A Study on the Mixed-Food Wastewater Treatment in an Anaerobic Packed Bed Reactor and Sludge Bed Reactor (혐기성 고정상반응기와 슬러지 Bed반응기에서 혼합-식품폐수처리에 관한 연구)

  • The characteristics of food wastewater treatment in an anaerobic packed bed reactor (APBR) with polyurethane as a packing material and sludge bed reactor (ASBR) was studied. The reactor of 9cm-ID, 150cm-height was fed in a continuous mode from bottom of reactor. For the purpose of constant temperature of reactor, water jacket was installed. The used packing materials was polyurethane sponge foam. Methane which was produced by decomposed organics collected at the top of the reactor for using as a fuel. The substrates used were synthetic, mixed and food wastewater. For the acclimatization of microorganisms, mixed wastewater was used. The major analyses were gas production, COD, pH and volatile acids. Based upon the completed works, the results are as follows: When food wastewater was fed the quantity of produced gas was less than that of synthetic wastewater, but food process saw higher methane content than synthetic process. As well as COD removal efficiency of food process reached at about 85%. In aspect of effluent volatile acid, food process showed low concentration of below 500 mg/l, therefore anaerobic reaction stabled. Conclusively food wastewater used can be digested by anaerobic treatment, especially anaerobic packed bed reactor showed 82% of COD removal, 75% of methane content, 10 l of gas production, and anaerobic sludge bed reactor did 79% of COD removal, 75% of methane content, 81 of gas production at 4 kgCOD/m$^3$day, 36$\circ$C.

Fruitbody Development of Pleurotus ostreatus via Bottle Cultivation Using Recycled Substrate

  • This study was carried out to determine the possibility of bottle cultivation utilizing recycled oyster mushroom culture waste as a cultivating substrate for P. ostreatus. Total nitrogen percentage was 0.76%, 1.13%, 1.16%, 1.36%, and 1.38% in the 1-, 2-, 3-, 4-, and 5-time mixed substrate, respectively; 0.95%, 1.04%, 1.34%, 1.36%, and 1.25% in the 1-, 2-, 3-, 4-, and 5-time postharvest substrate, respectively; and 0.72% and 0.68% in the 2- and 3-time nonadditive substrate, respectively. Weight of the fresh fruiting body harvest was 115 g, 120 g, 117 g, 118 g, and 114 g on 1-, 2-, 3-, 4-, and 5-time mixed substrate, respectively; and 105 g and 45 g on 2- and 3-time nonadditive substrate, respectively. The first mixed substrate (fresh) and recycled substrates generated no significant difference in the weight of fresh fruiting bodies harvested.

Property and Microstructure Evaluation of Pd-inserted Nickel Monosilicides (Pd 삽입 니켈모노실리사이드의 물성과 미세구조 변화)

  • A composition consisting of 10 nm-Ni/1 nm-Pd/(30 nm or 70 nm-poly)Si was thermally annealed using rapid thermal for 40 seconds at $300{\sim}1100^{\circ}C$ to improve the thermal stability of conventional nickel monosilicide. The annealed bilayer structure developed into $Ni(Pd)Si_x$, and the resulting changes in sheet resistance, microstructure, phase, chemical composition, and surface roughness were investigated. The silicide, which formed on single crystal silicon, could defer the transformation of $NiSi_2$, and was stable at temperatures up to $1100^{\circ}C$. It remained unchanged on polysilicon substrate compared with the sheet resistance of conventional nickel silicide. The silicides annealed at $700^{\circ}C$, formed on single crystal silicon and 30 nm polysilicon substrates exhibited 30 nm-thick uniform silicide layers. However, silicide annealed at $1,000^{\circ}C$ showed preferred and agglomerated phase. The high resistance was due to the agglomerated and mixed microstructures. Through X-ray diffraction analysis, the silicide formed on single crystal silicon and 30 nm polysilicon substrate, showed NiSi phase on the entire temperature range and mixed phases of NiSi and $NiSi_2$ on 70 nm polysilicon substrate. Through scanning probe microscope (SPM) analysis, we confirmed that the surface roughness increased abruptly until 36 nm on 30 nm polysilicon substrate while not changed on single crystal and 70 nm polysilicon substrates. The Pd-inserted nickel monosilicide could maintain low resistance in a wide temperature range and is considered suitable for nano-thick silicide processing.

  • This experiment was carried out to investigate the growth and yield responses of hydroponically grown tomato as affected by the different substrates using perlite and mixtures with perlite. Substrates used in this experiment were perlite (fine and coarse granule), peatmoss, rice hull and carbonized rice hull. The results obtained were as follows ; The best results in terms of total fruit yield and average fruit weight obtained in the single treatment of perlite, followed by perlite mixture with rice hull and carbonized rice hull. Leaf area was also higher in the plots of perlite mixture and optimum mixing ratio of perlite, peatmoss and carbonized rice hull was determined as 5:3:2 by volume. The more the rice hull was added to the mixed substrates, the less in fruit production was observed. However, adding perlite to other substrates brought higher fruit yield. Single treatment of rice hull showed the lowest fruit yield but enhanced in soluble solids contents over 6.0 $^{\circ}$Brix at each cluster.

Effect of Disodium Fumarate on In vitro Rumen Fermentation of Different Substrates and Rumen Bacterial Communities as Revealed by Denaturing Gradient Gel Electrophoresis Analysis of 16S Ribosomal DNA

  • Two experiments were conducted to investigate the effects of disodium fumarate on the in vitro rumen fermentation profiles of different substrates and microbial communities. In experiment 1, nine diets (high-forage diet (forage:concentrate, e.g. F:C = 7:3, DM basis), medium-forage diet (F:C = 5:5, DM basis), low-forage diet(F:C = 1:9, DM basis), cracked corn, cracked wheat, soluble starch, tall elata (Festuca elata), perennial ryegrass and rice straw) were fermented in vitro by rumen microorganisms from local goats. The results showed that during 24 h incubations, for all substrates, disodium fumarate increased (p<0.05) the gas production, and tended to increase (p<0.10) the acetate, propionate and total VFA concentration and decrease the ratio of acetate to propionate, whereas no treatment effect was observed for the lactate concentration. The apparent DM loss for tall elata, perennial ryegrass and rice straw increased (p<0.05) with the addition of disodium fumarate. With the exception of tall elata, perennial ryegrass and rice straw, disodium fumarate addition increased the final pH (p<0.05) for all substrates. In experiment 2, three substrates (a high-forage diet, a medium-forage diet and a high concentrate diet) were fermented by mixed rumen microbes in vitro. A polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) technique was applied to compare microbial DNA fingerprints between substrates at the end of 24 h incubation. The results showed that when Festuca elata was used as substrate, the control and disodium fumarate treatments had similar DGGE profiles, with their similarities higher than 96%. As the ratio of concentrate increased, however, the similarities in DGGE profiles decreased between the control and disodium fumarate treatment. Overall, these results suggest that disodium fumarate is effective in increasing the pH and gas production for the diets differing in forage: concentrate ratio, grain cereals and soluble starch, and in increasing dry matter loss for the forages (tall elata, perennial ryegrass and rice straw) in vitro, whereas its effect on changes of ruminal microbial community may largely depend on the general nature of the substrate.

A Study on the Driving Characteristics of Microbial Fuel Cell Using Mixed Strains in Domestic Wastewater (생활폐수 내 혼합균주를 이용한 미생물 연료전지의 구동 특성에 관한 연구)

  • The use of fossil fuels is a major contributor to the increase atmospheric greenhouse gas emissions. As such problems arise, interest in new and renewable energy devices, particularly fuel cells, is greatly increasing. In this study, various characteristics of mixed strains were observed in wastewater collected by the Jeonju Environment Office to investigate the effects of microorganisms on voltage generation and voltage generation of substrates, electrode materials, electrons, electron transport media, and ash microbial fuel cells. As a result of separately measuring the voltage generated during inoculation, the inoculation voltage of Escherichia coli K12 (E. coli K12) was 0.45 V, and the maximum inoculation voltage of the mixed strain was 1.2 V. Thereafter, voltage values were collected using a digital multimeter and the amount of voltage generated over time was measured. In the case of E. coli K12, the maximum voltage reached 0.45 V, and the cell voltage was maintained above 0.23 V for 140 hours. In contrast, for the mixed strain, the maximum voltage reached 1.2 V and the voltage was slowly decreased to 0.97 V. In addition, the degree of microbial adsorption to the electrod surface after the inoculation test was confirmed using a scanning electron microscope. Therefore, these results showed the possibility of purifying pollutants at the same time as power generation through the production of hydrogen ions using microorganisms and wastewater.

  • The strain FJ1 isolated from a rotten wood showed high activity to hydrolysis of cellulosic materials. The strain produced largely enzymes related in hydrolysis of cellulose and hemicellulose, such as CMCase, xylanase, ${\beta}-glucosidase$ , and avicelase. The culture conditions(pH, temperature, inoculation concentration) and substrate specificity to various cellulosic materials were examined to elevate productivity of the enzymes. The enzyme activities of CMCase and xylanase were 13.5U/ml and 24.3U/ml in agitation culture using Mandel's medium, respectively. The high activity of the enzymes was earned when mixed cellulosic materials of rice straw, sawdust, and pulp as substrates, indicating that the strain FJ1 could use crystalline substrates.

Effect of oxygen pressure on properties of $NdBa_2Cu_3O_{7-{\delta}}$ films on $SrTiO_3$ (100) substrates grown by pulsed laser deposition

  • We report a successful fabrication of high-$J_C$ $NdBa_2Cu_3O_{7-{\delta}}$ (NdBCO) films on (100) $SrTiO_3$ substrates by pulsed laser deposition (PLD) in high oxygen pressures ranging from 400 to 800 mTorr. Fabricated NdBCO films exhibited only c-axis orientation, good out-of-plane and in-plane textures, and also excellent superconducting properties, including critical temperature ($T_C$) and critical current density ($J_C$) of above 90 K and the highest of $3.1MA/cm^2$ at 77 K in self-field, implying that NdBCO is a perspective alternative to YBCO for coated conductor. In low oxygen pressures ranging from 100 to 200 mTorr, however, the films showed a-, c-mixed orientation and degraded $T_{C,zero}$ values due to the formation of $Nd_{1+x}Ba_{2-x}Cu_3O_{7-{\delta}}$-type solid solutions with an excessive substitution of $Nd^{3+}$ ions for the $Ba^{2+}$ sites.

Effect of OHθ and o-lodosobenzoate Ions on Dephosphorylation of Organo Phosphororus Ester in CTAX Micelle (CTAX 미셀 용액속에서 유기인 에스테르 화학물의 탈인산화 반응에 대한 OHθ 및 o-lodosobenzoate 이온의 영향)

  • This study deals with micellar effects on dephosphorylation of diphenyl-4- nitrophenylphosphate (DPNPPH), diphenyl-4-nitrophenylphosphinate (DPNPlN) and isopropylphenyl-4-nitrophenyl phosphinate (IPNPlN) mediated by $OH^\Theta$ or o-iodosobenzoate ion $(IB^\Theta)$ in aqueous and CTAX solutions. Dephosphorylation of DPNPPH, DPNPIN and IPNPIN mediated by $OH^\Theta$ or o-iodosobenzoate ion $(IB^\Theta)$ is relatively slow in aqueous solution. The reactions in CTAX micellar solutions are, however, much accelerated because CTAX micelles can accommodate both reactants in their Stem layer in which they can easily react, while hydrophilic $OH^\Theta\;(or\;IB^\Theta)$ and hydrophobic substrates are not mixed in water. Even though the concentrations $(>10^{-3}\;M)\;of\;OH^\Theta\;(or\;IB^\Theta)$ in CTAX solutions are much larger amounts than those $(6\times10^{-6}\;M)$ of substrates, the rate constants of the dephosphorylations are largely influenced by the change of concentration of the ions, which means that the reactions are not followed by the pseudo first order kinetics. In comparison to effect of the counter ions of CTAX in the reactions, CTACI is more effective on the dephosphorylation of substrates than CTABr due to easier expelling of $Cl^\Theta$ ion by $OH^\Theta\;(or\;IB^\Theta)$ ion from the micelle, because of easier solvation of $Cl^\Theta$ ion by water molecules. The reactivity of IPNPlN with $OH^\Theta\;(or\;IB^\Theta)$ is lower than that of DPNPlN. The reason seems that the 'bulky' isopropyl group of IPNPIN hinders the attack of the nucleophiles.

Usage of Enzyme Substrate to Protect the Activities of Cellulase, Protease and α-Amylase in Simulations of Monogastric Animal and Avian Sequential Total Tract Digestion

  • Cellulase from Aspergillus niger, (${\alpha}$-amylase from Bacillus sp. and protease from Bacillus globigii were used as enzyme sources in this study to examine how their respective substrates protect them in two kinds of simulated gastrointestinal tract digesting processes. Avian total digest tract simulation test showed that filter paper, Avicel and cellulose resulted in 7.7, 6.4 and 7.4 times more activity than of unprotected cellulose, respectively. Protease with addition of casein, gelatin or soybean protein showed no significant protection response. Starch protected amylase to be 2.5 times activity of the unprotected one. Monogastric animal total tract digestion simulation test showed that filter paper, Avicel and cellulose resulted in 5.9, 9.0 and 8.8 times activity of unprotected cellulase, respectively. Casein, gelatin and soybean protein resulted in 1.2, 1.3 and 2.0 times activity of unprotected protease, respectively. Starch did not protect amylase activity in monogastric animal total tract simulation. Protection of mixed enzymes by substrates in two animal total tract simulation tests showed that filter paper in combination with soybean protein resulted in 1.5 times activity of unprotected cellulose, but all substrates tested showed no significant protection effect to protease. Soybean protein and starch added at the same time protected the amylase activity to be two times of the unprotected one. Test of non-purified substrate protection in two animal total digest tract simulation showed that cellulase activity increased as BSA (bovine serum albumin) concentration increased, with the highest activity to be 1.3 times of unprotected enzyme. However, BSA showed no significant protection effect to protease. Amylase activity increased to 1.5 times as BSA added more than 1.5% (w/v). Cellulase activity increased to 1.5 times as soybean hull was added higher than 1.5%. Amylase had a significant protection response only when soybean hull added up to 2%. Protease activity was not protected by soybean hull to any significant extent.
