Sulforaphane is Superior to Glucoraphanin in Modulating Carcinogen-Metabolising Enzymes in Hep G2 Cells

  • Glucoraphanin is the main glucosinolate found in broccoli and other cruciferous vegetables (Brassicaceae). The objective of the study was to evaluate whether glucoraphanin and its breakdown product sulforaphane, are potent modulators of various phase I and phase II enzymes involved in carcinogen-metabolising enzyme systems in vitro. The glucosinolate glucoraphanin was isolated from cruciferous vegetables and exposed to human hepatoma cell line HepG2 at various concentrations (0-25 ${\mu}M$) for 24 hours. Glucoraphanin at higher concentration (25 ${\mu}M$) decreased dealkylation of methoxyresorufin, a marker for cytochrome P4501 activity; supplementation of the incubation medium with myrosinase (0.018 U), the enzyme that converts glucosinolate to its corresponding isothiocyanate, showed minimal induction in this enzyme activity at concentration 10 ${\mu}M$. Quinone reductase and glutathione S-transferase activities were unaffected by this glucosinolate; however, supplementation of the incubation medium with myrosinase elevated quinone reductase activity. It may be inferred that the breakdown product of glucoraphanin, in this case sulforaphane, is superior than its precursor in modulating carcinogen-metabolising enzyme systems in vitro and this is likely to impact on the chemopreventive activity linked to cruciferous vegetable consumption.

Intraspecific Protoplast Fusion of Citric Acid Producer, Candida lipolytica (구연산 생성 Candida lipolytica의 원형질체 융합)

  • In order to develope a protoplast fusion system for citric acid and SCP producing Candida lipolytica, the optimal conditions for the formation and regeneration of protoplast were examined and the protoplast fusion was performed. At the optimal conditions of growth phase and Zymolyase treatment, frequencies of protoplast formation were 98%. Approximately 20-30% of protoplasts were regenerated on the regeneration minimal medium containing 3% agar and 30mM $CaCl_2$ with the overlay of the same medium. The fusion frequencies, 4-5${\pm}$10$^{-4}$, were accomplished by the treatment of two nutritionally complementary auxotrophic protoplasts, L-14 ($lys^-$) and T-24 (X$30^-$), with 30% PEG 6000 containing 100mM $CaCl_2$ at $30^{\circ}C$ for 20 minutes.

Ablation rate study using short pulsed laser subjected to Alumina medium (알루미나 세라믹 소재의 초단파 레이저 어블레이션량 연구)

  • In this paper, ablation rate of $Al_2O_3$ ceramics by femtosecond laser fluence is derived with experimental method. The automatic three axis linear stage makes laser optics to move with high spatial resolution. With 10 times objective lens, minimal pattern width of $Al_2O_3$ is measured in the focal plane. Ablated surface area is shown as linear tendency increasing number of machining times with various laser power conditions. Machining times is most sensitive condition to control $Al_2O_3$ pattern width. Also, the linear increment of pattern width with laser power change is investigated. In high machining speed, the ablation volume rate is more linear with fluence because pulse overlap is minimized in this condition. Thermal effect to surrounding medium can be minimized and clean laser process without melting zone is possible in high machining speed. Ablation volume rate decelerates as increasing machining times and multiple machining times should be considered to achieve proper ablation width and depth.

  • Certain metabolic aspects of halothane's cardiac depressant action on the contractility of the myocardium were elucidated from a sudy of the effect of pyruvate on halothane-depressed rat atria. Approximately 6 mg% halothane was required to maintain a 50% depression of the contractility of rat atria suspended in a modified Krebs-Ringer bicarbonate glucose medium, pH 7.4, $30^{\circ}C$ for a 2 hr. period. Pyruvate was found to restore partially the contractility of halothane-depressed atria. The maximally effective concentration of pyruvate was 2.5 mM. There was minimal pyruvate effect on the force of contraction of control atria. The effect of pyruvate on halothane-depressed atria was shown to be due to the pyruvate and not the sodium ion of the sodium pyruvate. Pyruvate was found to produce no increase in the contractility of atria depressed by hypertonic medium, but caused a further depression. Selected aspects regarding the action of halothane on glucose metabolism in myocardial cells are discussed. The results are consistent with the hypothesis that at least a part of the negative inotropic action of halothane is due to an inhibition of glucose uptake or utilization in the glycolytic pathway.

Isolation and Phosphate-Solubilizing Characteristics of PSM, Aeromonas hydrophila DA33

  • bacterium having high abilities to solubilize in-organic phosphate was isolated from cultivated soils. The strain was identified as Aeromonas hydrophila DA33, based on the physiological and biochemical properties. The optimum temperature and initial pH to solubilize insoluble phosphate in sucrose minimal medium were 3$0^{\circ}C$ and pH 5.0, respectively. In these conditions, phosphate-solubilizing activities of the strain against two types of insoluble phosphate were quantitatively determined. When glucose was used for carborn source, the strain had a marked mineral phospahte solubilizing activity. Inorganic phospahte solubilization was directly related to the pH drop by the strain. Analysis of the culture medium confirmed the production of gluconic acid as the main organic acid released by Aeromonas hydrophila DA33.

  • In recent years, researchers have put a considerable effort to decrease the emission of harmful gaseous pollutants to the atmosphere. In order to remove simultaneously $SO_2$ and $NO_X$ from the flue gas of small and medium-sized ship, we designed minimal wet scrubber inside a compact multistage modular system. In this study we proceed experiment of elemental technology at each stage of the scrubber. The each stage is oxidation of NO which is the main component of $NO_X$, and removal of $SO_2$, respectively. $NaClO_2$ was used to oxidize NO gas, and NaOH was used to remove $SO_2$ gas. The maximum NO conversion efficiency and the $SO_2$ removal efficiency are both indicate 100%.

Improved Production of Curdlan with Concentrated Cells of Agrobacterium sp.

  • The addition of a limited concentration of yeast extract to a minimal salt medium (MSM) enhanced cell growth and increased the production of curdlan whereas nitrogen-limitation was found to be essential for the higher production of curdlan by Agrobacterium sp. ATCC 31749. As the amount of the inoculum increased, the cell growth as well as the production of curdlan also increased in the MSM without a nitrogen source. The cell growth and production of curdlan increased as the initial pH of the medium decreased as low as 5.0. The conversion rate and concentration of curdlan from 2% (w/v) glucose in the MSM with concentrated cells under nitrogen deletion was 67% and 13.4 g/L, respectively. The highest conversion rate of curdlan under the conditions optimized in this study was 71% when the glucose concentrations was 1% (w/v).

  • Formation of conidium, an asexual organ on the hyphae, was examined from ten species of Pleurotus. Conidiospores of them were distinguished into two types of spores; an elliptical and a globose spores. Dikaryotic hyphae of ten species and monokaryotic hyphae of three species were observed to produce conidiospore. Conidia were observed on the hyphae grown on mushroom complete agar medium but were not on mushroom minimal agar medium. Two aconidial mutants were obtained by the ultraviolet irradiation.

Cloning and Expression Analysis of Phenylalanine Ammonia-Lyase Gene in the Mycelium and Fruit Body of the Edible Mushroom Flammulina velutipes

  • Phenylalanine ammonia-lyase (PAL) gene is known to be expressed in plants, and is involved in the differentiation, growth and synthesis of secondary metabolites. However, its expression in fungi remains to be explored. To understand its expression in mushroom fungi, the PAL gene of the edible mushroom Flammulina velutipes (Fvpal) was cloned and characterized. The cloned Fvpal consists of 2,175 bp, coding for a polypeptide containing 724 amino acids and having 11 introns. The translated amino acid sequence of Fvpal shares a high identity (66%) with that of ectomycorrhizal fungus Tricholoma matsutake. Distinctively, the Fvpal expression in the mycelium was higher in minimal medium supplemented with L-tyrosine than with other aromatic amino acids. During cultivation of the mushroom on sawdust medium, Fvpal expression in the fruit body correspondingly increased as the mushroom grew. In the fruiting body, Fvpal was expressed more in the stipe than in the pileus. These results suggest that F. velutipes PAL activity differs in the different organs of the mushroom. Overall, this is first report to show that the PAL gene expression is associated with mushroom growth in fungi.

Optimal Culture Conditions for Production of Environment-Friendly Biosurfactant by Pseudomonas sp. EL-G527 (Pseudomonas sp. EL-G527에 의한 환경친화성 생물계면활성제의 생산최적조건)

  • A biosurfactant-producing microorganism was isolated from activated sludge by enrichment culture when grown on a minimal salt medium containing n-hexadecane as a sole carbon source. This microorganism was identified as Pseudomonas sp. and it was named Pseudomonas sp. EL-G527. It's optimal culture condition is 2% n-hexadecane, 0.2% NH$_4$NO$_3$, 0.3% KH$_2$PO$_4$, 0.3% $K_2$HPO$_4$, 0.02% MgSO$_4$ㆍ7$H_2O$, 0.0025% CaCk$_2$ㆍ6$H_2O$, 0.0015% FeSO$_4$ㆍ7$H_2O$ in 1$\ell$ distilled water and initial pH 7.0. Cultivation was initiated with a 2% inoculum obtained from starter cultures grown in 30 $m\ell$ of the same medium in 250 $m\ell$ flask. They were cultivated at 3$0^{\circ}C$ in reciprocal shaking incubator and the highest biosurfactant production was observed after 4 days.
