Prodution and Properties of the Insoluble Penicillinase from Streptomyces (방선균이 분비하는 불용성 Penicillinase)

  • A Streptomyces sp. strain AS-727 which was capable of producing penicillinase, was isolated from soil. The enzyme production was affected by the carbon and nitrogen sources added. Among them so far tested, glucose (or maltose) and sodium nitrate increased the enzyme production. And the amount of enzyme prodced reached maximum in 4 days cultivation. The optimla pH and temperature of the penicillinase was between pH 6.0 to 8.0 and 4$0^{\circ}C$ respectively. The stabel pH range of the enzyme was stable at 4$0^{\circ}C$, but it lost about 30% and 40% of the the activity respectively when it was treated at 6$0^{\circ}C$ and 8$0^{\circ}C$ for 60 minutes. The activity of the enzyme was inhibited by Z $n^{++}$, but A $g^{+}$, $Co^{++}$, $_Mn^{++}$, $Ca^{++}$, P $b^{++}$ did not affected enzyme activity. Peculiarly, the enzyme protein precipitated by freezing or addition of ammonium sulfate, urea, sodium chloride and some organic solvents as etanol, methanol, acetone was not dissolved in deionized water or any buffer solution.n.n.ion.n.n.

Phenotypic Characterization of Methylotrophic N2-Fixing Bacteria Isolated from Rice (Oryza sativa L.) (벼(oryza sativa L.)에서 분리한 Methylotrophic N2-Fixing Bacteria의 형태학적 특성)

  • In this study, we compared the levels of methylotrophic bacterial community diversity in the leaf, stem, grain, root and rhizosphere soil sainples of four rice cultivars collected from three regions of Korea. Thirty five pigmented and five non-pigmented isolates showing characteristic growth on methanul were obtained. When phylotypes were defined by performing numerical analysis of 42 characteristics, four distinct clusters were formed. While two clusters, I and IV diverged on the basis of nitrate and nitrite reduction, other two clusters, comprising only pink pigmented colonies, diverged on the basis of cellulase activity. Out of the two reference strains used in the analysis, Methyhbacterium extorquens AM1 diverged from all the clusters and M. fujisawaense KACC 10744 grouped under cluster III. All the isolates were positive for urease, oxidase, catalase and pectinase activity and negative for indole production, MR and VP test, $H_2S$ production, starch, and casein hydrolysis. No clusters were found to possess thermotolerant isolates, as no growth of the isolates was observed at $45^{\circ}C$. Two strains in cluster I were found to possess gelatin hydrolysis and methane utilizing properties respectively. Most of the isolates in all the four clusters utilized monosaccliarides, disaccharide and polyols as carbon source. Six isolates showed considerable nitrogenase activity ranging from 86.2 to $809.9nmol\;C_2H_4\;h^{-1}\;mg^{-1}$ protein.

Simple one-step synthesis of carbon nanoparticles from aliphatic alcohols and n-hexane by stable solution plasma process

  • This paper examines a simple one-step and catalyst-free method for synthesizing carbon nanoparticles from aliphatic alcohols and n-hexane with linear molecule formations by using a stable solution plasma process with a bipolar pulse and an external resistor. When the external resistor is adopted, it is observed that the current spikes are dramatically decreased, which induced production of a more stable discharge. Six aliphatic linear alcohols (methanol-hexanol) containing carbon with oxygen sources are studied as possible precursors for the massive production of carbon nanoparticles. Additional study is also carried out with the use of n-hexane containing many carbons without an oxygen source in order to enhance the formation of carbon nanoparticles and to eliminate unwanted oxygen effects. The obtained carbon nanoparticles are characterized with field emission-scanning electron microscopy, energy dispersive X-ray spectroscopy, and Raman spectroscopy. The results show that with increasing carbon ratios in alcohol content, the synthesis rate of carbon nanoparticles is increased, whereas the size of the carbon nanoparticles is decreased. Moreover, the degree of graphitization of the carbon nanoparticles synthesized from 1-hexanol and n-hexane with a high carbon (C)/oxygen (O) ratio and low or no oxygen is observed to be greater than that of the carbon nanoparticles synthesized from the corresponding materials with a low C/O ratio.

Artificial Control of ZnO Nanorods via Manipulation of ZnO Nanoparticle Seeds (산화아연 나노핵의 조작을 통한 산화아연 나노로드의 제어)

  • Synthesis and characterization of ZnO structure such as nanowires, nanorods, nanotube, nanowall, etc. have been studied to multifunctional application such as optical, nanoscale electronic and chemical devices because it has a room-temperature wide band gap of 3.37eV, large exiton binding energy(60meV) and various properties. Various synthesis methods including chemical vapor deposition (CVD), physical vapor deposition, electrochemical deposition, micro-emulsion, and hydrothermal approach have been reported to fabricate various kinds of ZnO nanostructures. But some of these synthesis methods are expensive and difficult of mass production. Wet chemical method has several advantage such as simple process, mass production, low temperature process, and low cost. In the present work, ZnO nanorods are deposited on ITO/glass substrate by simple wet chemical method. The process is perfomed by two steps. One-step is deposition of ZnO seeds and two-step is growth of ZnO nanorods on substrates. In order to form ZnO seeds on substrates, mixture solution of Zn acetate and Methanol was prepared.(one-step) Seed layers were deposited for control of morpholgy of ZnO seed layers by spin coating process because ZnO seeds is deposited uniformly by centrifugal force of spin coating. The seed-deposited samples were pre-annealed for 30min at $180^{\circ}C$ to enhance adhesion and crystallinnity of ZnO seed layer on substrate. Vertically well-aligned ZnO nanorods were grown by the "dipping-and-holding" process of the substrates into the mixture solution consisting of the mixture solution of DI water, Zinc nitrate and hexamethylenetetramine for 4 hours at $90^{\circ}C$.(two-step) It was found that density and morphology of ZnO nanorods were controlled by manipulation of ZnO seeds through rpm of spin coating. The morphology, crystallinity, optical properties of the grown ZnO nanostructures were carried out by field-emission scanning electron microscopy, high-resolution electron microscopy, photoluminescence, respectively. We are convinced that this method is complementing problems of main techniques of existing reports.

Microwave Mediated Production of FAME from Waste Cooking Oil : Optimization of Process Parameters by RSM (마이크로웨이브를 이용한 폐식용유로부터 FAME의 제조 : RSM에 의한 공정변수 최적화)

  • In this study, the optimization of the biodiesel production process from waste oil using microwave with response surface methodology (RSM) was conducted. The microwave irradiation time and power in addition to the alcohol/oil mole ratio were chosen as process parameters. Also the fatty acid methyl ester (FAME) content (over 96.5%) and kinematic viscosity (1.9~5.5 cSt) were selected as response values. From basic experiments, the range of quantitative factors were set as following; 4~6 min, 400~600 W, and 7~9 for the microwave irradiation time and power, and alcohol/oil molar ratio, respectively. The optimum conditions for the methanolysis were 5.0~5.1 min, 481.3~525.5 W, 7.9~8.4, and 2.0 or 3.0 mg KOH/g for the microwave irradiation time and power, methanol/oil molar ratio, and each acid value, respectively. The FAME content and kinematic viscosity were predicted as 97.49~96.34% and 4.01~4.12 cSt, respectively, under the condition above. Under the optimum experimental conditions, the results showed that the FAME content and kinematic viscosity of 97.82~96.42% and 4.07~4.16 cSt, respectively were measured and the mean error rates were 0.22% and 0.98%, respectively.

Study of Optimized Extraction Conditions for Simultaneous Anti-inflammatory and Antioxidant Activity of Artemisia iwayomogi using Response Surface Methodology (반응표면분석을 이용한 한인진의 항염 및 항산화 복합 활성 최적 추출 조건에 관한 연구)

  • This study was conducted to find a solvent, a extraction method and optimized conditions for Artemisia iwayomogi extract. which is effective to anti-inflammatory and antioxidant activity. A. iwayomogi extract by distilled water, ethanol, methanol, hexane and ethyl acetate using ultrasonic, high-pressure and supercritical extraction was investigated for NO inhibitory and DPPH radical scavenging activity. Extract obtained by ethanol and ultrasonic extraction method showed high NO production inhibitory activity, DPPH free radical scavenging activity and yield. Response surface methodology (RSM) was applied to find a optimized ultrasonic extraction conditions. Results showed that the optimum conditions for the higher yield were ethanol solvent of 45.71% concentration with extraction time and ultrasonic power of 63.33min and 308.84 W, respectively. This condition predicted 15.85% yield, but real yield was $16.40{\pm}0.28%$. The optimum conditions for simultaneous anti-inflammatory and antioxidant activity were established as ethanol concentration (80.81%), extraction time (90.00 min) and ultrasonic power (400.00 W). NO production inhibitory and antioxidant activity were $89.77{\pm}1.37%$ and $60.12{\pm}0.39%$, respectively. These results showed similar to the predicted values of 94.54%, 58.03% respectively.

Anti-oxidant, Anti-inflammation and Anti-microbial Effects of Hoangtonogak Plus Extracts (황토노각플러스 추출물의 항산화, 항염 및 항미생물 효능)

  • This study evaluated the possibility of Hoangtonogak Plus extracts as a bioactive ingredients for cosmetic products. Methanol(MN) and hot-water(WN) extracts were analysed by DPPH/ABTS radical scavenging activity, FRAP value for anti-oxidant activity, MTT assay for cell viability, inhibition of NO production and iNOS protein expression for anti-inflammatory effect, paper disc diffusion method for anti-microbial activity against Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli.. The contents of total polyphenol of MN and WN extracts were 2.92±0.01 mgGAE/g and 1.67±0.02 mgGAE/g, respectively. DPPH, ABTS and FRAP values of MN extracts were higher than WN at each concentration. No significant cytotoxicity was observed in RAW264.7 cells. Furthermore, NO production of MN and WN at 1 mg/mL concentration was measured as 11.69 μM, 20.4 μM, respectively. In addition, MN extracts showed anti-microbial effect only on S. epidermidis. Also MN extracts suppressed iNOS protein level in a concentration-dependent manner. According to our results, the MN extracts demonstrated its potential as a natural source of antioxidant with anti-microbial and anti-inflammatory properties.

Ficus vasculosa Wall. ex Miq. Inhibits the LPS-Induced Inflammation in RAW264.7 Macrophages

  • Ficus vasculosa Wall. ex Miq. (FV) has been used as a herbal medicine in Southeast Asia and its antioxidant activity has been shown in previous studies. However, it has not yet been elucidated whether FV exerts anti-inflammatory effects on activated-macrophages. Thus, we aimed to evaluate the ameliorative property of FV methanol extract (FM) on lipopolysaccharide (LPS)-induced inflammatory responses and the underlying molecular mechanisms in RAW264.7 macrophages. The experimental results indicated that FM decreased the production of inflammatory mediators (NO/PGE2) and the mRNA/protein expression of iNOS and COX-2 in LPS-stimulated RAW264.7 cells. FM also reduced the secretion of interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α and monocyte chemoattractant protein (MCP)-1 in LPS-stimulated RAW264.7 cells. Results also demonstrated that FM improved inflammatory response in LPS-stimulated A549 airway epithelial cells by inhibiting the production of cytokines, such as IL-1β, IL-6 and TNF-α. In addition, FM suppressed MAPK activation and NF-κB nuclear translocation induced by LPS. FM also upregulated the mRNA/protein expression levels of heme oxygenase-1 and the nuclear translocation of nuclear factor erythroid 2-related factor 2 in RAW264.7 cells. In an experimental animal model of LPS-induced acute lung injury, the increased levels of molecules in bronchoalveolar lavage (BAL) fluid were suppressed by FM administration. Collectively, it was founded that FM has anti-inflammatory properties on activated-macrophages by suppressing inflammatory molecules and regulating the activation of MAPK/NF-κB signaling.

Anti-inflammatory Effects of Houttuynia cordata and Lespedeza cuneata on Lipopolysaccharide-stimulated RAW264.7 Cells (마우스 대식세포 RAW264.7에서 어성초와 야관문의 항염증 효과)

  • In the present study, we prepared hot water extracts and the subsequent organic solvent fractions of methanol extracts of Houttuynia cordata (HC) and Lespedeza cuneata (LC), and investigated their anti-inflammatory effects on lipopolysaccharide-stimulated RAW264.7 cells. Among the treated samples, hexane, chloroform, and ethyl-acetate fractions of HC and LC inhibited nitric oxide (NO) production in a dose-dependent manner, and decreased inducible nitric oxide synthase (iNOS) protein expression. And, we analyzed the flavonoid contents of the ethyl-acetate fraction of HC and LC, and chose apigenin for the further experiments because apigenin was one of flavonoids commonly found in HC and LC. Apigenin dramatically inhibited NO production in a dose-dependent manner without affecting cell viability and decreased iNOS and cyclooxygenase-2 (COX-2) expression. In addition, apigenin suppressed the phosphorylation of p38 and Jun N-terminal kinase (JNK) indicating that apigenin exerts anti-inflammatory activity via the mitogen-activated protein kinase (MAPK) signaling pathway. Subsequently, we conducted RNA-sequencing analysis to detect differentially expressed genes upon apigenin treatment. Among the down-regulated genes, four cytokine genes (interleukin (IL)-1α, IL-1β, IL-6, and colony stimulating factor 2 (CSF2)) were selected for the further analysis, and the reduction of their expression by apigenin was confirmed with quantitative real-time polymerase chain reaction. Overall, our results suggest that Houttuynia cordata and Lespedeza cuneata have the anti-inflammatory effects and apigenin can be the one of key molecules responsible for their anti-inflammatory activities.

Study on Enhancement of Ammonia Generation for Effective Collision Frequency (유효충돌빈도를 고려한 암모니아 생성 증대기법 연구)

  • Research, such as developing alternative energy in the transportation field, including aviation, is being actively conducted to solve the issue of current climate change. Interest in ammonia fuel as a carbon free energy (CFE) source is increasing due to the ease of liquefaction and transportation and similarity in energy density to that of methanol. However, explosiveness and toxicity of ammonia make it difficult to handle. Therefore, in this study, stable ammonia production was attempted using relatively easy-to-handle urea water solution (UWS). High temperature steam was used to promote the hydrolysis of ammonia. In order to determine the causes for ammonia production below the theoretical equivalent ratio, it was suggested that there were not enough collisions to promote the hydrolysis based on the kinetic theory of gases. The hydrolysis of unreacted isocyanic acid (HNCO) was tested according to the change in water supply. As a result, an increased amount of ammonia produced was confirmed. The increased amount of ammonia produced in a certain section was dependent on the steam temperature and the flow rate of water supplied.
