DLPC Liposome에 미치는 도라지 추출성분의 비타민 C 첨가에 의한 항산화력 상승효과 (Enhancement of Antioxidation Effect of Platycodon grandiflorum with Vitamin C on the DLPC Liposomes)

  • 본 연구는 불포화 인지질인 DLPC liposome에 도라지(Platycodon grandiflorum A. DC, PG)의 용매별 분획물 일정 량을 가하고, 산화제 benzoyl peroxide를 가한 후 시료의 항산화 작용을 비교 분석하였다. 도라지 용매별 추출물 중 ethylether 분획물인 PGMEE와 ethylacetate 분획물인 PGMEA는 $\alpha$-tocopherol의 경우 보다 훨씬 그 산화지수가 낮아두 분획물에 의한 강한 항산화력을 측정할 수 있었으며, 잘알려진 강력한 항산화제인 BHT(butylated hydroxytoluence)의 경우와 비슷한 항산화 효과를 나타내었다. 한편 PGMEE 분획층에 비타민 C를 일정량을 첨가함으로서 상호 보강 작용에 의한 항산화 상승효과를 본 결과, Vit C를 첨가하지 않았을 때보다 산화지수가 횔씬 낮아졌다. BHT 함유의 DLPC liposome의 경우도 Vit C에 의한 비슷한 추과를 나타내었다. 이상의 결과에 의하여 도라지의 PGMEE 분획층에는 BHT와 비슷한 강한 항산화 효과를 가진 물질의 존재가 분명하다고 사료되며 비타민 C 첨가로 인한 항산화 상승 효과도 본 연구결과에 의해 뚜렷이 알 수 있었다. 이와 같은 연구에 의해 여러 질병들의 원인이 될 뿐만 아니라 세포막 손상을 일으키는 활성 산소 또는 유리기의 생성억제 등 균형된 식생활을 통한 식품 섭취의 중요성이 매우크다고 인식되며 특히 식품으로서 애용되고 있는 도라지의 인지질 세포막에 미치는 항산화 효과가 식품으로서 약이 되는 기전 규명에 기초자료가될 것을 바라는 바이다.

  • 고농도의 ethanol을 갖는 청주를 생산하기 위하여 일본양조협회 Saccharomyces cerevisiae 10호 균주로부터 ethanol 내성 균주 Saccharemyces cerevisiae SE211균주를 분리하였다. 분리 균주는 10%(v/v) ethanol이 포함되어 있는 배지에서의 성장속도가 빨랐으며, 높은 ethanol 농도에서도 성장이 가능하였다 또한 ethanol내성, 내삼투압성, 내열성 등이 원균주에 비해 우수하였다. 선발균주의 세포막 성분을 분석한 결과 sterol, phospholipid의 함량은 원균주와 차이가 없었으나, 지방산의 조성비는 차이를 보였다 또한 분리균주는 외부자극에 대한 저항성을 나타내는 물질로 알려진 세포내 trehalose의 함량이 원균주에 비해 증가하였다 따라서 선발균주의 ethanol내성 및 ethanol배지에서의 높은 성장속도는 세포막의 fatty acid및 세포내 trehalose함량을 변화시키는 능력이 높은 것에 기인한다고 생각되었다.

Effects of Local Anesthetics on Rotational Mobility of n-(9-Anthroyloxy)stearic Acid in Neuronal Membranes

  • To elucidate the molecular mechanism of pharmacological action of local anesthetics, we studied membrane actions of tetracaine, bupivacaine, lidocaine, prilocaine and procaine. Fluorescence polarization of n-(9-anthroyloxy)stearic acid (n-AS) was used to examine the effects of these local anesthetics on differential rotational mobility of different positions of the number of synaptosomal plasma membrane vesicle (SPMV) phospholipid carbon atoms. The four membrane components differed with respect to 3, 6, 9 and 16-(9-anthroyloxy)stearic acid (3-AS, 6-AS, 9-AS and 16-AP) probes, indicating that differences in the membrane fluidity might be present. Degrees of the rotational mobility of 3-AS, 6-AS, 9-AS and 16-AP were different depending on depth of hydrocarbon interior. In a dose-dependentmanner, tetracaine, bupivacaine, lidocaine, prilocaine and procaine decreased anisotropy of 3-AS, 6-AS, 9-AS and 16-AP in the hydrocarbon interior of the SPMV. These results indicate that local anesthetics have significant disordering effects on hydrocarbon interior of the SPMV, thus affecting the transport of $Na^+$ and $K^+$ in nerve membranes and leading to anesthetic action.

Structure-Activity Relationships of 9-mer Antimicrobial Peptide analogue of Protaetiamycine, 9Pbw2

  • 9Pbw2 is a 9-mer analog of protaetiamycine derived from the larvae of the beetle Protaetia brevitarsis. Previously, we designed four 9-mer peptide analogues to optimize the balance between the hydrophobicity and cationicity of the peptides and to increase bacterial cell selectivity. Among them, 9Pbw2 has high antibacterial activity without cytotoxicity. The results obtained in previous study suggest that the bactericidal action of 9Pbw2 may be attributed to the inhibition of the functions of intracellular components after penetration of the bacterial cell membrane. In order to understand structure-activity relationships, we determined the three-dimensional structure of 9Pbw2 in 200 mM DPC micelle by NMR spectroscopy. 9Pbw2 has one hydrophobic turn helix from $Trp^3$ to $Arg^8$ and positively charged residues at the N- and C-terminus. This result suggested that positively charged residues from position at the C-terminus in 9Pbw2 may be important for the primary binding to the negatively charged phospholipid head groups in bacterial cell membranes and hydrophobic residues in the middle portion face toward the acyl chains of the hydrophobic lipid in the bacterial cell membrane.

Rana box를 포함한 Brevinin-1 및 Thanatin의 구조-상생활성 상관관계 (Structure-antibiotic Acitivity Relationships of Brevinin-1 and Thanatin Containing Rana Box)

  • In order to investigate structure-antibiotic activity relationships of brevinin-1 and thanatin containing Rana box composed of basic loop formed by disulfide bridge in their arboxy terminus, thanatin, brevinin 1 and their analogues (T-B1, T-B2 and B-T) in which their Rana box sequence exchanged was designed and synthesized by the solid phase method using Fmoc-chemistry. The basic sequence of Rana box of thanatin had more significant effect on both antibacterial and antifungal activity than that of brevinin 1. The tail sequence (QRM) of thanatin was found to be important in its antibacterial and antifungal activity. Rana box sequence of brevinin-1 did not have a significant effect on its antitumor and phospholipid vesicle-aggregating activities. Brevinin-1 showed stronger $\alpha$-helical structure in the membrane-mimicking environment such as SDS micelle than thanatin. A remarkable increase in a-helicity of bervinin-1 plays more important role in antibiotic activity than that of thanatin. Furthermore, antibacterial activity of thanatin against E. coli resulted from the disruptive effect against the outer cell membrane of E. coli.

  • 동물실험을 통하여 새로이 밝혀진 항암제(anti-initiator/anti-ptomotor)인 CLA는 grilled ground beef에서 처음 분리되었다. CLA는 grilled ground beef 외에도, cheese 및 이들 관련식품에 많이 존재한다. CLA는 반추동물의 위에 서식하는 혐기성 bacteria에 의해 linoleic acid로부터 생성되며, 식품 가공 중에서도 생성된다. 이것은 또한 in vivo에서 linoleic acid의 carbon centered free radical 형태의 산화에 의해 생성되기도 한다. CLA는 아주 강력한 항산화제임이 밝혀져, 지금까지 알려지 있지 않았는 free radical에 대응하여 membrane을 보호하는 in situ defense mechanism 역할을 한다. 이는 또한 cytochrome P450 isozyme의 활성을 저해하는 반면, ODC 효소 활성 역시 저해한다. 그래서, 적어도 CLA의 이 세가지 biological activity가 CLA 항암기작에 관여하는 것으로 생각된다.

Effect of phenothiazine derivatives on the thermotropic phase transition of liposomal phospholipid membrane

  • The effect of phenothiazine derivatives on the thermotropic transition of liposomal lipid bilayer made of dipalmitoyl phosphatidylchline and dipalmitoyl phosphatidic acid was investigated with differential scanning calorimetry. The thermograms of the liposomal bilayer incorporated with levomepromazine, chlopromazine, prochloperazine, perphenazine and fluphenazine were obtained and the size of cooperative unit of the transition were calculated from the ratio of the van't Hoff enthalpy change to the calculated enthalpy change of the transition. The results showed that incorporation of phenothiazine derivatives into the liposomal bilayer reduced the transition temperature at which the transition from solid state to liquid-crystalline state occurs, and broadened the thermogram peaks. Phenothiazine derivatives also significantly reduced the size of cooperative unit of the transition. The effect of the drugs was proportional to the concentration of the drug in the bilayer. This means that phenothiazine derivatives might have significant fluidizing effects on the biomembrane. The sizes of cooperative unit were successfully corrlated with phar-macological activities of the drugs and the surface pressure increases of lipid monolayer by these drugs. These correlations might be ascribed to a possible hydrophobic nature of interaction between the biomembrane and the drugs involved in their pharmacology.

  • This paper presents the effect of the supramolecular complex of GA (Glycyrrhizic acid) and Mt(menthol) and GA: Mt (4: 1) obtained on their basis can restore functional dysfunction of the liver mitochondria in alloxan diabetes, ie, inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular compound GA: Mt (4: 1) in alloxan diabetes were more pronounced than those of menthol, GA and its GK: Mt (2: 1) and GA: Mt (9: 1) compounds. According to the results obtained, the concentration of GA did not affect the peroxidation of lipid membranes of the liver mitochondria. However, a concentration of 15 μM of GA was found to reduce LPO (lipid peroxidation) formed by the effect of Fe2+ / ascorbate on the mitochondrial membrane by 58.0 ± 5.0% relative to control. A Scanning Calorimetric Study of the Effect of Clover Saponin on Liposomal Phospholipid Membrane

  • The effect of clover saponin on the phase transition of liposomal lipid bilayers of dimyristoyl phosphatidylcholine was investigated with differential scanning calorimetry. The thermograms of the liposomal bilayers incorporated with the clover saponin were obtained, and the enthalpy changes and the sizes of cooperative unit of the transition were calculated. The results showed that incorporation of the clover saponin into the liposomal bilayers effectively reduced the transition temperature at which the transition from solid state to liquid-crystalline state occurs, and broadened the thermogram peaks. It also reduced the size of cooperative unit of the transition. These results indicate that the clover saponin might have significant effect on the fluidity of biological membranes.

갓김치 Chlorophylls 및 Carotenoids의 항산화 효과 (Antioxidative Effect of Cholorophylls and Carotenoids in Mustard Leaf Kimchi Activity)

  • Antioxidative effects of crude chlorophylls and carotenoids extracts from mustard leaf kimchi on the lipid peroxidation in rat liver homogenate, egg phosphatidyl choline (EPC) liposome and superoxide anion radical were examined. The extracts were found to inhibit the increase of the thiobarbituric acid (TBA) value and show the effect of antioxidative activity on the liposomal phospholipid membrane. The oxidation index of EPC liposome was markedly decreased in the prescence of the extracts. The antioxidative activity of the extracts from mustard leaf kimchi was not related with fermentation period of the kimchi. The extracts from mustard leaf showed the similar antioxidative activity of $\alpha$-tocopherol within in the given level of addition. However, the oxidation index. When the effect of the extracts from mustard leaf kimchi on free radical scavenging was observed by the determination of the superoxide anion radical scavenging activity, it had similar value to that of $\alpha$-tocopherol.
