Graft Copolymerization of Chitosan and Mono(2-methacryloyl oxyethyl) Acid Phosphate and Its Antifungal Effect (키토산과 모노(2-메타크릴로일 옥시에틸)산 포스페이트 그라프트공중합과 그의 항균효과)

  • New type of chitosan deriertives was synthesised by graft copolymerization of mono(2-methacryloyl oxyethyl) acid phosphate into chitosan. The reaction conditions showing the highest percentage of grafting were an initiator concentration of $3.5{\times}10^{-3}M$, monomer concentration of 0.19 M, and reaction temperature of $40^{\circ}C$, while rate of grafting showed same value after elapsing 4hours of reaction time. The antifungal activity of chitosan depending on content of free amine and kind of monomer was examined against Candida albicans, Trichophyton rubrum and Trichophyton uiolaceum by shake flask method. The pH value of buffer solution to show the highest antifungal activity was 5.75, and the selectivity of mold strain was observed.

THE EFFECT OF SPRUE DESIGN ON THE MARGINAL REPRODUCIBILITY OF CAST TITANIUM CROWNS (주입선 형태가 티타늄 주조관의 변연재현성에 미치는 영향)

  • There has been a great interest in the use of titanium for fixed and removable prostheses in recent because of its excellent biocompatibility. However the high melting temperature and chemical reactivity of titanium necessitates casting systems different from those used in conventional casting. The current titanium casting systems are based on an electric-arc design for melting the metal in an argon atmosphere and its exclusive investment. Despite the new developments in Ti casting systems, inadequate mold filling and internal porosity are frequently observed casting defects. Therefore, the study on the fabrication technique including sprue design to solve these casting defects is still necessary. In this study to evaluate the effect of sprue design on the castability of simulated cast titanium crowns, 10 cylindrical cast crowns for each group with four different sprue design(Single group. Double group, Runner bar group. Reservoir group) were fabricated. An impression of the entire casting margin was made and cut at $90^{\circ}$ intervals, and the sections were photographed in a microscope at $100{\times}$ magnification to record marginal discrepancy. The internal porosities of the cast crowns were disclosed by radiographs. Within the limits of this study. the following conclusions were drawn. 1. The overall mean marginal discrepancies for each group were as follows: Double group, $43.65{\mu}m$; Reservoir group, $50.27{\mu}m$; Single group, $54.17{\mu}m$; Runner bar group, $58.90{\mu}m$ (p<0.05). 2. The mean of marginal discrepancies for wax patterns was $10.65{\mu}m$. 3 The numbers of internal porosity showed the most in Runner bar group followed by Single group, Reservoir group, and Double group.

Determination of Salable Shelf-life for Wrap-packaged Dry-aged Beef during Cold Storage

  • We investigated microbial and quality changes in wrap-packaged dry-aged beef after completion of aging and subsequent storage in a refrigerator. After 28 days of dry aging (temperature, $4^{\circ}C$; RH, approximately 75%; air flow velocity, 2.5 m/s), sirloins were trimmed, wrap-packaged, and stored at $4^{\circ}C$ for 7 days. Analyses of microbial growth, pH, volatile basic nitrogen (VBN), 2-thiobarbituric acid-reactive substance (TABRS), and instrumental color, myoglobin, and sensory evaluation were conducted on days 0, 3, 5, and 7. The results show that the number of total aerobic bacteria (TAB), yeast, and lactic acid bacteria increased with an increase in storage days, whereas no change in the growth of mold was observed during 7 days of storage. Based on the legal standard for TAB count, the estimated shelf-life of wrap-packaged dry-aged beef was predicted to be less than 12.2 days. However, the shelflife should be less than 6.3 days, considering the result of sensory quality (odor, taste, and overall acceptance). No significant change in visible appearance was also observed during 7 days of storage. The results suggest that the present quality indicators for meat spoilage (pH, VBN, and TBARS) should be re-considered for dry-aged beef, as its characteristics are different from those of fresh and/or wet-aged beef.

  • For the purpose of disinfection and efficient storage, Korean Paldal variety rice was hulled into the unpolished and polished ones and packed in the kraft paper bags, irradiated with relatively high doses, 500, 800, and 1,000 krad from the $CO^{60}$ source and stored at room temperature, a variety of changes were observed as follows. 1) With 1,000 krad irradiation, mold was almost sterilized. 2) Fat acidity increased during storage and continued to increase more with increased doses. 3) The content of riboflavin eras severely reduced with higher doses. 4) The results of organoleptic test were featured by a yellow color and a keen irradiation odor appearing in the boiled rice. 5) In the irradiated rice starch with 3,000 krad, hydrolysis with ${\alpha}$-amylase was not effective while accelerated with diastase.

Outbreak of Bioaerosols with Continuous Use of Humidifier in Apartment Room

  • The effect of continuous humidifier use on the bioaerosol concentration in an indoor environment was investigated. An ultrasonic humidifier was operated for 10 hr per day for 15 days in an apartment room. During this time period, viable bioaerosol samples were taken using a single-stage Andersen sampler containing culture media plates for bacteria and fungi. The culture plates were then incubated at room temperature for 2~7 days depending on the media. The counts for the air sample plates were corrected for multiple impactions using the positive hole conversion method and are reported as the colony forming units per cubic meter of air (CFU/$m^3$). While the bacterial concentration measured using the tryptic soy agar (TSA) did not show any significant change during the first 3 days, the concentration increased from the $6^{th}$ day (6979 CFU/$m^3$) and reached a maximum on the $9^{th}$ day (46431 CFU/$m^3$). The concentration then decreased to 2470 CFU/$m^3$ on the $12^{th}$ day, at which point the fungal concentration increased rapidly to 14424~16038 CFU/$m^3$. Also, while the fungal concentration showed a significant change until the $9^{th}$ day of humidifier use, fungal growth was observed on the wallpaper and increased rapidly from the $12^{th}$ day. However, the bacterial concentration increased rapidly after the fungi were removed by remediation. The major fungal species identified in the samples were Penicillium representing 34%, Aspergillus representing 31%, Cladosporium representing 24%, and Alternaria representing 1%. The results also indicated that a relative humidity over 80% was easily achieved with continuous humidifier use. Yet, maintaining a high humidity in a room can cause a rapid outbreak of microbial growth.

AN ELECTROCHEMICAL STUDY ON SURFACE FINISH OF DENTAL AMALGAM (아말감의 표면연마에 관한 전기화학적 연구)

  • The purpose of this study was to observe characteristic properties of amalgam through the polarization curves and SEM images from 4 type amalgams (Amalcap, Shofu spherical. Dispersalloy and Tytin) with 3 different surface finish procedures (polishing, burnishing and carving) by using the potentiostats (EG & GPARC) and SEM (Jeol JSM-35). After each amalgam alloy and Hg was triturated as the direction of the manufacturer by means of mechanical amalgamator (Samki), the triturated mass was inserted into the cylndrical metal mold which was 12 mm in diameter and 10 mm in height and was pressed with $100kg/cm^2$. 4 specimens of each type amalgam were burnished with egg burnisher and another 4 specimens of each type amalgam were carved with Hollenback carver. Above 8 specimens and remaining untreated 4 specimens were stored at room temperature for about 7 days. Untreated 4 specimens of each type amalgam were polished with abrasive papers (Deer) from #400 to #1200 and finally on the polishing cloth with $0.5{\mu}m$ and $0.06{\mu}m$ $Al_2O_3 $ powder suspended water. Anodic polarization measurements was employed to compare the corrosion behaviours of the amalgams in 0.9% saline solution at $37^{\circ}C$. The open circuit potential was determined after 30 minutes immersion of specimen in electrolyte. The scan rate was 1 mV/sec and the surface area of amalgam exposed to the solution was $0.64cm^2$ for each specimen. All the potentials reported are with respect to a saturated calomel electrode (SCE). SEM images of each specimen were taken after + 800 mV (SCE) polarization. The results were as follows: 1. The corrosion potential of high copper amalgam was more anodic than that of low copper amalgam. 2. The polished amalgam were more resistant to corrosion than any other burnished and carved amalgam. 3. In the case of polishing, current density of high copper amalgam was lower than that of low copper amalgam.

  • By the recently developed GMP(Glass Molding Press) process, aspheric glass lenses are widely used in many optical applications such as digital cameras, optical data storages and electrical devices etc. The GMP process can economically produce complex shaped glass lenses with high precision and good repeatability. This study deals the optimization of molding conditions for aspheric glass lenses in progressive GMP process through Design Of Experiment(Taguchi method). Tree main factors for molding conditions were selected based on pressure, temperature and cooling time at 1st cooling stage. From the analysis of experiments which were preformed with 3-cavity glass mold, it was revealed that the cooling time was the most sensitive parameter for form accuracy(PV) in progressive GMP process.

Isolation and Characterization of an Alkaline Cellulase Produced by Alkalophilic Bacillus sp. HSH-810 (알칼리성 Cellulase를 생산하는 호알칼리성 Bacillus sp. HSH-810의 분리 및 효소 특성)

  • A bacterium producing alkaline cellulase was isolated from soil, leaf mold and compost, and was identified as alkalophilic Bacillus sp. HSH-810 by morphological, cultural and biochemical determination. The optimum cul-ture condition of Bacillus sp. HSH-810 for the growth and alkaline cellulase production was $30^{\circ}C$ and pH 10.0. The maximum alkaline cellulase production was obtained when 1.0%(w/v) CMC, 0.5%(w/v) peptone, 0.02%(w/v) $CaCl_2$ and 0.02(w/v) $CoCl_2$ were used as carbon source, nitrogen source and mineral source, respectively. The optimum pH and temperature of the enzyme activity were pH 10.5 and $50^{\circ}C$, respectively. This enzyme was fairly stable in the pH range of 6.0-13.0 and at $50^{\circ}C$. For the effect of surfactants, the activity of alkaline cellulase was stable in the presence of sodium-$\alpha$-olefin sulfonate (AOS), sodium dodecyl sulfonate (SDS), Tween 20 and Tween 80, but inhibited by the presence of 0.1 linear alkyl-benzene sulfonate (LAS) sig-nificantly.

Prediction of Cooling Performance for Indirect Evaporative Cooling System Using Danpla Sheet (단프라시트를 적용한 간접식 증발냉각 장치의 냉각 성능 예측)

  • Previous plastic heat exchangers are expensive because the mold must be newly manufactured depending on the air conditioning space. On the other hand, danpla is so thin that the heat exchange performance is excellent. Moreover, danpla can be used easily in ventilation systems in view of fabrication. This study proposes correlations for the cooling performance of an indirect evaporative cooling system. The experimental apparatus consisted of a heat exchanger, spray nozzle, fan, thermostat, pump, and measuring sensors for temperature, humidity, and airflow rate. The results showed that the effectiveness decreased gradually as the airflow rate increased. In addition, there was an optimal condition in terms of effectiveness. The performance prediction correlations were determined using the experimental data from various conditions. The proposed correlations showed performance accuracies within 4 % error.

Isolation and physiological characteristics of cellulolytic bacteria (섬유소 분해세균의 분리 및 생리적인 특성)

  • Three hundred and one cellulolytic bacterial were isolated from the 148 screening sources such as decomposed wood, soil, compost and leaf mold. Among them, strain KL-6 was found to have the highest of cellulase activity, and identified as species belonged to the genus Cellulomonas. Strain KL-6 was decompose up to 90% of the filter paper (whatman No. 1) substrate within 50 hours, and showed the colony halo formation (11 cm). The activities of CMCase (67 unit/ml), FPase (70 unit/ml) and ${\beta}-glucosidase$ (0.68 unit/ml) were obtained when this strain was cultured for 50 hrs at $30^{\circ}C$. Glucose was not found in detectable amounts at the FP medium. The optimum composition of nutrient medium for the cell growth by strain KL-6 was sucrose 0.5%, yeast extract 0.1%, $(NH_4)_2HPO_4\;0.1%$, $K_2HPO_4\;0.1%$, $MgSO_4{\cdot}7H_2O\;0.01%$, $CaCl_2\; 0.01%$, NaCl 0.6%, $CaCO_3\;0.1%$ and the optimum pH and temperature were 7.0 and $30^{\circ}C$, respectively.
