In this paper we propose a microfluidic chip that measures the mechanical stiffness of cell membrane using impedance measurement. The microfluidic chip is composed of PDMS channel and a glass substrate with electrode. The proposed device uses patch-clamp technique to capture and deform a target cell and measures impedance of deformed cells. We demonstrated that the impedance increased after the membrane stretched and blocked the channel.
In cases of the microfluidic channel, the electrokinetic influence on the transport behavior can be found. The externally applied body force originated from the electrostatic interaction between the nonlinear Poisson-Boltzmann field and the flow-induced electrical field is applied in the equation of motion. The electrostatic potential profile is computed a priori by applying the finite difference scheme, and an analytical solution to the Navier-Stokes equation of motion for slit-like microchannel is obtained via the Green's function. An explicit analytical expression for the induced electrokinetic potential is derived as functions of relevant physicochemical parameters. The effects of the electric double layer, the zeta potential of the solid surface, and the charge condition of the channel wall on the velocity profile as well as the electroviscous behavior are examined. With increases in either electric double layer or zeta potential, the average fluid velocity in the channel of same charge is entirely reduced, whereas the electroviscous effect becomes stronger. We observed an opposite behavior in the channel of opposite charge, where the attractive electrostatic interactions are presented.
In the detection of pathogenic microorganisms ATP-bioluminescence reaction is a fascinating method. ATP(adenosine triphosphate) is an energy source of all kinds of living organism and ATP-bioluminescence reaction uses this ATP. However, ATP exists not only in the cells but also outside the cells. Therefore ATP-bioluminescence reaction only with intracellular ATP is very important in pathogenic microorganism detection. Because of that reason we developed a microfluidic channel containing Dielectrophoretic zone which capture microorganisms and eliminating and washing extracellular ATP with ATP-degarading enzymes, adenosine phosphate deaminase and apyrase. Microorganisms are captured by pDEP force at the DEP electrode zone and only extracellular ATPs are washed and eliminated outside the zone.

We have designed, fabricated and tested an integrated microfluidic chip with a Planar Hall Effect (PHE) sensor. The sensor was constructed by sequentially sputtering Ta/NiFe/Cu/NiFe/IrMn/Ta onto glass. The microfluidic channel was fabricated with poly(dimethylsiloxane) (PDMS) using soft lithography. Magnetic nanoparticles suspended in hexadecane were used as ferrofluid, of which the saturation magnetisation was 3.4 emu/cc. Droplets of ferrofluid were generated in a T-junction of a microfluidic channel after hydrophilic modification of the PDMS. The size and interval of the droplets were regulated by pressure on the ferrofluid channel inlet. The PHE sensor detected the flowing droplets of ferrofluid, as expected from simulation results. The shape of the signal was dependent on both the distance of the magnetic droplet from the sensor and the droplet length. The sensor was able to detect a magnetic moment of $2{\times}10^{-10}$ emu at a distance of 10 ${\mu}m$. This study provides an enhanced understanding of the magnetic parameters of ferrofluid in a microfluidic channel using a PHE sensor and will be used for a sample inlet module inside of integrated magnetic lab-on-a-chip systems for the analysis of biomolecules.
열영동은 매질의 온도 구배에 의해 입자가 이동하는 현상이다. 본 논문에서는 미세유체 채널에서 입자의 열영동 현상에 대해서 논의한다. 흐름이 없는 비유동 채널에서 열원인 백금 와이어에 가해지는 전압에 비례해서 열영동에 의한 마이크로 입자의 이동이 더 크게 나타남을 확인하였다. 전압에 따른 백금 와이어 주변 온도 변화는 Callendar-van Dusen 식을 이용하여 예측하였다. 동일한 시스템에서 나노 입자의 열영동 현상을 관찰한 결과, 나노 입자도 마이크로 입자와 유사한 열영동 거동을 보임을 확인하였다. 마지막으로 Y 모양 미세유체 채널을 제작하고 백금 와이어 열원을 채널 내에 설치하여, 채널을 흐르는 현탁액 내의 입자의 열영동 현상을 구현하고, 이를 기반으로 현탁액의 흐름을 제어할 수 있음을 보인다.

Purpose: The development of an efficient in vitro cell culture device to process various cells would represent a major milestone in biological science and engineering. However, the current conventional macro-scale in vitro cell culture platforms are limited in their capacity for detailed analysis and determination of cellular behavior in complex environments. This paper describes a microfluidic-based culture device that allows accurate control of parameters of physical cues such as pressure. Methods: A microfluidic device, as a model microbioreactor, was designed and fabricated to culture Saccharomyces cerevisiae and Chlamydomonas reinhardtii under various conditions of physical pressure stimulus. This device was compatible with live-cell imaging and allowed quantitative analysis of physical cue-induced behavior in yeast and microalgae. Results: A simple microfluidic-based in vitro cell culture device containing a cell culture channel and an air channel was developed to investigate physical pressure stress-induced behavior in yeasts and microalgae. The shapes of Saccharomyces cerevisiae and Chlamydomonas reinhardtii could be controlled under compressive stress. The lipid production by Chlamydomonas reinhardtii was significantly enhanced by compressive stress in the microfluidic device when compared to cells cultured without compressive stress. Conclusions: This microfluidic-based in vitro cell culture device can be used as a tool for quantitative analysis of cellular behavior under complex physical and chemical conditions.

A simple, label-free microfluidic cell purification method is presented for separation of cancer cells by exploiting difference in cell adhesion. To maximize the adhesion difference, three types of polymeric nanostructures (50nm pillars, 50nm perpendicular and 50nm parallel lines with respect to the direction of flow) were fabricated using UV-assisted capillary moulding and included inside a polydimethylsiloxane (PDMS) microfluidic channel bonded onto glass substrate. The adhesion force of human breast epithelial cells (MCF10A) and human breast carcinoma (MCF7) was measured independently by injecting each cell line into the microfluidic device followed by culture for a period of time (e.g., one, two, and three hours). Then, the cells bound to the floor of a microfluidic channel were detached by increasing the flow rate of medium in a stepwise fashion. It was found that the adhesion force of MCF10A was always higher than that of MCF cells regardless of culture time and surface nanotopography at all flow rates, resulting in a label-free detection and separation of cancer cells. For the cell types used in our study, the optimum separation was found for 2 hours culture on 50nm parallel line pattern followed by flow-induced detachment at a flow rate of $300{\mu}l/min$.
Simple and highly efficient droplet merging method is proposed, which enables two nanoliter or picoliter droplets to merge regularly in a straight microchannel. Using a cross channel with inflows of one oil phase through the main channel and two water phases through the side channels, two droplets of different sizes can be generated alternatingly in accordance with flow rate difference of the water phases. It is shown that for a fixed oil phase flow rate, the flow rate of one water phase required for alternating droplet generation increases linearly with the flow rate of another water phase. By this method, the droplets are merged with 100 % efficiency without any additional driving forces.
Thermoplastic fusion bonding is widely used to seal polymer microfluidic devices and optimal bonding protocol is required to obtain a successful bonding, strong bonding force without channel deformation. Besides, UV modification of the PMMA (poly-methyl methacrylate) is commonly used for chemical or biological application before the bonding process. However, study of thermal bonding for the UV modified PMMA was not reported yet. Unlike pristine PMMA, the optimal bonding parameters of the UV modified PMMA were $103^{\circ}C$, 71 kPa, and 35 minutes. A very low aspect ratio micro channel (AR=1:100, $20{\mu}m$ depth and $2000{\mu}m$ width) was successfully bonded (over 95%, n>100). Moreover, thermal bonding of multi stack PMMA chips was successfully demonstrated in this study. The results may applicable to fabricate a complex 3 dimensional microchannel networks.
