Classification of Cordyceps Species Based on Protein Banding Pattern (단백질 분석을 기초로한 Cordyceps속 동충하초의 분류)

  • In order to find relationship within and between entomopathogenic species, analysis of protein band pattern in mycelia of 25 isolates was conducted by UPGMA. The results allowed differentiation of three groups on 85% similarity coefficient. Similarity coefficient within C. militaris was $0.787{\sim}1.000$, C. kyushuensis was 0.958-1.000 and C. pruinosa was 0.993-1.000. C210 and C298 isolates which had somewhat immersed perithecia, comparable to other C. militaris isolates, had 91% similarity. C108, C225-1 and C228 isolates pathogenic on Lepidopterous larvae had 89% similarity. Closely related species to C. militaris were C. kyushuensis and C. pruinosa. And similarity between C. pruinosa and C. kyushuensis was 88%. Similarity between C. bifusispora formed conidia on media and Paecilomyces tenuipes was 89%. C. scarabaeicola pathogenic specifically on adult Scarabaeidae had 82% similarity with above two species. C118 identified as C. militaris showed different protein banding patterns.

Evaluation of Conventional Prediction Models for Soil Thermal Conductivity to Design Horizontal Ground Heat Exchangers (수평형 지중열교환기 설계를 위한 토양 열전도도 예측 모델 평가)

  • Among the various thermal properties, thermal conductivity of soils is one of the most important parameters to design a horizontal ground heat exchanger for ground-coupled heat pump systems. It is well known that the thermal conductivity of soil is strongly influenced by its density and water content because of its particulate structure. This paper evaluates some of the well-known prediction models for the thermal conductivity of particulate media such as soils along with the experimental results. The semi-theoretical models for two-component materials were found inappropriate to estimate the thermal conductivity of dry soils. It comes out that the model developed by Cote and Konrad provides the best overall prediction for unsaturated sands available in the literature. Also, a parametric analysis is conducted to investigate the effect of thermal conductivity, water content and soil type on the horizontal ground heat exchanger design. The results show that a design pipe length for the horizontal ground heat exchanger can be reduced with an increase in soil thermal conductivity. The current research concludes that the dimension of the horizontal ground heat exchanger can be reduced to a certain extent by backfilling materials with a higher thermal conductivity of solid particles.

Optimization of Submerged Cultivation of Hericium erinaceum (Hericium erinaceum 액체배양의 최적화)

  • Recently, it has been known that Hericium erinaceum is a one of the very useful functional materials with great attention in mushroom processing industry. In present study, a liquid culture which was not studied systematically until now, was conducted as a method of cultivation for H. erinaceum, and also examined the characteristics of the liquid culture and conditions of process optimization. A good basal medium was selected through the cultivation of 16 species mushroom media and the optimum condition for medium and cultivation were chosen by response surface method. From these results, the optimum condition of medium for mushroom was 3% glucose, 0.2% yeast extract/peptone(1:1) and 0.1% $KH_2PO_4/MgSO_4$(1:1) and also the optimal culture condition was obtained at inoculum of 13.42%, temperature of $22.3^{\circ}C$ and pH of 5.7. The mycelial dry weight of 9 g/I was obtained under these conditions and this amount was about 1.7 times higher than that which were cultivated in basal medium for 8 days.

Purification of Methioninase from Pseudomonas putida and Its Effect on the Uptake of ^11C-Methionine in Vivo. (Pseudomonas putida 유래 Methioninase의 정제 및 생체내 ^11C-Methionine 섭취에 미치는 영향)

  • Purification of methioninase resulted in a yield of 69%, and SDS-PAGE analysis of the purified product revealed a single band of approximately 43 kDa in molecular weight. in vitro experiments with cancer cells incubated in methionine-free media demonstrated an increase in $^{11}$ C-methionine uptake to 25.8$\pm$1.1% at 6 hr, 31.8$\pm$0.8% at 24 hr, and 62.2$\pm$0.6% at 48hr, compared to controls. Treatment of the cancer cells with purified methioninase showed no decrease in survival after a 2 hr incubation with 0.01 U/ml, but survival of RR1022 cells decreased 30% after 24 to 48 hr incubation. SKOV-3 cells showed a 5% and 14% decrease in survival with 0.1 and 1 U/ml methioninase after 24 hr. After 48hr survival decreased 15% and 24% with 0.1 and 1 U/ml methioninase. Measurements of $^{11}$ C-methionine uptake in RR1022 cells demonstrated no change at 2 hr, but a 13.7$\pm$4.7% and 40.7$\pm$2.6% increase in uptake at 24 and 48 hr, respectively. SKOV-3 cells also showed no change at 2 hr, but had a 17.7$\pm$7.2% and 38.9$\pm$4.9% increase in $^{11}$ C-methionine uptake after 24 hr and 48 hr treatment with methioninase, respectively. $^{11}$ C-methionine PET imaging revealed clear visualization of both the tumors and contralateral infectious lesions. Administration of rMET appeared to result in a slight increase in tumor:nontumor contrast on $^{11}$ C-methionine PET images. Injection of purified methioninase also produced PET images where tumor uptake was higher than that of infectious lesions.

Effects of Swim Training on Tuberculosis infection in the Mouse Model at Different Temperature (온도 차이에 따른 수영훈련이 결핵균 감염정도에 미치는 영향)

  • Tuberculosis is the leading infectious disease in the world. It is urgent to develop new vaccine and treating drugs. Besides vaccines, we want to know the effects of regular swim training on TB infection in the mouse model. This study was designed to examine the effects of regular swim training on lung and spleen TB counts and $INF-\gamma$ activity in the trained mice at different temperature. The trained mice underwent a 10-wk endurance swim training (5 times/wk) in water at $29\~33^{\circ}C$ (WWG) and $21\~23^{\circ}C$(CWG) for 60 min. And they were divided into 3 groups according to the regular swim training (CG; control, WWG; warm water group, and CWG; cold water group). Mice were challenged by aerosol infection with M. tuberculosis H37Rv using an inhalation device (Glas-Col, Terre Haute, Ind.) calibrated to deliver bacteria into lungs. Three weeks after immunization, the mice were challenged. Four weeks after challenge, the mice were sacrificed and the numbers of viable bacteria in lung and spleen were determined by plating serial dilution of whole organ homogenates on nutrient Middlebrook 7H11 agar (Difco, Detroit, MI). Colonies were counted after four weeks incubation at $37^{\circ}C$. All data were expressed as mean, standard deviation by using SPSS package program (win 10.0). The result through the statistical analysis of this data were summarized as follows; In the weight changes, there were significant differences among CG, WWG, and CWG following the swim training at different temperature, and CWG was the lowest. In the change of $INF-\gamma$ following the swim training, there were significant differences (p<.05) among CG, WWG, and CWG after stimulated with media and CFP. In MTB counts, there were significant differences (p<.05) between CG and WWG in the lung. And also there were significant differences (p<.05) among CG, WWG, and CWG. These results suggest that regular swim training suppress Th1 immune response caused by decreased $INF-\gamma$ level in the WWG, Also For the WWG, highly increased level of TB counts appear in the lung and spleen compare to CG.

Relationship between Half Cell Potential and Corrosion Amount Considering Saturated Cover depth and W/C ratios in Cement Mortar (습윤상태의 피복두께와 물-시멘트비를 고려한 반전위와 철근 부식량의 상관성)

  • Concrete is a construction material with porous media and corroded steel inside affects negatively to durability and structural safety. This study aims a derivation of quantitative relationship between measured HCP (Half Cell Potential) and corrosion amount considering cover depth and W/C (water to cement) ratio. For the work, cement mortar specimens with 3 different W/C ratios and 4 different cover depths are prepared, HCPs are measured with 3 different corrosion level. HCP measurement significantly increases in the saturated condition and linear relationship is observed between corrosion level and acceleration period. With increasing corrosion level and W/C ratio, and decreasing cover depth, HCP measurement increases. Considering total corrosion level and HCP measurements, relatively low COV(Coefficient of Variation) of 0.67 is evaluated through multi-linear regression analysis, however higher COVs over 0.90 can be obtained considering level of HCP measurement. In the room condition, corrosion level can be evaluated through measured HCP in the given conditions of cover depth, W/C ratio. diameter of steel inside.

Spatial Characteristics of Korean Residential Distribution and Occupational Composition in China, United States, and Japan (재중.재미.재일동포의 거주지 분포와 직업구성의 공간적 특성)

  • This study examines spatial characteristics of residential distribution and occupational composition of Korean emmigrants who live in China, United States, and Japan. The data used are The Conditions of Korean Emmigrants published by Ministry of Foreign Affairs in 1995. Analysis method used this data are as follows: 1) to clarify the tendency and spatial distribution of Korean emmigrants in each nation. 2) to grasp the residential distribution of Korean emmigrants in China, United States, and Japan where many Koreans have lived. 3) to analyze the occupational composition and its spatial characteristics of Korean emmigrants. The main findindgs obtained are summarized as follows: Farmer, forester, stock farmer, and fishery occupy over 50% of the employees of Korean emmigrants in China; traders and other employees occupy about three-fourths of the employees of Korean emmigrants in United States; and other employees occupy about 80% of the employees of Korean emmigrants in Japan. Therefore, the ratio of occupational composition of Korean emmigrants was influenced by emmigration motive, level of economic development of emmigration nation, restrictive condition for ethnic minority, and social status before emmigration etc. Specialized occupational composition in region where many Korean emmigrants in each nation lived was that the highest specialized occupation in region including primary city in population scale is trader; the highest specialized occupation in region including secondary city is manufacturing employee, and the highest specialized occupation in region including third city has transitional characteristics of each region including primary and secondary city. And professional occupation such as lawyer, doctor, religionist and educator appeared to the region including primary city. Finally, Korean emmigrants in United States and Japan contributed to the revitalization of inner city areas. And potential ethnic organization can be seen in church, mass media, and enterprise association in United States, but it can be seen in community shopping association in Japan, Because American society is based on Christianity but Japanese society is not.

An Analysis of the Research Trend of the Participants in Emotional Science in Korea (한국의 감성과학 연구참여분야 동향 분석)

  • This study aims to analyse the research trend of the participants in Emotional Science in Korea and to suggest a desirable direction of research. I analysed the 1,262 articles published in Korean Journal of the Science of Emotion and Sensibility from 2000 to 2011 according to fields of study and keywords. The findings from the study are as follows. In the fields of study regarding Clothing Design, Psychology, Medicine, Visual Media, Brain Science they are still the top rated categories amongst participants. With the participation of Industrial Engineering and Ergonomics they have shown a meaningful difference depending on governmental support such as G7 Project. The problem with Industrial Design is that the majority of studies have been performed alone, and they have many keywords which are not related with Emotional Science. However, 31 new fields of study have been added to the research making the findings more desirable considering that Emotional Science is typical interdisciplinary filed of study. I hope that this study will provide basic data which can be used to promote the interdisciplinary research between the different fields of study in order to generate significant synergy.

Identification of a New Agar-hydrolyzing Bacterium Vibrio sp. S4 from the Seawater of Jeju Island and the Biochemical Characterization of Thermostable Agarose (제주도 연안 해양에서 분리한 한천분해 미생물 Vibrio sp. S4의 동정 및 내열성 agarase의 생화학적 특성)

  • Agar-hydrolyzing bacteria were isolated from the coastal sea water of Jeju Island. One isolate, designated as S4, was selected for further study. The S4 cells were Gram-negative and rod-shaped with smooth beige surfaces and single polar flagellum. Cells were grown at $15-42^{\circ}C$, 0.5-5% (w/v) NaCl, between pH 6.0 and 9.0, and in media containing 0.5-5% (w/v) NaCl. The G+C content was 49.93 mol%. The major fatty acids (>15%) were $C_{18:1}{\omega}7c$, $C_{16:0}$ and Summed feature 3 (comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH). Based on 16S rRNA sequencing and biochemical and chemotaxonomic characteristics, the strain was designated as Vibrio sp. S4. In liquid culture supplemented with 0.1% agar the cell density and agarase activity reached a maximum level in 72 h, while agarase activity in the culture without agar was negligible, implying agarose expression is induced by agar. The optimum pH and temperature for the extracellular crude agarase of S4 were 7.0 and $45^{\circ}C$, respectively. However, it also exhibited 98.6% and 87.6% at $40^{\circ}C$ and $50^{\circ}C$, respectively, of the maximum activity seen at $45^{\circ}C$. The crude agarase hydrolyzed agarose into (neo)agarotetraose and (neo)agarohexaose.

Detection of Endolichenic Fungi Producing Antifungal Compound (항진균성 물질을 생산하는 지의류 내생 곰팡이의 탐색)

  • To isolate a novel antifungal compound, we obtained 100 kinds of endolichenic fungi from Korean Lichen & Allied Bioresources Center and examined their antifungal capability. Three fungi Usnea rigidula (2326), Parmotrema pseudotinctorum (2202) and Myelochroa sp. (2292) showed high antifungal activity against Candida albicans when they grew in both liquid and solid media. We extracted the culture supernatants of these three fungi with chloroform and then with ethyl acetate. Chloroform fraction exhibited the highest antifungal activities when those fractions were examined for the growth inhibition of Candida albicans with disc diffusion method. The chloroform faction was on further analysis with $C_{18}$ column chromatography to see whether the inhibitors are already known or not. Two peak fractions were collected from 4-day culture extract for Usnea rigidula and from 6-day culture extract for Parmotrema pseudotinctorum on the HPLC. A peak fraction from chloroform extracts of 4-day culture filtrate of Parmotrema pseudotinctorum showed higher antifungal activities against C. albicans and C. glabrata than another peak fraction. It appears that the antifungal materials are relatively nonpolar as usnic acid often found in lichenic fungi.
