엽 구성물질의 형태적 특성 비교)

  • The morphological properties of various structure materials of domestic and foreign reconstituted tobacco products(RTP) were investigated by using the Bauer-McNett classifier and the image analyzer. The results of the fiber classification showed the fraction of the bigger size structure materials was larger in a domestic RTP than that in two foreign RTPs. In case of fine fraction, the domestic RTP had bigger fine fraction than two foreign RTPs. Images of each structure materials showed the scrap in the foreign RTPs kept the original shape which were rare in the domestic RTP fractions. Those results deduced that the raw materials in a foreign RTP process might be treated separately depending on the mechanical and morphological properties, which could reduce the amount of fine generation and increase the efficiency in raw material treatment.

The Effect of Electron Beam Irradiation on Chemical and Morphological Properties of Hansan Ramie Fibers

  • The purpose of this study investigates the effects of electron beam(EB) irradiation on the chemical and morphological properties of Hansan ramie fiber. Hansan ramie fibers were irradiated with electron beam doses of 0, 1, 3, 5 and 10kGy. The effect of electron beam irradiation on the chemical components of fibers as well as the surface chemical and morphological properties were investigated using chemical component analysis methods based on TAPPI standards, XPS, and SEM. The results indicate that the surface layers can be removed under suitable EB irradiation doses. Alcohol-benzene extraction and lignin content increases gradually with an increase in EB irradiation and reaching a maximum at an EB dose of 3kGy, and decreases at 10kGy. The surface chemical changes measured by XPS corresponded to the chemical composition analysis results. The C1 peak and the O/C ratio decreased with the removal of the multi-layer and primary layer by EB irradiation. The SEM images show the inter-fibrillar structure etched by EB irradiation up to 5kGy. At 10kGy, the surface structure of the ramie fiber shows highly aligned and distinctive striations in a longitudinal direction. The removal of these exterior layers of the fiber was confirmed by changes in surface morphology as observed in SEM images.

Chemical Composition of Rice Hull and Morphological Properties of Rice Hull Fibers (왕겨의 화학적 조성 및 왕겨섬유의 형태적 특성)

  • The rice hull could be the one of the most abundant agricultural waste in Korea. Since the efficient utilization of agricultural waste or byproducts of food industry would be critical for the sustainable growth, this study conducted the investigation of the chemical composition and the morphological properties of rice hull and rice hull fiber. It was found that there was big difference between the outer surface and the inner surface in the chemical composition and the morphological properties. Expecially, the outer surface showed the rugged patterns in which most of silica of rice hull existed. Little or no silica was found in the inner surface and rice hull fiber. The average fiber length of rice hull fiber was 0.45mm which was shorter than that of hardwood fiber. Rice hull fiber showed a round long shape which is typical shape of non-wood fibers
