Hybridization of Quercus aliena Blume and Q. serrata Murray in Korea - Analyses of Morphological variation and Flavonoid chemistry -

  • This research was conducted in order to understand the hybridization between Quercus aliena Blume and Q. serrata Murray in Korea which show wide range of morphological variations within species and interspecific variations of diverse overlapping characteristics caused by hybridization. Morphological analysis (principal components analysis; PCA) of 116 individuals representing two species and their intermediates were performed. As a result, two species were clearly distinguished in terms of morphology, and intermediate morpho-types assumed to be hybrids between the two species were mostly located in the middle of each parent species in the plot of the principal components analysis. There was a clear distinction between two species in trichome distribution pattern which is an important diagnostic character in taxonomy of genus Quercus, whereas intermediate morpho-types showed intermediate state between two species' trichome distributions. Forty-two individuals representing two species and their intermediates were examined for leaf flavonoid constituents. Twenty-three flavonoid compounds were isolated and identified: They were glycosylated derivatives of flavonols, kaempferol, quercetin, isorhamnetin and myricetin. The flavonoid constituents of Q. aliena were five glycosylated derivatives: kaempferol 3-O-galactoside, kaempferol 3-O-glucoside, quercetin 3-O-galactoside, quercetin 3-O-glucoside, and Isorhamnetin 3-O-glucoside. The flavonoid constituents of Q. serrata had 20 diverse flavonol compounds including five flavonoid compounds found in Q. aliena. It was found that there is a clear difference in flavonoid constituents of Q. aliena and Q. serrata. Flavonoid chemistry is very useful in recognizing each species and putative hybrids. The flavonoid constituents of intermediates were a mixture of the two species' constituents and they generally showed similar characteristics to morpho-types. The hybrids between Q. aliena and Q. serrata showed morphologically and chemically diverse characteristics and it is assumed that there are frequent interspecific hybridization and introgression.

A Study on Recognition of Friction Condition for Hydraulic Driving Members using Neural Network

  • It can be effective on failure diagnosis of oil-lubricated tribological system to analyze operating conditions with morphological characteristics of wear debris in a lubricated machine. And it can be recognized that results are processed threshold images of wear debris. But it is needed to analyse and identify a morphology of wear debris in order to predict and estimate a operating condition of the lubricated machine. If the morphological characteristics of wear debris are identified by the computer image analysis and the neural network, it is possible to recognize the friction condition. In this study, wear debris in the lubricating oil are extracted from membrane filter (0.45 ${\mu}m$) and the quantitative value fur shape parameters of wear debris was calculated through the computer image processing. Four shape parameters were investigated and friction condition was recognized very well by the neural network.

First Report on Lophiostoma macrostomum and Pestalotiopsis lespedezae Isolated from Conifer Leaves of Pinus densiflora in Korea

  • In this study, we isolated endophytic fungi from the conifer leaves of Pinus densiflora inhabiting Mt. Gyehwasan in Korea. We identified the isolated fungal strains based on their morphological characteristics and phylogenetic analysis using DNA sequences of the internal transcribed spacer, small subunit rDNA, large subunit rDNA, and β-tubulin region. In the process of this study, we confirmed two endophytic fungi that have not been recorded previously in Korea, Lophiostoma macrostomum and Pestalotiopsis lespedezae. In this report, we described the morphological characteristics of these fungal strains and the results of their molecular analysis.

First Report of Arbuscular Mycorrhizal Fungi in Korea: Acaulospora delicata,Dentiscutata colliculosa, and Racocetra alborosea.

  • Spores of arbuscular mycorrhizal fungi (phylum Glomeromycota) were isolated from rhizosphere soil collected in Korea. We identified the morphological characteristics of the spores and performed a phylogenetic analysis using the rDNA 18S, 5.8S, and 28S regions. To the best of our knowledge, we confirm the presence of three species of glomeromycotan fungi previously not reported in Korea, namely Acaulospora delicata, Dentiscutata colliculosa, and Racocetra alborosea. We described the morphological characteristics and results of phylogenetic analysis of these species.

Acaulospora jejuensis, a New Species in Glomeromycota from Korea

  • New species of arbuscular mycorrhizal fungi (Glomeromycota), Acaulospora jejuensis, was isolated from rhizosphere soils of Miscanthus sinensis in the grassland in Jeju Island of Korea. The species was identified using the morphological characteristics of the spores and the molecular phylogenetic analysis using partial DNA sequences from small subunit rDNA (SSU), internal transcribed spacer (ITS), and large subunit rDNA (LSU). Phylogenetic analysis placed the fungal species in a distinct clade within genus Acaulospora. Also, the species exhibited the morphological characteristics distinct from the other members of the genus. Therefore, Acaulospora jejuensis was described as a novel species from Korea.

Penicillium mexicanum: An Unrecorded Fungal Species Isolated from Air Samples Collected in Korea

  • We report the first discovery of Penicillium mexicanum in Korea. Fungal strains were isolated from air samples collected in Taean-gun, Chungcheongnam-do, Korea. The strain was identified based on its morphological characteristics, as well as molecular phylogenetic analysis of the internal transcribed spacer (ITS), β-tubulin (BenA), and calmodulin (CaM) regions. This strain exhibited a high sequence similarity to the reference sequences of P. mexicanum. These findings enhance our understanding of fungal biodiversity in Korea and underscore the importance of continuous monitoring of fungal species.

Three Unrecorded Endophytic Fungal Species Isolated from Plants in Taean-gun, Korea

  • In the present study, we isolated endophytic fungi from different tissues of two plants, Pseudosasa japonica and Quercus serrata from Taean-gun, Korea. Morphological characteristics of the isolated fungal strains were observed. We identified the fungi based on the phylogenetic analysis using DNA sequences from the region of internal transcribed spacer, large subunit rDNA, β-tubulin, and translation elongation factor 1-α DNA. Three endophytic fungi that were not previously recorded in Korea, namely Geomyces asperulatus, Leptoxyphium fumago, and Tubakia oblongispora were identified. Here, we describe the morphological characteristics of these unrecorded fungi and present the results of the phylogenetic analysis.

Six Species of Penicillium Associated with Blue Mold of Grape

  • Grape fruits with blue mold symptoms were collected from house storages in different locations in Korea and were investigated for their association with Penicillium species. A total of 12 isolates of Penicillium were isolated from the collected fruits. Based on morphological and cultural characteristics and ${\beta}-tublin$ gene sequence data analysis, they were identified as P. bialowiezense, P. citrinum, P. echinulatum, P. expansum, P. solitum and unidentified Penicillium species. P. solitum was the predominant followed by P. expansum. P. bialowiezense and P. echinulatum were newly recorded in Korea. ${\beta}-Tubulin$ gene sequences could be used to distinguish each species of Penicillium and the molecular groups were correlated well with the morphological species. The unidentified species was supposed to be a new species, not previously reported in literature.

국내 도입 블루베리의 생육기 및 형태적 특성에 의한 유연관계 분석 (Analysis of Relationship Based on Growth Period and Morphological Characteristics in Blueberry (Vaccinium spp.))

  • 국내에서 주로 재배중인 28개 품종의 품종 식별을 위해 생육기 및 형태적 특성을 조사하여 유연관계를 분석하였다. 래빗아이 블루베리 품종인 'Austin', 'Brightwell', 'Powderblue', 'Southland', 'Tifblue'가 하이부쉬블루베리에 비해 수확 기간이 길고 늦은 것으로 나타났다. 꽃의 L/D율은 래빗아이 블루베리 품종인 'Austin', 'Brightwell', 'Powderblue', 'Tifblue'과 남부하이부쉬 블루베리인 'Brigitta' 품종의 경우 2.0 이상으로 나타나 수분에 불리할 것으로 판단되었다. 블루베리 품종별 형태적 특성 결과를 군집 분석한 결과 블루베리의 생육 및 과실 의형태적 특성에 의해 크게 2개의 그룹으로 구분할 수 있었다. 그룹 I은 래빗아이 블루베리 품종들이었으며 그룹 II는 하이부쉬 블루베리 품종들이었다. 따라서 생육기 및 형태적 특성만으로도 래빗아이 블루베리 품종을 식별할 수 있을 것으로 판단되었으며 하이부쉬블루베리 내 반수고, 북부, 남부 하이부쉬 블루베리의 구분은 되지 않았다.

  • The morphological analysis of wear debris can provide early a failure diagnosis in lubricated moving system. It can be effective to analyze operating conditions of oil-lubricated tribological system with shape characteristics of wear debris in a lubricant. But, in order to predict and recognize an operating condition of lubricated machine, it is needed to analyze and to identify shape characteristics of wear debris. Therefore, If the morphological characteristics of wear debris are recognized by computer image analysis using the neural network algorithm, it is possible to recognize operating condition of hydraulic driving members. In this study, wear debris in the lubricating oil are extracted by membrane filter (0.45$\mu\textrm{m}$), and the quantitative values of shape parameters of wear debris are calculated by the digital image processing. This shape parameters are studied and identified by the artificial neural network algorithm. The result of study could be applied to prediction and to recognition of the operating condition of hydraulic driving members in lubricated machine systems.
