A strain of Pseudomonas aeruginosa was isolated from the spoiled product and its characteristics on various formaldehyde-releasing preservatives were investigated. This strain, P. aeruginosa FR, could utilize 1.0% of imidazolidinyl urea and 0.2% of DMDM hydantoin as a sole carbon and nitrogen source in the minimal salts medium. With the growth of the strain in minimal salts medium containing imidazolidinyl urea, formic acid was initially accumulated according to the decrease of formaldehyde concentration. It was suggested that formaldehyde dehydrogenase was involved in this oxidation process and could catalyze formaldehyde, imidazolidinyl urea, DMDM hydantoin and quaternium-15, but not bronopol. MICs of this strain to each preservation were 0.03% in formaldehyde, 1.0% in imidazolidinyl urea, 0.2% in DMDM hydantoin, 0.2% in quaternium-15 and 0.1% of EDTA-2Na. But the MICs were diminished about ten times when 0.01% of EDTA-2Na was added to the preservative systems. In actual challenge test, the eyeliner and the pack which contained paraben and imidazolidinyl urea were not able to be protected from this strain, but when 0.05% EDTA-2Na was added the products were sufficiently preserved.
Bacterial growth inhibition by lead, zinc and cadmium was measured by using modified Tris minimal medium. The toxicity against Escherichia coli strain was in the order of zinc> cadmium> lead, and the Escherichia coli strain overexpressing iron-containing superoxide dismutase 2 of Streptomyces coelicolor A3(2) was found to have resistance to heavy metals.
Biodegradation of endocrine-disrupting bisphenol A was investigated with several white rot fungi (Irpex lacteus, Trametes versicolor, Ganoderma lucidum, Polyporellus brumalis, Pleurotus eryngii, Schizophyllum commune) isolated in Korea and two transformants of T. versicolor (strains MrP 1 and MrP 13). I. lacteus degraded 99.4% of 50 mg/l bisphenol A in 3 h incubation and 100% in 12 h incubation. which was the highest degradation rate among the fungal strains tested. T. versicolor degraded 98.2% of 50 mg/l bisphenol A in 12 h incubation. Unexpectedly, the transformant of the Mn-repressed peroxidase gene of T. versicolor, strain MrP 1, degraded 76.5% of 50 mg/l bisphenol A in 12 h incubation, which was a lower degradation rate than wild-type T. versicolor. The removal of bisphenol A by I. lacteus occurred mainly by biodegradation rather than adsorption. Optimum carbon sources for biodegradation of bisphenol A by I. lacteus were glucose and starch, and optimum nitrogen sources were yeast extract and tryptone in a minimal salts medium; however, bisphenol A degradation was higher in nutrient-rich YMG medium than that in a minimal salts medium. The initial degradation of endocrine disruptors was accompanied by the activities of manganese peroxidase and laccase in the culture of I. lacteus.
The influence of growth regulators (NAA and BA) and sucrose concentrations (0, 3, 5, 7, 9%) on in vitro rapid-propagation of virus-free sweet potato [Ipomoea batatas (L.) Lam.] was investigated with single-node or shoot-tip culture of two cultivars ('Matnami' and 'Shinhwangmi'). The survival rate and growth of shoot-tip explant was also investigated under the presence or absence of light (blue and red LED = 7:3, 150±5 μmol·m-2·s-1 PPFD) during minimal-growth in vitro conservation at 15℃. Vine length, vine diameter, fresh weight and dry weight were enhanced without callusing of explant in the MS medium supplemented with 0.2-0.5 mg·L-1 BA. The growth of single-node and shoot-tip explants were significantly enhanced with the increase of vine length, number of leaf, number of root, fresh weight, and dry weight in the solid medium containing 5% sucrose and 0.2 mg·L-1 BA. Vine elongation of shoot-tip explants were highest in the liquid medium containing 3% sucrose than the solid medium. The survival rate of minimal-growth in vitro conservation was 100% in 5 months under the presence of light (LED, 150±5 μmol·m-2·s-1 PPFD) at 15℃, but the explants in dark condition died in 3 months. The light was absolutely necessary for the in vitro conservation under minimal-growth conditions of virus-free sweet potato plantlets at 15℃, and the high density of explants (10 plantlets per Petri Dish) was increased the efficiency of mass conservation.

This experiment was performed to study the structure of esophageal mucousa and the histochemical properties of glycosaminoglycans of esophageal mucous cells in four teleostean species, i.e., Agramus agramus, Inimicus japonicus, Epinephelus chlorostigma and Helicolenus dactylopterus. To observe the structure of eosphageal mucosa, hematoxylin-eosin(H-E) staining was used. The glycosaminoglycans was stained with PAS(periodic acid schiff), alcain blue(AB) pH 2.5, AB pH 1.0, aldehyde fuchsin(AF) pH 1.7, AF pH 1.0, AB pH 2.5-PAS, AB pH 1.0-PAS and AF pH 1.7-alcian blue pH 2.5. As for the amount and histochemical composition of glycosaminglycans in Agramus agramus, most of mucous secreting columnar cell and mucous cells contain large and moderate amount of neutral glycosaminoglycans. A few of mucous cells having small amount of neutral glycosamino-glycans and minimal amount of sulfated glycosamino-glycans. In Inimicus japonicus, the esophageal mucous cells were composed of most of medium sized and large mucous cells with moderate amount of neutral glycosaminoglycanonly only, a few of medium sized and large mucous cells and most of small mucous cells with considerable amount of neutral glycosaminoglycans and minimal to small amount of nonsulfated glycosaminoglycans, and a feq of small mucous cells with small amount of neutral glycosaminoglycans and minimal amount of sulfated glycosaminoglycans. In Epinephelus chlorostigma, most of medium sized and large mucous cells were mixed small amount of neutral glycosaminoglycans with sulfated glycosaminoglycans, a few of which were contained with moderate or considerable amount of neutral glycosaminoglycans with sulfated glycosaminoglycans, while most of small mucous cells containing considerable amount of neutral glycosaminoglycans and small to moderate or considerable to minimal amount of nonsulfated glycosaminoglycans(sialomucin) a few of which containing a mixture of considerable amount of neutral glycosaminoglycans and considerable amount of nonsulfated glycosaminoglycans(sialomucin)only. In Helicolenus dactylopterus, most of medium sized and large mucous cells, mixing with moderate to considerable amount of neutral glycosaminoglycans and small to moderate amount of sulfated glycosaminoglycans, while most of small mucous cells with considerable amount of nonsulfated glycosaminoglycans (sialomucin), a few of which having a mixture of considerable amount of neutral glycosaminoglycans and considerable or small amount of nonsulfated glycosaminoglycans(sialomucin).
Peniphora sp. JS17, isolated from forest old tree, produced extracellular enzymes that decolorized human hair melanin. The JS17 strain had laccase and manganese peroxidase activity while it did not has lignin peroxidase activity. Batch culture indicated that the melanin decolorization activity of JS17 strain originated from laccase. The culture conditions to maximize the production of melanin bleaching enzymes from Peniophora sp. JS17 mycelia were investigated. Among the tested media for the laccase production, minimal medium (2% glucose, 0.2% malt extract, 0.1% $KH_2PO_4$, 0.4% $MgSO_4{\cdot}7H_2O$) showed the highest activity of laccase. Then, to optimize the culture condition for the laccase activity, the influence of various carbon and nitrogen sources was investigated in minimal medium. Among various carbon and nitrogen sources, 2% xylose and 0.4% tryptone showed the highest production of laccase, respectively. The enzyme was purified using $(NH_4)_2SO_4$ precipitation and Hitrap Q sepharose column, and the purified enzyme showed two isoenzymatic bands with molecular masses of about 70 kDa by SDS-PAGE. The melanin decolorization activity was 77% and 55% within 48 h in the presence of 1-hydroxybenzotriazole (HBT) and syringaldehyde, respectively, whereas only about 9% melanin decolorized in case of no mediator.
The ascomycete fungus Fusarium fujikuroi causes bakanae disease in rice and this disease has been reemerging in Korea. Other fungal species including F. graminearum and Magnaporthe oryzae are often associated with F. fujikuroi, hampering pure isolation of F. fujikuroi from rice. In this study, we modified a selective medium for F. fujikuroi as supplementing both pentachloronitrobenzene and hydrogen peroxide into minimal medium. This medium efficiently suppressed the vegetative growth of F. graminearum and M. oryzae, but did not significantly reduce F. fujikuroi growth, providing an efficient tool for isolating F. fujikuroi.
Bacillus sp. SB-007 was isolated from pea leaves harvested from the southwestern parts of South Korea through screening on a minimal medium containing 0.2% purified cutin for its ability to induce the cutinase production. However, no cutinase was produced when it was grown in a minimal medium containing 0.2% glucose. A proteomic approach was applied to separate and characterize these differentially secreted proteins. The expression level of 83 extracellular proteins of the cutinase-producing Bacillus sp. strain SB-007 incubated in a cutinase-induced medium increased significantly as compared with that cultured in a non cutinase-induced medium containing glucose. The extracellular proteome of Bacillus sp. SB-007 includes proteins from different functional classes, such as enzymes for the degradation of various macromolecules, proteins involved in energy metabolism, sporulation, transport/binding proteins and lipoproteins, stress inducible proteins, several cellular molecule biosynthetic pathways and catabolism, and some proteins with an as yet unknown function. In addition, the two protein spots showed little similarities with the known lipolytic enzymes in the database. These secreted proteome analysis results are expected to be useful in improving the Bacillus strains for the production of industrial cutinases.
Optimal conditions for the biodegradation of endocrine-disrupting bisphenol A (BPA) were examined for the white rot fungus Trametes versicolor isolated in Korea. T. versicolor degraded 100% of 50 mg/L bisphenol A during 12 hr in yeast extract-malt extract-glucose (YMG) medium. When BPA was added to the 5-day preincubated fungal culture in YMG medium, all BPA was removed in 2 hr. T. versicolor could efficiently degrade BPA at $35^{\circ}C$, pH 6 in YMG medium. T. versicolor could more easily remove BPA of $1{\sim}25\;mg/L$ than that of higher concentrations ($50{\sim}100\;mg/L$) in YMG medium. T. versicolor degraded 100% of 50 mg/L BPA for 36 h in a minimal medium, which is lower degradation rate than that in YMG medium. Optimal conditions for BPA biodegradation in the minimal medium were similar to those in YMG medium. When BPA (50 mg/L) was added into domestic wastewater, it could be completely removed by T. versicolor. During the biodegradation of BPA by T. versicolor in YMG medium, its estrogenic activity decreased.
Bacillus licheniformis was grown in minimal nutrient medium containing 1% (w/v) of distillers dried grain with soluble (DDGS), palm kernel meal (PKM), wheat bran (WB) or copra meal (CM), and the enzyme activity of endoglucanase, ${\beta}$-glucosidase, xylanase and reducing sugars was measured to investigate a possibility of using cost-effective agricultural residues in producing cellulolytic and hemicellulolytic enzymes. The CM gave the highest endoglucanase activity of 0.68 units/mL among added substrates at 48 h. CM yielded the highest titres of 0.58 units/ml of ${\beta}$-glucosidase, compared to 0.33, 0.23, and 0.16 units/mL by PKM, WB, and DDGS, respectively, at 72 h. Xylanase production was the highest (0.34 units/mL) when CM was added. The supernatant from fermentation of CM had the highest reducing sugars than other additional substrates at all intervals (0.10, 0.12, 0.10, and 0.11 mg/mL respectively). It is concluded that Bacillus licheniformis is capable of producing multiple cellulo- and hemicellololytic enzymes for bioethanol production using cost-effective agricultural residues, especially CM, as a sole nutrient source.
