Sensing Nitrate and Potassium Ions in Soil Extracts Using Ion-Selective Electrodes (이온선택성 전극을 이용한 토양추출물의 질산 및 칼륨이온 측정)

  • Automated sensing of soil macronutrients would allow more efficient mapping of soil nutrient spatial variability for variable-rate nutrient management. The capabilities of ion-selective electrodes for sensing macronutrients in soil extracts can be affected by the presence of other ions in the soil itself as well as by high concentrations of ions in soil extractants. Adoption of automated, on-the-go sensing of soil nutrients would be enhanced if a single extracting solution could be used for the concurrent extraction of multiple soil macronutrients. This paper reports on the ability of the Kelowna extractant to extract macronutrients (N, P, and K) from US Corn Belt soils and whether previously developed PVC-based nitrate and potassium ion-selective electrodes could determine the nitrate and potassium concentrations in soil extracts obtained using the Kelowna extractant. The extraction efficiencies of nitrate-N and phosphorus obtained with the Kelowna solution for seven US Corn Belt soils were comparable to those obtained with IM KCI and Mehlich III solutions when measured with automated ion and ICP analyzers, respectively. However, the potassium levels extracted with the Kelowna extractant were, on average, 42% less than those obtained with the Mehlich III solution. Nevertheless, it was expected that Kelowna could extract proportional amounts of potassium ion due to a strong linear relationship ($r^2$ = 0.96). Use of the PVC-based nitrate and potassium ion-selective electrodes proved to be feasible in measuring nitrate-N and potassium ions in Kelowna - soil extracts with almost 1 : 1 relationships and high coefficients of determination ($r^2$ > 0.9) between the levels of nitrate-N and potassium obtained with the ion-selective electrodes and standard analytical instruments.

  • Many studies have been done to measure and analyze color for various purposes. Visual assessment has lack of objectivity and the equipment for color measurement is very expensive. In this study, we developed a device for quantitative analysis of the color using optical elements. With the color sensor, the ratio of RGB was calculated by measuring the light intensity that is reflected from an object. Inverse transformation of optical signal was performed to detect the color density. The suggested color analyzer can detect color information as well as sample concentration. Results of this study are expected to be used in various medical fields such as pH indicator and urine analysis.

Shelf Life Extension of White Rice Cake and Wet Noodle by the Treatment with Chitosan (키토산 처리에 의한 흰떡과 생면의 저장성 연장)

  • The effect of treatments with alcohol, chitosan, or with both alcohol and chitosan for extending the shelf-life of Korean white rice cake and wet noodle was investigated by measuring quality changes such as total microbial count, pH, and sensory qualities. Total microbial counts for control, alcohol-treated, and 1% lactic acid-treated white rice cakes exceeded the initial putrefactive criterion level of $1{\times}10^{6}\;CFU/g$ at 6, 27 and 20 days of storage, respectively. However, total microbial count of the white rice cake treated with chitosan was still less than the criterion level even at 76 days of storage. In the case of wet noodle, total microbial counts of control and alcohol-treated groups exceeded the criterion level within 7 days of storage, while that of chitosan-treated group was far less than the criterion level even at the end of storage of 82 days. Chitosan treatment extended the shelf-lives of both white rice cake and wet noodle appreciably.

Evaluation of Biogas Production Performance and Archaeal Microbial Dynamics of Corn Straw during Anaerobic Co-Digestion with Cattle Manure Liquid

  • The rational utilization of crop straw as a raw material for natural gas production is of economic significance. In order to increase the efficiency of biogas production from agricultural straw, seasonal restrictions must be overcome. Therefore, the potential for biogas production via anaerobic straw digestion was assessed by exposing fresh, silage, and dry yellow corn straw to cow dung liquid extract as a nitrogen source. The characteristics of anaerobic corn straw digestion were comprehensively evaluated by measuring the pH, gas production, chemical oxygen demand, methane production, and volatile fatty acid content, as well as applying a modified Gompertz model and high-throughput sequencing technology to the resident microbial community. The efficiency of biogas production from fresh straw (433.8 ml/g) was higher than that of production from straw silage and dry yellow straw (46.55 ml/g and 68.75 ml/g, respectively). The cumulative biogas production from fresh straw, silage straw, and dry yellow straw was 365 l-1 g-1 VS, 322 l-1 g-1 VS, and 304 l-1 g-1 VS, respectively, whereas cumulative methane production was 1,426.33%, 1,351.35%, and 1,286.14%, respectively, and potential biogas production was 470.06 ml-1 g-1 VS, 461.73 ml-1 g-1 VS, and 451.76 ml-1 g-1 VS, respectively. Microbial community analysis showed that the corn straw was mainly metabolized by acetate-utilizing methanogens, with Methanosaeta as the dominant archaeal community. These findings provide important guidance to the biogas industry and farmers with respect to rational and efficient utilization of crop straw resources as material for biogas production.

Food Quality Characteristics of Instant Gruel Prepared with Peeled Krill Euphausia superba Meat (크릴(Euphausia superba) 육을 이용한 인스턴트 죽의 품질특성 평가)

  • Instant krill gruel was produced using peeled Antarctic krill Euphausia superba as a high nutritional ingredient and the food quality was investigated. The food quality of krill gruel was examined by measuring proximate composition, cholesterol, calorie, Hunter color value, total amino acids, fatty acids, fluoride, viscoelastic properties, and by sensory evaluation. The krill gruel had a moisture content of 87% and a pH of 6.65. The krill gruel contained 51 kcal/100 g, and 0.1% fat and 3.5 mg/100 g cholesterol. Its fatty acid composition exhibited high levels of unsaturated fatty acids. The levels of oleic acid and linolenic acid were high, and n-3, n-6, and n-9 fatty acid contents ranged from 1% to 6%. The total amino acid content was 2132 mg/100 g, and the levels of glutamic acid, aspartic acid, leucine, alanine, and arginine were particularly high. Essential amino acids accounted for over 30% of the total amino acids. Fluoride level in the krill gruel was 3.07 mg/kg. The viscoelastic properties of the krill gruel were determined as 6.28 Pa at shear stress of 2.51 Pa. In the recovery test, the elastic restoring force after deformation was low.

The Effect of Hydrogen Peroxide Bleaching on the Properties of Unbleached Hardwood Kraft Pulp Adsorbed with Birchwood Xylan

  • Xylans are polysaccharides present in large amounts in cell walls of land plants. However, during kraft cooking, a high portion of hemicelluloses including xylans are dissolved in the cooking liquor. In the current trend for a more effective utilization of biomass, attention has been paid to the exploitation of xylans as strength-enhancing additives for paper. It is believed that surface xylan adds flexibility to the cell wall/fiber surface, resulting in stronger fiber-fiber joints or greater contact area between the fibers. Accordingly, there is proposal for a new pulping process involving the extraction of xylan prior to pulping, followed by their re-adsorption on the unbleached pulp. A suitable bleaching process should be employed then, which ought to does not only improve the brightness of the pulp, but also remain the effect of the adsorption of xylan on pulp fibers. The objective of this research was to investigate the impact of hydrogen peroxide bleaching on the properties of unbleached hardwood kraft pulp pretreated with birchwood xylan by measuring optical properties (brightness, post color number, opacity) as well as physical properties (tensile index, tearing index, bulk) of handsheets made from the bleached pulp. In the meantime, the influence of process variables of peroxide bleaching including bleaching temperature, time, initial pH and $MgSO_4$ dosage were studied.

Advanced Transmittance and Surface-Morphology of CdS thin films prepared by chemical bath deposition using various complexing agents for solar cells (용액 증착법으로 증착된 CdS 박막의 제조와 고상과 액상 화합제에 따른 표면 특성 비교)

  • In the past few years, the deposition and characterization of cadmium sulfide semiconducting thin films has received a considerable amount of interest due to their potential application in the area of electronic and opto-electronic devices fabrications. Polycrystalline CdS thin films posses good optical transmittance, wide band-gap and electrical properties makes it as one of the ideal material for their application to solar cell fabrication. Cadmium sulfate thin films were deposited by the chemical bath deposition method using tartaric acid and triethanolamine as a complexing agent. Deposition parameters such as pH, temperature, deposition time and concentration of the reactant species were optimized so as to obtain reflecting, good adherent uniform thin films on the glass substrate. Reaction mechanism of the thin film formation is also reported. The crystallographic structure and the crystallite size were studied by the X-ray diffraction pattern. The optical band-gap of deposited film is identified by measuring the transmittance in the visible region. Temperature dependence of resistivity confirmed the semiconducting behavior of the film. Scanning electron micrographs (SEM) showed the presence of grain particles of size 50 nm.

  • Domestic sewerage treatment plant is operated by activated sludge method and its modified method by using microorganism. In most cases, a method of using microorganism is directly controlled by the operator based on individual judgment through factors of DO, pH and ORP. In addition, under aerobic condition in bioreactor, energy consumption including excessive air injection is learned to be somewhat plenty. In order to solve this problem, in most of the process, improvement of internal recycling and activated environmental factor of microorganism were researched extensively. However, as factors are changed depending on various conditions, it is not sufficient as an indicator of judgment. As such, a research on operation of bioreactor that measures metabolic change in short time by directly measuring activated condition of microorganism using NADH fluorometer is required in reality.It is considered that the method like this could supplement problem of energy consumption being occurred in the existing treatment method and operational optimization of bioreactor would be enabled by controlling optimal air volume. Therefore, in this study, in order to obtain optimal operational indicator of bioreactor, proper air volume control test was performed and through batch test and site evaluation, possibility of NADH sensor being utilized as operational control indicator of bioreactor is intended to be analyzed. In order to compare with measured value, DO, ORP that are operational control indicator of existing bioreactor were used. As MLSS concentration was increased through batch test, NADH value was increased and site evaluation also showed similar tendency to batch test. Resultantly, it could be confirmed that changing level of NADH fluorometer was a sensor that could measure bioreactor condition effectively and optimized scale of bioreactor is considered to be utilized.

Development of Active thin Film Optical Waveguide $C^{2+}$ -ion Sensor (능동형 박막 광도파로 칼슘 이온 센서의 개발)

  • A new functional organic thin film optical waveguide ion sensor is designed, which can select a specific ion, i.e., $Ca^{2+}$ -ion. The sensing membrane was composed of PVC-PVAC-PVA copolymer matrix based on anionic cation-selective chromoionophor(ETH5294), neutral ionophore(K23El), anionic site and plasticizer and it was coated on the etched glass substrate as embeded type optical waveguide itself. The sensor sensitivity dependence on waveguide length and thickness, contence of chromoionophore, and each mode was investigated. And this sensor could detect $Ca^{2+}$ ion in concentrations ranging from 1$\times$10­6~1M(with 0.05M tris-HCI buffer solution of pH7.4) by measuring the absorbance change at 514nm of light. Utilizing thin film membrane, the fast response time and high sensitivity are obtained. Also, it is expected that this sensor can be applied to various biochemical important ions.ons.

Experimental Study on Dispersion and Thermal Properties of Nanofluids based on POE Oil (POE 오일 나노유체의 열물성과 분산성 평가를 위한 실험적 연구)

  • To apply the nanofluids into the general vapor compression cycle, basically have to know the thermal properties including thermal conductivity and dynamic viscosity. And needs to show the dispersion characteristics for various nanofluids and concentrations. So, firstly this study showed experimentally the thermal properties for various concentration (0.1%~0.7%, as mass balance) and temperature($20^{\circ}C{\sim}40^{\circ}C$) on $Al_2O_3$, $TiO_2$, and CuO nanofluids using base fluid as POE oil that has used in the scroll compressor for various refrigeration system. From the results, the dynamic viscosity of nanofluids was considerably changed from the base POE oil. And, the dispersion characteristics of various nanofluids using the simple methods like as analyzing the RGB value or measuring the sinking height of nanofluids were showed experimentally. Through the results, the dispersion characteristics of $Al_2O_3$ nanofluid was better than those of $TiO_2$, and CuO nanofluids not considering the real refrigeration cycle rurming conditions.
