Changes of Physiological Functionalities during the Fermentation of Medicinal Herbs Doenjang (약용 식물 첨가 된장의 숙성 중 생리기능성의 변화)

  • In order to develop new functional doenjang, two types of medicinal herb doenjang were prepared with several medicinal herbs, bamboo salts and meju and then changes of its physiological functionalities were investigated during 6 months of fermentation. Antihypertensive angiotensin converting enzyme(ACE) inhibitory activities of medicinal herb doenjangs were decreased to 39.0∼5l.7% from 61.7∼72.5% after 6 months of fermentation. Fibrinolytic activity and electron donating ability of medicinal herb doenjangs were showed 10.4∼11.3 U and 96.3∼99.1% after 6 months of fermentation, respectively. SOD-like activity of medicinal herb doenjangs were also showed approximate 10% and tyrosinase inhibitory activity of 20$^{\circ}C$ -water extracts of medicinal herb doenjang(I) was increased up to 97.6% from 12.9% after 6 months of fermentation. Crude protein content and total sugar content of medicinal herb doenjangs were approximate 14% and 33%, respectively, which were higher than those of traditional doenjang.

Antioxidant Activity of Methanol Extracts from Cudrania tricuspidata Bureau according to Harvesting Parts and Time (이용부위 및 채취시기에 따른 꾸지뽕나무 메탄올 추출물의 항산화성)

  • To obtain the information on antioxidant activity by harvesting parts and time in Cudrania tricuspidata Bureau, total phenolic contents, flavonoid contents, superoxide dismutase (SOD) like activity and 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity of methanol extracts from Cudrania tricuspidata were investigated. Total phenolic contents was the highest in leaf [$30.2{\sim}38.8\;mg/g$ dry weight (DW)] and followed by root bark, stem bark, fruit, root except bark and stem except bark. Among harvesting time, total phenolic contents of leaf was the highest as $38.1{\sim}38.8\;mg/g$ DW in May and June, and then decreased to October. Root bark harvested in April and May contained $26.6{\sim}27.8\;mg/g$ DW total phenolic compounds and total phenolic content fell to a very low values at 1.8 mg/g DW in February. Flavonoid contents was very high in leaf and root bark. Especially, root bark harvested in April had the highest of flavonoid contents as 23.2 mg/g DW. As a result of simple linear regression analysis of total phenolic contents on flavonoid contents according to harvesting parts, it showed a high correlation (p < 0.001) with $0.57{\sim}0.97\;R^2$ (coefficient of determination). As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of DPPH absorbance, was $13.7{\sim}20.5\;{\mu}g/mL$ in leaf so DPPH radical scavenging activity was very high. But SOD like activity of $1,000\;{\mu}g$ extract/mL MeOH was a low value of $0.3{\sim}9.2%$ in all samples.

Electrical Stimulation Can Facilitate Vestibular Compensation Following Unilateral Labyrinthectomy in Rats

  • To investigate the effects of electrical stimulation on vestibular compensation, which is the recovery of vestibular symptoms following unilateral labyrinthectomy (UL), intermittent electrical stimulation was applied to the injured vestibular portion in Sprague-Dawley rats. Vestibuloocular and vestibulospinal reflexes, electrical activity and expression of c-Fos protein in medial vestibular nuclei (MVN) were measured with time following UL. Spontaneous nystagmus occurred with frequency of $2.9{\pm}0.2$ beats/sec at 2 hours after UL and disappeared after 72 hours. Electrical stimulation decreased the frequency of nystagmus significantly till 24 hours after UL. Roll head deviation was $107{\pm}9.7^{\circ}$ at 2 hours after UL and the deviation was maintained till 72 hours, but electrical stimulation decreased the deviation significantly 6 hours after UL. Resting activity of type I neurons in ipsilateral MVN to the injured vestibular side decreased significantly compared with control at 6 and 24 hours after UL, but the activity of type I neurons was recovered to control level by electrical stimulation at 24 hours after UL. Gain of type I neurons induced by sinusoidal rotation of 0.1 Hz decreased significantly till 24 hours after UL, but electrical stimulation restored the activity at 24 hours. The gain of type II neurons decreased significantly at 6 hours after UL, but electrical stimulation restored the activity. Expression of c-Fos protein was asymmetric between bilateral MVN till 24 hours after UL, but the asymmetry disappeared by electrical stimulation 6 hours after UL. These results suggest that electrical stimulation to the injured vestibular portion facilitates vestibular compensation following UL by restoration of symmetry of neuronal activity between bilateral vestibular nuclei resulting from increased activity in ipsilateral vestibular nuclei to the injured side.

  • Twenty nine substituted thiaxanthone derivatives were synthesized for schistosomicidal evaluation. The compounds, 1-morpholino-4-methyl-6-nitrothiaxanthone and 1-(2-diethyl-aminoethyl-amino)-4 methyl-6-nitrothiaxanthone dipicrate were found to possess promising schistosomicidal activity against Schistosoma mansoni in mice.

Screening of Korean medicinal plants for antitumor activity

  • Potential antinumor activity of sixteen Korean medicinal plants was tested against leukemia SN 36, murine tomor model. Survival studies by measuring life span indicated that Morus alba (Moraceae), Verbascum phlomoides (Scrophulariaceae), Curcuma longa (Zingbieraceae), Torilis japonica (Umbelliferae), Bupleurum falcatum (Umbelliferae) and Codonopsis pilosula (Campanulaceae) exhibited significant antitumor activity with the dose-schedule employed in the experiment. Methanol (70%) extracts of Platycodon gradiflorum (Campanulaceae), Bupleurum longeradiatum (Umbelliferae) and Asiasarum sieboldii (Aristolochiaceae) showed rather potent toxicity.

Correlation Between Electrical Activity of Type I Neuron and c-Fos Expression in the Medial Vestibular Nuclei Following Unilateral Labyrinthectomy in Rats

  • To search the correlations between electrical activity and c-Fos expression in the process of vestibular compensation, we examined the changes of those two parameters in the medial vestibular nuclei (MVN) of unilaterally labyrinthectomized (ULX) rats. Spontaneous nystagmus with fast component toward the intact side disappeared gradually within 48 hours. Fourty eight hours after ULX, directional preponderance of the eye movement induced by sinusoidal rotation of the whole body which represents the symmetry of bilateral vestibular functions showed less than 20% by rotation of 0.1, 0.2, and 0.5 Hz, indicating the recovery of symmetry in bilateral vestibular functions. Six hours after ULX, spontaneous electrical activity of type I neurons resulted in asymmetry between bilateral MVN, however, the asymmetry of the electrical activity was decreased 48 hours after ULX. Immunocytochemical staining revealed that ULX produced dramatic induction of c-Fos positive cells in the MVN bilaterally. The number of c-Fos immunoreactive cells in the contralateral MVN was significantly higher than those in the ipsilateral MVN (p<0.0001) 2 hours after ULX. Thereafter, the number of c-Fos positive cells decreased bilaterally and was slightly, but not significantly higher in the ipsilateral MVN at 48 hours after ULX. The present results suggest that both electrical activity of type I neurons and c-Fos expression in MVN following ULX will reflect underlying mechanisms of recovery process of vestibular compensation.

Antioxidant and Xanthine Oxidase Inhibitory Activities of Hot Water Extracts of Medicinal Herbs (한약재 열수추출물의 항산화 활성 및 Xanthine Oxidase 저해 활성)

  • In our study, as many as 29 edible medicinal herbs were selected for testing their ability in the effective treatment of gout based on oriental medicine theory. We extracted each medicinal herb (135 g) with 4 L of distilled water at $100{\sim}105^{\circ}C$ for 210 min. Thereafter, we evaluated both the antioxidant and xanthine oxidase inhibition activities of the extracts obtained. Among all the edible medicinal herbs used in our study, only the extract from Scutellaria baicalensis Georgi (Korean name: hwang-geum) showed (1) the maximum total phenolic content (TPC) (2.25 mg gallic acid equivalent/mL), (2) DPPH radical scavenging activity (94.04%), and (3) xanthine oxidase inhibition activity (87.75%). We also observed that TPC was relatively highly correlated with both the DPPH radical scavenging activity (r=0.63) and xanthine oxidase inhibition activity (r=0.77). Our results suggest that S. baicalensis G. may be a potent antioxidant source for the extraction and development of nutraceuticals that may be utilized for effective treatment of gout.

Insecticidal activity of flupyrazofos KH502 against Plutella xylostella: a CoMFA study (배추좀나방에 대한 flupyrazofos KH502의 살충활성 CoMFA)

  • In recent years, the diamondback moth (DBM), Plutella xylostella has become one of the most important pests for cruciferous plants in the world. A new type of the thiophosphoryl pyrazole insecticide, called KH502. Its outstanding insecticidal activity could be effective alternative against DBM. We investigated, using the comparative molecular field analysis (CoMPA) method, The structure-activity relationship of various thiophosphorylpyrazole derivatives and structure requirement for insecticidal activity. We found, the key substructures in pyrazole derivatives, the trifluoro-methyl group and the thiophosphoryl group. Both play an important role in insecticidal activity with the binding site. The three dimensional Quantitative Structure Activity Relationship (QSAR) analysis could provide useful information for the structural requirements of pyrazole insecticide as an insecticidal and the design of a new insecticide.

Biological Activities of Extracts from Scutellaria baicalensis, Zizyphus jujuba and Atractylodes macrocephala (황금, 산조인, 백출 추출물의 생리활성)

  • Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of extracts from Scutellaria baicalensis, Zizyphus jujuba and Atractylodes macrocephala. Methods : Three kinds of medicinal herbs were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidant activity and growth inhibition activity against cancer cells. Results : Electron donating abilities (EDA) of the water and ethanol extract from Zizyphus jujuba and Atractylodes macrocephala were over than 87% at 1,000 ${\mu}g/mL$, respectively. The highest nitrite scavenging abilities (NSA) of the water and ethanol extracts from Scutellaria baicalensis were 68.9 and 79.2% at 1,000 ${\mu}g/mL$, respectively. Atractylodes macrocephala extracts had shown the highest growth inhibition activity against both of MDA and A549 cells and the water extract had higher activity than ethanol extract. Conclusions : These results suggest that the extracts of medicinal herbs used for this experiment had specific biological activity and they can be used as natural antioxidant to prevent oxidative damage in normal cells.

  • A series of novel exomethylene cyclopropyl nucleosides have been synthesized starting from Feist's acid. Classical nucleophilic substitution conditions ($K_2CO_3$, 18-crown-6) of the tosylate 2 as well as Mitsunobu reaction (DEAD, $PPh_3$) of alcohol 1 with pyrimidine bases afforded a series of novel cyclopropyl nucleosides. Compound 4b displayed moderate anti-HBV activity without any cytotoxicity up to $100{\;}{\mu}M$.
