To optimally convert corn hull, a byproduct from corn processing, into bioethanol using Pachysolen tannophlius, we investigated the optimal conditions for hydrolysis and removal of toxic substances in the hydrolysate via activated carbon treatment as well as the effects of this detoxification process on the kinetic parameters of bioethanol production. Maximum monosaccharide concentrations were obtained in hydrolysates in which 20 g of corn hull was hydrolyzed in 4% (v/v) $H_2SO_4$. Activated carbon treatment removed 92.3% of phenolic compounds from the hydrolysate. When untreated hydrolysate was used, the monosaccharides were not completely consumed, even at 480 h of culture. When activated carbon.treated hydrolysate was used, the monosaccharides were mostly consumed at 192 h of culture. In particular, when activated carbon-treated hydrolysate was used, bioethanol productivity (P) and specific bioethanol production rate ($Q_p$) were 2.4 times and 3.4 times greater, respectively, compared to untreated hydrolysate. This was due to sustained bioethanol production during the period of xylose/arabinose utilization, which occurred only when activated carbon-treated hydrolysate was used.

Monofunctional biosynthetic peptidoglycan transglycosylase (MBPT) catalyzes the formation of the glycan chain in bacterial cell walls from peptidoglycan subunits: N-acetylglucosamine (NAG) and acetylmuramic acid (NAM). Bifunctional glycosyltransferases such as the penicillin binding protein (PBP) have peptidoglycan glycosyltransferase (PGT) on their C terminal end which links together the peptidoglycan subunits while transpeptidase (TP) on the N terminal end cross-links the peptide moieties on the NAM monosaccharide of the peptide subunits to create the bacterial cell wall. The singular function of MBPT resembles the C terminal end of PBP as it too contains and utilizes a similar PGT domain. In this article we analyzed the infectious and non infectious protein sequences of MBPT from 31 different strains of bacteria using a variety of bioinformatic tools. Motif analysis, dot-plot comparison, and phylogenetic analysis identified a number of significant differences between infectious and non-infectious protein sequences. In this paper we have made an attempt to explain, analyze and discuss these differences from an evolutionary perspective. The results of our sequence analysis may open the door for utilizing MBPT as a new target to fight a variety of infectious bacteria.
In the previous paper, two strains of cellulase producing microorganism were isolated from night soil samples using shaking culture. This report deals with the physiological and morphological tests carried out according to the methods of Toyama and Chung. 1. Two strains, Ku-3371 and Ku-4383 which should the best growth in Czapek.s liquid medium, were identified as Trichoderma viride. 2. These strains grew the best at about $30^{\circ}C$ and the optimum pH values of growth in Czapek's liquid medium was 4.0. 3. These mould strains utilized monosaccharide as its carbon source and can utilized sodium glutamate, peptone and nitrate form nitrogen but other inorganic nitrogen compoumd are unable to use as its nitrogen source.
Chemical composition of the Eucommia ulmoides bark, including extractives, proximate, mineral, fatty acid and monosaccharide compositions, was studied The most abundant mineral was calcium (533.17 mg/l00 g). $\alpha-linolenic$ acid (24.7%) and linoleic acid (24.3%), showed higher contents among the fatty acids. Major monosaccharides of E. ulmoides balk were arabinose (13.94 mg/g), xylose (18.91 mg/g) and glucose (119.7 mg/g). From the bark of E ulmoides, four compounds were isolated and their structures were elucidated as caffeic acid (I), kaempferol (II), quercetin (III) and isoquercitrin (IV) by spectroscopic analysis such as NMR and MS, including cellulose TLC and other chemical evidence such as hydrolyzation and acetylation. The antioxidant activities of four isolated compounds were evaluated by DPPH free radical scavenging, hydroxyl scavenging and reducing power assays. The results indicated that all the isolated compounds showed higher DPPH radical scavenging activity than $\alpha-tocopherol$ and BHT that were used as positive controls and these four compounds exhibited considerable reducing power and hydroxyl radical (OH) scavenging activity. Considering from the results above, it suggests that the E. ulmoides bark is a potential natural source of antioxidant material.
The objective of this study was to understand the conversion characteristics of glucose and xylose using the major monosaccharide standards for lignocellulosic biomass. The acid-catalyzed organosolv pretreatment conducted using ethanol was significantly different from the acid-catalyzed process conducted in an aqueous medium. 5-hydroxymethylfurfural (5-HMF), levulinic acid and furfural were produced from glucose conversion. The maximum yield of 5-HMF was 5.5%, at 200℃, when 0.5% sulfuric acid was used. The maximum yield of levulinic acid was 21.5%, at 220℃, when 1.0% sulfuric acid was used. Furfural was produced from xylose conversion and under 0.5% sulfuric acid, furfural reached the maximum yield 48.5% at 210℃. Ethyl levulinate and methyl levulinate were also formed from the glucose standard following the esterification reaction conducted under conditions of the combined conversion method, which proceeded under both ethanol-rich and water-rich conditions.
Enzyme-assisted hydrolysis is frequently used as a cost-effective and efficient method to obtain functional ingredients from bioresources. This study involved the enzyme-assisted hydrolyzation and purification of fucoidan from Ecklonia maxima stipe and the investigation of its anti-inflammatory activity in lipopolysaccharide (LPS)-induced RAW 264.7 cells. Fucoidans of Viscozyme-assisted hydrolysate from E. maxima (EMSFs) harvested in Jeju, Korea. Structural and chemical characterizations were performed using fourier transform infrared spectroscopy, scanning electron microscope, and monosaccharide analysis. Among fucoidans, EMSF6 was rich in fucose and sulfate and had a similar structural character to commercial fucoidan. EMSF6 showed a strong inhibitory effect on nitric oxide generation in LPS-induced RAW 264.7 cells and significantly decreased the production of LPS-induced pro-inflammatory cytokines, including interleukin-6, interleukin-1β, and tumor necrosis factor α. The anti-inflammatory potential of EMSF6 was mediated through the downregulation of inducible nitric oxide synthase and cyclooxygenase-2 expression. Thus, fucoidans from E. maxima stipe are promising candidates for functional food products.
BACKGROUND/OBJECTIVES: The dietary intake of foods with fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAP) is known to adversely affect patients with irritable bowel syndrome (IBS). However, the effects of FODMAP have been studied predominantly among Western populations. This study aimed to identify foods high in FODMAP content which form a part of the Korean adult diet and obtain basic data for the preparation of IBS guidelines. SUBJECTS/METHODS: An online survey of 1,000 adults from the general population in the age group of 20 to 40 years was performed. Data from 787 participants (men, 386; women, 401) were analyzed. The general characteristics of the participants, health status, IBS diagnosis using the Rome III diagnostic criteria, semi-quantitative food frequency questionnaire findings, and food items causing symptoms were analyzed. RESULTS: Overall, 169 participants (21.5%) had IBS. The contribution of the FODMAP nutrients in both IBS and healthy groups was as follows: fructan > lactose > excess fructose > sorbitol > mannitol > galacto-oligosaccharides (GOS). The fructan intake was 4.6 ± 2.2 g/day and 4.3 ± 2.5 g/day in the IBS and healthy groups (P = 0.014), respectively. In the IBS group, the ratio of the intake of fructan to the total FODMAP intake was 39.5%, 29.8%, and 5.8% through onions, garlic, and bananas, respectively. CONCLUSIONS: Fructan was the FODMAP nutrient most consumed by Korean adults. Therefore, given the difference in the dietary habits of each country's population, the dietary guidelines for IBS should be country specific.
To find antitumor components from the higher fungi of Korea, the mycelia of Favolus alveolarius (Fr.) Quelet were cultured in a liquid medium. The cultured mycelia were extracted with hot water twice, and a high molecular weight fraction was obtained by adding two volumes of ethanol to the extract. Two grams of Fraction A were obtained by dialyzing it. It was further separated into four fractions by gel filtration with Sepharose CL-4B, and they were designated Fractions B, C, D, and E. The results of the antitumor test showed that Fractions A, B, C, D and E had tumor inhibition ratios of 92.3, 78.5, 59.6, 77.4 and 62.2%, respectively. Anthrone test was carried out to determine the contents of total polysaccharide of the five fractions, and they had 46.3, 27.3, 65.3, 64.6, and 46.1%, respectively. The contents of the total protein of the five fractions were 29.4, 13.9, 14.3, 14.3, and 29.1%, respectively. Monosaccharide subunits of each fraction were analyzed with gas chromatography, and glucose, xylose, mannose, galactose and fucose were identified. Fraction A was examined for immunological effects. It increased the count of hemolytic plaque forming cells 12.8 times to that of the control group, and the population of macrophage in peritoneal cavity 3.2 times to that of the control group.

We recently reported a Philyra pisum lectin (PPL) that exerts mitogenic effects on human lymphocytes, and its molecular characterization. The present study provides a more detailed characterization of PPL based on the results from a monosaccharide analysis indicating that PPL is a glycoprotein, and circular dichroism spectra revealing its estimated ${\alpha}$-helix, ${\beta}$-sheet, ${\beta}$-turn, and random coil contents to be 14.0%, 39.6%, 15.8%, and 30.6%, respectively. These contents are quite similar to those of deglycosylated PPL, indicating that glycans do not affect its intact structure. The binding properties to different pathogen-associated molecular patterns were investigated with hemagglutination inhibition assays using lipoteichoic acid from Gram-positive bacteria, lipopolysaccharide from Gram-negative bacteria, and both mannan and ${\beta}$-1,3-glucan from fungi. PPL binds to lipoteichoic acids and mannan, but not to lipopolysaccharides or ${\beta}$-1,3-glucan. PPL exerted no significant antiproliferative effects against human breast or bladder cancer cells. These results indicate that PPL is a glycoprotein with a lipoteichoic acid or mannan-binding specificity and which contains low and high proportions of ${\alpha}$-helix and ${\beta}$-structures, respectively. These properties are inherent to the innate immune system of P. pisum and indicate that PPL could be involved in signal transmission into Gram-positive bacteria or fungi.
The cultural characteristics and antibacterial activities of Cordyceps militaris and Paecilomyces tenuipes were compared. The mycelial growth was the highest on MCM (Mushroom Complete Medium) for C. militaris and on YMA (Yeast Malt Agar) for P. tenuipes. But the mycelial density on MMM (Mushroom Minimal Medium) was lower than other on media. The optimum mycelial growth was observed at $25^{\circ}C$. C. militaris was low mycelial growth when it was transferred over 5 times generation. The carbon source for the optimum mycelial growth was fructose of monosaccharide, maltose of disaccharide and dextrin of polysaccharide. The calcium nitrate of organonitrogen was found the best mycelial growth on C. militaris, while the sodium nitrate observed to be well for mycelial growth on P. tenuipes. The ammonium tartrate was observed to be the best among the inorganonitrogen used for mycelial growth. Antibacterial activities were found out just C. militaris against Bacillus cereus of Gram (+).
