Geosmin and Morphological Characteristics of Anabaena circinalis, Obtained from the Bukhan River (북한강에서 출현한 Anabaena circinalis의 형태학적 특성 및 지오스민(geosmin) 발생 양상)

  • This study was carried out in the Bukhan River in the summer of 2014 and 2015, to identify the relationship between geosmin and the morphological changes in Anabaena. Identification of Anabaena was conducted using morphological and molecular analyses. Anabaena in this study was similar to Anabaena circinalis, A. crass, and A. spiroides with regard to regular coils, vegetative cell, akinete shape, and size, hoever, it was distinguishabl from A. crass and A. spiroides because of its larger trichome coil size. Additionally, the sequences of phycocyanin (PC) gene from Anabaena showed a 99% genetic similarity with A. circinalis NIES-1647 strain. The coil diameter of trichome ranged from 106 to $899{\mu}m$, and the diameter and abundance showed an insignificant positive correlation (r=0.544, p<0.05). The result of relationship between the coil diameter and the cell number per 360-degree rotation was kept at $33.8{\pm}5.2$ cells per $100{\mu}m$ diameter despite variable diameter. The average geosmin concentrations in 2014 and 2015 were investigated to be 99 ng/L and 35 ng/L, respectively. A. circinalis cell density contributed considerably to the change in geosmin and was positively correlated with geosmin concentration (2014; r=0.599, p<0.01, 2015; r=0.559, p<0.01). Our results suggest that geosmin and coil diameter could be estimated with the help of cell density.

Theoretical Calculation on Alpha Track Density by Using an Electrostatic Ion Spectrometer (정전기분광분석방법에 의한 알파입자비적밀도의 이론적 계산연구)

  • To develop a technique of theoretical alpha track density calculation for comparison with measured track density, an electrostatic ion spectrometer was specially designed and fabricated. The mobility spectrum of first radon daughter(Po-218) in the range of $0.07{\sim}5.0cm^2/V\;s$ from the radon chamber was measured using-the electrostatic ion spectrometer. Measurement was taken in a radon chamber operated using dry particle free air passed through silica gel, activated charcoal and molecular sieve filters. The mobility of a new-born Po-218 ion measured by the electrostatic ion spectrometer was determined to be $1.92cm^2/V\;s$. A comparison of the theoretical and measured alpha track densities was completed and uncertainties concerning the shape of the spectrum were analyzed. It was found that the discrepancies in track densities are primarily Que to the neglect of wall loss of ions in the theoretical track density calculation.

  • At the ratio [$H_2O$]/[TEOS]=1.7, the silica sol was synthesized by partial hydrolysis in the presence of acid catalyst. After stabilizing the silica sol by trimethylsilylation, the molecular weight and viscosity of the sol obtained at various reaction times were examined to determine a best spinnability of the sol. Gel fibers were prepared from the sol solution after removing solvent in the solution, and the gel fibers were heated at $1,000^{\circ}C$. The prepared silica fibers were in the shape of circular cross-section and their tensile strength and $SiO_2$ purity were $83{\pm}20kg/mm^2$ and about 99.997%, respectively.

  • Lophatheri Herba is the aerial part of Lophatherum gracile Bronghiart(淡竹葉, Gramineae). 25~75 cm in length. Stem: cylindrical with nodes, empty inside, externally pale yellowish green. Leaf: dehiscent of lanceolate lamina, shrunken and rolled, 5~20 cm long, 10~35 mm wide; surface: pale green ~ yellowish green, parallel-formed with veins of square reticulate, more distinct of appearance on the lower surface. Banbusae Caulis In Taeniam is the stringy strip derived from the stem with the peeled-off epidermis of Phllostachys nigra Munro var. henosis Stapf, and Phllostachys bambusoides Siebold et Zuccarini(竹葉, Gramineae). Irregular in size and shape, thin plane ~ strip-shaped, sometimes powdery, sometimes 1~3 mm thick. Outer surface: pale green ~ yellowish green, sometimes grayish white L. gracile and P. nigra have different origins although they show similar morphologic features. We were able to distinguish between L. gracile and P. nigra which are almost indistinguishable through this study. AFLP(Amplifide Fragment Length Polymorphism) was more suitable for identifying differences between L. gracile and P. nigra in comparison with other genetic analysis using chemical analysis. Therefore. molecular biological methods are believed to be useful for discovering origins of herbal medicines.

Anthracnose Caused by Colletotrichum acutatum in Robinia pseudoacacia (Colletotrichum acutatum에 의한 아까시나무 탄저병)

  • During the year 2014, black locust (Robinia pseudoacacia L.) had been observed with dark brown spots on the leaves at Andong, Cheongsong, Mungyeong in Korea. Symptoms initially appeared as small, black lesions on the leaves, and sometimes, the leaves become yellow and ultimately leads to fall off the leaves. The pathogenic fungus grown in potato dextrose agar was white or sometime gray with mycelia in tufts and from which numerous conidia were produced. The conidia were straight and fusiform in shape and measured $8.3-17.2{\times}2.5-4.1{\mu}m$. Internal transcribed spacer (ITS) rRNA sequence analysis for sequence similarity of the ITS region revealed 100% identity with nucleotide sequences for Colletotrichum acutatum. The morphological characteristics, pathogenicity and molecular data have been confirmed that the symptomatic pathogen was C. acutatum. This is the first report of anthracnose caused by C. acutatum on black locust in Korea.

  • Nematophagous fungi can capture, kill, and digest nematodes using a specific capturing organ. Of the nematophagous fungi, while Arthrobotrys flagrans and A. superba have been described previously, certain characteristics have not been described. For a detailed description of the two nematophagous fungi, the fungi were isolated from soil samples and produced in a pure culture. Morphological characteristics, such as predatory ability (according to the nematode species), shape, and size of predatory organ, conidia, and chlamydospore were investigated and they were used for identification of the fungal isolates along with molecular phylogenetic analysis. Furthermore, this study provides the classification key for 21 nematophagous species.

  • Visual system of Korean Salamander(Hynobius leechi) was morphologically studied. Fertilized eggs in egg sacs were collected and were developed in sterile saline solution. Various sized larvae of 5-30mm in length were fixed. Specimens were paraffin sectioned and were observed under light microscope. In 5mm length larva, lens rudiment induced by optic cup was combined with sensory ectoderm. The shape of lens was changed as spherical in 12mm length larva, but the retinal layer did not differentiated into three layers. The differentiation of retinal layer was clear in 14-16mm length larva. The central region of lens fibers was degenerated. Iris and ciliary body were formed from the marginal zone of optic cup in 20mm length larva. Choroid was thicker in elder eye of 30mm length larva and cartilage developed at outer region of optic cup. The outer segment of photoreceptor cell layer grew longitudinally. Optic nerve was connected to the ventral part of brain through cartilage.

Molecular Identification of Sarcocystis grueneri in Wild Korean Water Deer (Hydropotes inermis argyropus)

  • The cysts of Sarcocystis grueneri were detected and characterized from the cardiac muscles of the Korean water deer (Hydropotes inermis argyropus). Of the 38 heart muscle samples examined by light microscopy, 10 were found infected with the cysts of Sarcocystis sp. The cysts appeared oval to spherical shape and measured $110-380{\mu}m$ in length and $90-170{\mu}m$ in width. A phylogenetic tree of the 18S rRNA sequences (1.5 kb) revealed a close relationship of the infected cysts to genus Sarcocystis. The 18S rRNA sequence of the infected cysts showed 100% identity to S. grueneri and 97% to S. capracanis. Here, we first report the S. grueneri infections in the Korean water deer.

Sclerotium Rot of Pulsatilla koreana Nakai Caused by Sclerotium rolfsii (Sclerotium rolfsii에 의한 할미꽃 흰비단병)

  • Sclerotium rot on Pulsatilla koreana was observed in the exhibition field of Jinju Agriculture Technology Center in July 2013. The infected plants showed water-soaked, blighted and rotted symptoms. White mycelial mats spread over lesions, and then numerous sclerotia were formed on flower stalk and flower stem near the soil line. The sclerotia were globoid in shape, 1-3 mm in size and white to brown in color. The optimum temperature for mycelial growth and sclerotia formation on PDA was $30^{\circ}C$ and the hyphal width was $4-8{\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. For molecular identification, the complete ITS rDNA sequence of the causal fungus was sequenced and analyzed. On the basis of mycological characteristics, ITS rDNA sequence analysis, and pathogenicity to host plants, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report of sclerotium rot on Pulsatilla koreana caused by S. rolfsii in Korea.

Purification and Characterization of Peroxidase Isozyme C from Mung Bean (녹두의 Peroxidase Isozyme C의 생화학적 성장)

  • Peroxidase isozyme C was isolated from mung bean cotyledon and purified to homogeneity as ascertained by chromatography and polyacrylamide gel electrophoresis, and then crystallized. Purification procedures included ammonium sulfate precipitation and column chromatography on Sephadex G-75, DEAE-cellulose and DEAE-Sephadex A-50. Peroxidase isozyme C was purified about 63 fold with 5% recovery. Isozyme C showed optimal activity at pH 5.0 with o-dianisidine and at pH 6.0 with guaiacol as substrate, and the optimal temperature was $70^{\circ}C$. Molecular weight of 50,000 was estimated for the isozyme C by SDS-polyacrylamide gel electrophoresis. At $70^{\circ}C$, it took 30 min to inactivate the isozyme to 50%, and at $80^{\circ}C$, this isozyme was almost completely inactivated in 20 min. The Km value of isozyme C for o-dianisidine was 0.11mM and that for guaiacol was 60.98mM using hydrogen peroxide as cosubstrate, and the kinetic pattern showed a competitive cyanide inhibition with respect to substrate. The crystalline structure of isozyme C was rectangular in shape.
