In situ 타원법을 사용한 광기록매체용 Ge-Sb-Te 박막의 최적성장조건 연구 (Study of optimum growth condition of phase change Ge-Sb-Te thin films as an optical recording medium using in situ ellipsometry)

  • 타원법(ellipsometry)을 사용하여 광기록 매체용 Ge$_2$ Sb$_2$ Te$^{5}$ (GST) 박막의 성장과정에 따른 타원상수 Ψ와 $\Delta$를 측정하여. GST 박막의 최적성장조건을 연구하였다. 아르곤기체압력과 DC 출력 그리고 기판의 온도를 변화시키면서 GST 박막을 성장시켰다. 제작된 시료들의 분광타원 데이터를 모델링 분석하여 GST박막의 밀도분포를 구하고 한편으로는 GST 박막이 성장하는 동안 측정한 in situ 타원 성장곡선을 분석하여 박막의 밀도분포의 변화를 추적하였다. 아르곤기체압력이 7 mTorr일 때 박막의 상대적인 밀도분포가 고르게 되었고 DC출력이 증가함에 따라 그리고 기판의 온도가 증가함에 따라 GST 박막의 밀도 균일성은 크게 향상되었다. 주사형전자현미경(SEM)을 사용하여 최적 밀도 균일성을 가지는 성장조건(7 mTorr, 45 W, 15$0^{\circ}C$)에서 제작된 GST 시료가 가장 균일한 구조를 보여줌을 확인하였다. 균일한 밀도 분포를 가지는 GST 박막의 성장조건 확립을 통하여 여러번 기록/재생할 때 광기록 박막의 안정성을 유지하는데 크게 기여할 것이다.

Monitoring of Polycyclic Aromatic Hydrocarbons in Sediments and Organisms from Korean Coast

  • Surface sediments (0- 5 cm), mussels (Mytilus coruscus and M. edulis) and oysters (Crassostrea gigas) were sampled at 20 stations in Korean coast during a period of February to July 2000. Samples were analyzed for polycyclic aromatic hydrocarbons (PAHs) content using gas chromatography coupled to mass spectrometer detector (GC/MSD). The sediment $\sum PAH$ concentration varied from 7 to 1,214 ng/g dry weight and the level of carcinogenic PAHs of six species ranged from 1 to 563 ng/g dry weight in surface sediments. In organisms, the levels of $\sum PAH$ were in the range of 63-876 ng/g dry weight and the concentrations of carcinogenic PAHs of six species were in the range of 4-582 ng/g dry weight. The highest PAH concentrations in sediments and organisms were in samples from Station 5 in Pohang coast and Station 8 in Jinhae coast, respectively. $\sum PAH$ concentrations in sediments and organisms collected from Korean coast were slightly low or comparable to those in other countries. The contribution of ring aromatic groups to sum of 16 PAHs in sediments and organisms showed a similar pattern for most stations. In sediments, the predominant contributions were four and five ring aromatics like fluoranthene, pyrene, benzo[a]anthracene, chrysene, benzo[b]fluoranthene, benzo[k]fluoranthene and benzo[a]pyrene. In the organisms, four ring aromatics like fluoranthene, pyrene, benzo[a]anthracene and chrysene were predominant PAH species. Several molecular indices such as phenanthrene/anthracene, fluoranthene/pyrene and LMW/HMW ratio were used to identify the origin of PAH contamination in sediments. The results indicate that PAHs of Korean coast were mainly of pyro1ytic contaminants with slight petrogenic input. A high correlation within individual PAH compound in marine sediments was observed, while correlation in organisms showed a highly significant relationship for heavier molecular weight PAHs.

  • Photoelectrochemical (PEC) systems are promising methods of producing H2 gas using solar energy in an aqueous solution. The photoelectrochemical properties of numerous metal oxides have been studied. Among them, the PEC systems based on TiO2 have been extensively studied. However, the drawback of a PEC system with TiO2 is that only ultraviolet (UV) light can be absorbed because of its large band gap (3.2 - 3.4 eV). Two approaches have been introduced in order to use PEC cells in the visible light region. The first method includes doping impurities, such as nitrogen, into TiO2, and this technique has been extensively studied in an attempt to narrow the band gap. In comparison, research on the second method, which includes visible light water splitting in molecular photosystems, has been slow. Mallouk et al. recently developed electrochemical water-splitting cells using the Ru(II) complex as the visible light photosensitizer. the dye-sensitized PEC cell consisted of a dye-sensitized TiO2 layer, a Pt counter electrode, and an aqueous solution between them. Under a visible light (< 3 eV) illumination, only the dye molecule absorbed the light and became excited because TiO2 had the wide band gap. The light absorption of the dye was followed by the transfer of an electron from the excited state (S*) of the dye to the conduction band (CB) of TiO2 and its subsequent transfer to the transparent conducting oxide (TCO). The electrons moved through the wire to the Pt, where the water reduction (or H2 evolution) occurred. The oxidized dye molecules caused the water oxidation because their HOMO level was below the H2O/O2 level. Organic dyes have been developed as metal-free alternatives to the Ru(II) complexes because of their tunable optical and electronic properties and low-cost manufacturing. Recently, organic dye molecules containing multi-branched, multi-anchoring groups have received a great deal of interest. In this work, tri-branched tri-anchoring organic dyes (Dye 2) were designed and applied to visible light water-splitting cells based on dye-sensitized TiO2 electrodes. Dye 2 had a molecular structure containing one donor (D) and three acceptor (A) groups, and each ended with an anchoring functionality. In comparison, mono-anchoring dyes (Dye 1) were also synthesized. The PEC response of the Dye 2-sensitized TiO2 film was much better than the Dye 1-sensitized or unsensitized TiO2 films.

Comparison of apical extrusion of intracanal bacteria by various glide-path establishing systems: an in vitro study

  • Objectives: This study compared the amount of apically extruded bacteria during the glide-path preparation by using multi-file and single-file glide-path establishing nickel-titanium (NiTi) rotary systems. Materials and Methods: Sixty mandibular first molar teeth were used to prepare the test apparatus. They were decoronated, blocked into glass vials, sterilized in ethylene oxide gas, infected with a pure culture of Enterococcus faecalis, randomly assigned to 5 experimental groups, and then prepared using manual stainless-steel files (group KF) and glide-path establishing NiTi rotary files (group PF with PathFiles, group GF with G-Files, group PG with ProGlider, and group OG with One G). At the end of canal preparation, 0.01 mL NaCl solution was taken from the experimental vials. The suspension was plated on brain heart infusion agar and colonies of bacteria were counted, and the results were given as number of colony-forming units (CFU). Results: The manual instrumentation technique tested in group KF extruded the highest number of bacteria compared to the other 4 groups (p < 0.05). The 4 groups using rotary glide-path establishing instruments extruded similar amounts of bacteria. Conclusions: All glide-path establishment instrument systems tested caused a measurable apical extrusion of bacteria. The manual glide-path preparation showed the highest number of bacteria extruded compared to the other NiTi glide-path establishing instruments.

Aspergillus flavus Y2H001의 식물생육촉진과 Gibberellin A3의 생산 (Plant Growth Promotion and Gibberellin A3 Production by Aspergillus flavus Y2H001)

  • 경상북도 성주군의 농경지에서 자생하는 들깨를 채집하여 이로부터 형태학적으로 상이한 15개의 내생진균을 순수 분리하였다. 이들의 배양여과액을 이용하여 난장이벼의 유묘에 처리하여 식물생장촉진 활성을 조사한 결과, 이들 중 Y2H001균주가 식물생장활성이 가장 우수한 것으로 나타났다. Y2H001균주의 ITS영역 염기서열과 ${\beta}$-tubulin 유전자 염기서열을 사용하여 계통학적 유연관계를 확인하였으며, 이러한 분자계통학적 분석 및 형태학적 관찰을 통해 Aspergillus flavus로 동정되었다. 또한 A. flavus Y2H001균주의 배양여과액을 GC/MS를 통하여 분석하였고 식물생장을 촉진하는 원인물질로 $GA_3$ (1.954 ng/mL)를 생산하는 것을 확인하였다.

졸-겔법을 이용한 γ-Al2O3 합성 시 PVA와 HNO3 첨가에 따른 입자크기 제어 (Particle Size Control by the Addition of PVA and HNO3 in γ-Al2O3 Synthesis Using by Sol-Gel Method)

  • 세라믹 재료 중 알루미나(Al2O3)는 산업에서 널리 사용되는 세라믹 재료로서 최근의 기술발전에 따라 재료 크기가 작아지고 이에 따른 특성이 다양하여 그 중요성이 더해 가고 있다. 본 연구에서는 다양한 알루미늄 알콕사이드 중 Aluminum isopropoxide(AIP)를 출발 원료물질로 하여 졸-겔(Sol-Gel)법에 의해 가수분해 및 해교과정을 거쳐 boehmite 졸을 제조하고 이후 건조 및 하소시켜 γ-Al2O3를 제조하였다. 이러한 제조 과정 중 입자의 응집현상을 방지하기 위해 9,000 ~ 10,000, 31,000 ~ 50,000, 89,000 ~ 98,000, 130,000의 분자량을 갖는 4종류의 PVA(Polyvinyl alcohol)를 첨가하고 3종류 질산(0.1, 0.3, 0.5 몰비)을 첨가하여 입자에 미치는 영향을 확인하고자 하였다. 제조된 γ-Al2O3는 X선 회절분석기(XRD), X선 형광분석기(XRF), 입도분석기(PSA), 전계방사 주사전자현미경(FE-SEM) 등의 기기분석을 통하여 결정구조 및 조성, 입자크기, 그리고 입자형상을 확인하였다. 그 결과, 약 98.2 %의 순도를 갖는 γ-Al2O3가 합성되었으며 첨가되는 질산의 첨가비가 높을수록, 그리고 PVA 분자량이 클수록 입자크기가 감소하고 균일성이 높아지는 것을 확인할 수 있었다. 이러한 결과로부터, PVA와 질산의 첨가비 조절에 따라 γ-Al2O3의 제조공정 중 입자크기 제어가 가능할 것으로 사료된다.

  • 병원성 곰팡이에 대해 강한 길항성을 보이는 토양미생물 Bacillus subtilis HJ927을 Phytophthora capsici에 감염된 고추밭으로부터 분리해냈다. B. subtilis HJ927을 P. capsici와 함께 고추에 처리한 결과 P. capsici만을 접종한 처리구에 비해 크게 식물을 보호하는 것을 root mortality 측정결과 확인하였다. B. subtilis HJ927는 항곰팡이성 물질로 3-methylbutyric acid, 2-methylbutyric acid, 그리고 methyl 2-hydroxy, 3-phenyipropanoate를 분비해 내는 것을 HPLE와 GC-MS를 통해 분리 동정하였다. 또한 B. subtilis HJ927는 가수분해효소인 ${\beta}$-1,3-glucanase를 분비해 냄으로서 위 화합물과 함께 식물을 보호하는 것으로 분석되었다.

습식 인공열화 시 밀랍본 복제품의 제책 부위별 열화특성 (Studies on the Aging Characteristics in Different Parts of Beeswax-treated Duplicates during Humidified Artificial Aging)

  • In this study, to understand the aging factor and mechanism in different partitions of the beeswax-treated volumes, the duplicated beeswax-treated volume was artificially aged at $80^{\circ}C$ of temperature and 65% of relative humidity and then a physical and optical properties of an aged volumes was analyzed. Also, the degraded components of the beeswax samples isolated from different partitions of aged volumes was measured using a gas chromatography/mass spectroscopy (GC/MS). In results, the surface of beeswax-treated volume which is primarily affected by a main aging factors such as light, oxygen, moisture was more deteriorated than the inside of that volume. However, unlike inside of the book volume which was made from paper, the inside of beeswax-treated volume wax was also considerably deteriorated. The inside of the beeswax-treated volume is largely unaffected by the oxygen and humidity during aging due to the water repellency and the air permeation resistance of beeswax. Therefore, it is confirmed that aging factors and mechanisms in the inside of the volume are different from thats of the outside of the volume. This fact was also verified by the results of GC/MS analysis of an beeswax samples which was sampled from different partitions of aged volumes. As result as GC/MS analysis of the beeswax extracted from the outside of the aged volume, the low molecular compounds with a carbon length of $C_9-C_{20}$ (fatty acid, etc) were increased and the compounds with a carbon length of above $C_{34}$ (ester, etc) were also increased. But the compounds with a chain length of $C_{21}-C_{36}$ (hydrocarbon, alcohol, etc) were decreased. In case of the aged beeswax of inside, the low molecular compounds with a carbon length of $C_9-C_{20}$ (fatty acid, etc) and the compounds with a chain length of $C_{21}-C_{36}$ (hydrocarbon, alcohol, etc) were increased. While, the compounds with a carbon length of above $C_{34}$ (ester, etc) were decreased.

셀룰로오스질 유기고형폐물의 열분해에 관한 연구 (A Study on Pyrolysis of Cellulosic Organic Solid Wastes)

  • 셀룰로오스질 유기고형폐물로서 목피 및 톱밥과 순수한 셀룰로오스로서 여지를 $300^{\circ}C$ 질소기류하 또는 질소와 공기의 혼합기류하에서 열분해하고 생성물은 공기냉각, 수냉각, 드라이아이스-아세톤 냉각에 의해서 응축되는 성분과 비응축성 가스 및 탄화찌끼의 양을 조사하였다. 또 이중 드라이아이스-아세톤 냉각에 이해서 포집된 휘발성 액체 성분을 기체크로마토그래프를 이용하여 분리하고 이들 성분의 머무는 시간과 시린지반응에 의해 동정하였다. 질소기류하에서 열분해할 경우에는 타아르성분이 13.4 ∼ 29.6${\%}$, 수용성액체가 0.01 ∼ 0.12${\%}$, 휘발성액체가 0.24 ∼ 1.43${\%}$, 비응축성가스가 9.84 ∼ 42.4${\%}$, 탄화찌끼가 44.0 ∼ 65.81${\%}$이었고 혼합기류하에서 열분해할 경우에는 다른 성분이나 응축성 액체의 양이 감소되고 비응축성가스가 증가되었다. 질소기류하에서 포집된 휘발성액체는 Porapak Q 칼럼에 의해서 분해물질에 상관없이 19가지의 똑같은 성분으로 분리되었고 다만 각 성분의 상대적 양만이 차이를 나타내었다. 혼합기류하에서 열분해할 경우에는 6개성분만이 생성되었고 주로 methanol, formaldehyde 등의 분자량이 적은 물질만이 생성되었다. 분리된 19성분을 머무는 시간과 시린지 반응에 의해 동정해 본 결과 methanol, formaldehyde, acetone, acetaldehyde, acetic acid와 3개의 hydroxyl기를 가진 것으로 추측되는 성분이 동정되었다.

Clinical and molecular biological aspect of the hyaluronidases: basis and clinical overview for oriental medical application

  • Components of extracellular matrix and the matrix-degrading enzymes are some of the key regulators of tumor metastasis and angiogenesis. Hyaluronic acid (HA), a matrix glycosaminoglycan, is known to promote tumor adhesion and migration, and its small fragments are angiogenic. Until now, we have compared levels of hyaluronidase, an enzyme that degrade HA, in normal adult prostate, benign prostate hyperplasia and prostate cancer tissues and in conditioned media from epithelial explant cultures, using a substrate (HA)-gel assay and ELISA-like assay (Kim et al., unpublished results). The present review described an overall characterization of hyaluronidases and its application to human diseases. The hyaluronidases are a family of enzymes that have, until recently, deed thorough explication. The substrate for these enzymes, hyaluronan, is becoming increasingly important, recognized now as a major participant in basic processes such as cell motility, wound healing, embryogenesis, and implicated in cancer progression. And in those lower life forms that torment human beings, hyaluronidase is associated with mechanisms of entry and spread, e.g. as a virulence factor for bacteria, for tissue dissection in gas gangrene, as a means of treponema spread in syphilis, and for penetration of skin and gut by nematode parasites. Hyaluronidase also comprises a component of the venom of a wide variety of organisms, including bees, wasps, hornets, spiders, scorpions, sh, snakes and lizards. Of particular interest is the homology between some of these venom hyaluronidases and the enzyme found in the plasma membrane of mammalian spermatozoa, attesting to the ancient nature of the conserved sequence, a 36% identity in a 300 amino acid stretch of the enzyme protein. Clearly, hyaluronidase is of biological interest, being involved in the pathophysiology of so many important' human disorders. Greater effort should be made in studying this family of enzymes that have, until recently, been overlooked. Also, oriental medical application of the hyaluronidase will be discussed with respect to inhibition and suppression of inflammation and malignacy.
