여드름 원인균 Propionibacterium acnes에 대한 생약 추출물의 항균활성 측정 (Antibacterial Activity of Some Medicinal Plants against Propionibacterium acnes)

  • Acne, one of the most common skin disease, is a chronic inflammatory disease and is caused by uncertain and multiple factors. Propionibacterium acnes belongs to the human cutaneous normal flora and is a major etiologic agent of acne vulgaris. In this study, we evaluated the antibacterial effect of 19 medicinal plants. Antibacterial activity of extracts prepared from the 19 medicinal plants was investigated against bacteria related to acnes, Propionibacterium acnes. Among them, some medicinal plants inhibited the growth of the P. acnes. Minimum inhibitory concentration (MIC) of Polygonum aviculare (Herb) was 0.2 mg/ml against P. acnes. MIC of Dianthus chinensis (Aerial Part), Forsythia viridissima (Fruit), Lygodium japonicum (Spore) and Sophora flavescens (Root) were 0.4 mg/ml. Based on these results, Polygonum aviculare (Herb), Dianthus chinensis (Aerial Part), Forsythia viridissima (Fruit), Lygodium japonicum (Spore) and Sophora flavescens (Root) may be considered as a candidate for a good medicine for acne.

생약의 Protein Tyrosine Phosphatase 1B 저해활성 검색 (Screening of Medicinal Herbs for Inhibitory Activity against Protein Tyrosine Phosphatase 1B)

  • Protein tyrosine phosphatase 1B (PTP1B) is predicted to be therapeutic target in treatment of type 2 diabetes and obesity. Thus, in order to search for PTP1B inhibitors, we screened the inhibitory activity of PTP1B in the water extracts of 84 medicinal herbs. Among them, the extracts of Pini Folium, Magnoliae Cortex, Artemisiae asiaticae Herba, Schizonepetae Herba, Menthae Herba, Mume Fructus, Cimicifugae Rhizoma, and Amomi Cardamomi Fructus showed relatively significant (58-68%) inhibitory activity against PTP1B. Especially, the methylene chloride fraction of the methanol extract of Menthae Herba (81% inhibition at 30 ${\mu}g$/ml) showed more potent inhibitory activity against PTP1B than others.

한약재 추출물의 항균효과 검색 (Screening of Antimicrobial Activity for Medicinal Herb Extracts)

  • 천연식품보존료 개발 목적의 일환으로 항균효력이 있을 것으로 추정되는 한약재 중에서 목단피, 황백, 연교, 오미자, 자초, 길경 등 20종을 대상으로 물과 95% 에탄올로 항균성물질을 추출하여 각종 미생물에 대한 항균력을 검토하였다. 물 추출물의 경우, 모든 세균에 대하여 증식억제 효과가 나타난 것은 치자, 구기자, 오미자 등의 3종이었으며, 그중에서 오미자가 증식 억제력이 제일 강하였다. 에탄올 추출물의 경우, 대부분의 세균에 대하여 증식억제 효과가 나타난 것은 치자, 오미자, 자초 등의 3종이었으며, 목단피, 황백, 금은화 등의 3종은 그람양성균에만 효과가 나타났는데, 특히 황백의 경우, S. aureus에 대하여는 0.01%의 첨가로 균의 증식이 억제되었다. 한편 이들 한약재들의 물 추출물은 진균류에 대해서는 증식억제 효과가 좋지 않았으나, 에탄올 추출물의 경우, 목단피, 황백, 치자, 자초 등의 4종이 증식억제 효과가 우수하였고, 그중 황백이 균증식 억제력이 제일 강하였다. 전 시료를 통하여 에탄올 추출물이 물 추출물 보다 $2{\sim}100$배나 항균력이 높았으며, 특히 자초의 에탄올 추출물은 맛과 향 등의 관능검사결과 이미 이취가 적을 뿐만 아니라 항균력도 우수하여 천연식품 보존료로서 이용 가능성이 높았다.

Characterization of Biocompatible Lipid-Based Vesicles Contained with Medicinal Herb Extracts

  • In order to increase the medicinal herbs efficiency of drug delivery, vesicles contained with medicinal herbs were prepared by phosphatidylcholines and surface active agent. Vesicles loaded with medicinal herbs were characterized by UV-spectroscopy, Zetasizer. The antioxidant activity of vesicles was measured by DPPH assay and ABTS radical scavenging assays. Also, an analysis was conducted to determine the effects of anti-inflammatory of vesicles contained medicinal herbs. In addition, the whitening effects of vesicles contained medicinal herbs extract were studied via tyrosinase inhibition assay. The results of vesicles were as follows. Vesicles appeared an average diameter of approximatively 164-599 nm. All studied vesicles contained with medicinal herbs showed antioxidant, anti-inflammatory and whitening effects in a dose-dependent manner. Therefore, this experiment achieves its purpose of synthesizing of vesicles. In conclusion, we recommended that the vesicles loaded with medicinal herbs have ability for anti-aging materials. Specifically, it will apply to cosmetic ingredients.

Anti-aging Potential of Extracts Prepared from Fruits and Medicinal Herbs Cultivated in the Gyeongnam Area of Korea

  • Many recent studies have focused on maintaining a healthy life by preventing and/or postponing the aging process. Numerous studies have reported that continuous exposure to reactive oxygen species can stimulate skin aging and that excessive accumulation of fat can cause an impaired skin barrier and tissue structure alterations. Thus, the maintenance of antioxidant homeostasis and the suppression of adipose accumulation are important strategies for skin anti-aging. Here, we prepared three types of extracts [whole juice, acetone-perchloric acid (PCA), and ethanol] from 20 fruits and medicinal herbs native to the Gyeongnam area of Korea. The total phenolic content of each extract was analyzed, and we observed higher total phenolic contents in the medicinal herbs. Consistent with this, the results of the oxygen radical absorbance activity capacity assay indicated that the in vitro antioxidant activities of the medicinal herb extracts were stronger than those of the fruit extracts. The fruits and medicinal herbs had strong effects on cell-based systems, including $H_2O_2$-induced oxidative stress in human keratinocytes and 3T3-L1 lipid accumulation. Nishimura Wase persimmon, Taishu persimmon, wrinkled giant hyssop, sweet wormwood, Chinese cedar, red perilla, tan shen, hiyodori-jogo, and cramp bark may be natural anti-aging materials with effective antioxidant and anti-adipogenic activities. Taken together, our findings may provide scientific evidence supporting the development of functional foods and nutraceuticals from fruits and medicinal herbs.

약용식물의 열수추출물과 적정 조성추출물 및 그 발효물이 알콜대사 효소활성에 미치는 영향 (Effects of Aqueous Medicinal Herb Extracts and Aqueous Fermented Extracts on Alcohol-Metabolizing Enzyme Activities)

  • 숙취해소효과가 우수한 제품을 개발하기 위하여, 약용식물 25종의 열수추출물에 대하여 in vitro에서 ADH와 ALDH의 저해 및 활성작용이 우수한 약용식물 12종을 선발, 일정 비율로 혼합한 약용식물 추출액에 대하여 발효를 행한 후 in vitro 및 in vivo실험을 통하여 숙취해소효과를 검토하였다, 그 결과, 감초 -0.3%, 갈근 -3.2%, 구기자 -1.7%, 맥문동 4.8%, 속새 8.3%, 오가피 3.5%, 및 지구자 -2.1%를 보여 지구자를 비롯한 7종의 추출물이 90%이상의 강한 저해활성을 보였고, ALDH활성에서는 오가피가 166.3%로 활성이 가장 촉진되었고, 뒤이어 구기자, 속새 및 백복령도 1.5배 이상의 활성을 촉진하였으며 이 외에 맥문동, 인진쑥, 작약, 차전초, 지구자, 홍삼 및 흑두에서도 효소활성을 촉진하는 것으로 나타나는 등 갈근을 포함하여 약 10여종의 약용식물에서 130%이상의 활성을 증가시키는 결과를 보여주었다. 12종으로 조성 혼합된 약용식물 추출물에서는 ADH활성이 -20.22%인데 비하여 추출발효액은 -62.63%정도 ADH저해활성이 높았으며, ALDH활성은 추출물에서는 173.20%이었으며, 추출발효액은 280.17%로 ALDH활성이 추출액보다 약 1.6배가 증가됨을 볼 수 있었다. 또한 흰쥐를 이용하여 약용식물 추출발효물의 숙취해소효과를 보기 위해 알코올 투여 후 시간에 따른 혈중 알코올과 아세트알데히드의 농도를 측정한 결과, 알코올 섭취 후 3시간 경과시 추출 발효물 400 mg/kg 처리군이 시판 숙취개선음료 처리군보다 24.1% 정도 우수한 효능을 보였고, 추출발효물 공급 3시간 경과 시부터 7시간까지 혈중 아세트알데히드의 감소폭은 추출 발효물 400 mg/kg 처리군에서 -65.1%로 시판 숙취개선음료를 공급한 군에서 -57.5%를 나타냈다.

허브추출물의 항균 작용 (Antimicrobial Activity of Herb Extracts)

  • 아로마테라피에도 널리 이용되는 라벤더, 패츌리, 로즈마리, 유칼립투스 등의 허브를 이용하여 식품의 천연보존제로서의 항균활성의 특성에 대하여 알아보았다. 프랑스산 로즈마리추출물의 경우 C, albicans ATCC 10231에 대한 MIC는 29.1 mg/mL로 나타났으며 B. subtilis ATCC 6613과 E. coli ATCC 25922에 대한 MIC는 14.5 mg/mL로 나타났다. 프랑스산 로즈마리와 미국산 라벤더의 추출물은 40에서 121$^{\circ}C$ 범위에서 열 안정성을 나타냈으며 pH 안정성 실험에서는 중성에 좋은 안정성을 나타내었다. 액체 배양시 프랑스산 로즈마리와 미국산 라벤더의 추출액을 첨가하여 피검균의 생육억제 실험을 행한 결과 액체배지 20mL에 대해 2.9mg의 허브추출물을 첨가한 경우에 생육억제효과를 나타내었다.

  • Protaetia Brevitarsis Seulensis(white grub) has been traditionally used as medicinal stuff to treat blood stasis, occlusion of menstruation, tetanus and liver cancer in Asian countries (Korea, Japan, China, Taiwan, India and Myanmar). Especially, Donguibogam, which is traditional korean medicinal book, described the white grub as traditional medicine to treat hepatic diseases and vascular disorders. The white grub has been considered as highly nutritional food. The major constituents of white grub are rich in protein, healthy fats, iron, calcium. Recent studies announced that white grub has hepatoprotective effect and anti-microbacterial effect. However, the immuno-enhancing effect of white grub extracts in RAW 264.7 macrophage cells has not been studied yet. In this study, the various concentrations of white grub extract were examined to find immuno-enhancing effects on RAW 264.7 cells. Cytotoxicity was determined by MTT assay and immuno-enhancing effect of white grub extract was investigated by measuring nitric oxide (NO) production compared with only lipopolysaccharide (LPS) treatment. White grub extracts (0.001 - 10 mg/ml) did not show cytotoxicity. Additionally, white grub extracts (0.001 - 1mg/ml) had Immuno-enhancing effect on RAW 264.7 cells compared with only LPS treated group. These results might be provided proof to develop beneficial immuno-enhancing material for human health.

Escherichia coli에 대한 한약재추출물의 항균활성 (Antimicrobial activity of extracts from medicinal herbs against Escherichia coli)

  • The extracts from 40 different traditional medicinal herbs were used to investigate the antimicrobial activities against Escherichia coli. Among them, the extracts from Paeonia suffruticosa (PS), Siegesbeckia orientalis (SO), Schizandra chinensis (SC), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited high antimicrobial activities against E. coli, Minimum inhibitory concentrations (MIC) of the RJ extract against E. coli were 0.8 mg/ml. After heating treatment of these extracts, the antimicrobial activities against E. coli were significantly reduced in case of the CS extract. After alkaline or acid treatment of these extracts, the antimicrobial activities against E. coli were significantly increased in the PS extract but reduced in both SO and RJ extract. Since extracts from RJ and CS exhibited the highest antimicrobial activities, bacterial growth-inhibiting activities against E. coli by these two extracts were further examined. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 100, 300 or 500 ppm of RJ extract ranged from 0.1 to 0.2 compared to 0.35~0.65 in the absence of RJ extract, indicating that growth of E. coli was significantly inhibited within 24 hours by the addition of at least 100 ppm of RJ extract. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 300 or 500 ppm of CS extract ranged from 0.01 to 0.25 compared to 0.5~0.55 in the absence of CS extract, indicating that growth of E. coli was also significantly inhibited within 24 hours by the addition of at least 300 ppm of CS extract. In conclusion, these findings suggest that extracts from RJ and CS may play important roles for antimicrobial activities against E. coli causing various animal diseases.
