New Record of Potamogeton × anguillanus Koidz. (Potamogetonaceae) in Korea (가래과 가래속의 한반도 미기록식물: 얼치기대가래)

  • In this study, we report a unrecorded plant on the Korean peninsula, Potamogeton × anguillanus Koidz. from Yeondangcheon (stream) in Yeondang-ri, Nam-myeon, Yeongwol-gun, Gangwon-do province. Potamogeton × anguillanus is a natural hybrid between P. wrightii Morong. and P. perfoliatus L. It is distinguished by its characteristic that the petiole is shorter than P. wrightii and that the leaf blade is narrower and longer than P. perfoliatus also, it didn't fully wrap the stem. Considering that P. × anguillanus is a natural hybrid, the name is given to it as 'Eol-chi-gi-dae-ga-rae'. We provide descriptions of the morphological characteristics, photographs, and key to allied taxa in Korea of P. × anguillanus.

  • S-Adenosyl-L-Methionine(SAM) has an important role for DNA methylation and cell signaling. SAM was synthesized from methionine and ATP by SAM synthetase and play an pivotal function in the primary and secondary metabolism of cells. Recent studies have revealed in the effect of SAM in case of morphological differentiation in both eukaryotes and prokaryotes. We isolated SAM gene from Saccharomyces cerevisiae and cloned it into expression vector for E. coli respectively. An 1.15 kb SAM-s gene fragment was isolated by Low-strigency PCR using ORF primer. By the analysed primary sequence deduced from DNA sequence, this gene included conserved domains similar with other well-known SAM synthetase. First of all, SAM synthetase gene cloned pGEM-T vector and subcloned into histidine tagging system to purify the expressed protein using metal chelating resin. Typical characteristic analysis of this enzyme is underway.

  • Many isolated featureless domains were explicitly observed even at the air-water interface. We measured the surface pressure shift originating from the photo-isomerization of azobenzene units on the periphery of dendrimers. The maximum surface pressure was gradual1y increased and saturated by cyclic compression and decompression. By irradiation of 365 [nm] light, the surface pressure was increased, which was originated by the photo-isomerization process of the azobenzene group on the periphery from trans to cia form. The increase of the dipole moment ($\mu$), which may increase the interaction among Azo dendrimer molecules, made an important role on surface pressure shift. From the absorbance spectrum by UV irradiation and heat treatment, we can see that the absorbance in the UV region decreases with the increase of the UV irradiation time, but the peak at 350 m, characteristic of dendrimers in the LB monolayers, was not shifted until four irradiation cycles. This suggests that optical behavior and morphological change are affected by the functional group and the symmetric chain.

  • Colonial morphology of the various yeasts often encountered in sputum or other clinical specimens was investigated on the corn meal-potato-yeast extract agar medium (GJCPY) containing orange-yellow pigments extracted from Gardenia jasminoides fruits in hopes of differential identification on primary cultures. The results obtained are as follows. 1) Cryptococcus neoformans which is a medically important yeast and whose colony showed brown to purple brown on GJCPY medium was distinguishable not only from buff colored Cr. laurentii after one week incubation but also from Candida spp. 2) Colony color of Candida albicans, a most common species in sputum specimens and of Ca. parapsilosis, a rare isolate, remained unchanged even after 15 days incubation. 3) Ca. tropicalis, second common isolate from sputums and Ca. krusei, a rare isolate, formed a characteristic rough and wrinkled colonies that permit to differentiate them from others. 4) Rare isolates, Ca. guilliermondii and Ca. lusitaniae, turned to prussian blue within three days of incubation. 5) Torulopsis sp. and Saccharomyces cerevisiae showed glossy grayish blue or light blue after one week incubation. The findings clearly showed that Ga. jasminoides pigments medium was useful to the morphological differentiation of medically important yeasts that were often encountered in sputum or other clinical specimens.

Two unrecorded alien plants in South Korea: Holosteum umbellatum L. and Asperugo procumbens L. (한국 미기록 외래식물: 산형나도별꽃, 갈퀴지치)

  • Two alien plants Holosteum umbellatum L. and Asperugo procumbens L., belonging to Boraginaceae and Caryophyllaceae, respectively, have been first recorded in Hongseong-gun, Chungcheongnam-do and Incheon, South Korea. This initial investigation reports to inform the importance of detection and extermination of these potential weeds in surrounding areas. Furthermore, we report the morphological characteristic features of these two alien plants.

Induction of Intrinsic and Extrinsic Apoptosis Pathways in the Human Leukemic MOLT-4 Cell Line by Terpinen-4-ol

  • Terpinen-4-ol is a terpene found in the rhizome of Plai (Zingiber montanum (Koenig) Link ex Dietr.). In this study apoptogenic activity and mechanisms of cell death induced by terpinen-4-ol were investigated in the human leukemic MOLT-4 cell line. Terpinen-4-ol exhibited cytotoxicity in MOLT-4 cells, with characteristic morphological features of apoptosis by Wright's staining. The mode of cell death was confirmed to be apoptosis by flow cytometric analysis after staining with annexin V-FITC and propidium iodide. A sub-G1 peak in DNA histograms of cell cycle assays was observed. Terpinen-4-ol induced-MOLT-4 cell apoptosis mediated through an intrinsic pathway involving the loss of mitochondrial transmembrane potential (MTP) and release of cytochrome c into the cytosol. In addition, terpinen-4-ol also induced apoptosis via an extrinsic pathway by caspase-8 activation resulting in the cleavage of cytosolic Bid. Truncated-Bid (tBid) translocated to mitochondria and activated the mitochondrial pathway in conjunction with down-regulation of Bcl-2 protein expression. Caspase-3 activity also increased. In conclusion, terpinen-4-ol can induce human leukemic MOLT-4 cell apoptosis via both intrinsic and extrinsic pathways.

  • This study deals with effect of plasma treatment on the properties of unidirectional ligno cellulosic fabric Hildegardia Populofolia (HDP) fabric. Thermal stability of the fabric was determined by differential scanning calorimetry (DSC) and Thermo gravimetric analysis (DSC). Morphological properties was analyzed by SEM analysis and found that the surface was rough upon plasma treatment which provides good interfacial adhesion with matrix during composite fabrication. Thermal stability and mechanical properties of the plasma treated fabric slightly increases compare to alkali and untreated fabric. It was observed that tensile properties of the fabric increases upon plasma treatment due to the formation of rough surface. SEM analysis indicates formation of rough surface on plasma treatment which helps in increasing the interfacial interaction between the matrix (hydrophobic) and fabric (hydrophilic).

  • The effect of various thermomechanical treatments on the structure and rheological behaviour of Al-6.2wt%Si alloy in its solidification range were investigated using a Searle type high temperature viscometer. During continuous cooling, the viscosity increases gradually with increasing fraction of solidified alloy, until a critical fraction of solidified alloy is reached above which the viscosity sharply increases. The viscosity of the slurry, at a given volume fraction wolid, decreased with increasing shear rate. The size and morphology of primary solid particles during stirring is influenced strongly by shear rates, cooling rates, volume fraction and stirring time of solid. Morphological changes during stirring as a function of solid volume fractions, shear rate and processing time were also reported. In this study, the size of primary solid particles in these alloys consistently increases and the it`s aspect ratio decrease with the increase in fraction solid and decrease in shear rate. Crystal morphology changes from rosette type to spheroid type with the increase in shear rate and solid fraction.

  • 2-chloroethylethyl sulfide (CEES) is an alkylating agent that readily reacts with a wide variety of biological molecules causing metabolic abnormality. The mechanism of cell death during CEES injury is poorly understood. We have examined the effect of exposure of thymocytes with various concentrations of CEES to determine the pattern of cell death in thymocytes injury induced by CEES. In the present study, we show that two patterns of cell death occurred by either one of two mechanisms: apoptosis and necrosis. Exposure to low level of CEES (100 ${\mu}M$) for 5 h caused an induction of apoptosis on thymocytes, as identified by the following criteria: DNA fragmentation visualized by the characteristic "ladder" pattern was observed upon agarose gel electrophoresis and morphological features were revealed by microscopical observations. In contrast, exposure to high levels of CEES (500 ${\mu}M$) induce necrotic features such as cell lysis. Thus, depending on the concentrations, CEES can result in either apoptotic or necrotic cell damage. Our findings suggest that thymocytes which are not killed directly, but merely injured by low levels of CEES, are able to activate an internally-programmed cell death mechanism, whereas thymocytes receiving severe damages apparently can not.

Morphological Characteristics of Conidiogenesis in Cordyceps militaris

  • Conidial development of Cordyceps militaris was observed from germinating ascospores and vegetative hyphae through light and scanning electron microscopy (SEM). Ascospores were discharged from fresh specimens of C. militaris in sterile water as well as Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates. We observed ascospore germination and conidial formation periodically. Under submerged condition in sterile water, most part-spores germinated unidirectionally and conidia were developed directly from the tips of germinating hyphae of part-spores within 36 h after ascospore discharge, showing microcyclic conidiation. First-formed conidia were cylindrical or clavate followed by globose and ellipsoidal ones. Germination of ascospores and conidial development were observed on SDAY agar by SEM. Slimy heads of conidia on variously arranged phialides, from solitary to whorl, developed 5 days after ascospore discharge. Besides, two distinct types of conidia, elongated pyriform or cylindrical and globose, were observed in the same slimy heads by SEM. Conidia were shown to be uninucleate with 4,6-diamidino-2-phenylindole staining. Conidiogenous cells were more slender than vegetative hyphae, having attenuated tips. Microcyclic conidiation, undifferentiated conidiogenous hyphae (phialides), polymorphic conidia and solitary, opposite to whorled type of phialidic arrangement are reported here as the characteristic features of asexual stage of C. militaris, which can be distinguished from other Cordyceps species.
