구절초 추출물의 항비만 활성 (Antiobesity Activity of Chrysanthemum zawadskii Methanol Extract)

  • 구절초(Chrysanthemum zawadskii)는 국화과에 속하는 다년생 초본으로 일본, 중국 등에 분포하고 있으며 항산 화, 항염증, 항균 등의 생리활성이 알려져 있으나 항비만 활성에 대해서는 보고된 바 없다. 본 연구에서는 구절초 메탄올 추출물(CZME)의 항비만 활성을 췌장 lipase 효소 활성 억제능과 지방세포분화 및 지방 생성에 미치는 영향을 분석하였다. 그 결과 CZME는 농도 의존적으로 lipase 효소 활성을 억제시켰으며, insulin, dexamethasone, 3-isobutyl-1-methylxanthine (MDI)로 분화를 유도한 3T3-L1 preadipocyte를 이용하여 지방세포 분화 및 지방생성에 미치는 영향을 분석한 결과 지방세포 분화, 세포 내 지방 축적, TG 함량 등을 독성 없이 농도의존적으로 억제 하였다. 이러한 구절초의 지방세포 분화 억제능은 핵심 작용 인자인 C/EBPα, C/EBPβ, 그리고 PPARγ의 유전자 및 단백질 발현조절에서 기인함을 확인하였다. 뿐만 아니라 MDI로 분화를 유도한 3T3-L1 preadipocyte에서 생성된 지방의 분해능을 분석한 결과 지방세포 분화 억제뿐만 아니라 지방세포 내 중성지방 또한 유의적으로 분해시키는 것으로 나타났다. 이러한 결과는 구절초가 보유한 췌장 lipase 활성 저해능, 지방세포 분화 억제능, 지방세포내 지방 분해능을 통한 항비만 활성을 처음으로 밝혀낸 것이며 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.

북방산 개구리(Rana dybowskii)의포의 프로젝트론 생서에 대한 cAMP의 조절작용 (Role of cAMP in the Regulation of Progesterone Production and Secretion by Frog (Rana dybowskii) Follicles in vitro)

  • 북방산개구리의 여포를 인공배양하면서 여포의 progesterone(p$_4$) 의 생성양상과 cAMP의 조절작용을 조사하여 보았다. 배양중인 여포에 뇌하수체 추출물(frog pituitary homogenate,FPH)을 처리하였을 때 배양 한시간 부터 여포내 p$_4$의 양이 급격히 증가하였다. 그러나 생성된 p$_4$의 최대양이나(약 60-300pg/follicle),peak를 이루는 시간이(2시간 이후)개체에 따라 차이가 있었다. FPH의 처리를 받지않은 대조군에서는 배양기간에 관계없이 여포내에서 대략 10pg/follicle정도의 p$_4$가 측정되었으나 예외인 개체도 있었다. 배양액내로 분비된 p$_4$의 양은 여포내에 생성된p$_4$양의 약 60%정도 이었다. 배양액에 adenlate cyclass의 촉진제인 forskolin이나phosphodiesterase의 저해제인 3-isoburyl-1- methylxanthine(IBMX)을 동시에 혹은 따로 처리하면 p$_4$의 생성과 분비가 역시 증가하며 모든 양상이 FPH의 자극에 의한 것과 거의 같았다. 따라서 개구리여포의 스테로이드생성은 cAMP를 통하여 조절된다는 것과 여포세포내에는 cAMP의 생성과 분해에 관계하는 효소들이 있다는 것을 알았다.

참개구리와 옴개구리 여포의 프로제스테론 생성과 난자의 성숙 (Progesterone Production and Oocyte Maturatf on of Frog (Rona nigromaculata and Rana rugoBa) Follicles in vitro)

  • 참구개구와 옴개구리의 여포를 생체외 배양하면서 progesterone (P$_4$)생성과 난자의 성숙 및 cyclic(cAMP)의 조절작용을 조사하였다.참개구리의 여포에 뇌하수체 추출물(frog pituitary homogenate.FPH)을 처리하면 농도에 의존하여 여포의 P$_4$생성이 증가하였으며 난자의 성숙(핵붕괴)이 일어났다. 이들 여포들을 배양하면서 3시간 간격으로 호르몬이 여포에 축적된 양, 배양액에 분비된 양 및 난자의 성숙율을 조사한 결과 FPH처리군에서 P$_4$는 3-6시간에 최고치 (여포내,약 400pg/여포;분비량,약 800pg/여포)를 나타내었으며 난자의 핵붕괴는 9-12시간에 일어났다. 상기 여포들과 같은 조건으로 배양하면서 forskolin과 3-isobutyl-1-methylxanthine(IBMX)을 배양액에 처리하여 간접적으로 여포내 cAMP의 농도를 높여주면 FPH와 유사한 양사으로 호르몬의 생성을 촉진하였다. 그러나 난자의 성숙은 전혀 일어나지 않았다. 옴개구리의 여포를 배양하면서 FPH를 처리하였을 때는 아무 처리를 하지않은 대조군과 비교하여 거의 P$_4$의 생성을 촉진하지 않았으며 난자의 성숙도 유도하지 못했다, 그러나 이들에게 forskolin과 IBMX를 처리하면 P$_4$의 생성을 현저하게 촉진하여 다량의 P$_4$가 여포와 (약 800pg/여포) 배양액에 (1700pg/여포)축적되었다.

PHOSPHODIESTERASE 억제제 (PDE-1), SODIUM NITROPRUSSIDE, AMITRIPTYLINE, 및 CHLORPROMAZINE의 항-혈소판작용

  • Thrombin (0.25 U/ml : TB), 소-피부 collagen (200 $\mu\textrm{g}$/ml : CG), adenosine 5'-diphesphate (4,0 $\times$ $10^{-5}$M : ADP), 및 epinephrine (4,0 $\times$ 10 $^{-5}$M : EPI)의 가토-혈소판 응집과 단백인산화작용에 미치는 PDE-I (3-isobutyl-1-methylxanthine : IBMX, 및 KR 30075), amitriptyline (AP), chlorpromazine (CP), 및 sodium nitrogrosside (SNP)의 염향을 비교-검토하였다. 그 결과, KR은 2,2 $\times$ $10^{-7}$M 이하의 $IC_{50}$/에서 EPI > ADP > CG > TB 순으로 각각을 억제하였으며, SNP 보다도 강하였고; KR-30075보다 약하나 IBMX, AP, 및 CP도 각 응집재의 작용을 억제하였으며 특히 EPI에 대하여 $10^{-8}$M 이하의 $IC_{50}$/에서 유의한 억제력을 보였다. 각 응집제들은 41 kD 인산화는 유의하게 증가시키며 47 kD와 20 kD 단백인산화는 감소시켰는데; 모든 항응집성 약물이 41 kD 인산화-증가는 유의하게 억제하였다, 아울러, AP와 CP는 47 kD 단백인산화-감소에 영향을 미치지 않았으나 20 kD 단백인산화-감소는 억제하였다. PDE-I (IBMX와 KR)와 SNP는 47 kD와 20 kD 단백인산화-감소를 다소 약화시켰으며, 43 kD와 22 kD 단백인산화를 KR > IBMX > SNP순으로 유의하게 증가시켰고, KR의 22 kD 단백인산화작용은 현저하였다.

원두커피내 카페인 및 클로로겐산 함량 평가를 위한 동시분석법 (Simulateous Determination for the Contents of Caffeine and Chlorogenic Acid in Coffee Beans)

  • Caffeine is an alkaloid of the methylxanthine family known as a central nervous system stimulant, temporarily warding off drowsiness and restoring alertness in humans. There is a recommended upper limits of caffeine for health because a high dose can cause negative effects. Chlorogenic acid is a natural polyphenol compound known to have an antioxidant activity. In this study, the contents of caffeine and chlorogenic acid in coffee beans from different origins(Costa Rica, Indonesia, Vietnam) were determined by using liquid chromatography-tandem mass spectrometry(LC-MS/MS). The experiment offers more selectivity and sensitivity for those compounds compared with conventional methods such as UV/VIS spectrophotometry. The average concentrations of caffeine and chlorogenic acid in coffee beans origined in Costa Rica were 15.05 mg/g and 5.33 mg/g respectively. In the case of coffee beans origined in Indonesia, the average concentrations were 13.10 mg/g for caffeine and 3.75 mg/g for chlorogenic acid. Vietnamese coffee showed that the average concentrations were 17.79 mg/g for caffeine and 1.12 mg/g for chlorogenic acid. This study can contribute to a better understanding of the contents of caffeine and chlorogenic acid in various coffee beans in order to evaluate dietary intake.

15-Hydroxyprostaglandin Dehydrogenase Is Associated with the Troglitazone-Induced Promotion of Adipocyte Differentiation in Human Bone Marrow Mesenchymal Stem Cells

  • Adipocyte differentiation in human bone marrow mesenchymal stem cells (hBM-MSCs) is not as efficient as that in murine pre-adipocytes when induced by adipogenic agents including insulin, dexamethasone, and 3-isobutyl-1-methylxanthine (IDX condition). Therefore, the promotion of adipocyte differentiation in hBM-MSCs has been used as a cell culture model to evaluate insulin sensitivity for anti-diabetic drugs. In hBM-MSCs, $PPAR{\gamma}$ agonists or sulfonylurea anti-diabetic drugs have been added to IDX conditions to promote adipocyte differentiation. Here we show that troglitazone, a peroxisome proliferator-activated receptor-gamma ($PPAR{\gamma}$) agonist, significantly reduced the levels of anti-adipogenic $PGE_2$ in IDX-conditioned hBM-MSC culture supernatants when compared to $PGE_2$ levels in the absence of $PPAR{\gamma}$ agonist. However, there was no difference in the mRNA levels of cyclooxygenases (COXs) and the activities of COXs and prostaglandin synthases during adipocyte differentiation in hBM-MSCs with or without troglitazone. In hBM-MSCs, troglitazone significantly increased the mRNA level of 15-hydroxyprostaglandin dehydrogenase (HPGD) which can act to decrease $PGE_2$ levels in culture. These results suggest that the role of $PPAR{\gamma}$ activation in promoting adipocyte differentiation in hBM-MSCs is to reduce anti-adipogenic $PGE_2$ levels through the up-regulation of HPGD expression.

Effect of Co-culture with Spermatozoa on the Resumption of Meiosis in Porcine Germinal Vesicle Oocytes Arrested with Meiotic Inhibitors

  • In vitro maturation of porcine immature cumulus-enclosed oocytes can be enhanced by co-incubation with spermatozoa even before fertilization. The aim of this study was to determine whether the addition of spermatozoa into the culture medium can stimulate the meiosis resumption of porcine cumulus-enclosed oocytes arrested at germinal vesicle (GV). Cumulus-enclosed oocytes (CEOs) were collected from follicles of 3 to 5mm diameter. Porcine CEOs were cultured in tissue culture medium containing various meiosis inhibitors and spermatozoa. Oocytes were examined for evidence of GV and GV breakdown after 24 h culture. After 24 h culture $43.8\%$ of oocytes cultured in only TCM 199 remained at GV stage whereas $56.2\%$ of oocytes were able to resume meiosis. When porcine CEOs were cultured in the medium with meiosis inhibitor such as, dibutyryl cAMP (dbcAMP) and forskolin (Fo), more than $90\%$ of oocytes were not able to resume meiosis. However, co-culture of porcine CEOs with spermatozoa was able to overcome the inhibitory effect of dbcAMP and Fo. Irrespective of the presence of 3-isobutyl-1-methylxanthine (IBMX), no difference was observed in the proportion of oocyte reached germinal vesicle breakdown (GVBD). The present study suggests that dbcAMP and Fo prevent the spontaneous maturation of competent oocyte in culture after isolation from follicles and that mammalian spermatozoa contain a substance(s) that improves meiosis resumption in vitro of porcine cumulus-enclosed oocytes.

Inhibitory Effects of Pine Cone (Pinus densiflora) on Melanogenesis in B16F10 Melanoma Cells

  • The pathological condition of excessive melanogenesis causing freckles, melasma, senile lentigo, pigmented acne scars, and cancer has a critical impact on the wellness of individuals. The mechanism of melanogenesis is related to the expression of melanogenic enzymes. Here, we evaluated the inhibitory effect of pine cone (Pinus densiflora) extracts on melanogenesis. P. densiflora, the Korean Red Pine, is the predominant tree species in the cool, temperate forests of northeast Asia, occurring in pure stands across Korea, Japan, and parts of northern China and Russia. P. densiflora leaves, pollen, and bark have been widely used for traditional medicine, or edible purposes. However, pine cones are rarely used as natural raw materials, although they contain many bioactive phytochemicals. The pine cone ethyl acetate fraction (PEF) showed no toxicity to B16F10 cells at a concentration of less than $100{\mu}g/mL$. PEF inhibited the expression of microphthalmiaassociated transcription factor (MITF), tyrosinase and tyrosinase-related factors in B16F10 cells treated with 3-Isobutyl1-methylxanthine (IBMX). These results suggest that pine cones can be used as an effective natural melanogenesis inhibitory agent.

한국인에 대한 지문과 장문의 정량적 분석 (Regulation of Cumulus Expansion of Porcine Cumulus-Oocyte Complexes in vitro: Involvement of cAMP and Calcium)

  • The present experiments were carried out to investigate the mode of cAMP regulation of cumulus expansion in pig. Intracellular level of cAMP in the cumulus cells was modulated by culturing porcine cumulus oocyte complexes (COC's) with forskolin, an adenylate cyclase stimulator and 3-isobutyl-1-methylxanthine (IBMX), a phosphodiesterase inhibitor. The role of calcium in the hormone induced cumulus expansion process was also studied. Forskolin in the medium stimulated cumulus expansion from the concentration of 0.01 $\mu$M and induced full expansion at l-10 $\mu$M In contrast, IBMX in the medium (20-180 $\mu$M) failed to induce the expansion. Verapamil, a calcium ion transport blocker, suppressed follicle stimulating hormone(FSH)-induced cumulus expansion in a dose dependent fashion (0.002-0. 2 mM) when the COC's were exposed to the drugs during culture period (32 hr). But verapamil did not interfere with the triggering action of FSH during early four hours of culture period. The data presented here showed that adenylate cyclase in the porcine cumulus cells may play a key role in the regulation of the intracellular cAMP level and calcium ion may be involved in the later period of cumulus expansion process.

Optimization of adipogenic differentiation conditions for canine adipose-derived stem cells

  • Background: Canine adipose-derived stem cells (cADSCs) exhibit various differentiation properties and are isolated from the canine subcutaneous fat. Although cADSCs are valuable as tools for research on adipogenic differentiation, studies focusing on adipogenic differentiation methods and the underlying mechanisms are still lacking. Objectives: In this study, we aimed to establish an optimal method for adipogenic differentiation conditions of cADSCs and evaluate the role of peroxisome proliferator-activated receptor gamma (PPARγ) and estrogen receptor (ER) signaling in the adipogenic differentiation. Methods: To induce adipogenic differentiation of cADSCs, 3 different adipogenic medium conditions, MDI, DRI, and MDRI, using 3-isobutyl-1-methylxanthine (M), dexamethasone (D), insulin (I), and rosiglitazone (R) were tested. Results: MDRI, addition of PPARγ agonist rosiglitazone to MDI, was the most significantly facilitated cADSC into adipocyte. GW9662, an antagonist of PPARγ, significantly reduced adipogenic differentiation induced by rosiglitazone. Adipogenic differentiation was also stimulated when 17β-estradiol was added to MDI and DRI, and this stimulation was inhibited by the ER antagonist ICI182,780. Conclusions: Taken together, our results suggest that PPARγ and ER signaling are related to the adipogenic differentiation of cADSCs. This study could provide basic information for future research on obesity or anti-obesity mechanisms in dogs.
