This study was to investigate the effect on weight control and changes in serum glucose, total protein , GPT, GOT , cholesterol, triglyceride, calcium, and phosphorus contents of seventeen overweight female college students during experimental periods [before grape diet(0 day-, after grape diet (3 day), after control diet(3 day), after normal diet(30 day)]. The mean weight, height , BMI, and Rohrer index of the subjects at the onset of the study was 58.91$\pm$5.22kg, 159.94$\pm$6.01cm, 22.96$\pm$1.17, and 20.06$\pm$1.48 respectively. During the study subjects, conducted grape diets, control diets, and normal diets to control their weight. They analyzed their changes in anthrophometric variables. The mean weight was significantly decreased from 58.29$\pm$7.85kg to 57.2$\pm$8.23kg after normal diet(p<0.05). Therefore BMI (body mass index) and Rohrer index was also significantly decreased after experimental periods. The analysis of dietary intake showed that the grape diet induced unbalanced low nutrient intake, but the intake of total calories and other nutrients were almost recovered their onset of the study during control and normal diets. The concentration of serum glucose, cholesterol, triglyceride GOT, and GTP were significantly(p<0.05) decreased and significantly(p<0.05) increased again during experimental periods. Calcium concentrations were not affected by grape diet, but phosphorus concentration was significantly reduced after grape and control diet.
The metabolic patterns of C-1 and C-6-carbon atoms of glucose were observed in the tissue homogenates of the Ehrlich ascites tumor tissue which was incubated for 3 hours in the Dubnuff metabolic shaking incubator. $C^{14}-1-and\;C^{14}-6-glucose$ were used as tracers. The glucose media in which tissue homogenate was incubated was kept at a concentration of 200mg% glucose of carrier and appropriate amount of $C^{14}-1-or\;C^{14}-6-tracer$. At the end of 3 hour incubation, respiratory $CO_2$ samples trapped by alkaline which is placed in the tenter well of incubation flask were analyzed for the total $CO_2$ production rates and their radioactivities. The tissue homogenate samples after incubation were analyzed for their concentrations of glucose, lactate, pyruvate and glycogen and calculations were made on the glucose consumption rate, pyruvate and lactate accumulation rates. The following results were obtained. Data obtained in each group are as follows: 1. In the tissue homogenate, which was incubated with $C^{14}-1-glucose as a substrate, total $CO_2$ production rate averaged $19.0{\pm}5.0{\mu}M/hr/gm$ and the mean specific activity of respiratory $CO_2$ was $840{\pm}296\;cpm/mgC.$ Relative specific activity (RSA) which means the fraction of $CO_2$ derived from medium $C^{14}-1-glucose$ to total $CO_2$ production rate was calculated by ratio of SA of respiratory $CO_2$ and medium $C^{14}-1-glucose.$ RSA was $14.3{\pm}5.0%,$ Accordingly actual $CO_2$ production rate from medium $C^{14}-1-glucose$ showed a mean value of $2.79{\pm}1.35\;{\mu}m$ of which amount was equivalent to the mean value of total glucose consumption rate $(RGDco_2)$, namely, $5.1{\pm}1.3%.$ Lactate and pyruvate appearance rates averaged $7.13{\pm}1.26\;and\;0.21{\pm}0.02{\mu}M/hr/gm,$ respectively. Assuming that these 3 carbon compounds appeared in the medium were derived from glucose, calculations were made that relative glucose disappearance rate into lactate $(RGD_L)$ was $38.0{\pm}5.4%\;and\;RGD_P$ was $1.23{\pm}0.03%.$ Therefore, about 43.3% of the total glucose consumed were accounted for by conversion into the respiratory $CO_2$, lactate and pyruvate. 2. In the second group, which was incubated with $C^{14}-1-glucose$ as a substrate, glucose consumption rate, lactate and pyruvate appearance rates showed almost the same order as the values of the $C^{14}-1-glucose$ substrate group. However, RSA was remarkably decreased showing a mean value of $1.02{\pm}0.13%.$ This fact means that the C-6 carbon of glucose take the minor part in the oxidative metabolism of glucose. The glycogen level in both substrate tissue homogenate showed less than 0.3% of tissue weight. These low value suggested that there was an inhibition of carbohydrate synthesis in the Ehrlich ascites tumor tissue. 3. The catabolic pathway of glucose in the tumor tissue were analyzed on the basis of Bloom's principle from the values of RSA. It was found that in the tumor tissue more than 90% of $CO_2$ derived from glucose were oxidized via the alternate pathway other than principal EMP-TCA cycle such as hexose monophosphate pathway (HMP). From the data described above, it was assumed that in the Ehrlich tumor tissue anaerobic glycolysis proceeds normally although, the oxidation of products of anaerobic glycolysis via the TCA cycle is inhibited resulting in the accumulation of lactate and almost all of oxidative energy from glucose is released by oxidative pathway such as HMP.

Objective: The purpose of this study was to determine serum antioxidant nutrients and the relationship between serum antioxidants and risks of chronic diseases in obese Korean children Methods: Normal weight Korean school children (0=170), mean age of 11.5$\pm$1.5, and obese (body fat mass > $28\%$) children (0=176), mean age of 11.0$\pm$1.8, were recruited Fat mass ($\%$) was determnined by Bioelectrical Impedance (BEI), and body mass index (BMI) was calculated Fasting blood was collected to measure serum antioxidant nutrients, vitamin A, vitamin E and zinc. Serum lipid profiles including total cholesterol (TC), high density cholesterol (HDL) and triglyceride (TG), and blood glucose, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPI) were also determined. Differences in serum blood measurements between obese and normal children were assessed by independent t test. Pearson's correlation analysis was used to determine the relationship between variables. Results: Blood glucose, GPT, total cholesterol, and triglycerides concentrations were significantly higher among obese boys, compared to normal boys (p<005). Significantly lower concentrations of serum vitamin E, after adjustment for TG and TC, was shown in obese boys (0.26 mg/mg) and obese girls (0.31 mg/mg), compared to normal boys (0.36 mg/mg) and girls (0.38 mg/mg) (p<0.05). Fat mass ($\%$) was negatively con-elated with serum vitamin A and vitamin E. Conclusion: Obese Korean children showed insufficient serum vitamin E concentration and increased risk for diabetes, atherosclerosis, and liver disease. Since lower vitamin E concentration was negatively con-elated with atherogenic index, improved vitamin E status in children may decrease the risk of atherosclerosis later in life.

Solid lipid nanoparticles (SLNs) were freeze-dried to obtain a stable solid dosage form with the aid of various cryoprotectants such as trehalose, sucrose, glucose, fructose, and glycerol. Tricaprin(TC) and trilaurin(TL) were used as lipid matrices for SLNs and stabilizers were egg phosphatidylcholine and pegylated phospholipid. All cryoprotectants tested did not cause changes in mean particle size of SLNs when mixed with SLNs before freeze-drying. However, the mean particle sizes of reconstituted SLNs after freeze-drying were significantly different from those of the un-lyophilized original SLN dispersions depending on the types and concentration of cryoprotectants. Although the freeze-dried SLNs without any cryoprotectants were easily reconstituted by hand-shaking, the mean particle size drastically increased (> $8\;{\mu}m$ for TC SLNs and around $1\;{\mu}m$ for TL SLNs) compared to that of un-lyophilized original dispersion (97 nm for TC SLNs and 164 nm for TL SLNs). Trehalose and sucrose were the most effective additives to protect the SLNs during lyophilization. The reconstituted SLNs were physically stable for 24 hours when lyophilized with 12.5% trehalose, sucrose, glucose, fructose or glycerol.

The purpose of this study is to define the normal findings of cerebrospinal fluid (CSF) of the clinically healthy Beagle dogs and to provide basic information in diagnosis of neurologic disorders. CSF obtained from 13 clinically healthy dogs was examined for total and differential cell counts, total protein concentration, glucose and lactate dehydrogenase (LDH) concentration, specific gravity, turbidity, and protein electrophoresis. On gross examination, CSF samples evaluated were clear and colorless. Few red blood cells and nucleated cells were present. The mean concentration of glucose and LDH examined were 65.8 mg/dl and 2.7 mg/dl, respectively. The cellular components of CSF samples based on differential counts were monocytes (41.9%), activated macrophages (35.8%), lymphocytes (20.0%), neutrophils (1.6%), and eosinophils (0.7%). The fractions of electrophoretic protein in CSF were albumin (52.7%), alpha-globulin (16.5%), beta-globulin (24.8%), and gamma-globulin (3.0%). Results of albumin quota were ranged from 0.15 to 0.38. In conclusion, this study provided normal composition of CSF in Beagle dogs.
Blood glucose information is important for self regulation in daily life, but the frequency of self test remains to be only 17%(9 tests/month) in Korea, mainly due to pain during blood sampling. The present study tried to validate the clinical efficacy of the forearm as an alternative sampling site with minimized pain. Capillary blood was sampled both on the index finger($G_F$) and the forearm($G_A$), immediately followed by glucose measurements in 531 subjects, 25 who visited the Health Enhancement Center of C University Hospital, then venous blood($G_V$) was sampled for glucose test. The blood glucose concentration measured on the forearm was closer to the venous glucose than on the finger. The mean difference between $G_V$and $G_F$ was only 10 mg/dL well within the internationally accepted error limit. Error grid analyses of $G_F-G_V$, $G_A-G_V$ and $G_A-G_F$ revealed that the number of data points in regions A and B took 100%, 99.8%, and 97.9%, respectively. These results demonstrate the forearm blood glucose test is not only accurate but also clinically valid. Therefore, the forearm blood glucose test can be a useful way of self managing the chronic diabetes with minimized sampling pain.
For mass production of polyhydroxyalkanoates (PHA), high cell density cultures of Alcaligenes eutrophus by fed-batch culture under phosphate-limitation condition has been investigated. PHA accumulation by the regulation by the regulation of initial phosphate concentration could be automatically induced, and high density cell culture above 200 g/L also could be successfully produced. The production of Poly-$\beta$-hydroxybutyrate (PHB) and dry cell weight increased with increasing the initial phosphate concentration. When the initial concentrations of phosphate were in the ranges of 1.5~4.5 g-PO$_4$/L, PHB and dry cell weight obtained were 83~266 g/L and 61~216 g/L, respectively, and PHB productivity was in the ranges of 1.35~3.10 g/L.h. When a mixture of glucose and propionic acid is used as carbon sources, poly(3-hydroxybutyrate-co-poly-3-hydroxyvalerate), P(3HB-co-3HV), could be also successfully produced under phosphate limitation condition. When the mole ratio of propionic acid to glucose in the feeding solution is 0.22, a final dry cell weight of 150 g/L and a P(3-HB-co-3HV) of 90 g/L were produced. Morphological changes and size distribution of PHB granules synthesized in A. eutrophus under phosphate-limitation condition are determined by TEM during the course of fed-batch. Mean granule diameters of PHB produced are in the range of 0.36~0.39 $\mu$m, and mean cell size was elongated from 0.54~0.59 $\mu$m$\times$ 1.3~1.5 $\mu$m to 0.83~0.89 $\mu$m $\times$2.0~2.3 $\mu$m. Phosphate concentration in media did not affect size distribution of PHB granule and cell.
Twenty white rabbits anesthetized with nembutal (30 mg/kg) were employed in this experiment. Five of them served as controls; the remaining rabbits as experimental group were subjected to irreversible hemorrhagic shock. Shock was induced by bleeding the animals until mean blood pressure decreased to a level of 50-40 mmHg. This level of pressure was maintained for 3-4 hours, after which the drawn blood was reinfused. The reinfusion of blood caused the elevation of arterial pressure almost the control level for some minutes, after which a gradual and progressive decline of blood pressure became evident. This decline was thought to be the result from irreversible hemorrhagic shock. When mean blood pressure declined to less than 50 mmHg, chest was opened and samples of arterial blood and left ventricular muscle were taken. Left ventricular muscle and blood plasma were analyzed for potassium, sodium, chloride and water content. Blood glucose concentration was determined by Somogyi-Nelson's method. Extracellular and intracellular myocardial water and electrolyte content were calculated on the basis that electrolytes are distributed between plasma water and interstitial water according to Gibbs-Donnan equilibrium. In this calculation extracellular water was substituted for Na space. The findings obtained were as follows: 1. The concentration of blood glucose was 87mg% in the controls and it rose to 222 mg% in shock (P<0.01). 2. Plasma potassium elevated significantly from 3.3 mEq/l in controls to 8.0 mEq/l in shock (P<0.01), while small decreases in sodium (151-146 mEq/l) and chloride (102-96 mEq/l) were observed (P<0.3, P<0.1), 3. The changes of blood water content (83.1-84.3%) and cardiac water content (77.5-78.3 gm/100gm WT) were observed. 4. In control animals myocardial potassium levels which averaged 30.2 mEq/100 gmDT rose significantly to 40.3 mEq/100 gmDT in shock (P<0.01), while moderate decreases in sodium(16.3-14.3 mEq/100 gmDT) were observed in shock. 5. The calculated transmembrane resting potential of left ventricular muscle of control animals averaged 95 mV, while rabbits in shock averaged 77 mV. (P <0.01). The findings of this experiment do not correspond with the conclusions that myocardial depression seems to be the cause of irreversible hemorrhagic shock, because the excitability of heart muscle is elevated. From the point of view that the lowered transmembrane resting potential, the cause of death in terminal stage of irreversible hemorrhagic shock may be ventricular fibrillation. It can't be said, however, that the lowered transmembrane resting potential is responsible for the transition from reversible to irreversible hemorrhagic shock. The marked increase in blood glucose suggested that glycogenolysis in the liver is favorably active in shock.

Rockfish, Sebastes schlegelii (mean length $14.53{\pm}1.14cm$ and mean weight $38.36{\pm}3.45g$), were exposed for 4 weeks with the different levels of ammonia in the concentrations of 0, 0.1, 0.5, and 1.0 mg/L at 19 and $24^{\circ}C$. The indicators of growth performance such as daily length gain, daily weight gain, condition factor, and hematosomatic index were significantly reduced by the ammonia exposure and high temperature. The ammonia exposure induced a significant decrease in hematological parameters, such as red blood cell (RBC) count, white blood cell (WBC) count, hemoglobin (Hb), and hematocrit (Ht), whose trend was more remarkable at $24^{\circ}C$. Mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) were also notably decreased by the ammonia exposure. Blood ammonia concentration was considerably increased by the ammonia concentration exposure. In the serum components, the glucose, glutamic oxalate transaminase (GOT), and glutamic pyruvate transaminase (GPT) were substantially increased by the ammonia exposure, whereas total protein was significantly decreased. But, the calcium and magnesium were not considerably changed.
Continuous production of lactic acid from glucose by Lactobacillus rhamnosus with cell recycling using an acoustic cell settler was carried out. The performance of the system, such as the concentration of cell and product were compared with the control experiment without recycling. The acoustic settler showed cell separation efficiency of 67% during the continuous operation and the cell concentration in the fermentor with recycle exceeded that of the control by 29%. Com-pared with the control, tactic acid production was increased by 40%, while glucose consumption was only increased by 8%. The higher value of lactic acid production to substrate consumption (Yp/s, product yield coefficient) achieved by cell recycling is interpreted to indicate that the recycled cell mass consumes less substrate to produce the same amount of product than the control Within system environmental changes due to the longer mean cell residence time induced the cells maintaining the metabolic pathways to produce Less by-Product but more product, lactic acid.
