In this study, we tried to optimize the composition of medium and culture conditions for the total avermectin and avermectin B1 production from S. avermitilis, which is a natural producer of avermectin family. Among various carbon and organic nitrogen sources tested, fructose and malt extract were most effective on avermectin production. Next addition of polyethylene glycol and $K_{2}HPO_{4}$ in medium significantly improved the intracellular contents of avermectin. Thus the optimized medium composition was 50 g/L fructose, 30 g/L malt extract, 5 g/L casamino acid, 2.5 g/L PEG 3,350, and 1 g/L $K_{2}HPO_{4}$, which increased the avermection production from10 to 478 mg/L. The contents of avermectin B1 complex was about 50% of the total amount of avermectin.

This study was conducted to optim ize the composition of CEM (cabbage extract medium) and cryoprotectants on the growth of Lactobacillus plantarum, a probiotics growing in plant and milk. For this, we analyzed the growth characteristics of Lb. plantarum in CEM and subsequently optimized the medium composition by addition of carbon, nitrogen sources and buffering agents. Among carbon sources, glucose showed the best result to increase the cell density after dilution of CEM. When 0.5% yeast extract and 1% soy peptone were supplemented in the diluted CEM, Lb. plantarum grew up to the maximum cell density. Addition of buffering agents in CEM was not significantly effective to increase the cell density. Meanwhile, addition of 12% skim milk, 5% sucrose and 0.5% glycerol showed a cryoprotective effect against cell damage of Lb. plantarum during freeze drying process showing high survival rate after 150 days. This optimized CEM can be used for economical production of bacterial cells particularly originated from a plant-related ecosystem.

The effects of stearic(18 : 0, SA), oleic(18 : 1 cis, OA) and elaidic acid(18 : 1 trans, EA) on the cell growth, contents of cellular lipids, and the fatty acid composition of cellular and medium lipids in Hep-G$_2$cells were evaluated. The cells were incubated in serum-free medium containing 25, 50, 100 and 200$\mu$M of a fatty acid combined with albumin for 2 days. The fatty acid concentration up to 100$\mu$M showed the normal growth, but the cell growth decreased in the presence of 200$\mu$M fatty acid. The treatment of cells with 100$\mu$M of a fatty acid for two days significantly(p<0.05) increased the cellular triglyceride(TG) content in all fatty acid groups compared to control, but TG contents was not significantly different among all treatment group, but total cholesterol(TC) was the highest level in EA group. The level of free cholesterol(FC) and cholesteryl ester(CE) was similar to those of TC in all fatty acid treated group. The cellular phospholipid(PL) contents were similar between the control and all fatty acid groups. The treatment of cells with SA has no notable effects on the fatty acid composition of TG, CE and PL. The OA treatment caused significant increases in CE(51.2%) and PL(29.8%), but not in TG. The EA treatment resulted in 10.1, 10.7 and 7.8% of $C_{18:1\;trans}$ content in cellular TG, CE, and PL. The TG, CE and PL of medium were relatively similar between SA and OA groups. In EA treated group, TG, CE and PL of medium contained 17.0%, 0.7% and 5.6% of $C_{18:1\;trans}$, respectively.

The Cassava (Manihot esculenta Crantz) is a perennial woody shrub cultivated mainly in the tropics for its starchy tuberous roots. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. Thus, a study was undertaken to develop an efficient in vitro mass propagation protocol of Manihot esculenta Crantz. Young and actively growing stem segments were excised from adult plants of cassava. Samples were cut into a 3~4 cm nodal segments with single node after sterilization, and cultivated in the different medium supplemented with various plant growth regulators for 4 weeks. For shoot multiplication, single-node stem segments, approximately 1 cm in length, were taken from in vitro derived shoots and subcultured. After 4~6 weeks, the shoot generation rate was 55.6%, the shoot number and its length were 1.0/explant and 2.3 cm in the most favorable medium composition. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.
This study examines for the processes of making a architectural significant issue of 20 cases form of Beijjng's hotel through analyzing the space composition from early the 1950's to 1992 in China. In the methods of analyzing process, We study the literatures, research reports and architectural drawings including the site survey. We hope to establish the principle in morphological characteristics and space composition of Beijing's high-rise hotels through analyzing the plan of these hotels. Finally, We find out the principles after researching. Firstly, Beijing's hotels in China faced on the being difficulties through the process of national development especially in political and economical conditions of country and peoples. Consequently, size of the Beijing's hotel developed with the fact that is complicated from that is single styles focused on the medium-sized one, modernizing popularity and the high-rise hotel buildings appeared plentifully and the form appeared with tall building putting first. Secondly, there are many immanent and interlocking styles in hotel's architectural block plans, many composition styles in big sized hotels, in opposition immanent styles in small sized ones and there are many trend of concourse style in medium sized hotels. To the layout of hotel's room inside of the linear styles are many in big and medium sized hotels.
This study was conducted to provide basic data for setting a standard medium for winter mushroom cultivation. Investigation of medium composition in winter mushroom farms in Korea revealed that the types of medium used for each farm were slightly different and that the mixing ratio of the medium also varied. All farmers used corncob, rice bran, and beet pulp as the main media, and calcium carbonate and ground oyster shells were used at 1.0~3.8% to adjust the pH of the medium. Analysis of the physicochemical properties of the mixed media showed nitrogen content of 1.28~1.52%, carbon content of 45.7~48.5%, and C/N ratio of 30.7~37.9. The content of inorganic components was significantly different in each farm, depending on the type and amount of minerals in the mixed medium. Mycelial growth was the fastest at Farm 2, reaching 6.5 cm within 34 days of culture, followed by Farm 4 at 5.7 cm, whereas Farm 1, 3, and 5 showed similar growth. Mycelial density was high without any significant difference between farms. Survey of yield according to medium composition in each farm showed the highest yield in Farm 2 at 173.4 g/bottle, followed by Farm 1 at 168.4 g/bottle, whereas Farm 3 and 4 showed similar yield. However, Farm 4 had the lowest yield at 145.4 g/bottle. Therefore, mushroom yield was different depending on the medium composition in the farm, and thus selection of a medium that can be used universally in multiple mushroom farms was considered possible.
We successfully isolated human anagen hair follicles from human scalp skin by microdissection and tried to culture them under various conditions. First we confirmed negative effect of serum on human hair follicle organ culture. As a next step serum-free medium compositions, Philpott medium, IMDM, and DHGM (Dongguk hair growth medium) were tried. Philpott medium is a general medium for hair organ culture based on Williams' E medium and DHGM is a special self-developed medium containing high amino acids and vitamins (B group) composition. As results, hair follicle in Philpott medium and IMDM showed anagen phase morphological structure, but rapid loss of hair elongation, low alkaline phosphatase expression, and very low expression of CK19. It is thought these hair follicles rapidly regressed from apoptosis. However, hair follicles in DHGM showed long term anagen phase morphological structure, continuous hair elongation, high alkaline phosphatase, and CK19 expression. These results demonstrate that high amino acids and vitamins (B group) composition are essential to in vitro long term human hair follicle organ culture and this culture medium will be useful in basic study of hair biology or application study to the development of alopecia treatment drugs.
The optimum medium composition for the production of L-tryptophan with Klebsiella pnuemoniae pheA tyrA trpE trpR/pSC 101-trp$^{+}$ and the effect of precusors in the optimum medium were studied. The specific growth rate in the optimum medium was almost the same as that in the basal medium, the former showing 1.01 and the latter 1.07 hr $$^{-1}$, but the produced tryptophan was increased 45% in the optimum medium. The maximum amount of produced tryptophan was 159 mg/l within 14 hours. Tryptophan production was ceased by casamino acid addition over 4 g/l in medium, but cell maSS increased with its addition. Indole and anthranilate as precusors had toxic effect on growth and tryptophan production at experimented concentration range (over 20 mg/l), but L-serine had good effect on tryptophan production, resulting in 175 mg/l tryptophan within 14 hours.
A jungwangu, a type of medium-sized mortar, is a firearm with a barrel and a bowl-shaped projectileloading component. A bigyeokjincheonroe (bombshell) or a danseok (stone ball) could be used as a projectile. According to the Hwaposik eonhae (Korean Translation of the Method of Production and Use of Artillery, 1635) by Yi Seo, mortars were classified into four types according to its size: large, medium, small, or extra-small. A total of three mortars from the Joseon period have survived, including one large mortar (Treasure No. 857) and two medium versions (Treasure Nos. 858 and 859). In this study, the production method for medium mortars was investigated based on scientific analysis of the two extant medium mortars, respectively housed in the Jinju National Museum (Treasure No. 858) and the Korea Naval Academy Museum (Treasure No. 859). Since only two medium mortars remain in Korea, detailed specifications were compared between them based on precise 3D scanning information of the items, and the measurements were compared with the figures in relevant records from the period. According to the investigation, the two mortars showed only a minute difference in overall size but their weight differed by 5,507 grams. In particular, the location of the wick hole and the length of the handle were distinct. The extant medium mortars are highly similar to the specifications listed in the Hwaposik eonhae. The composition of the medium mortars was analyzed and compared with other bronze gunpowder weapons. The surface composition analysis showed that the medium mortars were made of a ternary alloy of Cu-Sn-Pb with average respective proportions of (wt%) 85.24, 10.16, and 2.98. The material composition of the medium mortars was very similar to the average composition of the small gun from the Joseon period analyzed in previous research. It also showed a similarity with that of bronze gun-metal from medieval Europe. The casting technique was investigated based on a casting defect on the surface and the CT image. Judging by the mold line on the side, it appears that they were made in a piece-mold wherein the mold was halved and using a vertical design with molten metal poured through the end of the chamber and the muzzle was at the bottom. Chaplets, an auxiliary device that fixed the mold and the core to the barrel wall, were identified, which may have been applied to maintain the uniformity of the barrel wall. While the two medium mortars (Treasure Nos. 858 and 859) are highly similar to each other in appearance, considering the difference in the arrangement of the chaplets between the two items it is likely that a different mold design was used for each item.

Wave propagation in gyrotropic or anisotropic medium is analyzed in terms of the eigenmodes of the medium, which are admixture of TE and TM waves. The field composition and the phase velocity of the modes are also determined. The results of the analysis are applied to thin film optical waveguide using such medium as substrate and/or film. Based on the characteristic equations for phase constants of the waveguide, the condition for TE-TM mode convection is derived, and wave propagation in the guide is represented in the form of Jones matrix, which allows a new interpretation in the conversion efficiency of the thin-film optical waveguides.
