In this work, the effect of the culture medium composition on microbial growth and ethanol production in Clostridium autoethanogenum culture was investigated to enhance the ethanol productivity. D-Ca-pantothenate, vitamin B12 (as vitamins), and sodium sulfide (as sulfur source) were selected as examined components, and the effects of components' concentration on cell growth and ethanol production was investigated. For D-Ca-pantothenate concentrations varing from 0.5, 5, 50 and 500 mg/L, a slight increase in the ethanol production was observed at the 0.5 mg/L, but negligible differences in microbial growth and ethanol production were measured for the concentration ranges examined. The effect of vitamin B12 concentrations from 0.1, 1.0, 10, and 100 mg/L on the microbial growth and ethanol production was investigated, and it was found that the ethanol production using a 0.1 mg/L of vitamin B12 concentration increased by 245% compared to that of using the basic medium concentration (10 mg/L). The effect of sodium sulfide concentrations (0.5, 5, and 10 g/L) on the microbial growth and ethanol production was also studied, and the inhibition of microbial growth was observed when the sodium sulfide usage was over 0.5 g/L. In conclusion, changes in D-Ca-pantothenate and sodium sulfide concentrations did not affect the ethanol production, whereas even a 100 times lower concentration of vitamin B12 than that of the basic medium improved the production.
In this work, we investigated the effect of the concentration of medium components on microbial growth and ethanol production in order to improve ethanol productivity in the Clostridium autoethanogenum culture process using syngas as a sole carbon source. Molybenum, nickel and cobalt (as heavy metal ions) were selected as examined components, and the effects of components concentration on the cell growth and ethanol production was examined. Among molybdenum concentrations of 0, 0.001, 0.01 and 0.1 g/L. a slight increase in ethanol production was observed at 0.001 g/L, but significant differences in the microbial growth and ethanol production were not observed in the examined concentration range. In the case of nickel concentration of 0, 0.001, 0.01 and 0.1 g/L, the change in the microbial growth and ethanol production was investigated, and it was found that the ethanol production using 0.001 g/L increased by 26% compared to that of using the basal medium concentration (0.01g/L). The effect of cobalt concentrations (0, 0.018, 0.18 and 1.8 g/L) on the microbial growth and ethanol production was also investigated, and the inhibition of microbial growth was observed when the cobalt usage was over 0.18 g/L. In conclusion, cobalt did not show any further improvement of ethanol production by changing concentration, however, molybdenum and nickel showed increases in the produced ethanol concentration compared to that of using 1/10 times of the basal medium concentration.

Interests in the field of rapid methods and automation in microbiology have been growing steadily on an international scale in recent years. International meetings concerned this problem have been held in elsewhere in the world countries since the past twenty years. But, unfortunately in the field of microbial examination in food hygiene, this problem have not yet been developed so much as in the field of clinical microbiology. Today, I would like to introduce you here present aspects of rapid and automation technologies, those which are manly carrying in milk and meats industries. My illustration will be given recent improved technologies using automatic apparatus and instruments along with process of microbial count procedure. Recent direct microbiological counting system (ChemeScan \ulcorner) as real time ultrasensitive analysis created by Cheminex Ltd., France is now most evolutional instrument to provide direct microbial counts, down to one cell, within 30 minutes. The results from these evaluations how a good correlation between the ChemScan system and the standard plate count method. This system will be successful application for not only in the field of pharmacology but also food microbiology. In addition, current identification of microbes by sophisticated instruments suitable for food microbiology, one of which Biology is manual system (BIOLOG\ulcorner), provides reference-level capability at a modes price. For the manual system, the color reactions in the microplate are read by eye and manually keyed into personal computer. Species identification appears on the computer screen within seconds, along with biotype patterns, a list of closely related species, and other useful statistics. In present this is useful application for microbial ecology and epidemiological survey. RiboPrinter system newly produced by DuPont is now focusing among microbiologists in the world, and is one of the biggest microbial characterization system using a DNA-based approach. The technology analyzer is bacterial culture for its genetic fingerprint or riboprint pattern. Finally Bio-cellTracer system for automatic measurement of fungal growth and Fukitori-Maseter, a Surface Hygiene Monitoring Kit by using swabe procedure in food processing environment are briefly illustrated in this presentation.

The objective of this study was to determine the effect of feeding direct-fed microbials (DFM) on the growth performance and prophylaxis of calf diarrhea during the pre-weaning period as an alternative to antibiotics. A multi-species DFM was formulated including three lactic acid bacteria (Lactobacillus salivarius Ls29, Pediococcus acidilactia Pa175, and L. plantarum Lp177), three Bacillus strains (B. subtilis T4, B. polymyxa T1 and SM2), one yeast, Saccharomyces boulardii, and a nonpathogenic E. coli Nissle 1917. Lactic acid bacteria and Bacillus strains were selected based on the antibacterial activity against various animal pathogens, especially pathogenic E. coli using agar diffusion methods in vitro. Test and control groups were fed milk replacer and calf starter supplemented with DFM ($10^9$ cfu each of eight species/d/head, n = 29) or with antibiotics (0.1% neomycin sulfate in milk replacer and Colistin 0.08% and Oxyneo 110/110 0.1% in calf starter, n = 15), respectively. Overall fecal score and the incidence rate of diarrhea were reduced in the DFM group compared to the antibiotics one. About 40% of calves in antibiotic group suffered from diarrhea while in DFM group only 14% showed diarrhea. There was no difference in the average daily gain and feed efficiency of two groups. The hematological levels of calves were all within the normal range with no significant difference. In conclusion, the feeding of multispecies DFM during the pre-weaning period could reduce calf diarrhea and there was no difference in the growth performance between the groups, thus showing the potential as an alternative to antibiotics.
Bacillus thuringiensis (Bt) is the most widely used microbial insecticide in the biological control market. Cultivation of the microorganism to high cell densities offers potential for enhancing the rate of formation as well as the concentration of the desired products In the fermentation broths in bioreactor. With this objective, we developed the new bioreactor incorporating ceramic membrane module for the retention of cell mass. Cell yield and spore formation of Bacillus thuringiensis was improved markedly by adopting this new bioreactor based on glucose -limited feeding operation. It was possible to grow the cell and the heat-resistant spore to above $1.2\;{\times}\;10^{10}\;CFU/ml$ density. With glucose-limited operation, we studied the growth behavior of Bacillus thuringiensis during the cell retention culture. Linear growth of Bacillus thuringiensis was observed under glucose-limited culture, which matched well with simple mathematical model of cell retention culture.
Various kinds of native herbage grasses like-Miscanthus sinensis, Arundinella hirta, Cymbopogen geirngii, Themeda japonica etc. are widely distributed in every Korean mountain. So we investigated the availability of native grass, Miscanthus sinensis as a substrate for the production of S. C. P. The results were obtained as follows. 1) At the alkali treatment, NaOH was the most effective with the exclusion of lignin, and pretreated Miscanthus with NaOH appeared to be a good substrate for the microbial growth. 2) When Miscanthus was treated with one to 10% NaOH, the microbial growth increased in proportion to the increased alkali concentration. Beyond 4% NaOH, a slight increase was observed. 3) Phosphoric acid, as a neutralizer, was the most effective in cell production after alkali treatment. 4) On the effect of incubation time, the productivity was best found at 60 hours, and the cell weight was 9.23 mg per 1 ml, and the microbial digestibility of substrate was 75.2%.
For the production of microbial cells from cellulosie materials by cellulore-assimilating bacteria, Cellulomonas flavigena GFB 24-1, isolated by authors, utilization of this organism on some microbiological properties was investigated. The results of these studies were summarized at follows; 1. When the organism was incubated in the growth medium at pH 7.0 for 50 hours, its growth was the most effective and the level of excreted total protein in the menstruum increased continuously during the stationary phase of cell growth. 2. The optimal enzyme activity was observed in the pH region of 5 to 7 and culture period of 40 to 50 hours. 3. The microbial degestibility of cellulosic wastes such as sawdust, rice hull, rice straw, peanut hull and used newspaper was less than 30%, whereas that of cellulose powder was 47.1% and rice straw was digested 77% by NaOH treatment. 4. Bacterial cells incubated in the growth medium were increased up to 8% of sustrate concentration and showed a decrease on further concentration. 5. The production of microbial cells from NaOH treated rice straw was obtained 10.6mg/ml of culture medium.
We prepared capsules containing Saccharomyces cerevisiae and Zoogloea ramigera cells for the removal of lead(II) and cadmium ions. Microbial cells were encapsulated and cultured in the growth medium. The S.cerevisiae cells grown in the capule did not leak through the capsule membrane. The dried cell density reached to 250 g/l on the basis of the inner volume of the 2.0 mm diameter capsule after 36 hour cultivation. The dry whole cell expolymer density of encapsulated Z.ramigera reached to 200 g/L. The capsule was crosslinked with triethylene tetramine and glutaric dialdehyde solutions. The cadmium uptake of encapsulated whole cell expolymer of Z.ramigera was 55mg Cd/g biosorbent. The adsorption line followed well Langmuir isotherm. The lead uptake of the encapsulated S. cerevisiae was about 30 mg Pb/g biomass. The optimum pH of the lead uptake using encapsulated S. cerevisiae was found to be 6. Freundlich model showed a little better fit to the adsorption data than Langmuir model 95 percent of the lead adsorbed on the encapsulated biosorbents was desorbed by the 1 M HCl solution. The capsule was reused 50 batches without loosing the metal uptake capacity. And the mechanical strength of the crosslinked capsule was retained after 50 trials.

The biodegradable and biocompatible poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), a copolymer of microbial polyester, was fabricated as nanofibrous mats by electrospinning. Image analysis of the electrospun nanofibers fabricated from a 2 wt% 2,2,2-trifluoroethanol solution revealed a unimodal distribution pattern of fiber diameters with an observed average diameter of ca. 185 nm. The fiber diameter of electrospun fabrics could be controlled by adjusting the electro spinning parameters, including the solvent composition, concentration, applied voltage, and tip-to-collector distance. Chondrocytes derived from rabbit ear were cultured on a PHBV cast film and an electrospun PHBV nano-fibrous mat. After incubation for 2 h, the percentages of attached chondrocytes on the surfaces of the flat PHBV film and the PHBV nanofibrous mat were 19.0 and 30.1 %, respectively. On the surface of the electrospun PHBV fabric, more chondrocytes were attached and appeared to have a much greater spreaded morphology than did that of the flat PHBV cast film in the early culture stage. The electro spun PHBV nanofabric provides an attractive structure for the attachment and growth of chondrocytes as cell culture surfaces for tissue engineering.

Deoxynucleoside kinases exist as heterodimeric pairs specific for deoxyadenosine/deoxyguanosine kinase (dAK/dGK) and deoxyadenosine/deoxycytidine kinase (dAK/dCK). The aspartic acid-84 in dGK was mutated to alanine, asparagine and glutamic acid by site-directed mutagenesis. The mutation resulted in a drastic decease in dGK activity compared to the unmodified cloned enzyme while it increased production of dAK activity. The mutated dak/dgk genes, which synthesize tandem deoxyadenosine/deoxyguanosine kinase, were inserted back to the Lactobacillus acidophilus and Lactococcus lactis by electroporation to determine the effect of site-directed mutation of he enzymes on the microbial growth. However, no significant change was observed in cell growth and lactic acid production between wild type and mutant lactic acid bacteria.
