  • The effect of dietary fat on the immunotoxicity of chloramphenicol was investigated in mice, sensitized and challenged with sheep red blood cells. Chloramphenicol suppressed more cell-mediated immunity than humoral immunity. The saturated fat diet elevated humoral and cell-mediated immunotoxicity, whereas the unsaturated fat diet decreased humoral immunotoxicity, but elevated cell-mediated immunotoxicity of chloramphenicol. Especially, the normal diet in saturated and unsaturated fat restored immunosuppressive effect of chloramphenicol compared to the saturated fat diet and the unsaturated fat diet.

  • This study analyzes targeting 584 senior citizens aged 65 and over in Gyeongsangnam-do to verify a mediated control effect of optimism and family support in stress and successful aging of the aged. Using SPSS Macro Way suggested by Preacher and Hayes. A study model is verified through a route model, and the verification of the study model is conducted in three stages. The first stage analyzes whether a control variable has a control effect in a relationship between predictor variable and standard variable, and the second stage confirms a relationship between predictor variable and mediated variable. In addition, the third stage confirms a mediated control effect. An analytical result is summarized as follows. First, as a result to verify a mediated effect between stress and successful aging, and optimism, it is confirmed that it supports a partially-mediated model. Second, as a result to verify a control effect of optimism and family support of successful aging, it is confirmed that everything has a control effect. Third, as a result to verify a mediated control effect of stress, optimism, successful aging, and family support, there is a mediated control effect. Based on these study results, practical undertone and future task are suggested, and a way to improve family support is presented for successful aging in advanced age.

  • Objective: This study focused on the relationships between emotionality, social competence and play behavior in a sample of 218 4- to 5-year olds attending daycare centers. Methods: All variables were measured by the daycare teachers of the surveyed children. The data were analyzed using Simple Regression and Multiple Regression through SPSS Win 25.0. Results: First, preschoolers' positive emotionality had a negative effect on their play disruption and play disconnection and a positive effect on play interaction. On the other hand, their negative emotionality had a positive effect on play disruption and play disconnection and a negative effect on play interaction. Second, their positive emotionality had a positive influence on social competence and their negative emotionality had a negative influence. Finally, the effect of their positive emotionality on play disruption was partially mediated, and their play disconnection and play interaction was totally mediated by social competence. On the other hand, the effect of preschoolers' negative emotionality on play disruption and play disconnection was totally mediated, and their play interaction was partially mediated by social competence. Conclusion/Implications: The results of this study imply that we need to help preschoolers promote emotionality and social competence together to change their play behavior positively.

  • Background: Fc receptor-mediated phagocytosis is a complex process involving the activation of kinases and phosphatases. FcgammaRIIB has been known to transduces inhibitory signals through an immunoreceptor tyrosine-based inhibitory motif (ITIM) in cytoplasmic domains. In this study, we examined the involvement of inositol-phosphatase in the Fc receptor-mediated phagocytosis. Methods: J774 cells were infected using vaccinia viral vector containing SH2 domain-containing inositol-phosphatase (SHIP) cDNA and stimulated with the sensitized sheep red blood cells. Results: Stimulation of J774 cells induced the tyrosine phosphorylation of SHIP which was maximal at 5 minutes. Phosphatidylinositol-3 (PI-3) kinase inhibitor (wortmannin) inhibits J774 cell phagocytosis of sensitized sheep red blood cells in a dose-dependent manner. Heterologious expression of SHIP in J774 cells inhibits phagocytosis of sensitized sheep red blood cells in a dose-dependency manner, but catalytically dead mutants of SHIP has no effect on phagocytosis. Conclusion: These results strongly suggest that the active signals mediated by PI-3 kinase are opposed by inhibitory signals through SHIP in the regulation of Fc receptor-mediated phagocytosis.

  • Objective: The aim of this study is to investigate the effect of Injin butanol fractions with Thin Layer Chromatography on Fas-mediated Apoptosis. Method: Injin-butanol fraction separated by TLC. MIT assay, cell cycle analysis, Caspase-3 protease assay, DNA fragmentation assay and quantitative RT-PCR were performed to evaluate the effects of TLC extraction of lnjin-butanol fraction on cell viability, cell cycle progression and apoptosis. Results: Scopoletin, luteolin, apigenin and unknown powder was isolated by TLC. Fas-mediated apoptosis analysis shows that scopoletin has inhibiting function on apoptosis. Caspase- 3 protease assay analysis shows that scopoletin inhibits activity of caspase-3. Quantitative RT-PCR analysis shows that no activity on caspase-3, but apoptosis inhibition cytokine -Bcl-2- is activated, and apoptosis activating cytokine -Bax- is unactivated. Conclusion: These results show that each fraction of Injin-butanol TLC extraction, especially scopoletin, acts as a protective function on liver cell viability, and inhibitory function on apoptosis.
