Memory Enhancing Effect of Codonopsis lanceolata by High Hydrostatic Pressure Process and Fermentation (초고압 발효 더덕 추출물의 인지능력 개선 효과)

  • Alzheimer's disease (AD), most common form of dementia is characterized that memory deficit and loss of cognitive function. The Codonopsis lanceolata (C.lanceolata) was treated by high hydrostatic pressure process and fermentation. This study was evaluated cognitive enhancing effect C.lanceolata extract by high hydrostatic pressure process and fermentation and compared with common C.lanceolata extract using Morris water maze and passive avoidance test. And their neuroprotective effect on glutamate induced oxidative stress in HT22 cell was investigated by MTT assay. High hydrostatic pressure process and fermented C.lanceolata extract (HFCE) and common C.lanceolata extract (CCE) (100 and 300 mg/kg) were administered to mice. Results showed HFCE enhanced cognitive function than CCE as shown by decrease in escape latency time. HFCE increased the latency time of the passive avoidance test compared to CCE. Furthermore, HFCE showed significant neuroprotective effect against glutamate cytotoxicity in HT22 cells. These results indicate that high hydrostatic pressure process and fermented more improve spatial cognitive ability of C. laanceolata.

A VIA-based RDMA Mechanism for High Performance PC Cluster Systems (고성능 PC 클러스터 시스템을 위한 VIA 기반 RDMA 메커니즘 구현)

  • The traditional communication protocols such as TCP/IP are not suitable for PC cluster systems because of their high software processing overhead. To eliminate this overhead, industry leaders have defined the Virtual Interface Architecture (VIA). VIA provides two different data transfer mechanisms, a traditional Send/Receive model and the Remote Direct Memory Access (RDMA) model. RDMA is extremely efficient way to reduce software overhead because it can bypass the OS and use the network interface controller (NIC) directly for communication, also bypass the CPU on the remote host. In this paper, we have implemented VIA-based RDMA mechanism in hardware. Compared to the traditional Send/Receive model, the RDMA mechanism improves latency and bandwidth. Our RDMA mechanism can also communicate without using remote CPU cycles. Our experimental results show a minimum latency of 12.5${\mu}\textrm{s}$ and a maximum bandwidth of 95.5MB/s. As a result, our RDMA mechanism allows PC cluster systems to have a high performance communication method.

Performance Analysis of Flash Memory SSD with Non-volatile Cache for Log Storage (비휘발성 캐시를 사용하는 플래시 메모리 SSD의 데이터베이스 로깅 성능 분석)

  • In a database system, updates on pages that are made by a transaction should be stored in a secondary storage before the commit is complete. Generic secondary storages have volatile DRAM caches to hide long latency for non-volatile media. However, as logs that are only written to the volatile DRAM cache don't ensure durability, logging latency cannot be hidden. Recently, a flash SSD with capacitor-backed DRAM cache was developed to overcome the shortcoming. Storage devices, like those with a non-volatile cache, will increase transaction throughput because transactions can commit as soon as the logs reach the cache. In this paper, we analyzed performance in terms of transaction throughput when the SSD with capacitor-backed DRAM cache was used as log storage. The transaction throughput can be improved over three times, by committing right after storing the logs to the DRAM cache, rather than to a secondary storage device. Also, we showed that it could acquire over 73% of the ideal logging performance with proper tuning.

  • Current rendering processors are organized mainly to process a triangle as fast as possible and recently parallel 3D rendering processors, which can process multiple triangles in parallel with multiple rasterizers, begin to appear. For high performance in processing triangles, it is desirable for each rasterizer have its own local pixel cache. However, the consistency problem may occur in accessing the data at the same address simultaneously by more than one rasterizer. In this paper, we propose a parallel rendering processor architecture resolving such consistency problem effectively. Moreover, the proposed architecture reduces the latency due to a pixel cache miss significantly. For the above two goals, effective memory organizations including a new pixel cache architecture are presented. The experimental results show that the proposed architecture achieves almost linear speedup at best case even in sixteen rasterizers.

Effect of Water and Ethanol Extracts Codonopsis lanceolata on Spatial Learning and Memory in Mice (더덕 물 추출물과 에탄올 추출물의 인지능 개선 활성 비교)

The water and 70% ethanol extracts (100, 300 and 500 mg/kg) were administered to mice. The neuroprotective effect on glutamate-induced cell death in HT22 cells was additionally investigated using MTT assay. Results showed 70% ethanol extract of Codonopsis lanceolata enhanced cognitive function than water extract, as shown by decrease in escape latency time in Morris water maze test. In passive avoidance test, 70% ethanol extract also increased the latency time compared to the water extract. Furthermore, 70% ethanol extract significantly protected neuronal cell against glutamate cytotoxicity and showed higher than neuroprotective effect of water extract. These results indicate that 70% ethanol extract more improve spatial cognitive ability and protected neuronal cells than water extract.

Comparison of scopolamine-induced cognitive impairment responses in three different ICR stocks

  • Cognitive impairment responses are important research topics in the study of degenerative brain diseases as well as in understanding of human mental activities. To compare response to scopolamine (SPL)-induced cognitive impairment, we measured altered parameters for learning and memory ability, inflammatory response, oxidative stress, cholinergic dysfunction and neuronal cell damages, in Korl:ICR stock and two commercial breeder stocks (A:ICR and B:ICR) after relevant SPL exposure. In the water maze test, Korl:ICR showed no significant difference in SPL-induced learning and memory impairment compared to the two different ICRs, although escape latency was increased after SPL exposure. Although behavioral assessment using the manual avoidance test revealed reduced latency in all ICR mice after SPL treatment as compared to Vehicle, no differences were observed between the three ICR stocks. To determine cholinergic dysfunction induction by SPL exposure, activity of acetylcholinesterase (AChE) assessed in the three ICR stocks revealed no difference of acetylcholinesterase activity. Furthermore, low levels of superoxide dismutase (SOD) activity and high levels of inflammatory cytokines in SPL-treated group were maintained in all three ICR stocks, although some variations were observed between the SPL-treated groups. Neuronal cell damages induced by SPL showed similar response in all three ICR stocks, as assessed by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, Nissl staining analysis and expression analyses of apoptosis-related proteins. Thus, the results of this study provide strong evidence that Korl:ICR is similar to the other two ICR. Stocks in response to learning and memory capacity.

The Effects of Sahyangsohapwon on the Affective Reactivity and the Acquisition of Two-way avoidance in AD Model Rats (사향소합원(麝香蘇合元)이 정서반응성(情緖反應性)과 Alzheimer's disease 모델 백서(白鼠)의 학습(學習)에 미치는 영향(影響))

  • The effects of Sahyangsohapwon on the affective reactivity of rats were studied with open-field behavior. Sample group was treated with the medicine for 8 weeks, whereas control group was treated with the vehicle. The effects of Sahyangsohapwon on the enhancement of learning and memory of AD model rats were studied with two-way avoidance task. Sample group electrically lesioned on nbM(nucleus basalis of Meynert) was treated with the medicine for 8 weeks, whereas control group with nbM lesion and sham group with the sham operation were treated with the vehicle. 1. In the open-field behavior task, the start latency from start box was measured $27.08{\pm}7.51sec$ in control group, $23.15{\pm}5.98sec$ in sample group. Rats in sample group showed a tendency of shortened latency going out to a strange place compared with those in control group, but with no statistical significance(p>0.05). 2. In the open-field behavior task, the number of locomotion crossing the grid lines was measured $84.54{\pm}3.55$ in control group, $116.93{\pm}6.41$ in sample group. There was an increased locomotion in sample group compared with control group with statistical significance(p<0.01). This can be interpreted as rats in sample group showed lowerd anxiety under a strange environment. 3. In the open-field behavior task, the rearing number was measured $7.46{\pm}0.57$ in control group, $10.13{\pm}0.95$ in sample group. There was an increased rearing in sample group compared with control group with statistical significance(p<0.05). This can also be interpreted as rats in sample group showed lowerd anxiety under a strange environment. 4. In the open-field behavior task, the number of crossing behavior was measured $5.54{\pm}1.50$ in control group, $9.20{\pm}1.67$ in sample group. There was a increasing tendency of crossing behavior in sample group compared with control group, but with no statistical significance(p<0.05). 5. In the open-field behavior task, the total activity was measured $97.54{\pm}4.70$ in control group, $136.27{\pm}792$ in sample group. There was an increased total activity in sample group compared with control group with statistical significance(p<0.01). This can also be interpreted as rats in sample group showed lowerd anxiety under a strange environment. 6. In the analysis of effects on the learning and memory in AD model rats with two-way avoidance task, the response latency was measured $6717{\pm}134msec$ in the 1st session, $5416{\pm}160msec$ in the 2nd session, $5252{\pm}148msec$ in the 3rd session in control group. It was measured $6724{\pm}155msec$ in the 1st session, $4642{\pm}139msec$ in the 2nd session, $4914{\pm}148msec$ in the 3rd session in sample group and $4357{\pm}144msec$ in the 1st session, $3125{\pm}115msec$ in the 2nd session, $3091{\pm}98msec$ in the 3rd session in sham group. There were differences between sham group and nbM lesioned groups with statistical significance in post hoc analysis(p<0.000). And in the 2nd session, there was a reduction of latency in sample group compared with control group with statistical significance (p<0.000). This showed that sample group had better learning capacity than control group. 7. In the analysis of effects on the learning and memory in AD model rats with two-way avoidance task, the number of avoidance response was measured $5.85{\pm}1.41$ in the 1st session, $14.23{\pm}2.89$ in the 2nd session, $15.69{\pm}2.56$ in the 3rd session in control group. It was measured $7.92{\pm}1.94$ in the 1st session, $16.83{\pm}2.29$ in the 2nd session, $15.42{\pm}2.81$ in the 3rd session in sample group and $14.38{\pm}1.62$ in the 1st session, $22.88{\pm}0.89$ in the 2nd session, $23.88{\pm}1.64$ in the 3rd session in sham group. There were differences between sham group and nbM lesioned groups with statistical significance in post hoc analysis(p<0.001). But between control and sample group, there was no significant difference. With the experimental results above, Sahyangsohapwon can be supposed to have the enhancing effects on the affect reactivity and learning with memory of AD model rats induced by electrolyte injury of nbM.

Effect of Gagamjeongji-hwan and Evodiae Fructus on Memory Impairment and Neuronal Damage Induced by Focal Ischemia in the Rat (국소 전뇌 허혈 모델 백서에서 가감정지환(加減定志丸)과 오수유(吳茱萸)가 기억증진과 신경세포보호에 미치는 효과)

  • This study was conducted to determine the effect of Gagamjeongji-hwan (JJH)(Jiajiandingzhi-wan) and Evoidae Fructus (EF) on learning and memory disturbance and neuronal damage induced by focal ischemia in the rat. Rats were used for testing in the following three. Morris Water Maze, Cholineacetyltransferase (ChAT) immunohistochemistry, acetylcholine esterase (AchE) histochemistry. JJH+ISCH group (ischemia-induced rats pretreated with JJH) and EF+ISCH group (ischemia-induced rats pretreated with EF) significantly reduced the latency of swimming time, compared with those of ISCH group (ischemia-induced rats) in morris water maze acquisition test. JJH+ISCH group attenuated ischemia.induced learning and memory damage in morris water maze retention test. The density of ChAT neurons of the JJH+ISCH and EF+ISCH group in the hippocampal CA1 area was increased, compared to that of SAL+ISCH group (ischemia-induced rats pretreated with SAL). The density of AchE neurons of the JJH+ISCH and EF+ISCH group in the hippocampal CA1 and CA3 area was increased, compared to that of SAL+ISCH group. These results suggest that Gagamjeongji-hwan (JJH) and Evodiae Fructus (EF) may have significant protective effects on ischemia-induced brain damage and memory impairments.

  • Recently, the demand for high performance non-volatile memory storage devices that can replace existing hard disks has been increasing in environments requiring high performance computing such as data-centers and social network services. The performance of such non-volatile memory can greatly depend on the interface between the host and the storage device. With the evolution of storage interfaces, the non-volatile memory express (NVMe) interface has emerged, which can replace serial attached SCSI and serial ATA (SAS/SATA) interfaces based on existing hard disks. The NVMe interface has a higher level of scalability and provides lower latency than traditional interfaces. In this paper, an evaluation and analysis are conducted of the performance of NVMe storage devices through various workloads. We also compare and evaluate the cost efficiency of NVMe SSD and SATA SSD.

Effect of Hfe Deficiency on Memory Capacity and Motor Coordination after Manganese Exposure by Drinking Water in Mice

  • Excess manganese (Mn) is neurotoxic. Increased manganese stores in the brain are associated with a number of behavioral problems, including motor dysfunction, memory loss and psychiatric disorders. We previously showed that the transport and neurotoxicity of manganese after intranasal instillation of the metal are altered in Hfe-deficient mice, a mouse model of the iron overload disorder hereditary hemochromatosis (HH). However, it is not fully understood whether loss of Hfe function modifies Mn neurotoxicity after ingestion. To investigate the role of Hfe in oral Mn toxicity, we exposed Hfe-knockout ($Hfe^{-/-}$) and their control wild-type ($Hfe^{+/+}$) mice to $MnCl_2$ in drinking water (5 mg/mL) for 5 weeks. Motor coordination and spatial memory capacity were determined by the rotarod test and the Barnes maze test, respectively. Brain and liver metal levels were analyzed by inductively coupled plasma mass spectrometry. Compared with the water-drinking group, mice drinking Mn significantly increased Mn concentrations in the liver and brain of both genotypes. Mn exposure decreased iron levels in the liver, but not in the brain. Neither Mn nor Hfe deficiency altered tissue concentrations of copper or zinc. The rotarod test showed that Mn exposure decreased motor skills in $Hfe^{+/+}$ mice, but not in $Hfe^{-/-}$ mice (p = 0.023). In the Barns maze test, latency to find the target hole was not altered in Mn-exposed $Hfe^{+/+}$ compared with water-drinking $Hfe^{+/+}$ mice. However, Mn-exposed $Hfe^{-/-}$ mice spent more time to find the target hole than Mn-drinking $Hfe^{+/+}$ mice (p = 0.028). These data indicate that loss of Hfe function impairs spatial memory upon Mn exposure in drinking water. Our results suggest that individuals with hemochromatosis could be more vulnerable to memory deficits induced by Mn ingestion from our environment. The pathophysiological role of HFE in manganese neurotoxicity should be carefully examined in patients with HFE-associated hemochromatosis and other iron overload disorders.
