BACKGROUND/OBJECTIVES: Epigenetic regulation by nutrients can influence the development of specific diseases. This study sought to examine the effect of individual nutrients and nutrient families in the context of preventing chronic metabolic diseases via epigenetic regulation. The inhibition of lipid accumulation and inflammation by nutrients including proteins, lipids, vitamins, and minerals were observed, and histone acetylation by histone acetyltransferase (HAT) was measured. Correlative analyses were also performed. MATERIALS/METHODS: Nutrients were selected according to information from the Korean Ministry of Food and Drug Safety. Selected nutrient functionalities, including the attenuation of fatty acid-induced lipid accumulation and lipopolysaccharide-mediated acute inflammation were evaluated in mouse macrophage Raw264.7 and mouse hepatocyte AML-12 cells. Effects of the selected nutrients on in vitro HAT inhibition were also evaluated. RESULTS: Nitric oxide (NO) production correlated with HAT activity, which was regulated by the amino acids group, suggesting that amino acids potentially contribute to the attenuation of NO production via the inhibition of HAT activity. Unsaturated fatty acids tended to attenuate inflammation by inhibiting NO production, which may be attributable to the inhibition of in vitro HAT activity. In contrast to water-soluble vitamins, the lipid-soluble vitamins significantly decreased NO production. Water- and lipid-soluble vitamins both exhibited significant inhibitory activities against HAT. In addition, calcium and manganese significantly inhibited lipid accumulation, NO production, and HAT activity. CONCLUSIONS: Several candidate nutrients and their family members may have roles in the prevention of diseases, including hepatic steatosis and inflammation-related diseases (i.e., nonalcoholic steatohepatitis) via epigenetic regulation. Further studies are warranted to determine which specific amino acids, unsaturated fatty acids and lipid-soluble vitamins or specific minerals influence the development of steatosis and inflammatory-related diseases.
광주지역의 한 종합병원 수검자를 대상으로 백혈구 수와 대사증후군과의 관련 요인을 분석하고자 단면 연구를 시행한 결과 대사증후군의 유병률은 남성이 25.2%, 여성은 13.3%로 나타났으며, 연령별 유병률은 남성은 40대에서, 여성은 60세 이상에서 가장 높은 유병률을 보였고 남녀 모두 백혈구수가 증가 할수록 대사증후군의 유병률이 높게 나타났다. 대사증후군과 백혈구수와의 로지스틱 회귀분석의 결과에서는 남, 여 모두 백혈구 수가 증가 할수록 대사증후군의 위험도가 증가하였다. 결론적으로 본 연구에서는 비록 정상범위 일지라도 증가된 백혈구 수는 대사증후군과 연관성이 있다는 것을 확인할 수 있으며, 대사증후군 기준을 충족시키는 항목수가 많을수록 백혈구 수는 증가하는 경향을 보였고, 백혈구 수를 네 개의 군으로 나누었을 때 백혈구 구분수가 증가할수록 대사증후군의 유병률도 증가하고 있었다. 백혈구수와 백혈구 감별계산이 대사증후군을 예측하는 인자로 가치가 있는지는 광범위한 대단위 연구와 시간적 순서에 의해 인과관계를 규명하는 코호트연구가 필요하다고 생각된다.
Purpose : In this study, using Oyster Shell Thermal Therapy for metabolic diseases, we analyzed the effect of immune and inflammation-related variables and respiratory health-related variables of test subjects to verify the effect of improving respiratory health. Methods : In this study, 26 patients with metabolic diseases were divided into an experimental group (N=13) and a control group (N=13). After Oyster Shell Thermal Therapy (four weeks/three times a week/1 hour per time), metabolic disease-related variables and immune and respiratory health-related variables were measured and compared between the two groups. The conclusion of this study is as follows: Results : After the four-week Oyster Shell Thermal Therapy, in terms of changes in the metabolic disease-related variables, the control group exhibited a higher increase in TC and LDL-C levels than the experimental group. In the case of glucose, the experimental group showed a decrease after the experiment (p<.05). After the four-week thermotherapy, a statistically significant interactive effect occurred in natural killer (NK) cells among the immune-related variables. According to the results of a post-experimental analysis, the control group showed a higher decrease in NK cells than the experimental group (p<.05). After the 4-weeks thermotherapy, the experimental group showed a greater increase in maximum oxygen intake of the respiratory health-related variables than the control group. Conclusion : Based on a comprehensive review of the study results, the subjects who underwent the four-week Oyster Shell Thermal Therapy exhibited positive physical changes in metabolic disease-related variables as well as immune and respiratory health-related variables, which demonstrates the effectiveness of Oyster Shell Thermal Therapy on immune and respiratory health. Accordingly, it is recommended to conduct long-term Oyster Shell Thermal Therapy with various models in terms of the size and shape.
Elevated serum concentration of inflammation markers is known as an independent risk factor of metabolic syndrome (MS) and dietary intake is an important factor to control MS. The purpose of this study was to investigated the hypothesis that inflammatory indices are associated with dietary intake and diet quality index-international (DQI-I) in subjects with MS. A cross-sectional study was conducted on 156 men and 73 postmenopausal women with MS, defined by three or more risk factors of the modified Adult Treatment Panel III criteria. Serum levels of high sensitive C-reactive protein (hs-CRP), adiponectin were examined and nutrients intake and DQI-I were assessed using a semiquantitative food frequency questionnaire. The total DQI-I score was significantly higher in female subjects ($65.87{\pm}9.86$) than in male subjects ($62.60{\pm}8.95$). There was a positive association between hs-CRP and polyunsaturated fatty acid intake (p < 0.05) and a negative association between adiponectin and lipid (p < 0.05), total sugar (p < 0.01), and total fatty acids (p < 0.05). When the subjects were divided into 5 groups by quintile according to serum adiponectin and hs-CRP level, there was no association between DQI-I score and hs-CRP levels. Moderation score of DQI-I was significantly higher in highest quintile group than the lower quintile groups. Therefore, our results provide some evidence that dietary intake and diet quality are associated with inflammation markers and dietary modification might be a predictor to decrease risk for metabolic syndrome complications. However further research is needed to develop the dietary quality index reflecting the inflammatory change by considering the dietary habit and pattern of Koreans.
면역 체계와 대사 체계는 항상성을 유지하는데 중요한 요소이다. 면역 반응과 대사 조절은 연관성이 높아, 정상적인 대사가 교란되면 대사 질환이 발생하며, 면역 반응에도 변화가 발생하였다. 마찬가지로, 비만은 면역 반응과 높은 관련이 있다. 에너지 대사의 불균형으로 발생하는 비만은 인슐린 저항성, 제2형 당뇨병, 지방간 질환, 동맥경화증, 고혈압 등의 대사 질환과 관련이 있다. 알려진 바로는, 비만은 낮은 수준의 염증이 만성화된 상태가 특징이다. 비만 환경에서, 면역세포의 미세 환경은 대식세포, 자연살해세포, T세포 같은 면역세포의 독특한 활성화 표현형에 의해 염증성이 되었다. 또한, 면역 세포는 세포 간의 기전, 사이토카인을 매개하는 기전을 통해 상호작용하여 비만으로 인한 염증 반응을 강화한다. 이러한 현상은 기존의 췌장 리파아제나 알파-아밀라아제 같은 체내 효소의 억제나 지방전구세포의 분화를 억제를 표적으로 하는 일반적인 비만의 약리학적 치료 외에 면역세포 활성화 조절을 표적으로 하는 비만의 약리학적 치료 전략을 시사한다. 본 논문에서는 대식세포, 자연살해세포, T세포의 활성화 표현형과 비만 환경 내이들의 양상에 대해 정리하였다. 또한, 본 논문에서는 현재까지 확인된 면역세포의 활성화 조절을 통한 비만을 완화하는 약리학적 물질에 대해서 정리하였다.
비만은 감염원 없이 진행되는 낮은 수준의 만성적인 전신성 염증상태로 간주되며, 만성대사질환의 원인이 된다. 본 연구에서는 미나리를 발효하여 만든 식초가 3T3-L1 지방전구세포의 분화 및 RAW 264.7 세포에서의 항염증 효과에 미치는 영향을 확인하였다. MDI (IBMX, dexamethasone, insulin)에 의해 분화가 유발된 3T3-L1 지방전구세포에 대한 미나리 식초의 분화 억제능을 Oil-red-O staining을 통해 확인하였으며, western blot법을 통해 지방세포 형성 시 중요한 전사인자인 peroxisome proliferator-activated receptor-${\gamma}$ (PPAR-${\gamma}$)와 CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$)의 발현을 감소시키는 것으로 나타났다. 또한, 미나리 식초가 lipopolysaccharide (LPS)에 의해 염증이 유발 된 RAW 264.7 세포의 nitric oxide (NO) 생성을 억제시켰으며, inducible NO synthase (iNOS)와 cyclooxygenase-2 (COX-2)의 단백질 발현을 감소시키는 것을 확인하였다. 이러한 결과들을 통해 미나리 발효 식초는 지방세포 분화와 염증 억제 효과를 가지고 있음을 확인하였다. 따라서 미나리 발효 식초는 대사성 질환을 예방할 수 있는 천연물 소재로 이용 가능할 것으로 생각된다.
목 적: 양전자방출단층검사(Fluorine-18 2-Deoxy-D-Glucose positron emission tomography : $^{18}F-FDG$ PET)는 가시적 영상분석과 반정량적 섭취계수 분석 방법을 사용하여 폐의 염증성 질환과 폐암을 구별 및 진단하는 데 유용하다. 일반적으로 표준화섭취계수(Standardized Uptake Value : SUV)가 폐 질환의 진단에 사용되지만, 이는 폐 조직에 있어서 반정량적, 정량적 분석을 통한 동적인 정보를 포함하지 않는다. 그러므로, 본 연구는 $^{18}F-FDG$ PET 검사에서 폐 질환의 동적 섭취 분석이 보다 정확한 대상질환의 진단을 유용하게 할 수 있을 것이라는 가정하에 시간-방사능 곡선(Time-Activity Curve: TAC), 표준화섭취계수-동적곡선(Standardized Uptake Value-Dynamic Curve : SUV-DC), 패트락 분석법(Patlak analysis : Glucose Metabolic Rate(MRGlu))을 토대로 얻은 분석방법을 이용하여 진단적 유용성을 평가하였다. 대상 및 방법: 폐 질환을 가진 17명의 환자를 대상을 하여, 각각 60분간 Dynamic $^{18}F-FDG$ PET검사를 시행하였다. 획득한 정보를 통하여 폐질환의 형태를 따라 관심영역(Region of Interest: ROI)를 그린 후, 반정량적 분석인 TAC, SUV-DC와 정량적 분석인 Patlak analysis를 각각의 군에 따라 분석하여 비교 분석하였다. 결 과: Dynamic $^{18}F-FDG$ PET을 통한 분석결과, TAC 형태는 초기 혈류상에서 폐의 염증성 질환과 폐암의 구분이 어려웠으나, 시간이 지날수록 폐암의 곡선이 염증성 질환의 경우보다 확연히 증가하였다. SUV-DC의 경우는 TAC 형태와 거의 유사한 형태를 가졌다. Patlak analysis 분석결과, 대동맥 영역에서는 폐의 염증성 질환이 폐암보다 높은 혈액 방사능을 보였으나 시간이 지남에 따라 염증성 질환의 혈액 방사능이 극히 낮아졌다. 반면, 병변 조직에서는 폐암이 가장 높은 섭취를 보였으며 폐의 염증성 질환은 중간 정도에 머물렀다. 결 론: TAC와 SUV-DC 분석에서 폐암과 폐의 염증성 질환은 각각 고유한 그래프 형태를 띠었으며 Patlak analysis에서 역시 주목할 만한 차이를 보였다. 따라서 이러한 분석법을 토대로 좀더 깊이 있는 연구가 진행된다면 비침습적으로 폐의 질환을 구별하는 데 보다 적절하고 유용한 진단적 정보를 얻을 수 있을 것이다.

Aged garlic extract (AGE) is known to have a protective effect against immune system, endothelial function, oxidative stress and inflammation. We examined the effects of exercise with and without aged garlic extract administration on body weight, lipid profiles, inflammatory cytokines, and oxidative stress marker in high-fat diet (HFD)-induced obese rats. Forty-five Sprague-Dawley rats were fed either a HFD (HFD, n = 40) or a normal diet (ND, n = 5) for 6 weeks and thereafter randomized into ND (n = 5), HFD (n = 10), HFD with AGE (n = 10), HFD with Exercise (n = 10), or HFD with Exercise+AGE (n = 10) for 4 weeks. AGE groups were administered at a dose of 2.86 g/kg body weight, orally. Exercise consisted of running 15-60 min 5 days/week with gradually increasing intensity. AGE (P<0.01), Exercise, and Exercise+AGE (P<0.001) attenuated body weight gain and food efficiency ratio compared to HFD. Visceral fat and liver weight gain were attenuated (P<0.05) with all three interventions with a greater effect on visceral fat in the Exercise+AGE than AGE (P<0.001). In reducing visceral fat (P<0.001), epididymal fat (P<0.01) and liver weight (P<0.001), Exercise+AGE was effective, but exercise showed a stronger suppressive effect than AGE. Exercise+AGE showed further additive effects on reducing visceral fat and liver weight (P<0.001). AGE significantly attenuated the increase in total cholesterol and low-density lipoprotein-cholesterol compared with HFD (P<0.05). Exercise+AGE attenuated the increase in triglycerides compared with HFD (P<0.05). Exercise group significantly decrease in C-reactive protein (P<0.001). These results suggest that AGE supplementation and exercise alone have anti-obesity, cholesterol lowering, and anti-inflammatory effects, but the combined intervention is more effective in reducing weight gain and triglycerides levels than either intervention alone.
Objective: Interleukin-6 (IL-6) is a T cell-derived B cell stimulating factor which plays an important role in inflammatory diseases. In this study, the pharmacokinetic properties of LMT-28 including physicochemical property, in vitro liver microsomal stability and an in vivo pharmacokinetic study using BALB/c mice were characterized. Methods: LMT-28 has been synthesized and is being developed as a novel therapeutic IL-6 inhibitor. The physicochemical properties and in vitro pharmacokinetic profiles such as liver microsomal stability and Madin-Darby canine kidney (MDCK) cell permeability assay were examined. For in vivo pharmacokinetic studies, pharmacokinetic parameters using BALB/c mice were calculated. Results: The logarithm of the partition coefficient value (LogP; 3.65) and the apparent permeability coefficient values (Papp; 9.7×10-6 cm/s) showed that LMT-28 possesses a moderate-high cell permeability property across MDCK cell monolayers. The plasma protein binding rate of LMT-28 was 92.4% and mostly bound to serum albumin. The metabolic half-life (t1/2) values of LMT-28 were 15.3 min for rat and 21.9 min for human at the concentration 1 μM. The area under the plasma drug concentration-time curve and Cmax after oral administration (5 mg/kg) of LMT-28 were 302±209 h·ng/mL and 137±100 ng/mL, respectively. Conclusion: These data suggest that LMT-28 may have good physicochemical and pharmacokinetic properties and may be a novel oral drug candidate as the first synthetic IL-6 inhibitor to ameliorate mammalian inflammation.
Baicalin, a main constituent of the rhizome of Scutellaria baicalensis, is metabolized to baicalein and oroxylin A in the intestine before its absorption. To understand the role of intestinal microflora in the pharmacological activities of baicalin, we investigated its anti-inflammatory effect in mice treated with and without antibiotics. Orally administered baicalin showed the anti-inflammatory effect in mice than intraperitoneally treated one, apart from intraperitoneally administered its metabolites, baicalein and oroxylin A, which potently inhibited LPS-induced inflammation. Of these metabolites, oroxylin A showed more potent anti-inflammatory effect. However, treatment with the mixture of cefadroxil, oxytetracycline and erythromycin (COE) significantly attenuated the anti-inflammatory effect of orally administered baicalin in mice. Treatment with COE also reduced intestinal bacterial fecal ${\beta}$-glucuronidase activity. The metabolic activity of human stools is significantly different between individuals, but neither between ages nor between male and female. Baicalin was metabolized to baicalein and oroxylin A, with metabolic activities of $1.427{\pm}0.818$ and $1.025{\pm}0.603$ pmol/min/mg wet weight, respectively. Baicalin and its metabolites also inhibited the expression of pro-inflammatory cytokines, TNF-${\alpha}$ and IL-$1{\beta}$, and the activation of NF-${\kappa}B$B in LPS-stimulated peritoneal macrophages. Of them, oroxylin A showed the most potent inhibition. Based on these findings, baicalin may be metabolized to baicalein and oroxylin A by intestinal microflora, which enhance its anti-inflammatory effect by inhibiting NF-${\kappa}B$ activation.

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
