microRNA for determining the age-related myogenic capabilities of skeletal muscle

  • Skeletal muscle exhibits a loss of muscle mass and function with age. Decreased regenerative potential of muscle stem/progenitor cells is a major underlying cause of sarcopenia. We analyzed microRNAs (miRNA) that are differentially expressed in young and old myoblasts, to identify novel intrinsic factors that play a degenerative role in aged skeletal muscle. miR-431, one of decreasing miRNAs in old myoblasts, improved the myogenic differentiation when overexpressed in old myoblast, but suppressed their myogenic capability in knockdowned young myoblasts. We found that miR-431 directly binds to 3` untranslated regions (UTR) of Smad4 mRNA, and decreases its expression. Given that SMAD4 is one of the downstream effectors of TGF-β, a well-known degenerative signaling pathway in myogenesis, the decreased miR-431 in old myoblast causes SMAD4 elevation, thus resulting in defective myogenesis. Exogenous expression of miR-431 greatly improved the muscle regeneration in the cardiotoxin-injured hindlimb muscle of old mice by reducing SMAD4 levels. Since the miR-431 seed sequence is conserved in human SMAD4 3'UTR, miR-431 regulates the myogenic capacity of human skeletal myoblasts in the same manner. Our results suggest that age-associated miR-431 is required for the maintenance of the myogenic capability in myoblasts, thus underscoring its potential as a therapeutic target to slow down muscle aging.

간호대학생의 생명의료윤리의식과 비판적 사고성향이 간호전문직관에 미치는 영향 (The Influence of Biomedical Ethics and Critical Thinking Disposition on Nursing Professionalism in Nursing College Students)

  • 본 연구는 간호대학생의 생명의료윤리의식과 비판적 사고성향이 간호전문직관에 미치는 영향을 알아보고자 시도되었다. 본 연구는 2021년 5월부터 6월까지 충청남도 S대학의 간호대학생 144명을 대상으로 구조화된 설문을 통해 데이터를 수집하였다. 자료의 분석은 기술통계, t-test, ANOVA, Scheffe test, Pearson's 상관관계 분석, 다중회귀분석을 이용하였다. 본 연구의 결과 간호대학생의 간호전문직관은 생명의료윤리의식과 비판적 사고성향에 유의한 양의 상관관계(r=.400, p<.001, r=.431, p<.001)가 있었다. 간호대학생의 간호전문직관에 영향을 미치는 요인은 학년(𝛽=0.303, p<.001), 생명의료윤리의식(𝛽=0.287, p<.001)과 비판적 사고성향(𝛽=0.294, p<.001)이었고, 설명력은 31.2%였다. 본 연구의 결과로 간호대학생의 간호전문직관을 높여주기 위해 생명의료윤리의식과 비판적 사고성향을 높여주는 것이 필요하며, 그 방안으로는 학생의 학년을 고려한 생명의료윤리의식과 비판적 사고성향 프로그램의 개발과 적용이 있다.

남태평양 해양의 잠재력과 우리나라의 새로운 연구개발 투자 분야에 대한 고찰 (An Overview of Potentials in the South Pacific and the New R&D Investment Challenges)

  • With the draining of land resources, and rising new economic and resources expansionism, finding a new and yet to be developed ${\ulcorner}treasure{\lrcorner}$ is often a fierce and competitive challenge among many nations. This is especially true for Korea confronted with its urgent needs to find new resources and strategy to maintain the current pace of societal demand and economic development in this often impatient and rapidly progressing world. The ocean world in the South Pacific provides potentials to quench both orders with promises of new ${\ulcorner}treasure{\lrcorner}$ of living and ecosystem resources such as new fisheries and new bioactive products as well as providing alternative aesthetic, recreational resources and critical informations on various societal problems such as extreme weather conditions and early wining of the consequences resulting from global environmental changes.

Potential Utility of FDG PET-CT as a Non-invasive Tool for Monitoring Local Immune Responses

  • Purpose: The tumor microenvironment is known to be associated with the metabolic activity of cancer cells and local immune reactions. We hypothesized that glucose metabolism measured by 2-deoxy-2-($^{18}F$)fluoro-D-glucose ($^{18}F-FDG$) positron emission tomography (PET)-computed tomography (CT) ($^{18}F-FDG$ PET-CT) would be associated with local immune responses evaluated according to the presence of tumor infiltrating lymphocytes (TILs). Materials and Methods: We retrospectively reviewed 56 patients who underwent $^{18}F-FDG$ PET-CT prior to gastrectomy. In resected tumor specimens, TIL subsets, including cluster of differentiation (CD) 3, CD4, CD8, Forkhead box P3 (Foxp3), and granzyme B, were subjected to immunohistochemical analysis. The prognostic nutritional index (PNI) was calculated as: ($10{\times}serum$ albumin value)+($0.005{\times}peripheral$ lymphocyte counts). Additionally, the maximum standard uptake value ($SUV_{max}$) was calculated to evaluate the metabolic activity of cancer cells. Results: The $SUV_{max}$ was positively correlated with larger tumor size (R=0.293; P=0.029) and negatively correlated with PNI (R=-0.407; P=0.002). A higher $SUV_{max}$ showed a marginal association with higher CD3 (+) T lymphocyte counts (R=0.227; P=0.092) and a significant association with higher Foxp3 (+) T lymphocyte counts (R=0.431; P=0.009). No other clinicopathological characteristics were associated with $SUV_{max}$ or TILs. Survival analysis, however, indicated that neither $SUV_{max}$ nor Foxp3 held prognostic significance. Conclusions: FDG uptake on PET-CT could be associated with TILs, especially regulatory T cells, in gastric cancer. This finding may suggest that PET-CT could be of use as a non-invasive tool for monitoring the tumor microenvironment in patients with gastric cancer.

Effects of Antibiotic Growth Promoter and Characterization of Ecological Succession in Swine Gut Microbiota

  • Ever since the ban on antibiotic growth promoters (AGPs), the livestock death rate has increased owing to pathogenic bacterial infections. There is a need of developing AGP alternatives; however, the mechanisms by which AGP enhances livestock growth performance are not clearly understood. In this study, we fed 3-week-old swine for 9 weeks with and without AGPs containing chlortetracycline, sulfathiazole, and penicillin to investigate the effects of AGPs on swine gut microbiota. Microbial community analysis was done based on bacterial 16S rRNA genes using MiSeq. The use of AGP showed no growth promoting effect, but inhibited the growth of potential pathogens during the early growth stage. Our results showed the significant increase in species richness after the stabilization of gut microbiota during the post-weaning period (4-week-old). Moreover, the swine gut microbiota was divided into four clusters based on the distribution of operational taxonomic units, which was significantly correlated to the swine weight regardless of AGP treatments. Taxonomic abundance analysis indicated a negative correlation between host weight and the abundance of the family Prevotellaceae species, but showed positive correlation to the abundance of the family Spirochaetaceae, Clostridiaceae_1, and Peptostreptococcaeae species. Although no growth performance enhancement was observed, the use of AGP inhibited the potential pathogens in the early growth stage of swine. In addition, our results indicated the ecological succession of swine gut microbiota according to swine weight. Here, we present a characterization of swine gut microbiota with respect to the effects of AGPs on growth performance.

국내산 녹차 품종과 일본산 녹차 품종의 항산화 효능 비교 (Evaluation of Antioxidant Activities of Selected Green Tea Cultivars from Korea and Japan)

  • 국내 육성 품종 JW07-190, JW07-192과 일본산 도입 품종 Meiryoku (MR), Ryoufuu (RF), Yamanoibuki (YI) 대해서 80% 메탄올 추출물을 공시 재료로 ESR을 이용하여 항산화 효능을 평가하였다. 국내 육성 품종인 JW07-192에서 DPPH radical, alkyl radical, hydroxyl radical에 대해 비교적 높은 radical 소거능을 보였고, 모든 녹차 품종에서 강력한 DPPH radical 소거능을 보였다. 전체적으로, 국내 육성 품종인 JW07-192가 일본 도입 품종과 비교하였을 때 더 좋은 항산화 효능을 나타내었다. 총 폴리페놀 함량은 국내 육성 계통인 JW07-192가 $431.57{\pm}28.45\;mg/g$로 가장 높은 함량을 나타냈고, 플라보노이드 함량은 도입 품종인 YI가 $313.44{\pm}6.31$로 가장 높은 함량을 보였다.

할맥과 압맥의 칠반 및 호화특성 (Cooking and Pasting Properties of Split and Pressed Barley)

  • 국내 시판 할맥과 압맥제품의 성분 및 취반특성을 알아보고자 보리쌀 24제품(할맥 9개, 압맥 15개)을 수집하여 단백질, $\beta$-glucan, 아밀로오스 함량, 백도, 취반 및 호화특성 등을 조사하였다. 또한 일본산 가공 보리쌀 7제품(할맥 3개, 압맥 4개)도 구입하여 비교 분석하였다. 국내산 할맥과 압맥제품들의 단백질, $\beta$-glucan, 아밀로오스 함량은 비슷하였으며, 일본산 제품들의 경우에도 같은 경향을 나타내었다. 국내 제품의 경우 일본 제품에 비하여 단백질함량이 높고 백도가 낮은 특징을 보였으며 국내산과 일본산 모두 백도는 압맥이 할맥보다 높은 경향을 나타내었고 단백질과 백도는 부의 상관관계(r=-0.5112, p<0.0l)를 나타내었다. 국내산 할맥제품의 흡습성은 342~436, 퍼짐성은 할맥제품이 449~608%로 높고 용출고형물은 낮아 압맥제품보다 좋은 취반특성을 나타내었다. 특히, 찰성보리쌀을 가공한 제품이 더 좋은 취반특성을 나타내었다. 일본산 제품의 경우에도 할맥의 퍼짐성이 431~518%로 압맥보다 높은 경향을 나타내었다. 국내산 할맥과 압맥 제품의 호화개시온도는 압맥과 할맥제품에서 비슷하였으며 찰성보리쌀을 가공한 할맥 제품이 다른 제품에 비하여 낮은 호화개시온돈 높은 강하점도와 낮은 치반점도를 나타내었다. 일본 제품은 국내산 제품보다 낮은 호화개시온도와 높은 치반점도를 나타내었다.
