Characteristics of Digestion Dynamics of Rice and Oat Straw Relating to Microbial Digestion in the Rumen of Sheep Given High-Concentrate Diets

  • Rumination behavior, in vivo digestibility of cell wall constituents, particle size reduction in the rumen, and retention time in the digestive tract of sheep were examined using rice and oat straw as roughage sources. The in sacco digestibility, rumen fermentation, and microbial population and internal adenosine 5-triphosphate (ATP) content were also determined under feeding conditions of high-roughage and high-concentrate diets. Chewing number and time in rumination behavior were higher with rice straw than with oat straw, while the in sacco and in vivo DMD of rice straw were consistently lower than those of oat straw. Rice straw also showed higher frequency of thinner and longer particles in the rumen contents and lower retention time in the whole digestive tract as compared to those of oat straw. Rice straw was more effective to maintain the ruminal pH than oat straw, being reflected in higher internal ATP content of large-type protozoa on the high- concentrate diet. Changes in the ruminal microflora by shifting from the low- to the high- concentrate diet were also different between rice and oat straw.

Microencapsulation of Mitragyna leaf extracts to be used as a bioactive compound source to enhance in vitro fermentation characteristics and microbial dynamics

  • Objective: Mitragyna speciosa Korth is traditionally used in Thailand. They have a high level of antioxidant capacities and bioactive compounds, the potential to modulate rumen fermentation and decrease methane production. The aim of the study was to investigate the different levels of microencapsulated-Mitragyna leaves extracts (MMLE) supplementation on nutrient degradability, rumen ecology, microbial dynamics, and methane production in an in vitro study. Methods: A completely randomized design was used to assign the experimental treatments, MMLE was supplemented at 0%, 4%, 6%, and 8% of the total dry matter (DM) substrate. Results: The addition of MMLE significantly increased in vitro dry matter degradability both at 12, 24, and 48 h, while ammonia-nitrogen (NH3-N) concentration was improved with MMLE supplementation. The MMLE had the greatest propionate and total volatile fatty acid production when added with 6% of total DM substrate, while decreased the methane production (12, 24, and 48 h). Furthermore, the microbial population of cellulolytic bacteria and Butyrivibrio fibrisolvens were increased, whilst Methanobacteriales was decreased with MMLE feeding. Conclusion: The results indicated that MMLE could be a potential alternative plant-based bioactive compound supplement to be used as ruminant feed additives.

Analysis of a Microbial Community Denitrying Nitrate to Nitrogen Gas in a Nitrate-Contaminated Aquifer

  • Little study has been published specifically addressing the dynamics of nitrate reducing bacteria (NBR) during the bioremediation of nitrate-contaminated aquifer. In our previous study we successfully quantified fumarate-enhanced microbial nitrate reduction rate in a nitrate-contaminated aquifer by using a series of single-well push-pull tests (PPTs). In this study we analyzed the suspended population during PPTs. To monitor changes in the microbial community, PCR amplification of 16S rDNA genes and denaturing gradient gel electrophoresis (DGGE) were used to study the dynamics of the bacterial community in detail. Before the stimulation of NBR, the dominant DGGE bands obtained by PCR were affiliated with V-Proteobacteria consisting of Acinetobacter spp. and Pseudomonas fluorescens. However, as NBR biostimulation proceeded, the dominant patterns of DGGE bands changed, and they were affiliated with Azoarcus denitrificans Td-3 and Flavobacterium xanthum. Azoarcus denitrificans Td-3 is known to completely reduce nitrate to nitrogen gas. The series of single-well push-pull tests in this study should prove useful for conducting rapid, low-cost feasibility assessments for in situ denitrification and provide important information about which microorganisms play a key role in bioremediation of a nitrate contaminated aquifer.

식품중 미생물 위해성평가 방법론 연구 (Study on the Methodology of the Microbial Risk Assessment in Food)

  • 최근 국내에서는 Escherichia coli O157:H7, Listeria monocytogenes등의 미생물에 대한 건강위해성이 강조되면서 미생물 위해성평가에 대한 필요성이 제기되고 있고, U.S.FPA, U.S.DA, FAO/WHO를 비롯한 국제기구 및 외국 유관기관들에서도 미생물 위해성평가 방법을 식품관리에 활용하고자 방법론 연구에 주력하고 있다. 미생물 위험성은 화학물질과 달리 인체건강에 대한 영향이 즉각적이고, 심각하게 나타나 정량적인 위해성평가가 용이하지 않고 유해화학물질과는 다른 평가방법이 요구된다. 식품중 미생물의 위해성평가는 크게 4단계로 구분되는데, 미생물관련질환 추세파악 및 미생물 관련질병에 관한 역학조사 등을 활용하는 위험성확인 단계와 실제 식품원료, 식품가공, 수송, 포장단계 중 식품의 물리적, 화학적 조건에 따른 미생물 변화를 고려하여 식품중 미생물에 대한 노출을 정량화하는 노출평가 단계, 미생물의 용량에 따른 질병발생에 근거하여 용량-반응관계를 규명하는 용량-반응평가 단계, 규명된 모델을 활용하여 모든 평가결과를 통합함으로 위해 도치 예측과 불확실성 분석 등을 수행하는 위해도결정단계로 구성되어 있다. 미생물 용량-반응평가는 크게 비역치(Nonthreshold)와 역치(Threshold) 평가 방법론으로 구분되는데, 비역치 평가방법론은 단일 병원균이 감염을 일으킬 수 있다는 것과 감염을 일으킬 수 있는 확률이 독립적이라는 가정을 전제로 하고, 역치평가방법론은 미생물이 감염을 일으키기 위해서 각기 개별 역치가 존재하는데 어느 정도의 미생물수가 모여 서로 작용해야 독성유발물질을 만들어 낸다는 가정을 전제로 한다. 현재 받아들여지고 있는 비역치 모델로는 Exponential, Beta-poisson, Gompertz, Gamma-weibull 모델 등이 있으며, 역치모델로는 Log-normal, Log-logistic모델 등이 있다. 본 연구에서는 인체 volunteer자료를 활용하여 용량-반응자료를 입력하고 용량-반응자료를 토대로 적합한 수학적 모델을 찾아내어, 선별한 모델의 적합도 검정을 실시하는 방법론 연구를 실시하였으며, 노출평가 자료와 용량-반응평가 결과를 연계하여 위해도를 결정하는 과정에 대해 연구하였다 이 밖에도 모델(Food MicroModel)을 이용하여 식품의 염도, 수분활성도, 온도, pH등의 조건에 따른 미생물의 성장률, 사멸률 등 변화를 예측할 수 있는 방법론 연구를 통해 식품의 최적 보관 조건등을 찾아내는 방법을 습득하였다. 미생물 위해성평가는 외국에서도 아직 초기 연구단계에 있으며 현재로서 사후조사자료인 역학자료보다 건강한 성인남자를 대상으로 한 volunteer 자료를 우선적으로 활용하고 있으나 노약자나 민감그룹에 대한 실험은 현실적으로 불가능하여 동물실험을 이용한 평가방법을 연구중에 있다. 추후 연구방향으로는 국내 volunteer들을 대상으로 한 미생물별 용량-반응결과를 토대로 population sensitivity를 비교할 수 있는 기초자료를 확보함으로써 미생물에 대한 인구집단의 반응 민감성 차이를 비교하고 시료채취 후 즉각적인 실험실적 분석이 가능토록하여 정확한 인체노출평가를 수행함으로써 미생물 위해성평가방법론을 식품미생물관리에 적용하는 것이다.

송이 균환(菌環) 주변(周邊)의 토양미생물(土壤微生物)과 토양효소(土壤酵素)의 동태(動態) (Dynamics of Soil Microflora and Soil Enzymes around the Fairy-rings of Tricholoma matsutake)

  • 송이가 토양 생태계에 미치는 영향을 파악하기 위하여 균환 통과부, 자실체 발생부, 활성균환부 및 균환 전진예정부로 나누어 송이 균환의 발달에 따른 토양미생물과 토양효소의 동태를 조사하였다. 송이 균환부는 진균류와 방선균류의 수에서 일반 산림토양에 비하여 적었으며 균환 발달에 따라서도 차이가 있었다. 송이 균환이 점유하고 있는 자실체 발생부와 활성균환부는 토양 미생물의 양이 균환 전진예정부에 비하여 약 ${\frac{1}{3}}$이하이었으며, 상대적으로 송이 균이 우세한 지위를 차지하고 있는 것으로 여겨졌다. 탈수소효소 활성은 송이의 활성균환부와 균환 전진예정부간에 뚜렷한 차이를 나타내어 송이의 균근적인 특성이 명확히 드러났다. 송이 균환부 주변 토양의 탈수소효소 활성은 자실체 발생초기인 8월 말에 가장 높았던 반면, 인산가수분해효소는 3월에서 6월로 갈수록 높아졌다가 8월에는 감소하였지만 10월에 다시 높은 활성을 보였다. 이를 통하여 송이 자실체가 형성된 이후에도 송이 균의 활력이 충분히 유지되는 것으로 추론할 수 있었다.

Interactions between Entodinium caudatum and an amino acid-fermenting bacterial consortium: fermentation characteristics and protozoal population in vitro

  • Ruminal protozoa, especially entodiniomorphs, engulf other members of the rumen microbiome in large numbers; and they release oligopeptides and amino acids, which can be fermented to ammonia and volatile fatty acids (VFAs) by amino acid-fermenting bacteria (AAFB). Studies using defaunated (protozoa-free) sheep have demonstrated that ruminal protozoa considerably increase intraruminal nitrogen recycling but decrease nitrogen utilization efficiency in ruminants. However, direct interactions between ruminal protozoa and AAFB have not been demonstrated because of their inability to establish axenic cultures of any ruminal protozoan. Thus, this study was performed to evaluate the interaction between Entodinium caudatum, which is the most predominant rumen ciliate species, and an AAFB consortium in terms of feed degradation and ammonia production along with the microbial population shift of select bacterial species (Prevotella ruminicola, Clostridium aminophilum, and Peptostreptococcus anaerobius). From an Ent. caudatum culture that had been maintained by daily feeding and transfers every 3 or 4 days, the bacteria and methanogens loosely associated with Ent. caudatum cells were removed by filtration and washing. An AAFB consortium was established by repeated transfers and enrichment with casamino acids as the sole substrate. The cultures of Ent. caudatum alone (Ec) and AAFB alone (AAFB) and the co-culture of Ent. caudatum and AAFB (Ec + AAFB) were set up in three replicates and incubated at 39℃ for 72 h. The digestibility of dry matter (DM) and fiber (NDF), VFA profiles, ammonia concentrations, pH, and microscopic counts of Ent. caudatum were compared among the three cultures. The co-culture of AAFB and Ent. caudatum enhanced DM degradation, VFA production, and Ent. caudatum cell counts; conversely, it decreased acetate: propionate ratio although the total bacterial abundance was similar between Ec and the Ec + AAFB co-culture after 24 h incubation. The ammonia production and relative abundance of C. aminophilum and P. anaerobius did not differ between AAFB alone and the Ec + AAFB co-culture. Our results indicate that Ent. caudatum and AAFB could have a mutualistic interaction that benefited each other, but their interactions were complex and might not increase ammoniagenesis. Further research should examine how such interactions affect the population dynamics of AAFB.

Stability of Partial Nitrification and Microbial Population Dynamics in a Bioaugmented Membrane Bioreactor

  • Bioaugmentation of bioreactors focuses on the removal of numerous organics, with little attention typically paid to the maintenance of high and stable nitrite accumulation in partial nitrification. In this study, a bioaugmented membrane bioreactor (MBR) inoculated with enriched ammonia-oxidizing bacteria (AOB) was developed, and the effects of dissolved oxygen (DO) and temperature on the stability of partial nitrification and microbial community structure, in particular on the nitrifying community, were evaluated. The results showed that DO and temperature played the most important roles in the stability of partial nitrification in the bioaugmented MBR. The optimal operation conditions were found at 2-3 mgDO/l and $30^{\circ}C$, achieving 95% ammonia oxidization efficiency and nitrite ratio ($NO_2^-/{NO_x}^-$) of 0.95. High DO (5-6 mg/l) and low temperature ($20^{\circ}C$) had negative impacts on nitrite accumulation, leading to nitrite ratio drop to 0.6. However, the nitrite ratio achieved in the bioaugmented MBR was higher than that in most previous literatures. Denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH) were used to provide an insight into the microbial community. It showed that Nitrosomonas-like species as the only detected AOB remained predominant in the bioaugmented MBR all the time, and coexisted with numerous heterotrophic bacteria. The heterotrophic bacteria responsible for mineralizing soluble microbial products (SMP) produced by nitrifiers belonged to the Cytophaga-Flavobacterium-Bacteroides (CFB) group, and $\alpha$-, $\beta$-, and $\gamma$- Proteobacteria. The fraction of AOB ranging from 77% to 54% was much higher than that of nitrite-oxidizing bacteria (0.4-0.9%), which might be the primary cause for the high and stable nitrite accumulation in the bioaugmented MBR.

  • 토양으로 부터 분리한 Bacillus sp. WRD-l의 토마토 생육에 미치는 영향을 조사하기 위하여 미생물 배양액을 100배, 300배, 500배의 희석 비율에 따라 토마토재배지에 관주를 실시하여 토마토 생육 및 토양미생물상 변화를 관찰 하였다. 대조구로는 실제 미생물의 영향을 확인하기 위하여 Bacillus sp. WRD-1 배양액을 멸균하여 사용하였다. 토마토 생육은 전체적으로 100배> 300배> 500배> 대조구 순으로 희석 비율과 비례하였다. 시간별 토양미생물 수는 세균과 방선균의 수는 증가하는 경향을 보였으며, 곰팡이수는 감소하는 경향을 보였다. 이러한 결과로 토양활성개량미생물제로의 개발 가능성을 확인하였다.

Effects of Different Additives on Fermentation Characteristics and Protein Degradation of Green Tea Grounds Silage

  • This study evaluated the fermentation characteristics and protein degradation dynamics of wet green tea grounds (WGTG) silage. The WGTG was ensiled with distilled water (control), or lactic acid bacteria (LAB), enzyme (E), formic acid (FA) and formaldehyde (FD) prior to ensiling. Three bag silos for each treatment were randomly opened at 0, 3, 7, 14, 28 and 60 days after anaerobic storage. For all the treatments, except for FA, there was a rapid decline in pH during the first 7 days of ensiling. LAB treatment had higher lactic acid content, lower ammonia-N ($NH_3$-N) and free-amino nitrogen (FAA-N) contents than other treatments (p<0.05). E treatment had higher lactic acid, water-soluble carbohydrates (WSC) and non-protein nitrogen (NPN) content than the control (p<0.05). FA treatment had higher $NH_3$-N and FAA-N content than the control (p<0.05). FD treatment had lower NPN and FAA-N content than the control, but it did not significantly inhibit the protein degradation when compared to LAB treatment (p>0.05). Results indicate that LAB treatment had the best effect on the fermentation characteristics and protein degradation of WGTG silage.

Dietary rambutan peel powder as a rumen modifier in beef cattle

  • Objective: The experiment was conducted to study the effect of rambutan (Nephelium lappaceum) fruit peel powder (RP) on feed consumption, digestibility of nutrients, ruminal fermentation dynamics and microbial population in Thai breed cattle. Methods: Four, 2-year old (250±15 kg) beef bull crossbreds (75% Brahman×25% local breed) were allotted to experimental treatments using a 4×4 Latin square design. Four dietary supplementation treatments were imposed; non-supplementation (control, T1); supplementation of RP fed at 2% of dry matter intake (DMI) (low, T2); supplementation of RP fed at 4% of DMI (medium, T3) and supplementation of RP fed at 6% of DMI (high, T4). All cattle were given a concentrate supplement at 1% of body weight while Napier grass was provided as a free choice. Results: The findings revealed that RP supplementation did not negatively affect (p>0.05) DMI of Napier grass, while RP intake and total DMI were the greatest in the RP supplementation at 4% and 6% DMI. Nevertheless, the nutrients (dry matter, organic matter, crude protein, neutral detergent fiber, and acid detergent fiber) digestibilities were not changed in the RP supplementation groups. Rumen fermentation parameters especially those of total volatile fatty acids, acetate and butyrate were not significantly changed. However, the propionate concentration was remarkably increased (p<0.05) in the RP supplementation. Notably, the ratio of acetate to propionate, the number of protozoa, as well as the methane estimation were significantly reduced in the RP supplemented groups (4% and 6% of DMI), while the counts of bacteria was not altered. Conclusion: Supplementation of RP (4% of DMI) improved rumen propionate production, reduced protozoal population and methane estimation (p<0.05) without a negative effect on feed consumption and nutrients total tract digestibilities in beef cattle. Using dietary rambutan fruit peel powder has potential promise as a rumen regulator.
