Electroreflectance 측정에 의한 Si이 첨가된 $Al_{0.32}Ga_{0.68}As$에서의 $E_1$ 전이에 대한 연구 (A Study on $E_1$Transition in Si-Doped $Al_{0.32}Ga_{0.68}As$by Electroreflectance Measurement)

  • Silicon doped $Al_{0.32}Ga_{0.68}As$ were growth by molecular beam epitaxy. Electroreflectance(ER) spectra of the $E_1$ transition of Schottky barrier Au/n-$Al_{0.32}Ga_{0.68}As$ have been measured at various modulation voltage($V_{ac}$) and dc bias voltage($V_{bias}$). from the $E_1$peak, band gap energy of the $Al_{0.32}Ga_{0.68}As$ is 1.883 eV which corresponds to an Al composition of 32%. As modulation voltage($V_{bias}$) is changed, a line shape at the $E_1$transition does not change, but its amplitude varies linearly. The amplitude of $E_1$signal decrease with increasing the forward dc bias voltage($V_{bias}$), but the line shape does not change. It suggests that the low field theory rather than Franz-Keldysh oscillation is Required to interpret spectra. Also, spectra at the $E_1$transition were broadened with increasing the reverse dc bias voltage($V_{bias}$) which suggests the presence of Field-induced broadening.

Establishment of Hertwig's Epithelial Root Sheath/Epithelial Rests of Malassez Cell Line from Human Periodontium

  • Human Hertwig's epithelial root sheath/epithelial rests of Malassez (HERS/ERM) cells are epithelial remnants of teeth residing in the periodontium. Although the functional roles of HERS/ERM cells have yet to be elucidated, they are a unique epithelial cell population in adult teeth and are reported to have stem cell characteristics. Therefore, HERS/ERM cells might play a role as an epithelial component for the repair or regeneration of dental hard tissues; however, they are very rare population in periodontium and the primary isolation of them is considered to be difficult. To overcome these problems, we immortalized primary HERS/ERM cells isolated from human periodontium using SV40 large T antigen (SV40 LT) and performed a characterization of the immortalized cell line. Primary HERS/ERM cells could not be maintained for more than 6 passages; however, immortalized HERS/ERM cells were maintained for more than 20 passages. There were no differences in the morphological and immunophenotypic characteristics of HERS/ERM cells and immortalized HERS/ERM cells. The expression of epithelial stem cell and embryonic stem cell markers was maintained in immortalized HERS/ERM cells. Moreover, immortalized HERS/ERM cells could acquire mesenchymal phenotypes through the epithelial-mesenchymal transition via TGF-${\beta}1$. In conclusion, we established an immortalized human HERS/ERM cell line with SV40 LT and expect this cell line to contribute to the understanding of the functional roles of HERS/ERM cells and the tissue engineering of teeth.

  • Epithelial-to-mesenchymal transition (EMT)는 embryogenesis에서 중요한 역할을 하며 tumor metastasis, invasion에도 관여함으로써 tumor progression 및 aggressiveness에 기여한다. EMT는 EMT hallmark인 epithelial E-cadherin의 발현 감소와 mesenchymal-like cell morphology를 획득함으로써 epithelial cell polarity를 잃어버리는 특징을 가지고 있다. $O_2{^-}$, $H_2O_2$, $OH^-$와 같은 활성산소가 EMT를 유도하는 것으로 알려져 있다. Snail이 E-cadherin의 발현을 억제함으로써 ROS에 의한 EMT에 관여하는 것으로 알려져 있으나, 그 기작은 완전히 밝혀져 있지 않다. 본 연구에서는, noninvasive breast tumor cell line인 MCF-7 세포에 Egr-1을 과발현시킨 후 그 영향을 조사하였다. Egr-1이 과발현되면, MCF-7 세포는 epithelial cell polarity를 잃고 spindle-shaped로 변화되므로, Egr-1이 EMT를 유도할 가능성이 대두되었다. 또한 Snail이 Egr-1에 의한 EMT에 관여함을 확인하였다. 나아가, 본 연구진은 Egr-1-Snail axis가 ROS에 의해 활성화 되고, ROS에 의한 EMT에서 중요한 역할을 함을 발견하였다.

Probing neutral gas clouds and associated galaxies in the early universe

  • Neutral (HI) gas clouds associated with galaxies are responsible for fuelling the star-formation in the universe. In literature, the extremely strong damped Lyman-alpha absorbers (or ESDLAs) have been known to be sensitive to the effects of HI-H2 transition and star-formation in galaxies. Yet, ESDLAs are rare to probe due to the smaller cross section they subtend on the sky (similar to galaxies). In my talk, I will focus primarily on my study of the nature of ESDLAs that are observed as absorption signature along the line-of-sight (LOS) of a quasar (QSO). I will further look at the HI-H2 transition and interesting results relevant to diffuse molecular gas and the multi-phase medium (gas in different ionization states) that are associated with ESDLAs. Furthermore, I will also discuss how the ESDLA environments differ from the high star-forming and molecular environments detected in blind optical and radio surveys consecutively.

Herschel FIR Observations of Molecule Lines in L1448-MM

  • L1448-MM, known as a class 0 YSO with a prominent outflow, was observed with the Photodetector Array Camera and Spectrometer (PACS) aboard Herschel Space Observatory by the key program, DIGIT (Dust, Ice, Gas in Time, PI: Neal Evans). The PACS covers various molecular and atomic line transitions such as CO, OH, $H_2O$, [OI], and [CII] at wavelengths from 55 to 210 ${\mu}m$. The line emission of $H_2O$, [OI], mid-J CO, and the OH fundamental transition distributes along the outflow direction although high-J CO and other OH emission peaks at the central spatial pixel. According to our excitation analysis, the CO gas has two temperature components: 300 K and 750 K, which are attributed to PDR and shock, respectively. However, the $H_2O$ gas with the rotation temperature ($T_{rot}$) of 200 K seems only affected by shock. Interestingly, the relative strength of OH transitions suggests the IR pumping process in L1448-MM. We also mapped L1448-MM in CO J=2-1 with the SRAO 6m telescope to compare with the FIR line transition maps.

공정인자들이 나노임프린트 리소그래피 공정에 미치는 영향에 대한 분자동역학 연구 (Molecular Dynamics Study on the Effect of Process Parameters on Nanoimprint Lithography Process)

  • Molecular dynamics simulations of nanoimprint lithography NIL) are performed in order to investigate effects of process parameters, such as stamp shape, imprinting temperature and adhesive energy, on nanoimprint lithography process and pattern transfer. The simulation model consists of an amorphous $SiO_{2}$ stamp with line pattern, an amorphous poly-(methylmethacrylate) (PMMA) film and an Si substrate under periodic boundary condition in horizontal direction to represent a real NIL process imprinting long line patterns. The pattern transfer behavior and its related phenomena are investigated by analyzing polymer deformation characteristics, stress distribution and imprinting force. In addition, their dependency on the process parameters are also discussed by varying stamp pattern shapes, adhesive energy between stamp and polymer film, and imprinting temperature. Simulation results indicate that triangular pattern has advantages of low imprinting force, small elastic recovery after separation, and low pattern failure. Adhesive energy between surface is found to be critical to successful pattern transfer without pattern failure. Finally, high imprinting temperature above glass transition temperature reduces the imprinting force.

Anti-metastatic mechanism of mountain cultivated wild ginseng in human cancer cell line

  • Objective : Ginseng is one of most widely used herbal medicine. Ginseng showed anti-metastasis activities. However, its molecular mechanisms of action are unknown. So we want to report the wild ginseng repress which plays key roles in neoplastic epithelial-mesenchymal transition process. Methods : Treatment of the human colorectal carcinoma LOVO cells and human gastric carcinoma SNU601 cells with the increased concentrations of cultivated wild ginseng extracts resulted in a gradual decrease in the AXIN2 gene expression. Results : Metastasis-suppressor genes, maspin and nm23 was not affected by the treatment of ginseng extracts in LOVO cells. Moreover, the mountain cultivated wild ginseng or mountain wild ginseng are similar in their inhibitory effects on the expression of AXIN2 gene, but are substantially stronger than cultivated ginseng. Conclusion : We described the novel mechanism of wild ginseng-induced anti-metastasis activity by repressing the expression of AXIN2 gene that plays key roles in epithelial-mesenchymal transition process.

HOW DO MASSIVE STARS FORM? INFALL & OUTFLOW IN DENSE CORES IN THE MILKY WAY

  • Massive stars are some of the most influential objects in the Universe, shaping the evolution of galaxies, creating chemical elements and hence shaping the evolution of the Universe. However, the processes by which they form and how they shape their environment during their birth processes are not well understood. We use $NH_3$ data from "The $H_2O$ Southern Galactic Plane Survey" (HOPS) survey to define the positions of dense cores/clumps of gas in the southern Galactic plane that are likely to form stars. Then, using data from "The Millimetre Astronomy Legacy Team 90 GHz" (MALT90) survey, we search for the presence of infall and outflow associated with these cores. We subsequently use the "3D Molecular Line Radiative Transfer Code" (MOLLIE) to constrain properties of the infall and outflow, such as velocity and mass flow. The aim of the project is to determine how common infall and outflow are in star forming cores, and therefore to provide valuable constraints on the timescales and physical process involved in massive star formation. Preliminary results are presented here.

KPNA3 promotes epithelial-mesenchymal transition by regulating TGF-β and AKT signaling pathways in MDA-MB-231, a triple-negative breast cancer cell line

  • Karyopherin-α3 (KPNA3), a karyopherin-α isoform, is intimately associated with metastatic progression via epithelial-mesenchymal transition (EMT). However, the molecular mechanism underlying how KPNA3 acts as an EMT inducer remains to be elucidated. In this report, we identified that KPNA3 was significantly upregulated in cancer cells, particularly in triple-negative breast cancer, and its knockdown resulted in the suppression of cell proliferation and metastasis. The comprehensive transcriptome analysis from KPNA3 knockdown cells indicated that KPNA3 is involved in the regulation of numerous EMT-related genes, including the downregulation of GATA3 and E-cadherin and the up-regulation of HAS2. Moreover, it was found that KPNA3 EMT-mediated metastasis can be achieved by TGF-β or AKT signaling pathways; this suggests that the novel independent signaling pathways KPNA3-TGF-β-GATA3-HAS2/E-cadherin and KPNA3-AKT-HAS2/E-cadherin are involved in the EMT-mediated progress of TNBC MDA-MB-231 cells. These findings provide new insights into the divergent EMT inducibility of KPNA3 according to cell and cancer type.

MOLECULAR GAS AND RADIO JET INTERACTION: A CASE STUDY OF THE SEYFERT 2 AGN M51

  • We observed multiple CO transition lines and the HCN(1-0) line at ~ 1" (~ 34 pc) or higher resolution toward the Seyfert 2 nucleus of M51 using the IRAM Plateau de Bure Interferometer (PdBI) and the Submillimeter Array (SMA). All the images show very similar overall molecular gas distribution; there are two discrete clouds at the eastern and western sides of the nucleus, and the western cloud exhibits an elongated distribution and velocity gradient along the radio jet. In addition, high HCN(1-0)/CO(1-0) brightness temperature ratios of about unity have been observed, especially along the radio jet, similar to those observed in shocked molecular gas in our Galaxy. This strongly indicates that the molecular gas along the jet is shocked, that the radio jet and the molecular gas are interacting, and the jet is entraining both diffuse (CO) and dense (HCN) molecular gas outwards from the circumnuclear region. This is the first clear imaging of the outflowing molecular gas entrained by the AGN jet, and showing the detailed physical status of outflowing molecular gas. Since a relatively high HCN(1-0)/CO(1-0) ratio has been observed in the high velocity wing of ultraluminous infrared galaxies, it can also be explained by a similar mechanism to those we describe here.
