Thrips in Medicinal Crops in Korea:Identification and Their Damages (국내 주요 약용작물에 발생하는 총채벌레의 종류와 피해)

  • Damages of medicinal crops by thrips and identification of thrips species were investigated by periodic surveys on major medicinal crops, Platycodon grandiflorum, Schizandra chinensis, Codonopsis lanceolata, Lycium chinense, Bupleurum falcatum, Ledebouriella seseloides, Angelica gigas, Glycyrrhiza uralensis, Liriope platyphylla, Atractyloides sp., once or twice a month in Suwon, Eumseong, Jecheon, Pyeongchang, Chyungyang from May to October in 2012. Thrips and their damages were found at all of the medicinal crops investigated, mostly occurring at early growing stages of each medicinal crop and causing apical meristem region withered, thereby inhibiting the growth of plants. For species identification of thrips collected from each medicinal crops, DNA from each individual was extracted and ITS2 and COI regions were amplified by PCR. As a result, Frankliniella intonsa, Thrips tabaci, and Megalurothrips distalis have been identified as dominant species on medicinal crops in open field, whereas Frankliniella occidentalis was restricted to Lycium chinense in which protected cultivation. This study suggests that the occurrence of thrips species on medicinal crops seems to be affected by host specificity and environmental factors such as cultivation types.

  • Glyphosate(N-[phosphonomethyl]glycine) applied to the assimilate-exporting leaves or sprayed to the whole plants of tomato(Lycopersicon esculentum Mil var. Moneymaker). Glyphosate induced the rapid accumulation of shikimic acid within 24 h. The accumulation of shikimic acid companied with chlorophyll loss in meristematic leaves, i.e. apical leaves. The chlorosis was acropetal in apical region of young growing leaf. The degradation of chlorophyll seems to be a secondary or tertiary effect of glyphosate. However, the level of shikimic acid accumulated was reduced except for roots and apical leaves from 5 days after treatment. The accumulating levels are considerably differed through the applicated regions. The level of shikimic acid is highest at the apical meristem 4 days after the application to 3rd old leaf.

Induction of Chlorophyll Deficient Mutant Plant of Cymbidium kanran by EMS Treatment (EMS처리에 의한 한란의 엽록소 결핍 돌연변이 식물체의 유도)

  • Chlorophyll mutants were produced by treating the rhizome of Cymbidium kanran with mutagen, EMS(ethyl methan sulfonate). The germination ratio of Cymbidium kanran seeds was 5.5 times higher when the seeds were treated with ultrasonic treatment for 20 minutes than untreated control. Fifty to sixty percent of the rhizomes became dark brown when they were cultured in a liquid growth medium containing 0.2% EMS for three weeks. When the dark-brown rhizomes were cultured in a solified MS medium, new rhizomes were developed from a part of the old ones. Chlorophyll mutant rhizomes were obtained from a meristem tissue by a subculturing the cuts of these new rhizomes for a year. Of the chlorophyll mutants, a zigzag-striped type of rhizome was dominant and light-yellow and albino ones were also produced. While the zigzag-striped type rhizomes were differentiated into green and striped plant, the light yellow and the white rhizomes produced yellow-striped and albino plants repectively.These results indicate that the EMS treatment on the rhizome is an effective means to induce a chlorophyll mutant. We believe that this method may be useful to produce variegated plants chlorophyll mutants from other orchids.

  • This study was conducted to obtain some basic information needed for the propagational system of high quality garlic trough the culture of healthy tissues. non shoot-tips of bulbil obtained in mid May were cultured on MS medium containing 8% sucrose supplemented with 0.1 mg/L NAA, in vitro bulbli were formed, but the shoots were formed at the early to middle in June. Multiple shoots were induced by the culture of receptacles on MS medium supplemented with 0.1 mg/L NAA and 10mg/L BA..Among the flower bud, bulbil and receptacle, receptacle showed most suitable in terms of shoot formation efficiency, More than 50 shoots per single involucre were produced under the optimum condition. Results indicate that in vitro culture of involucre has a high potential for the micropropagation of high quality seed bulbs.

  • This experiment was conducted to find out the optimum cultural conditions for the propagation of M.9 apple rootstocks such as EMLA M.9 and NAKB T-337 in vitro. Murashige and Skoog (MS) medium supplemented with $1.0mg{\cdot}L^{-1}$ BA, $0.1mg{\cdot}L^{-1}$ IAA, $30g{\cdot}L^{-1}$ sucrose, and $8g{\cdot}L^{-1}$ agar was suitable for shoot proliferation. Removing of apical meristem and horizontal placing of explants on medium increased shoot proliferation significantly. The best rooting was obtained on 1/2 MS medium supplemented with 0$0.5mg{\cdot}L^{-1}$ IBA, $20g{\cdot}L^{-1}$ sucrose, and $8g{\cdot}L^{-1}$ agar.

A New Cymbidium Orchid Variety "Daegook" bred by In Vitro Mutagenesis (조직배양 돌연변이 기술에 의한 Cymbidium 난 품종 "대국" 육종)

  • Cymbidium is one of the largest genus in the orchid family and a number of hybrids have been bred in the world. During mass-propagating the Cymbidium "Dongyang" using the meristem culture technology, a useful leaf mutant was selected from the protocom like bodies. The new Cymbidium variety by in vitro mutangesis from "Dongyang" was named as 'Daegook' in 1998. Compared to Dongyang, "Daegook" mutant has white or yellow stripes along the margin of leaves and flowers. The plant length of "Daegook" was shorter than "Dongyang" and the mean length and width of leaf in "Daegook" was 40 cm and 1.6 cm, respectively. The new variety, "Daegook", is expected to be a popular Cymbidium variety among consumer as a ornamental orchid mutant for pot culture by its fine and unique stripes and growth characters.

Overexpression of RICE FLOWERING LOCUS T 1 (RFT1) Induces Extremely Early Flowering in Rice

  • RICE FLOWERING LOCUS T 1 (RFT1) is a major florigen that functions to induce reproductive development in the shoot apical meristem (SAM). To further our study of RFT1, we overexpressed the gene and examined the expression patterns of major regulatory genes during floral transition and inflorescence development. Overexpression induced extremely early flowering in the transgenics, and a majority of those calli directly formed spikelets with a few spikelets, thus bypassing normal vegetative development. FRUITFULL (FUL)-clade genes OsMADS14, OsMADS15, and OsMADS18 were highly induced in the RFT1-expressing meristems. OsMADS34 was also induced in the meristems. This indicated that RFT1 promotes the expression of major regulatory genes that are important for inflorescence development. RFT1 overexpression also induced SEPALLATA (SEP)-clade genes OsMADS1, OsMADS5, and OsMADS7 in the greening calli before floral transition occurred. This suggested their possible roles at the early reproductive stages. We found it interesting that expression of OsFD1 as well as OsFD2 and OsFD3 was strongly increased in the RFT1-expressing calli and spikelets. At a low frequency, those calli produced plants with a few leaves that generated a panicle with a small number of spikelets. In the transgenic leaves, the FUL-clade genes and OsMADS34 were induced, but SEP-clade gene expression was not increased. This indicated that OsMADS14, OsMADS15, OsMADS18, and OsMADS34 act immediately downstream of RFT1.

Selection of (Ac/Ds) insertion mutant lines by abiotic stress and analysis of gene expression pattern of rice (Oryza sativar L.) (비생물학적 스트레스 관련 벼 Ac/Ds 삽입 변이체의 선발 및 유전자 발현 분석)

  • Transposon-mediated insertional mutagenesis is one of powerful strategy for assessing functions of genes in higher plants. In this report, we have selected highly susceptible and tolerance plant by screening about high salt (3% NaCl) and cold stresses ($4^{\circ}C$) from F2 seeds of 30,000 Ac/Ds insertional mutagenesis lines in rice (Oryza sativa L. cv. Dongjin). In order to identify the gene tagging, insertion of Ds element was analyzed by Southern blot and these results revealed that 19 lines were matched genotype of selected lines with phenotype from the first selected 212 lines, and 13 lines have one copy of Ds elements. The Franking Sequence Tags (FSTs) of selected mutant lines showed high similarities with the following known function genes: signal transduction and regulation of gene expression (transpoter, protease family protein and apical meristem family protein), osmotic stress response (heat shock protein, O-methyltransferase, glyceraldehyde-3-phosphate dehydrogenase and drought stress induce protein), vesicle trafficking (SYP 5 family protein) and senescence associated protein. The expression pattern of 19 genes were analyzed using RT-PCR under the abiotic stresses of 9 class; 250mM NaCl, osmotic, drought, 3% $H_2O_2$, $100{\mu}M$ ABA, $100{\mu}M$ IAA, 0.1 ppm 2,4-D, $4^{\circ}C$ cold and $38^{\circ}C$ high temperature. Isolated knock-out genes showed the positive response about 250 mM NaCl, drought, $H_2O_2$, PEG, IAA, 2,4-D, ABA treatment and low ($4^{\circ}C$) and high temperature ($38^{\circ}C$). The results from this study indicate that function of selected knock-out genes could be useful in improving of tolerance to abiotic stresses as an important transcriptional activators in rice.

Morphological Adaptation of Zostera marina L. to Ocean Currents in Korea (한국산 거머리말(Zostera marina L.)의 해류에 대한 형태적 적응)

  • The main purpose of this research is to prepare and provide basic materials for the propagational strategy of eelgrass by investigating on the morphological adaptation of Korean Zostera marina to ocean currents. An eelgrass plant mainly consists of rhizome, leaf sheath, leaves and roots. The rhizome is the horizontal stem of the plant that serves as the backbone from which the leaves and roots emerge. The leaf sheath is the bundle at the base of the leaves that holds the leaves together, protecting the meristem, the primary growth point of the shoot. Leaves originate from a meristem which is protected by a sheath at the actively growing end of the rhizome. As the shoot grows, the rhizome elongates, moving across or within the sediment, forming roots as it progresses. The aggregated leaves from the leaf sheath are found to have two cell layers on one side and multiple layers of airy tissues called aerenchyma on the other. The aerenchyma tissues are developed in multi-layered cell structures surrounding the veins which are formed in the leaf sheath. Generative shoots are made of rhizomes, which are circular or ovoidal, stem, and spathe and spadix. The transverse section of rhizome and the stem and central floral axis is found to be circular, ovoid and in the shape of convex respectively, and the vascular bundle, which is a part of transport system, has one large tube in the center and two small tubes on both sides. The layers of collenchyma cells numbered from 12 to 15 in the stem, and from 7 to 12 in the rhizome. The seed coat is composed of sclereids, small bundles of sclerenchyma tissues, which prevent the influx of sea water from the outside and help endure the environmental stress. In conclusion, alternative multi-layer structure in circular, convex type aggregated leaf base are interpreted to morphological adaption as doing tolerable elastic structure through movement of seawater. The generative shoots develop long slim stem and branches in circular or ovoidal shapes to minimize the adverse impacts of sea current, which can be interpreted as the plant's morphological adaptation to its environment.

Effect of Medium Composition and Growth Regulators on Mass Propagation of Virus-Free Plant from the Meristem Cultures of 'Jarang' Grape ('자랑' 포도 생장점 배양으로부터 무병주 식물 대량번식에 미치는 배지 구성물질과 생장 조절제의 영향)

  • This study was performed to clarify the effect of medium compositions and plant growth regulators on the shoot, root formation and growth of 'Jarang' grape for mass propagation of virus-free plant. The formation and growth of shoot were considerably favorable in half-concentration of MS medium. However, the formation of adventitious root per explants (avg. 2.1) was effective in higher concentration (two times) of MS medium. For sucrose concentration, 1% for the shoot formation, 3% for the adventitious root formation and 1% for the growth were observed as yield significant results. With the addition of 0.05% of activated carbon, the shoot growth was improved, and it was effective for the adventitious root formation and growth as well. A pH of 6.8 in the medium was the most suitable for mass propagation; the results showed significant enhancement in the number of nodes and the length of the shoot, 3.9 and 1.3 cm, respectively. The shoot growth was the most vigorous in BA 1.0 mg/L due to the impact of the growth regulator on the mass propagation in it. Consequently, 16.9 shoots per explant were formed in NAA 1.0 mg/L so good results were obtained.
