Isolation and Characteristics of Bacteria Showing Biocontrol and Biofertilizing Activities (생물방제 및 생물비료 활성을 가지는 세균의 분리 및 특성)

  • To develop multifunctional microbial inoculant, microorganisms with antagonistic activity and biofertilizing activity were screened. Pantoea agglomerans and Bacillus megaterium from our laboratory culture collection, and strain MF12 from soil near poultry farm in Miryang were selected. On the basis of morphological, physiological studies and 16S rDNA sequence analysis, isolate MF12 was identified as the Bacillus pumilis. Three strains were studied for insoluble phosphate solubilization, indole-3-acetic acid (IAA) and siderophore production, ammonification ability, hydrolytic enzyme production and antifungal activity against phytopathogenic fungi. P. agglomerans did not produce any visible clear zone on agar plate containing 0.5% $Ca_3(PO_4){_2}$ as a sole phosphorus source. However, this strain could solubilize insoluble phosphate in liquid medium. All strains produced IAA ranged from $3{\sim}639{\mu}g/ml$ depending on culture time and had ammonification ability. Among three strains, only P. agglomerans produced siderophore. P. agglomerans produced pectinase and lipase, B. megaterium produced amylase, protease and lipase while B. pumilis produced protease and lipase. P. agglomerans showed antifungal activities against phytopathogenic fungi, Fusarium oxysporum and Colletotrichum gloeosporioides. B. pumilis showed antifungal activities against Botrytis cinerea, Sclerotinia sclerotiorum and Phythium ultimum.

Isolation, Identification, and Characterization of Ornithine-Producing Enterococcus faecalis OA18 from Kefir Grain (케피어그레인으로 제조한 요쿠르트로부터 Enterococcus faecalis OA18 균주의 분리 및 특성규명)

  • Lactic acid bacteria (LAB) OA18 was isolated from yogurt prepared by using Kefir Grain as a starter. The OA18 strain was a Gram-positive, cocci-type bacterium, and able to grow anaerobically with $CO_2$ production. The OA18 strain grew well on MRS broth supplemented with 50 mM arginine at $30-37^{\circ}C$ and pH of 7.0-9.0. The optimum temperature and pH for growth are $37^{\circ}C$ and pH 7.0. The isolate fermented ribose, D-glucose, cellobiose, D-trehalose, but not L-xylose, D-melibiose, and inositol. The 16S rRNA gene sequence of the isolate showed 99.8% homology with the Enterococcus faecalis 16S rRNA gene (Access no. AB012212). Based on the biochemical characteristics and 16S rRNA gene sequence analysis data, it was identified and named as E. faecalis OA18. The E. faecalis OA18 strain showed a high ornithine-producing capacity in the presence of arginine and also showed an antimicrobial activity against Streptomyces strains such as Streptomyces coelicolor subsp. Flavus, S. coeruleorubidus, S. coeruleoaurantiacus, S. coelicolor, S. coeruleoprunus. The cell growth of E. faecalis OA18 strain was maintained in MRS broth with a NaCl concentration of 0-7%.

Isolation and Characterization of Acetic Acid Bacteria for Producing "Makgeolli Seed-Vinegar" (막걸리 종초 제조에 적합한 초산균의 분리 및 발효특성)

  • Acetic acid bacteria strains were isolated from commercial natural vinegar. Ten isolated strains were identified using 16S rRNA gene sequencing data after evaluating the ethanol- and sulfur-tolerance. Eight of the strains isolated were identified as Acetobacter pasteurianus. A. pasteurianus JAC002, JAC005, and JAC008 strains, which showed a high ethanol tolerance, were selected for making "Makgeolli seed-vinegar". Rice wine vinegars were manufactured with the selected strains through fermentation, and their physicochemical properties and antimicrobial activities were evaluated. A. pasteurianus JAC002 strain showed the highest oxidation ability to acetic acid from ethanol on the twentieth day of fermentation, resulting in 4.21% total acidity, 3,791.77 mg% acetic acid content, and 2,931.78 mg% ethanol consumption content. Rice wine vinegar manufactured with the A. pasteurianus JAC002 strain showed increased antimicrobial activities against Staphylococcus aureus (KACC1927) and Escherichia coli (KACC10115). As a result, A. pasteurianus JAC002 strain was found to be the most suitable strain for "Makgeolli seed-vinegar".

Fibrinolytic, Immunostimulating, and Cytotoxic Activities of Microbial Strains Isolated from Kochujang (고추장 분리 균주의 혈전용해능, 면역활성능 및 세포독성 효과 조사)

  • This study was carried out to investigate the functional activities of microorganisms isolated from kochujang, such as fibrinolytic, immunostimulating, and cytotoxical activities, and to apply these microorganisms to kochujang products. Ninety-one microbial strains with proteolytic activity were selected from 294 strains isolated from traditional and commercial kochujang. Three strains (TPP 0014, TPP 6013, and TPP 6015) with high fibrinolytic activity were tested for their immunostimulating and cytotoxical activities. For the assessment of macrophage activation, cytokines such as tumor necrosis factor, $interleukin-1{\alpha}$ and nitrogen oxide were measured with the murine macrophage cell line RAW 264.7. In addition, the cytotoxical activities of the three strains were examined by MTT assay on the colon cancer cell line SNU-C4 and normal cell line CHO-K1. Using an API identifying kit, two of the microbial strains (TPP 0014 and TPP 6015) were identified as Bacillus stearothermophilus and the other strain (TPP 6013) was identified as B. amyloliquefacience.

Isolation of dhlA Gene Responsible for Degradation of 1, 2-dichloroethane from Metagenomic Library Derived from Daecheong Reservoir (대청호로부터 제작한 메타지놈 라이브러리에서 1, 2-dichloroethane의 분해에 관여하는 dhlA 유전자의 분리)

  • Traditional screening techniques have missed up to 99% of microbial resources existing in the nature. Strategies of direct cloning of environmental DNAs comprising tine genetic blueprints of entire microbial metagenomes provide vastly more genetic information than is contained in the culturable. Therefore, one way to screening the useful gene in a variety of environments is the construction of metagenomic DNA library. In this study, the water samples were collected from Daecheong Reservoir in the mid Korea, and analyzed by T-RFLP to examine the diversity of the microbial communities. The crude DNAs were extracted by SDS-based freezing-thawing method and then further purified using an $UltraClean^{TM}kit$ (MoBio, USA). The metagenomic libraries were constructed with the DNAs partially digested with EcoRI, BamHI, and SacII in Escherichia coli DH10B using the pBACe3.6 vector. About 14.0 Mb of metagenomic libraries were obtained with average inserts 13 ${\sim}$ 15 kb in size. The genes responsible for degradation of 1, 2-dichloroethane (1, 2-DCE) via hydrolytic dehalogenation were identified from the metagenomic libraries by colony hybridization. The 1, 2-dichloroethane dehalogenase gene (dhlA) was cloned and its nucleotide sequence was analyzed. The activity of the 1, 2-DCE dehalogenase was highly expressed to the substrate. These results indicated that the dhlA gene identified from the metagenomes derived from Deacheong Reservoir might be useful to develop a potent strain for degradation of 1, 2-DCE.

Ecological Characteristics of Actinomycetes from Mercury and Chrome Polluted Soil (수은, 6가크롬 오염토양으로부터 분리된 방선균군의 생태학적 특성)

  • Ecological characteristics of microbial populations inhabiting heavy metal polluted soil were investigated. The samples were collected from 293 sites around an factory and industry at Gyeoungsangbuk-do. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, $Cr^{6+}$, CN), seven sites have been seriously contaminated by mercury and chrome. A quantitative evaluation of microbial populations in mercury and chrome contaminated soil was examined by using plate count method. Bacterial numbers in polluted soil samples ranged from $7.4X10^5\;to\;9.3X10^7\;cfu\;g^{-1}$, about $10\sim100$ fold less than the count for the unpolluted soil. Moulds were not detected in chrome polluted soil. The log values of actinomycetes of each contaminated soil samples were log ranged from 6.18 to 7.52. The ratio of actinomycetes was similar to unpolluted soil. The investigation showed actinomycetes to be the major microbial population inhabiting the mercury and chrome polluted soil. Thirty-one isolates among the total isolates were examined for antibacterial activity. These isolates were identified based on a phylogenetic analysis using 16S rRNA gene nucleotide sequences, they were categorized in three major phylogenetic groups, belong to the Streptomyces (6 strains), Saccharopolyspora (3 strains), Nocardiodes (1 strain). On the phylogenetic tree, the clade consisting of five isolates were distantly related to all of the established Streptomycetes genera, indicating the possibility as members of new species.

Media Optimization of Corynebacterium glutamicum for Succinate Production Under Oxygen-Deprived Condition

  • Corynebacterium glutamicum is one of the well-studied industrial strain that is used for the production of nucleotides and amino acids. Recently, it has also been studied as a possible producer of organic acids such as succinic acid, based on its ability to produce organic acids under an oxygen deprivation condition. In this study, we conducted the optimization of medium components for improved succinate production from C. glutamicum under an oxygen deprivation condition by Plackett-Burman design and applied a response surface methodology. A Plackett-Burman design for ten factors such as glucose, ammonium sulfate, magnesium sulfate, potassium phosphate ($K_2HPO_4$ and $KH_2PO_4$), iron sulfate, manganese sulfate, biotin, thiamine, and sodium bicarbonate was applied to evaluate the effects on succinate production. Glucose, ammonium sulfate, magnesium sulfate, and dipotassium phosphate were found to have significant influence on succinate production, and the optimal concentrations of these four factors were sequentially investigated by the response surface methodology using a Box-Behnken design. The optimal medium components obtained for achieving maximum concentration of succinic acid were as follows: glucose 10 g/l, magnesium sulfate 0.5 g/l, dipotassium phosphate ($K_2HPO_4$) 0.75 g/l, potassium dihydrogen phosphate ($KH_2PO_4$) 0.5 g/l, iron sulfate 6 mg/l, manganese sulfate 4.2 mg/l, biotin 0.2 mg/l, thiamine 0.2 mg/l, and sodium bicarbonate 100 mM. The parameters that differed from a normal BT medium were glucose changed from 40 g/l to 10 g/l, dipotassium phosphate ($K_2HPO_4$) 0.5 g/l changed to 0.75 g/l, and ammonium sulfate ($(NH_4)_2SO_4$) 7 g/l changed to 0 g/l. Under these conditions, the final succinic acid concentration was 16.3 mM, which is about 1.46 fold higher than the original medium (11.1 mM) at 24 h. This work showed the improvement of succinate production by a simple change of media components deduced from sequential optimization.

Condition of ex situ Bioremediation of Polycyclic Aromatic Hydrocarbons in Marine Sediments (해양퇴적토내 다환방향족탄화수소 생분해 증진 조건 연구)

  • Polycyclic aromatic hydrocarbons (PAHs) are a kind of toxic environmental pollutants and has been accumulated usually in marine sediments. Due to their potential hazardous to human, removal of PAHs from environments has been great concern. In the present study, the effect of microbial inoculation and the supplementation of mixed form cyclodextrin (M-CD) was assessed in the pre-sterilized or nonsterilized microcosms for optimizing operational conditions for ex situ bioremediation of sediments contaminated by PAHs. Activity of electron transport system (ETSA) was increased by the addition of M-CD regardless of inoculation of microorganisms in microcosms without sterilization. The degradation rate of PAHs in sterilized microcosms was app. 9-20% by the inoculation of single strain and 24-37% by the inoculation of microbial consortium supplemented with 1% M-CD, respectively. The degradation was not observed in microcosms without sterilization under the same conditions. The proportion of inoculated microorganisms also decreased in nonsterilized microcosms. Signals of inoculated bacteria were decreased to detection limit after 2 days in the microcosms without M-CD. In conclusion, microbial inoculation with appropriate carbon sources and removal of natural flora and grazers are required for the efficient ex situ bioremediation of sediments contaminated by PAHs in bioslurry reactor.

Comparison of Antifungal Activity, Plant Growth Promoting Activity, and Mineral-Solubilizing Ability of Bacillus sp. Isolated from Rhizosphere Soil and Root (근권 토양과 뿌리로부터 분리된 Bacillus sp.의 항진균 활성, 식물 생장 촉진 활성 및 미네랄 가용화능 비교)

  • The purpose of this study was to evaluate the antifungal activity, plant-growth-promoting activity, and mineral solubilization ability of 10 species of phytopathogenic fungi to select a Bacillus sp. from rhizosphere soils and roots that can be used as a microbial agent. The antifungal activity for phytopathogenic fungi varied based on the Bacillus sp. Among the selected strains, DDP4, DDP16, DDP148, SN56, and SN95 exhibited antifungal activity for nine or more species of phytopathogenic fungi. Regarding nitrogen-fixation ability, all Bacillus sp. showed similar levels of activity, and siderophore production ability was relatively high in ANG42 and DDP427. The indole-3-acetic acid production abilities were in the range of 1.83-67.91 ㎍/ml, with variations in activity based on the Bacillus sp. One strain with a high activity was selected from each species, and their mineral solubilization abilities were examined. Most Bacillus sp. could solubilize phosphoric acid and calcium carbonate, and DDP148 and SN56 could solubilize silicon and zinc, respectively. These results suggested that Bacillus sp. can be considered potential multi-purpose microbial agents for plant growth promotion and disease prevention.

Biodegradation of VOC Mixtures using a Bioactive Foam Reactor II: Analysis of Microbial Community (계면활성제 미생물반응기의(혼합 VOCs) 생분해 II: 미생물의 군집해석)

  • A toluene-degrading bacterial strain was isolated from a mixed culture that was maintained using toluene as a sole carbon and energy source. The isolated bacterium was classified as Pseudomonas sp. TBD4 based on the close relationship to bacteria belonging to this genus. A bottle study to determine biodegradation rates of individual aromatic compounds showed that the biodegradation was faster in the order of toluene, benzene, styrene, and p-xylene. However, when various mixtures were subjected to TDB4, styrene was degraded at the highest rate, indicating that both toluene and p-xylene could stimulate the degradation of other substrates whereas styrene played as an inhibitor. In addition, the mixed culture and TDB4 were inoculated to the bioactive foam reactor (BFR), and the reactor performance and the corresponding change of microbial community were monitored using the fluorescent in situ hybridization (FISH) method. When an inlet concentration of the VOC mixture increased to greater than 250 ppm, the overall removal efficiency dropped significantly. The FISH measurement demonstrated that the ratio of TDB4 to the total bacteria also decreased to less than 20% along with the decline in removal efficiency in the BFR. As a result, the periodic addition of the pre-grown TDB4 might have been beneficial to achieve a stable performance in the BFR operated over an extended period.
