한국산 색가오리과(Dasyatidae) 어류 1미기록종, Pteroplatytrygon violacea (First Record of the Pelagic Stingray, Pteroplatytrygon violacea (Dasyatidae, Myliobatiformes) from Korea)

  • 매가오리목, 색가오리과에 속하는 Pteroplatytrygon violacea 1개체(전장 1,058 mm)가 2017년 7월 6일 제주도 북서쪽 연안에서 처음으로 유자망에 의해 채집되었다. 이 종은 넓은 주둥이를 갖는 점, 5쌍의 아가미구멍이 있는 점, 꼬리에 1개의 가시가 있는 점, 꼬리의 배쪽에 있는 피습이 꼬리끝까지 이르지 않는 점, 꼬리의 등쪽에는 피습이 없는 점, 그리고 배의 표면은 전체적으로 어두운 자주색을 띠는 것이 특징이다. 이러한 형태적 특징에 따라서 이 종을 P. violacea로 동정하였으며, 표본의 COI 유전자 염기서열을 이용하여 종동정을 확인하였다. 이 미기록종은 배 표면의 체색으로 인해 기존에 붙여진 이름에 따라 종명과 속명을 각각 "보라색가오리"와 "보라색가오리속"으로 제안한다.

Morphological, molecular, and chromosomal identification of dwarf haploid parthenosporophytes of Tauya basicrassa (Phaeophyceae, Laminariales) from the Sea of Okhotsk

  • Morphological, molecular and chromosomal studies were carried out on Tauya basicrassa, an endemic kelp species distributed on the northern continental coast of the Sea of Okhotsk in Russia. The sporophytes of T. basicrassa grow up to 3-6 m long, 1.8-2.2 m wide, and 6.5-7 kg wet weight. The thallus has a blade with very thick narrow basal portion and thinner and much broader upper portion, which usually splits into 3 bullated lobes. A dwarf laminariacean alga, which did not show any morphological similarity to the other species of the order Laminariales, was found from the same locality. The blade of this alga is thin and soft, reached 26-34 cm long and 6-6.5 cm wide and had 4 longitudinal rows of bullations that covered the entire blade. Molecular analysis showed that the dwarf alga has 100% sequence identity in plastid-encoded RuBisCo spacer, mitochondrial cytochrome c oxidase subunit 1 and nuclear-encoded rDNA genes with normal sporophytes of T. basicrassa, indicating that they are different life forms of the same species. Fluorescent DAPI staining showed that the nucleus in the normal sporophyte was 50-65% larger than those of the dwarf ones. Chromosome count using acetocarmine staining showed n = ca. 20 for the normal sporophytes of T. basicrassa and n = ca. 10 for the dwarf one. These results suggest that the dwarf thallus is a haploid parthenosporophyte of T. basicrassa, which developed in nature. This is the first evidence of parthenosporophytes of the laminariacean algae occurring naturally in the field.

Morphological and Molecular Diagnosis of Necator americanus and Ancylostoma ceylanicum Recovered from Villagers in Northern Cambodia

  • Human hookworm infections caused by adult Ancylostoma spp. and Necator americanus are one of the most important tropical diseases. We performed a survey of intestinal helminths using the Kato-Katz fecal examination technique targeting 1,156 villagers residing in 2 northern provinces (Preah Vihear and Stung Treng) of Cambodia in 2018. The results revealed a high overall egg positive rate of intestinal helminths (61.9%), and the egg positive rate of hookworms was 11.6%. Nine of the hookworm egg positive cases in Preah Vihear Province were treated with 5-10 mg/kg pyrantel pamoate followed by purging with magnesium salts, and a total of 65 adult hookworms were expelled in diarrheic stools. The adult hookworms were analyzed morphologically and molecularly to confirm the species. The morphologies of the buccal cavity and dorsal rays on the costa were observed with a light microscope, and the nucleotide sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene were analyzed. The majority of the hookworm adults (90.7%) were N. americanus, whereas the remaining 9.3% were Ancylostoma ceylanicum, a rare hookworm species infecting humans. The results revealed a high prevalence of hookworm infections among people in a northern part of Cambodia, suggesting the necessity of a sustained survey combined with control measures against hookworm infections.

  • 2020년 5월 제주도 남동부 해역에서 봉고네트로 성대과 자어 1개체(전장 5.14 mm)가 채집되었다. 본종은 mtDNA COI 염기서열 분석을 통해 우리나라에서 처음 보고되는 달재속 1 미기록종, Lepidotrigla longifaciata로 확인되었다. 본종의 전기자어의 형태적 특징은 긴 주둥이, 큰 입, 부채 모양의 큰 가슴지느러미를 가지며, 흑색소포는 복강과 목덜미에만 관찰되었다. 우리나라에서 처음 발견된 본종의 신국명으로 '긴머리 달재'를 제안한다.

Molecular Identification of Diphyllobothrium latum from a Pediatric Case in Taiwan

  • Human diphyllobothriasis is a parasitic disease caused by ingestion of larvae (plerocercoids) in raw or undercooked fish and commonly found in temperate areas. Rare cases were reported in tropical or subtropical areas especially in children. The first documented case of pediatric diphyllobothriasis in Taiwan had been reported 11 years ago. Here, we report another 8-year-old girl case who presented with a live noodle-like worm hanging down from her anus, with no other detectable symptoms. We pulled the worm out and found the strobila being 260 cm in length. Examination of gravid proglottids showed that they were wider than their lengths, containing an ovoid cirrus sac in the anterior side and the rosette-shaped uterus. Eggs extracted from the uterus were ovoid and operculated. Diphyllobothrium latum was confirmed by molecular analysis of the mitochondrial DNA cytochrome c oxidase subunit 1 (cox1) gene. The girl was treated with a single oral dose of praziquantel, and no eggs or proglottids were observed from her stool in the subsequent 3 months. The reemergence of human diphyllobothriasis in non-endemic countries is probably due to prevalent habit of eating imported raw fish from endemic areas. This pediatric case raised our concern that human diphyllobothriasis is likely underestimated because of unremarkable symptoms.

  • We presented detailed morphological descriptions of the eggs, larvae and juvenile of Acanthaphritis unoorum based on specimens collected with bongo nets from Korean waters during the period May 2017-July 2018. We collected 18 individuals including eggs (n= 4, 0.77-0.85 mm in egg diameter), preflexion larvae (n= 6, 4.11-6.31 mm in standard length, SL), flexion larvae (n= 4, 6.60-7.82 mm SL), postflexion larvae (n= 3, 8.94-13.46 mm SL), and one juvenile (n= 1, 14.67 mm SL). The mitochondrial (mt) DNA 16S rRNA sequences of the eggs, and the cytochrome c oxidase subunit I (COI) sequences of the larvae were identical to those of A. unoorum adults (genetic distances <0.01). The A. unoorum larvae and the juvenile that we collected were morphologically similar to those of Dactylopsaron dimorphicum, but the A. unoorum specimens were readily distinguishable by the presence of lateral melanophores. This is the first record of A. unoorum in Korean waters. We propose a new Korean name for A. unoorum: "O-ri-bu-ri-nuntung-i".

Genetic Diversity of Hard Ticks (Acari: Ixodidae) in the South and East Regions of Kazakhstan and Northwestern China

  • To date, there is no report on the genetic diversity of ticks in these regions. A total of 370 representative ticks from the south and east regions of Kazakhstan (SERK) and Xinjiang Uygur Autonomous Region (XUAR) were selected for molecular comparison. A fragment of the mitochondrial cytochrome c oxidase subunit I (cox1) gene, ranging from 631 bp to 889 bp, was used to analyze genetic diversity among these ticks. Phylogenetic analyses indicated 7 tick species including Hyalomma asiaticum, Hyalomma detritum, Hyalomma anatolicum, Dermacentor marginatus, Rhipicephalus sanguineus, Rhipicephalus turanicus and Haemaphysalis erinacei from the SERK clustered together with conspecific ticks from the XUAR. The network diagram of haplotypes showed that i) Hy. asiaticum from Almaty and Kyzylorda Oblasts together with that from Yuli County of XUAR constituted haplogroup H-2, and the lineage from Chimkent City of South Kazakhstan was newly evolved; and ii) the R. turanicus ticks sampled in Israel, Almaty, South Kazakhstan, Usu City, Ulugqat and Baicheng Counties of XUAR were derivated from an old lineage in Alataw City of XUAR. These findings indicate that: i) Hy. asiaticum, R. turanicus and Ha. erinacei shared genetic similarities between the SERK and XUAR; and ii) Hy. marginatum and D. reticulatus show differences in their evolution.

한국의 희귀 나비가오리[Gymnura japonica (나비가오리과, 연골어강)]의 분류학적 재검토 (Taxonomic Review of a Rare Butterfly Ray Gymnura japonica (Gymnuridae, Chondrichthyes), in Korea)

  • We collected a total of four butterfly ray specimens (Gymnura japonica, 213.4-695.0 mm in total length) in Korea from 2016 to 2021 and investigated their morphological and molecular characteristics in order to clarify their taxonomic status. These features are summarized as follows. Disc lozenge-shaped, 1.8-2.0 times broader than long. Tail very short, post-cloaca length 23.9-28.2% in disc width. Snout short, no rostral cartilage. Clasper short, no hook. Dorsal surface uniform yellow or brownish grey, with or without rounded light yellow spots. An analysis of 434 base-pair sequences of mitochondrial DNA cytochrome c oxidase subunit I showed that all four specimens corresponded to G. japonica from Japan (Kimura-2-parameter distance = 0-0.2%), suggesting that the color patterns found may be due to intraspecific color variation. G. japonica resembles Gymnura poecilura but differs in that it has a shorter tail length to disc width (23.9-28.2% in G. japonica vs. 40.1-48.3% in G. poecilura). This study revealed that G. japonica occurred in areas affected by the Tsushima Warm Current, tentatively suggesting that G. japonica may be an indicator species for monitoring marine ecosystem changes due to climate change.

  • 2020년 11월 한국 제주도 남부에서 봉고네트로 채집된 놀래기과 고생놀래기속 후기자어 1개체를 형태 분석한 결과, Thalassoma quinquevittatum로 확인되었다. 자치어 시기에 형태적으로 매우 유사한 색동놀래기 T. amblycephalum와 등쪽 경사, 눈의 모양, 흑색소포의 분포에서 구별된다. 미토콘드리아 DNA COI 염기서열 548 bp을 분석한 결과, T. quinquevittatum 성어와 0.2~0.5% 유전적 차이를 보였고, 같은 속의 T. amblycephalum, T. cupido, T. lunare, T. purpureum, T. lucasanum, T. lutescens와는 각각 12.3%, 13.7%, 13.2%, 12.1%, 13%, 11.2%의 유전적 차이를 보였다. 국내 처음 보고되는 본 종의 새로운 국명으로 "다섯줄색동놀래기"를 제안한다.

Identification of Heterodera glycines (Tylenchida; Heteroderidae) Using qPCR

  • The soybean cyst nematode, Heterodera glycines, is a major plant-parasitic nematode that has caused important economic losses to Korea's soybean production. Four species of cyst nematodes, H. schachtii, H. glycines, H. trifolii, and H. sojae, all belong to schachtii group are coexist in field soil in Korea. The rapid identification of the nematode is crucial for preventing crop damage and in decision making for controlling this nematode. This study aimed to develop a species-specific primer set for quantitative PCR (qPCR) assay of H. glycines. The specific primer set (HGF1 and HGR1) for H. glycines was designed based on the cytochrome c oxidase subunit I (COI) sequence of mitochondrial DNA. After optimization, it is possible to identify the H. glycines using a qPCR assay with DNA extracted from a single cyst and single second-stage juvenile (J2). The specificity was confirmed by the absence of SYBR fluorescent signals of three other Heterodera species. A serial dilution of DNA extracted from a single cyst was obtained for the sensitivity test. The result showed that the standard curve of the test had a highly significant linearity between DNA concentration and Ct value (R2 = 0.996, slope = -3.49) and that the detection limit concentration of DNA of the primer set was 10 pg of DNA per reaction. Our findings suggested that H. glycines could be distinguished from H. sojae and other Heterodera species when a qPCR assay is used with a specific primer set.
