Screening for Antioxidative Activity of Oriental Medicinal Plant Materials (시판 한약재에 대한 항산화 활성 검정)

  • Antioxidative effect of 42 medicinal plant extracts was screened to search natural antioxidants. The extracts of Terminalia chebula, Caesalpinia sappan, Sanguisorba officinalis, Rubus coreanus, and Alpinia katsumadai showed strong inhibition effect on DPPH radical, and LDL oxidation. Those of Eugenia caryophyllata, Alpinia officinarum, Zingiber officinale, Xanthium strumarium, Sophora japonica, Aristolochia contorta, and Alpinia katsumadai exhibited potent antioxidative activity on linoleic acid peroxidation. Among 42 medicinal plant materials, the extract of Terminalia chebula showed the highest scavenging activity $(3.08\;{\mu}g/ml)$ on DPPH radical which was higher than that $(13.52\;{\mu}g/ml)$ of ${\alpha}-tocopherol$. The extract of Terminalia chebula showed also stronger inhibition activity on LDL oxidation than that of ${\alpha}-tocopherol$ did.

Antifungal and Insecticidal Activity of Methanol Extracts from 17 Medicinal Plants against Biological Deterioration of Organic Cultural Heritage (약용식물 17종 메탄올 추출물의 유기질문화재 생물학적 열화원에 대한 항진균 및 살충활성 조사)

  • To investigate bioactive materials for development of natural conservative agent on organic cultural heritage, methanol extracts from 17 medicinal plants were screened for antifungal and insecticidal activity against 6 wood rot fungi and adult of Lasioderma serricorne. Antifungal activity of extracts was tested by using paper disc soaking method against wood-rot fungi. Among these extracts, the most significant antimicrobial activity was observed from the extract of Coptis japonica at 5 mg/disc against all wood rot fungi tested. The insecticidal activity of extracts was examined by topical application against L. serricorne adults. The extracts from Asarum sieboldii gave 83.3 % mortality at $25{\mu}g/adult$ for 24hr. From these result, the methanol extract from C. japonica and A. sieboldii described could be useful for conservation of organic cultural heritage against biological deterioration by wood rot fungi and L. serricorne.

Anti-inflammatory Effect of Leaves Extracts from Aralia cordata through Inhibition of NF-κB and MAPKs Signaling in LPS-stimulated RAW264.7 Cells

  • Aralia cordata (A. cordata), which belongs to Araliaceae, is a perennial herb widely distributed in East Asia. We evaluated the anti-inflammatory effect of stems (AC-S), roots (AC-R) and leaves (AC-L) extracted with 100% methanol of A. cordata and elucidated the potential signaling pathway in LPS-stimulated RAW264.7 cells. The AC-L showed a strong anti-inflammatory activity through inhibition of NO production. AC-L dose-dependently inhibited NO production by suppressing iNOS, COX-2 and $IL-{\beta}$ expression in LPS-stimulated RAW264.7 cells. AC-L inhibited the degradation and phosphorylation of $I{\kappa}B-{\alpha}$, which donated to the inhibition of p65 nuclear accumulation and $NF-{\kappa}B$ activation. Furthermore, AC-L suppressed the phosphorylation of ERK1/2 and p38. These results suggested that AC-L may utilize anti-inflammatory activity by blocking $NF-{\kappa}B$ and MAPK signaling pathway and indicated that the AC-L can be used as a natural anti-inflammatory drugs.

Antioxidant, Antimicrobial and Anti-inflammatory of Mixed Medicinal Herb Extract (한약재 복합 추출물의 항산화, 항균 및 항염 효과)

  • Objectives : This paper aimed to verify the applicability of mixed extract ofAngelica gigasNakai,Cnidium officinaleMakino,Paeoniala ctifloraPall,Rechmannia glutinosaLibosch,Scutellaria baicalensisGeorgi, which were prescribed for improving inflammation in Donguibogam, as the materials for beauty food and functional medicinal herb cosmetics by manufacturing such mixed extract and evaluating the biological activity of the extract.Methods : The mixed medicinal herb water extract(MMW) and ethanol extract(MME) were freeze-dried to be used as the specimen. We performed electron donating ability, lipid acidification inhibitory activity, anti-inflammatory activity against skin flora, MTT assay, NO inhibitory activity and the protein expression inhibitory activity of iNOS and COX-2.Results : For anti-oxidation experimentation, the electron donating abilities of MMW and MME were above 60.0% and 90.0% at 500 μg/ml, respectively. In the inhibition rate of lipid peroxidation, MMW and MME showed 43.1% and 52.1% at 1,000 μg/ml, respectively. As a result of antimicrobial activity, both the MMW and MME showed significant clear zones forPropionibacterium acnesat 4 mg/disc, but did not indicated the clearzones forStaphylococcus aureus, Escherichia coliandStaphylococcus epidermidis. Anti-inflammatory activity by NO assay showed LPS-induced NO was significantly inhibited in a concentration-dependent manner. Also, the expression of iNOS and COX-2 proteins were significantly inhibited following treatment with MMW and MME of 50 μg/ml.Conclusions : Accordingly, it can be concluded that mixed medicinal herb extract has the potential to beused as a functional food and cosmetic material.

Antioxidant and Antibacterial Activities of the Byproducts of Abies holophylla Extract (전나무 부산물 추출물의 항산화 및 항박테리아 활성)

  • Background: The extract of Abies holophylla is used as an ingredient in cosmetics. This study assessed the antioxidant and antibacterial activities of the material remaining after the extract is used. Methods and Results: The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azinobis (3-ethyl benzothiazoline)-6-sulfonic acid (ABTS) radical scavenging abilities were assessed to determined the free radical scavenging activity. The total phenol and flavonoid contents were determined to measure the antioxidant activity. The DPPH and ABTS radical scavenging activities of the resudual extract were higher (95.61 - 99.42% and 74.26 - 77.98% in water extract respectively) than those of the positive control. In 50% EtOH extract, the total phenol content was $389.84mg{\cdot}GAE/m{\ell}$, and the total flavonoid was $0.15mg{\cdot}QE/m{\ell}$. The minimum inhibition concentration degree for antibacterial activity against Staphylococcus aureus was < 8 to < $125{\mu}g/m{\ell}$ compared to that of the positive control in all extracts. The clear zone against S. aureus was found to be $12.2{\pm}3.8mm$. Conclusions: The A. holophylla byproducts were found to have antioxidant and antibacterial activities. Therefore, the materials remaining after the A. holophylla extract is used in cosmetics has potential functional uses.

Anti-Oxidative and Anti-Inflammatory Activities of Seven Medicinal Herbs including Tetrapanax papyriferus and Piper longum Linne (통초, 필발을 포함한 7종 한약재 추출물의 항산화 및 항염증 활성)

  • In this study, we analyzed the anti-oxidative and anti-inflammatory activities of seven medicinal herbs. All extracts of the tested herbs, Euryale ferox Salisbury, Echinops setifer Iljin, Amomum cardamomum Linne, Tetrapanax papyriferus, Illicium verum Hook. f., Typha orientalis Presl, and Piper longum Linne, exhibited potent anti-oxidative activity as confirmed by DPPH radical scavenging capacity. Lipopolysaccharide (LPS) induced nitric oxide (NO) production, in the RAW 264.7 cell line, was also ameliorated by all extracts' treatments in a dose dependent manner. NO suppressive activity originated from the inhibition of inducible nitric oxide synthase (iNOS) protein expression by the extracts. Three extracts, E. ferox S., I. verum Hook. f., and P. longum L., possessed suppressive activity against, not only iNOS, but also cycloxygenase 2 (COX-2) protein expression. These three extracts may then serve as potential candidates for non steroidal analgesic inflammation drugs (NSAIDs). Furthermore, all extracts induced anti-oxidative enzyme, heme oxygenase 1, protein expression. Taken together, these results provide an important new insight into the fact that various medicinal herbs possess potent anti-oxidative and anti-inflammatory activities and might be utilized as promising agents in the field of health products. Further studies for the identification of the active compounds from medicinal herbs are clearly needed.

Anticancer and Antioxidant Activity of Allergen-Removed Extract in Rhus verniciflua Stokes

  • Allergen-removed-extract was produced from Rush verniciflua by two phase methods. Phase one was high temperature treatment of Rush verniciflua tree to get allergen-removed-extract. Phase two was extraction of solution from phase one product using water or organic solvents. The solutions from above method show high antioxidant activity, anticancer activity, and improvement in lung function, but did not contain urushiol family compounds.

Inhibitory Activity of Medicinal Plants against Differentiation of Osteoclasts (생약의 파골 세포 분화 저해활성 검색)

  • Bone is continuously remodeled by osteoblasts and osteoclasts. Osteoclasts play an important role in bone metabolism by resorbing the bone matrix. Thus, the compounds inhibiting osteoclasts can improve bone diseases such as osteoporosis. The methanol extracts of 159 herbal medicines were screened for the inhibitory activity against differentiation of osteoclasts. Among the tested extracts, Achuranthis Radix and Corydalis Tuber showed relatively strong inhibitory activity against differentiation of osteoclasts, whereas they have no significant effect on proliferation of osteoclasts.
