Continuous Production of Pullulan by Aureobasidium pullulans HP-2001 with Feeding of High Concentration of Sucrose

  • In this study, glucose, sucrose, and dextrin were found to be better carbon sources for the production of pullulan by Aureobasidium pullulans HP-2001. Maximal production of pullulan with 200 g/l sucrose as a carbon source was 54.2 g/l. The highest yield of pullulan from sucrose was 0.40, when the sugar concentration was 100 g/1. Optimal conditions for the continuous production of pullulan by A. pullulans HP-2001 in a 7-1 bioreactor were determined by studying the effects of composition of feed solution, dilution rate, and concentration of sucrose in the feed solution. Pullulan concentration and productivity with 100 g/l glucose and 2.5 g/l yeast extract were 38.1 g/l and 0.53 g/l h for 72 h, respectively, in a batch culture of A. pullulans HP-2001. When the substituted medium contained 100 g/l sucrose, 2.5 g/l yeast extract, and mineral salts, which is the same composition as the medium for the production of pullulan, the pullulan concentration and productivity were 74.9 g/l and 0.55 g/l h for 120 h, respectively. The production of pullulan at the steady state increased with a dilution rate up to 0.015/h, and its concentration was 78.4 g/l with a weight average molecular weight ($M_w$) of $4.0{\times}10^5$. Unlike a batch culture, however, the decline of the $M_w$ and the number average molecular weight ($M_n$) of pullulan was not found in the continuous culture of A. pullulans HP-2001. When the concentration of sucrose in the feed solution was 200 g/l, 113.5 g/l of pullulan was obtained at the steady state. The steady state was maintained longer in the continuous culture fed with the feed solution containing 200 g/l sucrose than when fed with the feed solutions containing either 100 or 150 g/l sucrose.

Growth Temperature-Dependent Conversion of De novo-Synthesized Unsaturated Fatty Acids into Polyhydroxyalkanoic Acid and Membrane Cyclopropane Fatty Acids in the Psychrotrophic Bacterium Pseudomonas fluorescens BM07

  • A psychrotrophic bacterial strain, Pseudomonas fluorescens BM07, synthesized unsaturated fatty acids (UFA) from fructose in response to lowering of growth temperature, and incorporated them into both polyhydroxyalkanoic acid (PHA) and membrane lipid. The blocking of PHA synthesis by adding 5 mM 2-bromooctanoic acid to the growth medium, containing 70 mM fructose, was found to be a useful means to profile the composition of membrane lipid by gas chromatography. As the growth temperature changed from 35 to $50^{\circ}C$, the total content of two UFA, 3-hydroxy-cis-5­dodecenoic acid ($C_{12:1}$) and 3-hydroxy-cis-7-tetradecenoic acid ($C_{14:1}$), in PHA increased from 31 to 44 $mol\%$. The growth at lower temperatures also led to an increase in the level of two major UFA, palmitoleic acid (C16:1 cis9) and cis-vaccenic acid (C18:1 cis11), in membrane lipid. A fraction of these membrane-lipid UFA was converted to their corresponding cyclopropane fatty acids (CFA). The CFA conversion was a function of culture time, exhibiting biphasic increase before and after entering the stationary phase. However, pH changes in growth media had no effect on the CFA conversion, which is contrary to the case of E. coli reported. The cells grown at $30^{\circ}C$ responded to a cold shock (lowering the medium temperature down to $10^{\circ}C$) by increasing the level of C16:1 cis9 and C 18: I cis II up to that of $10^{\circ}C$-grown control cells and concomitantly decreasing the relative level of cis-9,10­methylenehexadecanoic acid (the CFA converted from C16:1 cis9) from 14 to 8 $mol\%$, whereas the 10-grown cells exhibited little change in the lipid composition when exposed to a warmer environment of $30^{\circ}C$ for 12 h. Based on this one- way response, we suggest that this psychrotrophic strain responds more efficiently and sensitively to a cold shock than to a hot shock. It is also suggested that BM07 strain is a good producer of two unsaturated 3-hydroxyacids, $C_{12:1}\;and\;C_{141:1}$.

Studies on the $\beta$-Galactosidase from Thermphilic Bacterium - Physiological Characteristics of the Selected Thermophile - (고온성 세균의 $\beta$-Galactosidase에 관한 연구 ( I ) - 분리고온균의 생리적 특성 -)

  • This experiment was carried out to elucidate the thermotolerant properties of a thermophilic bacterium which isolated from soils of the hot springs area and selected for the $\beta$-galactosidase production. Biochemical and physiological characteristics of this strain were studied, including the investigation on the fatty acid composition for its neutral fats. The results obtained were summarized as follows. 1. This bacterium was identified as a strain belong to the genus Thermus. 2. Optimal temperature and pH for growth of this strain were $65^{\circ}C$ and pH 6.5 respectively, and it was found to be an absolute thermophilic bacterium which could not grow at 37$^{\circ}C$. 3. No growth was obtained in the medium which contained more than 1.0% of sodium chloride. 4. The tolerable concentration of antobiotics were 10$\mu\textrm{g}$ of penicillin G per $m\ell$ of medium and 0.5$\mu\textrm{g}$ of chloramphenicol per $m\ell$ respectively 5. This strain had autotrophilic requirements for calcium-pantothenate and pyridoxine-HCO as an-essential factor and for niacin as a stimulative factor. 6. Fatty acid composition of neutral fats of the strain was palmitic acid. 60.20%; lauric acid, 11.8%; myristic acid, 7.56%, behenic acid, 4.25%; capric acid, 1.77%; stearic acid, 2.13%; arachidic acid, 1.53%; and others unidentified, 10.7%.

Fatty acid composition of neutral fats of the strain was palmitic acid, 60.20%; lauric acid, 11.80%; myristic acid, 7.56%; behenic acid, 4.25%; capric acid, 1.77%; stearic acid, 2.13%; arachidic acid, 1.53%; and others unidentified, 10.7%.

  • The Wondong caldea is a deeply eroded structure that offers spectacular exposures through the core and margins of a resurgent caldera. The Wondong Tuff and the postcollapse intrusions range from medium-silica rhyolite to rhyodacite in composition and the postcollapse lava and tuff, preresurgent and resurgent intrusions also range from medium-silica rhyolite to an-desite, which jump to gap dacite composition. The continuous compositional zonations generally define a large stratified magma system in the postcollapse and resurgent magma chamber. Isotopic and trace element evidence suggest that the compositional zonations might have resulted from the differentiations from crystal fractionations of a parental andesitic magma, accompanying a little contamination from the crustal assimilations near the chamber roof and wall. But chemically and isotopically distinct late intusions might have resulted from emplacement of any different magma batch.

  • Serratia marcescens CK-3, decomposing chitin which is a mar component of cell wall in phyitopathogenic fungi, was isolated from the continuous cropping rhizosphere of pepper and cucumber and its enzymatic property was examined. S. marcescens CK-3 was found tn have an tagonistic effects against, Fusarium axysporum and Rhizoctonia solani and to have complex enzyme system such as chitinase, laminarinase, and proteinase. The preferable composition of the medium for production of chitinase was fond and was as follows : colloidal chitin 1.5%, tryptone 0.5%, glucose 1.0%, peptone 0.2%, $MgSO_4{\cdot}7H_2O\;0.1%,\;K_2HPO_4\;0.1%,\;and\;NaCl\;0.1%$(w/v), pH 6.8. The maximum enzyme production was observed after culture of 72 hours at $30^{\circ}C$ using a medium containing the above chemical composition. The optimal pH and temperature for in vitro activity of chitinase from S. marcescens CK-3 were pH 7.5 and $50^{\circ}C$, respectively. The enzyme activity in-creased by metal ions such as$Ag^+$ and $Mn^{++}$.

  • To study the follicular atretic mechanism in mammalian ovary, the medium-sized (3.0-6.0mm) follicles of porcine ovary were morphologically classified as nonnal and atretic. Steroid concentrations in the follicular fluid were analyzed by radioimmunoassay, and the proteins in that fluid were electrophoretically separated. Concentrations of progesterone in the atretic follicular fluid of follicular phases were higher than those of normal ones (p < 0.05). Concentrations of testosterone were high in normal luteal and atretic follicular-phases follicles. The concentrations of estradiol remained significanily lower in atretic follicular-phases follicles than normal. After sodium dodecyl sulfate-polyacrylamide gel electrophoresis of follicular fluid proteins, four kinds of proteins (20K, 32K, 33K, 38K) were detected, and those proteins were not present in sera. According to the ovarian cycle, proteins of MW of 112K and 141K were identified. In atretic follicular fludies, MW of 23K and 24K were specifically detected. From the above results, we can conclude that, as ovarian cycle changes, steroid contents and protein composition in atretic follicular fluid are different from the normal developing follicular fluid. To further understand the physiologic roles of the proteins present in the atretic follicular fluids, these proteins need to be characterized and identified.

Effects of Artificial Culture Medium of Wild Ginseng on the Physico-chemical Characteristics of Pork (돈육의 이화학적 품질 특성에 미치는 산삼 배양액 급여 영향)

  • A total of 120 pigs (Berkshire) were used to investigate the effect of dietary supplementation with artificial culture medium of wild ginseng (CWG) on the physico-chemical characteristics of pork About $60{\pm}3kg$ pigs were randomly assigned to 4 pens based on sex and diet (C: commercial diet feed or T: commercial diet+1 L CWG per day for 70 days). Pigs were slaughtered at approximately 110 kg live weight, and proximate composition and physico-chemical characteristics were measured in pork loin. The moisture content, hardness and chewiness of pork were higher in gilt fed CWG than in gilt fed the control diet, but the water-holding capacity of pork and $L^*$ values of fat color were lower. The shear force and $a^*$ values of pork were higher in barrow fed CWG than in harrow fed the control diet. The crude fat content, hardness, cohesiveness, chewiness of pork and $a^*\;and\;b^*$ values of fat color were lower in barrow than in gilt, but the pH was higher. These results imply that the proximate composition and physico-chemical characteristics of pork could be affected by dietary supplementation with CWG and the sex of the pig, while the texture properties and at values of pork may be improved with dietary CWG.

The origin of the dolomite of the Pungchon Formation near Taebaeg City, Kangwondo, Korea (강원도 태백시 일대에 분포하는 풍촌층 돌로마이트의 성인)

  • The objective of this study is to investigate the origin of the dolomite in the Pungchon Formation of the Choseon Supergroup near Taebaeg City, Kangwondo, Korea. The Pungchon Formation is composed of limestone, dolomitic limestone, and dolomite with thin beds of flat pebble conglomerate (FPC) and mudrock. Texturally, the dolomite in the Pungchon Formation can be divided into four types; 1) coarse-sized, xenotopic dolomite in massive dolomite, 2) medium-siEed, idiotopic dolomite in flat pebble conglomerate, 3) xenotopic dolomite replacing ooids, algalnodules, and echinoderms, and 4) the dolomite in mottled fabric. The dolomite in mottled fabric can be subdivided into three types; a) coarse-sized, xenotopic saddle dolomite cement, b) medium-sized, idiotopic, cloudy-centered, clear-rimmed (CCCR) dolomite, and c) coarse-sized, idiotopic dolomite. The carbon isotopic composition of the Pungchon dolomite is in the range of $-2.8-1.4\%_{\circ}(PBD)$, suggesting that the carbon isotopic composition was buffered by the preexisting marine carbonates. Lighter oxygen isotopic values ($\delta^{18}O-15.7-8.7\%_{\circ}, PBD$) indicate that the Pungchon dolomite may have formed under high temperature in a burial diagenetic environment. The higher initial $^{87}Sr/^{86}Sr$ ratio of the Pungchon dolomite (0.7010-0.7161) than that of the coeval Cambrian seawater (0.7088-0.7092) indicates that dolomitizing fluids had been modified from the isotopic exchange with continental crust. Low Sr and Na contents(<200 ppm) of dolomite agree well with previously reported data for burial dolomite. Hifh Fe and Mn contents of the dolomite support the idea that the Pungchon dolomite may have formed in a deep burial diagenetic environment.

Keratinase Production by Recalcitrant Feather Degrading Pseudomonas Geniculata and Its Plant Growth Promoting Activity (난분해성 우모분해 Pseudomonas geniculata에 의한 케라틴 분해효소 생산 및 식물성장 촉진 활성)

  • We investigated the optimal conditions for keratinase production by feather-degrading Pseudomonas geniculata H10 using one variable at a time (OVT) method. The optimal medium composition and cultural condition for keratinase production were determined to be glucose 0.15% (w/v), beef extract 0.08% (w/v), $KH_2PO_4$ 0.12% (w/v), $K_2HPO_4$ 0.02% (w/v), NaCl 0.07% (w/v), $MgSO_4{\cdot}7H_2O$ 0.03%, $MgCl_2{\cdot}6H_2O$ 0.04% along with initial pH 10 at 200 rpm and $25^{\circ}C$, respectively. The production yield of keratinase was 31.6 U/ml in an optimal condition, showing 4.6-fold higher than that in basal medium. The strain H10 also showed plant growth promoting activities. This strain had ammonification activity and produced indoleacetic acid (IAA), siderophore and a variety of hydrolytic enzymes such as protease, lipase and chitinase. Therefore, this study showed that P. geniculata H10 could be not only used to upgrade the nutritional value of feather wastes but also useful in situ biodegradation of feather wastes. Moreover, it is also a potential candidate for the development of biofertilizing agent applicable to crop plant soil.
