Application study of random forest method based on Sentinel-2 imagery for surface cover classification in rivers - A case of Naeseong Stream - (하천 내 지표 피복 분류를 위한 Sentinel-2 영상 기반 랜덤 포레스트 기법의 적용성 연구 - 내성천을 사례로 -)

  • Understanding the status of surface cover in riparian zones is essential for river management and flood disaster prevention. Traditional survey methods rely on expert interpretation of vegetation through vegetation mapping or indices. However, these methods are limited by their ability to accurately reflect dynamically changing river environments. Against this backdrop, this study utilized satellite imagery to apply the Random Forest method to assess the distribution of vegetation in rivers over multiple years, focusing on the Naeseong Stream as a case study. Remote sensing data from Sentinel-2 imagery were combined with ground truth data from the Naeseong Stream surface cover in 2016. The Random Forest machine learning algorithm was used to extract and train 1,000 samples per surface cover from ten predetermined sampling areas, followed by validation. A sensitivity analysis, annual surface cover analysis, and accuracy assessment were conducted to evaluate their applicability. The results showed an accuracy of 85.1% based on the validation data. Sensitivity analysis indicated the highest efficiency in 30 trees, 800 samples, and the downstream river section. Surface cover analysis accurately reflects the actual river environment. The accuracy analysis identified 14.9% boundary and internal errors, with high accuracy observed in six categories, excluding scattered and herbaceous vegetation. Although this study focused on a single river, applying the surface cover classification method to multiple rivers is necessary to obtain more accurate and comprehensive data.

Determination of Bumetanide in Human Plasma by a Validated HPLC Method and Its Application to Single-dose Pharmacokinetics (사람 혈장 중 부메타니드의 HPLC 분석법 검증 및 단일 용량 투여에 의한 약물동태 연구)

  • A high-performance liquid chromatographic method was employed for the determination of bumetanide in human plasma. After addition of internal standard (IS, naproxen) and acidification of the plasma with 1 M hydrochloric acid, the drug and IS were extracted into dichloromethane. The organic phase was back-extracted into 1 M sodium bicarbonate solution and 50 ${\mu}l$ of the aqueous phase was injected onto a reversed-phase C18 column with a mobile phase consisting of methanol: water: glacial acetic acid = 65 : 35 : 1. The samples were detected utilizing a fluorescence detector (excitation wavelength 235 nm, emission wavelength 405 nm). The method was specific and validated with a lower limit of 5 ng/mL. Intra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of the method was demonstrated by analysis of plasma after oral administration of a single 2 mg dose to 24 healthy subjects. From the plasma bumetanide concentration vs. time curves, the mean AUC was $246.5{\pm}73.8\;ng{\cdot}hr/mL$ and $C_{max}$ of $132.1{\pm}40.9$ ng/mL reached 1.2 hr after administration. The mean biological half-life of burnet ani de was $1.1{\pm}0.2$hr. Based on the results, this simple and validated assay method could readily be used in any pharmacokinetic or bioequivalence studies using humans.

  • There are two methods in estimating the remaining life of in-service airfield concrete pavement. They are a method simply using the past accumulated traffic and a method using the theoretical mechanistic analysis. Since the former method is somewhat far from the actual condition, the latter method is widely used by most engineers and researchers. The most essential component of the latter method is the fatigue model of the concrete slab. A fatigue model for airfield concrete pavement is developed in this study by a series of fatigue tests using 30 concrete cylinder specimens obtained from a 10 year old in-service airfield concrete slab. Strengths for the stress ratio calculation were obtained from the split tensile test of the cores sliced. Fatigue test mode was repeated split tensile test. The R2 of developed fatigue model was 0.5. Specimens taken from another airport had been tested for validation of the model. The results showed a good fit to the model. It was also found that the fatigue life predicted from the model was a tittle greater when the stress ratio is greater than 80 percent than other fatigue models developed earlier in America.

Validation of analysis of urinary fluoride by ion selective electrode method (이온선택전극법에 의한 소변 중 불소 이온 분석법 검증)

  • A simple and sensitive analytical method for fluoride in urine by ion selective electrode (ISE) method was presented. Traditional buffer for fluoride determination using ISE is acetate-based one. Researchers have pointed out some drawbacks of the buffer for fluoride ISE analysis, and some other buffers including citrate-ammonium buffer and MES buffer have been studied for accurate determination of fluoride in urine here. These buffers provided promising results in environmental field, and this author focused on overcoming the interference of co-existing aluminium. The results show that MES-CyDTA buffer gave the best recovery with accuracy of 95-97.5% and precision of 1.9-7.9% for reference sample of 1.8-7.8 mg/L fluoride in urine, with smaller amount of samples and shorter analysis time compared with the traditional method which used acetate buffer. The method was applied to field samples, and which showed urinary of $0.98{\pm}0.38mg/g$ creatinine for workers in electric cable manufacturing factory (n=15) and $0.59{\pm}0.30mg/g$ creatinine for non-exposed workers (n=12).

Validation of an analytical method of oxyresveratrol for standardization of Mulberry (Morus alba L.) branch extract as a functional ingredient (상지추출물의 기능성원료 표준화를 위한 지표성분 옥시레스베라트롤 분석법 검증)

  • An HPLC method was developed to quantitate a marker, oxyresveratrol (ORT), for the standardization of mulberry branch extracts (MBE) as a functional ingredient. HPLC was performed on a $C_{18}$ column with a gradient elution using 0.05% $H_3PO_4$ and acetonitrile at a flow rate of 0.8 mL/min, and detected at 320 nm. The HPLC method was validated according to Korea Food and Drug Administration (KFDA) guideline of analytical procedures with respect to specificity, linearity, accuracy and precision. Calibration curve of ORT showed high linearity ($R^2=1$), and limits of detection and quantification were 0.3 and $1.0{\mu}g/mL$, respectively. Relative standard deviation values from intra-and inter-day precision were less than 3.52 and 4.70%, respectively. Recovery rate ranged from 97.64% to 103.69%, and ORT content in MBE was approximately 3.78%. These results suggest that the HPLC method developed for the analysis of ORT in MBE is simple, efficient, and could contribute to the quality control of MBE.

Development and Validation of an HPLC Method for the Pharmacokinetic Study of Pentoxifylline in Human Serum (체내동태 연구를 위한 혈청 중 펜톡시필린의 HPLC 정량법 개발 및 검증)

  • A selective and sensitive reversed-phase HPLC method for the determination of pentoxifylline in human serum was developed, validated, and applied to the pharmacokinetic study of pentoxifylline. Pentoxifylline and internal standard, chloramphenicol, were extracted from the serum by liquid-liquid extraction with dichloromethane and analyzed on a Luna CI8(2) column with the mobile phase of acetonitrile-0.034 M phosphoric acid (25:75, v/v, adjusted to pH 4.0 with 10 M NaOH). Detection wavelength of 273 nm and flow rate of 0.8 mL/min were used. This method showed linear response over the concentration range of 10-500 ng/mL with correlation coefficients greater than 0.999. The lower limit of quantification using 0.5 mL of the serum was 10 ng/mL, which was sensitive enough for pharmacokinetic studies of pentoxifylline. The overall accuracy of the quality control samples ranged from 89.3 to 92.7% for pentoxifylline with overall precision (% C.V.) being 4.1-9.2%. The relative mean recovery of pentoxifylline for human serum was 105.8%. Stability (stock solution, short and long-term) studies showed that pentoxifylline was not stable during storage. But three freeze-thaw cycles and extracted serum samples were stable. This method showed good ruggedness (within 15% C.V.) and was successfully applied for the analysis of pentoxifylline in human serum samples for the pharmacokinetic studies of orally administered $Trental^{\circledR}$ tablet (400 mg pentoxifylline), demonstrating the suitability of the method.

Simultaneous Analysis of 17 Organophosphorous Pesticides in Blood by Automated Head Space-SPME GC/MS (HS-SPME-GC/MS에 의한 혈액중 17종 유기인계 농약의 동시분석법)

  • HS-SPME-GC/MS was studied and optimized for the determination of 17 orgarnophosphorous pesiticides (OPPs: chlorpyrifos, chlorpyrifos-methyl, demeton-s-methyl, diazinon, dimethoate, EPN, fenitrothion, fenthion, malathion, methidathion, monocrotophos, parathion, phenthoate, phosphamidon, sulfotep, terbufos, triazophos) in blood. Optimum SPME parameters were selected: choice of SPME fiber (85 ${\mu}m$ polyacrylate), pH effect (0.5 N HCl), salt effect ($Na_2SO_4$, 0.2 g; 20%), headspace incubation temperature ($80^{\circ}C$), headspace incubation time (1 min), headspace adsorption time (30 min) and GC desorption time (2 min). These parameters were optimized using HS-SPME autosampler coupled with gas chromatography-mass spectrometry (GC-MS). Method validation was carried out in terms of linearity, limit of detection (LOD), limit of quantitation (LOQ) and recovery in blood. The assay was linear over 0.5~5.0 mg/l ($r^2$=0.955~1.000). Limit of detection (LOD) and limit of quantitation (LOQ) in blood were determined 0.03~0.3 mg/l (S/N=3) and 0.1~1.1 mg/l (S/N=10), respectively. Relative recovery with 0.5, 1 and 5 mg/l (in blood) were 90.8%, 98.5% and 94.1%, respectively. This method will be applied to the determination of the orgarnophosphorous pesticides in postmortem blood. The proposed protocol can be an attractive alternative to be used in routine toxicological analysis.

Computation of Flows Around a High Speed Catamaran

  • A numerical study is carried out to clarify the characteristics of flow fields and breaking phenomena around a high speed catamaran hull advancing on calm water. Computations are carried out for Froude numbers between 0.2 and 1.0 and for ratios of the distance between hulls to the catamaran length varying between 0.2 and 0.5 for a mathematically defined Wigley hull. A Navier-Stokes solver which includes the nonlinearities of free surface conditions is employed. Computations are performed in a rectangular grid system based on the Marker & Cell method. For validation, present computation results are compared with existing experimental results. As an application, the results of the displacement catamaran are used for the breaking analysis.

A study of the GPS/Levelling in Jeju island using PGPS (제주지역에서 PGPS를 이용한 GPS 수준측량 적용)

  • This study describes the validation of the GPS/Levelling using GPS data that were obtained from three Permanent GPS stations of the Jeju island. Each orthometric height of three Permanent GPS stations was calculated from the Bench mark of levelling with Traverse-survey and relative baseline processing The test result shows that the error of closure of orthometric height was about 1.6cm and one of ellipsoidal height was about 1.5cm. This result represents that the geoidal height and undulations are regular in the Jeju island. According to the test results, it is identified that one can calculate the orthometric height using Permanent GPS station data rather than traditional levelling method which has a some complexity especially in Jeju island.

Sequence driven features for prediction of subcellular localization of proteins

  • Predicting the cellular location of an unknown protein gives a valuable information for inferring the possible function of the protein. For more accurate prediction system, we need a good feature extraction method that transforms the raw sequence data into the numerical feature vector, minimizing information loss. In this paper, we propose new methods of extracting underlying features only from the sequence data by computing pairwise sequence alignment scores. In addition, we use composition based features to improve prediction accuracy. To construct an SVM ensemble from separately trained SVM classifiers, we propose specificity based weighted majority voting. The overall prediction accuracy evaluated by the 5-fold cross-validation reached 88.53% for the eukaryotic animal data set. By comparing the prediction accuracy of various feature extraction methods, we could get the biological insight on the location of targeting information. Our numerical experiments confirm that our new feature extraction methods are very useful for predicting subcellular localization of proteins.
