Gentiana scabra Bunge var buergeri Max, can be widely used such as cut-flower, ornamental and medicinal plant among the native plants. Microscopic and Scanning electron microscopic observation plant were carried out to estimates the characteristics correlated with germinability and germination process, and the results were as follows. The seeds were germinated just after harvest, and it means that there is no or little dormancy of the seeds. Matured seeds showed higher germinability than those of immatured. As the storage period was longer, immatured seeds showed shorter longevity than those of matured. The average longevity was about 190 days. According to the observation results of morphological changes during the germination, seed coat was expanded with water absorption immediately after seeding, and radicals were observed 2 days after seeding. Root hairs were formed behind the growing tip of the root on 4 days after seeding and cotyledon were unfolded on 6 days after seeding. In the microscopic observation of leaf tissues, it could be easily distinguished between midrib, and leaf blade, and also observed upper epidermis, palisade parenchyma and spongy parenchyma consisted as a sing1e layer. Palisade parenchyma was consisted of a single layer of cell. Stomata were restricted lower surface, arranged as anamocytic type, and surrounded by guard cells. In the microscopic anatomy of stem were consisted of cortical layer, forming a single layer of epidermis and parenchyma, and vascular bundle which was consisted of continuous cylinder of vascular tissues.

Objectives : This study was designed to establish a characteristic discrimination of internal and external morphological standard of original plants and herbal states in Pogostemonis and Agastachis Herba. Methods : In this studies, the external-internal morphological standards were determined by using stereoscope and butanol series. Results : 1. The external characteristics: Pogostemon cablin has hairs and brown-like in stem, elliptical fruit. On the other hand, Agastache rugosa has no hairs and red-like in stem, obovatic trigone fruit. 2. The physical characteristics: Pogostemon cablin is gray in whole, has hairs in stem and numerous hairs of ash in leaf. On the other hand, Agastache rugosa is yellow-green in whole, has no hairs in stem. Specially the latter has deep-green colour and numerous hairs presenting mostly at lower epidermis in leaf. 3. The physical characteristics in currents: Pogostemon cablin is brown, has hairs and round-like stem. On the other hand, Agastache rugosa is green or yellow-green, has no hairs and tetragon in stem. 4. The internal characteristics: Pogostemon cablin has progressed spongy tissue in epidermal cell of leaf and many rank of epidermal cell in stem. On other hand, Agastache rugosa has I rank palisade tissue in leaf and few rank of epidermal cell in stem. In the external shape, it was possible that herbs were distinguished according to artificial cIassification and that same genus-degree of relatedness among herbs could be distinguished by more precise and active observation. In the shape of real herbs, I compared current herbs in market with original herbs which were just collected or were on the course of drying. In addition, it was possible that the internal shape could be identified by using microscope after butanol series. Conclusion : Though it was impossible to make distinction of herbs which are not current in my search contents, this search contents will be a standard for applying herbs in the future. An Additional standard establishment including physiochemical reaction and gene research is required in order to supplement the fault of this search.
Present study was performed to describe the morphological and molecular characterization of Toxocara tanuki (Nematoda: Ascaridae) from Korean raccoon dog, Nyctereutes procyonoides koreensis, naturally infected in the Republic of Korea (Korea). Juvenile and adult worms of T. tanuki were recovered in 5 out of 10 raccoon dogs examined and the larval worms were detected in 15 out of 20 muscle samples (75%). Small lateral alae were observed on the cranial end of the body in male and female adults and 2 long spicules (3.0-3.5 mm) were characteristically observed in the posterior end of males. In SEM observation, 18 pairs of proximal precloacal, a precloacal median, a postcloacal median and 5 pairs of postcloacal papillae were uniquely revealed in the posterior portion of males, but the proximal papillae were not shown in the lateral ends of females. Molecular analysis on the 18S rRNA partial DNA sequences was revealed the same finding in both samples, adult worms and muscle larvae, which are closely related to T. tanuki. In conclusion, it was confirmed for the first time that T. tanuki is indigenously distributed, the Korean raccoon dog is acted as the natural definitive host of this nematode in Korea and the morphological characteristics of T. tanuki were shown in specific structure for single postcloacal median papilla in male.
High temperature corrosion behavior of AISI-type 316L stainless steel for the MCFC(molten carbonate fuel cell) bipolar application was studied by immersion test and penetration attack method in anode environment ($650^{\circ}C$, $Li_2CO_3/K_2CO_3=62/38$ mol%, $H_2/CO_2=80/20$ vol%) without or with different $CaCO_3$ content. Not only immersion test method but also morphological observation of samples in the carbonate melts are adopted as experimental methods. With aid of the morphological observation of cross section of samples immersed in a carbonate melt was possible to obtain penetration attack. The concentration effect of $CaCO_3$ inhibitor was investigated in order to verify the optimum concentration for practical application in MCFC stack operation. The corrosion rate in the presence of $CaCO_3$ was proven to be decreased as a function of $CaCO_3$ concentration. The corrosion rate in the presence of $CaCO_3$ was measured with a value of 6.9 mpy which is 2.4 times lower than that of inhibitor-free electrolyte. The cross section microscopy revealed that the internal penetration by oxidation in molten carbonate is very severe. In this case, the attack was occurred not only dissolution loss in the electrolyte by corrosion reaction but also weight gain through oxide layer by internal penetration.

Twenty-nine culture strains belonging to the genus Alexandrium Halim (Dinophyceae) were established from water column or sediments in Korea. Seventeen isolates were identified as A. tamarense (Lebour) Balech, eight isolates as A. sp. cf. catenella and one as A. catenella (Whedon et Kofoid) Balech according to the presence or absence of a ventral pore, the shape of the posterior sulcal plate and the sulcal width. Three isolates were unable to be identified due to considerable distortion of thecal plates and lack of enough materials, but typical of A. tamarense and/or A. catenella. The overall cell shape of A. tamarense was usually longer than wide. The posterior sulcal plate was definitely longer than wide dorsoventrally, and sulcus extended posteriorly without apparent widening. They were distributed in three major coasts of Korea. In contrast, the cell shape of A. sp.cf. catenella was generally anterior-posteriorly flattened. The transversal axis of the posterior sulcal plate was always longer than the longitudinal, or both axes were nearly equal in length. Its sulcus was broader than that of A. tamarense and widened in the direction of antapex about 1.5 times. This morphotype existed in nearshore and offshore waters of the southern Korea sea. One of A. catenella isolates from Jinhae Bay showed no conspicuous differences with A. sp. cf. catenella except for the consistent absence of a ventral pore.
In the course of an investigation on the morphological variation of Korean striped bittering, Acheilognathus vamatsutae Mori, two kinds of morrholoflical types were found. Some morphological characters were compared between them and multivariate analysis was conducted. Since the difference of the barbel length between them was very significant (p < 0.01,1-test), they should be designated as L-and 5-type Hshes: fishes with long barbels and those with should barbels, respectively. Lateral line scales, snout length and prepectoral length showed a tendency that the observation could be distinguished from each other (p < 0.05, Duncan's multiple range tests. There were no individuals over 70 mm BL in S-type and so S-type fishes were somewhat dwarf than L-type fishes. The two types were also clearly distinguished by multivariate analyses using cluster and discriminant analyses. According to the observation on the four populations of the Naktong River, the blue-green stripe on the body side and the white band on the margin of anal fin in males of S-type fishes were well developed through the year but those of males of L-type fishes completely disappeared or became slender during nonbreedins season as that of females. Of the seven localities surveved, there were no places that the two types cohabit. S-type fishes are limited only in the Kumho and Wichon River of the Naktong River system but L-type fishes are distributed more widely In the Hongchon, Mangvons, Somjin, Mirang and Panbyon River.
To establish an easy classification method of bamboos observation of the shapes of vascular bundle sheath of Sasa, one of the important natural resources available in Korea, was made on the basis of vascular bundle sheath of bamboo culms because Sasa has a unique vascular bundle sheath as discussed by Grosser and Liese et al. Morphological characteristics of Saw observed are summarized as follows: 1. The thickness of culm wall showed no distinctive difference between upper and low parts of culms of Sasa grown at the ground level. 2. In relation to the taxonomical classification of bamboo, Sasa showed a and a' type but did not e.f.h. and e' type was not identified. 3. Two typical types of bamboo vascular bundle sheath, i.e. b.c.d.e. and g type and d and e type, were found in Sasa but sometimes b.c.g. type was not observed. 4. The results mentioned above seem to be an important key for classification of Sasa.

This study about morphological observation of pre- or post ovulated eggs obtained from artificially maturated female eels. Female eels were divided with two groups as wild eels from nature and feminized eels from farm. Artificial maturation had been conducted with the established methods in this laboratory, and then maturate eggs sampled at fixed 3 times and these were observed with stereomicroscope. Rate of increased body weight (RIW) were measured with 2 times. Egg diameters and development of oil droplets are determined for standardization of egg maturation degree, and the transparency of egg cytoplasm and the homogeneous degrees of egg size were referred to determine of egg quality. Rate of increased body weight (RIW) were good in range about 10 % at final salmon pituitary extracts (SPE) injection time and in range about 20 % at $17{\alpha}$, $20{\beta}$-dihydroxy-4-pregnen-3-one (DHP) injection time. Good matured egg for fertilization had $900-1000{\mu}m$ diameter, and they had about 50 oil droplets in size about $100{\mu}m$ diameter. There were not predominant differences at egg size and oil droplet development between wild female and feminized eels.

Objectives : The consumption of licorice is large in Korea, but domestic production is insufficient due to the lack of adaptability. This study aimed to provide a morphological basis for adding interspecific hybrid licorice with improved adaptability to pharmacopoeia. Methods : This study was to establish identification criteria for the original plants, external and internal morphology of the authentic herbal medicines (Glycyrrhiza uralensis, G. glabra and G. inflata), market products and artificially interspecific hybrid forms of licorice. For this purpose, previous studies were investigated and visual and histological observations were carried out. We focused on the internal morphology by microscopic observation for securing objectivity. Finally, we proposed the identification keys for precise classification of each part. Results : 1) Original plant : Licorice species in the compendium were distinguished by the number of leaflets, presence of hair on the fruit, curvature and swelling of the fruit. 2) External morphology : Licorice species were distinguished by degree of powderiness, tearing gap, radial structure in the cross section and existence of protrusion of outer epidermis. 3) Internal morphology : Licorice species were distinguished by the degree of development of phloem fiber bundle, development of obliterated sieve, whether the secondary medullary ray are branched. In the case of interspecific hybrids, the characteristics of both species used for hybridization were mixed in all observation methods. Conclusions : These results suggest that the interspecific crossbred licorice is suitable for the pharmacopoeial standard. Therefore, it can be applied as a herbal medicine through additional supplementary study.
Purpose: This study investigates the alterations in A549 human non-small-cell lung cancer (NSCLC) cells exposed to Citrus junos extract (CJE). We further examine the antiproliferative and apoptotic effects of CJE on NSCLC cells. Methods: Inhibition of proliferation was examined by applying the 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) colorimetric assay on CJE-treated A549 NSCLC cells. The lactate dehydrogenase (LDH) assay was performed to measure the degree of toxicity of CJE on NSCLC cells. The effect on migratory proliferation was confirmed using the scratch wound healing assay. The antiproliferative effect of the CJE on human lung cancer cells was verified through morphological observation, fluorescence microscopy, and caspase-3 colorimetry. Results: Exposure of NSCLC cells to CJE resulted in a dose- and time-dependent decrease in cell activity and increased toxicity to the cells. In addition, microscopic observation revealed a reduced ability of the cancer cells to migrate and proliferate after exposure to the CJE, with simultaneous morphological apoptotic changes. Fluorescence staining and microscopic examination revealed that this death was a process of self-programmed cell death of NSCLC cells. Compared to unexposed NSCLC cells, the expression of caspase-3 was significantly increased in cells exposed to CJE. Conclusion: Exposure of A549 human NSCLC cells to CJE inhibits the proliferation, increases the cytotoxicity, and decreases the ability of cells to migrate and grow. Moreover, the expression of caspase-3 increases after CJE treatment, suggesting that the apoptosis of NSCLC cells is induced by a chain reaction initiated by caspase-3. These results indicate that Citrus junos is a potential therapeutic agent for human non-small-cell lung cancer.
