Using Physical Properties of Molten Glass to Estimate Glass Composition

  • A vitrification process is under development in KEPRI for the treatment of low-and medium-level radioactive waste. Although the project is for developing and building Vitrification Pilot Plant in Korea, one of KEPRI's concerns is the quality control of the vitrified glass. This paper discusses a methodology for the estimation of glass composition by on-line measurement of molten glass properties, which could be applied to the plant for real-time quality control of the glass product. By remotely measuring viscosity and density of the molten glass, the glass characteristics such as composition can be estimated and eventually controlled. For this purpose, using the database of glass composition vs. physical properties in isothermal three-component system of SiO$_2$-Na$_2$O-B$_2$O$_3$, a software TERNARY has been developed which determines the glass composition by using two known physical properties(e.g. density and viscosity).

Medium Composition of Aspergillus oryzae PF for the Production of Proteolytic Enzyme (단백질 분해효소 생산을 위한 Aspergillus oryzae PF균주의 배지조성)

  • The most favorable nitrogen source for the production of protease by Aspergillus oryzae PF was 2% soybean flour among sodium nitrate, ammonium sulfate, defatted soybean, skim milk, casein, peptone, and yeast extract. The production of protease from A. oryzae PF was higher at the concentration of 2% lactose than at variable concentration of glucose, sucrose, soluble starch, corn starch, potato starch, wheat starch, rice starch, cellulose, and gum arabic. Protease production was affected by the concentration of KH2PO4, Triton X-100, CaCo3, and MgSO4, and it was the highest at the highest at the concentration of 3% KH2PO4, 0.01% Triton X-100, 0.3% CaCO3, and 0.06% MGSO4.

  • Alkaline cellulolytic enzymes from cultured medium of Coprinus cinereus 2249 were purified with gel and ion-exchange chromatography and characteristics of those enzyme proteins were investigated. A fiber length distribution and a crystallinity of cellulose and sugar composition of enzyme treated Mixed Office Wastepaper(MOW) and Unbleached Kraft Pulp(UKP) were analysed. The conclusion could summarized as follows; \circled1 Alkaline and acidic, endo- and exo-glucanases were purified from cultured medium of Coprinus cinereus 2249. \circled2 The approximate molecular weight of alkaline endo-glucanase was 42 kDa, and also that of alkaline exo-glucanase was 50 kDa. A fiber length distribution and a crystallization of cellulose and sugar composition of enzyme treated MOW and UKP were not so much changed with original paper and pulp.

The Fermentation Characteristics of the Interspecific Protoplast Fusant of S. peucetius and S. platensis (Streptomyces peucetius subsp. caesius와 S. platensis사이의 원형질체 융합균주의 발효특성)

  • An interspecific fusant strain, Streptomyces MS1 was obtained by protoplast fusion between S. peucetius subsp. caesius and S. platensis. We studied on the fermentation characteristics of the fusant strain. The fermentation products of the fusant MS1 was identical with S. peucetius, but its production of anthracycline was more stable than S. peucetius under various fermentation conditions in regard to acidogenesis of fermentation broth. The optimal medium composition for anthracycline production by fusant MS1 as follows: sucrose 2.0%, glucose 1.0%, soytone 0.7%, $CaCO_3$ 0.2%, $KH_2PO_4$ 0.013%, casamino acids 0.01%, $K_2SO_4$ 0.025%, $MaCl_2\;6H_2O$ 1.024%, 5M $CaCl_2\;5H_2O$ 0.4%, 1N NaOH 0.7%, 20% L-proline 1.5%. In this condition, the productivity of anthracycline was $80{\sim}100\;{\mu}g/ml$.

  • Induction of symbiosis between N. muscorum and cultured tobacco cells associately cultured on nitrogen-free medium and effects of polyamines on culture condition were carried out. Analysis of amino acid composition in associate cultures showed increase in total amino acid amounts than single culture of cultured tobacco cells and methionine was markly increased in associative culture on N-free medium treated with 1 mM spermine. These results indicated that the composition of amino acids increased effectively in associative cultures by nitrogen fixation of N. muscorum.scorum.

  • An important modelling requirement for multimedia systems is the representation of temporal constraints among several different media that ordinarily arise in multimedia application. The multimedia system which integrates various types of data is required to express voice, image, text and graphic data with simultaneousness or an arbitrary time intervals. The purpose of this study is to analyze the temporal reationships and synchronization among several different kinds of medium. Moreover, it proposes a Multimedia Display-Time Composition Model (MDTC Model) which guarantees a representation of any kind of temporal synchronization among more than two kinds of medium. The proposed model is applied to a company advertizing system. Finally, a multimedia authoring tool based on the MDTC Model is implemented.

  • The action of phospholipid on the rhodotorucine A(RH.A) acceptance by heterbasidiomyceteous yeast Rhodosporidium toruloides mating type a cells and the effect of medium composition during sexual differentiation were investigated. Activation of trigger peptidase(TPase)was very sensitive to the originated phospholipid from R. toruloides and was more sensitive to phospholipid liposome made up of phospholipi. Phospholopod present on the membrance of mating type a cells consists of phospatidylglycerol(PG), phosphatidylethanolamine(PE), phospatidylcholine(PC), phospatidylinositol(PI), and phosphatidylserine(PS) of 12.9, suprisingly 45.4, 11.0, and 13.9%, respectively. As the result of using C-1 and N-1 mediums which limited C and N sources capable of inhibiting the synthesis of phospholipid, it resulted inhibiting sexual dlfferentiation and production of Rh.A from mating type Acells.

  • Selective culture of actinomycetes from soil microbes and their antibiotic producing characters by agar-disk method were examined. Some of the organisms which produced antibiotics on agar disk did not produce antibiotics in liquid culture. Further examination indicated that production of antibiotic was dependent on the composition of medium. Many streptomycestes produced antibacterial substances in tryptic soy broth but others produced antifungal antibiotics in V-8 broth. Production of antibacterial substances by Streptomyces sp. was also dependent on the medium composition.

Comparison of Liquid and Solid-State Fermentation Processes for the Production of Enzymes and Beta-Glucan from Hulled Barley

  • Solid-state fermentation using hulled barley was carried out to produce enzymes and β-glucan. The one-factor-at-a-time experiments were carried out to determine the optimal composition of the basal medium. The modified synthetic medium composition in liquid-state fermentation was determined to be 70 g/l hulled barley, 0 g/l rice bran, 5 g/l soytone, and 6 g/l ascorbic acid. Optimal pretreatment conditions of hulled barley by solid-state fermentation were evaluated in terms of maximum production of fungal biomass, amylase, protease, and β-glucan, which were 1.26 mg/g, 31310.34 U/g, 2614.95 U/g, and 14.6% (w/w), respectively, at 60 min of pretreatment condition. Thus, the solid-state fermentation process was found to enhance the overall fermentation yields of hulled barley to produce high amounts of enzymes and β-glucan.

Optimizing Fermentation Medium Composition for Bacterial Cellulose Production by a Newly Isolated Gluconacetobacter sp. RKY5

  • In this study, we investigated the optimal medium composition for bacterial cellulose (BC) production by Gluconacetobacter sp. RKY5. Among the various kinds of carbon sources, glycerol was the most efficient as a sole carbon source and its optimal concentration for BC production was 15 g/L. The optimal concentration of yeast extract as a nitrogen source for BC production was found to be 8 g/L. $K_{2}HPO_{4}$ and acetic acid were selected respectively as a phosphate source and a secondary substrate, and both optimal concentrations were 3 g/L. The amount of produced BC was 4.59 g/L in a static culture and 6.5 g/L in a shaking culture condition with 150 rpm. These values were 2.1 and 2.7 times higher than those in a static (2.16 g/L) and a shaking (2.41 g/L) cultures using HS medium generally used for BC production.
