Conversion of water-insoluble components of the basidiocarps of ganoderma lucidum to water-soluble components by hydrolyzing with chitinase

  • We investigated the optimum conditions for conversion of water-insoluble components of basidiocarps of Ganoderma lucidum to water-soluble components by hydrolyzing with chitinase. We also tried it with Ganoderma luciclum residue remaining after extracting hot water-soluble components of Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chitinase, 2% Ganoderma lucidum or 6% Ganoderma lucidum residue, at pH 3 and at $ 35^{\circ}C$), the contents of total water-soluble components (polysaccharide or protein) were measured, and it was found that the contents of water-soluble components increased to 1.5-2.7 fold. Michaelis constant, $K_m$ and maximum rate, $V_max$ calculated by Lineweaver-Burk plot for hydrolysis of Ganoderma lucidum were 1.75% and 0.02%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 53.15% and 0.53%/min respectively The protein-bound polysaccharide was isolated after hydrolysis and molecular weights were measured by Sepharose CL-4B gel filtration and compared with the molecular weights of polysaccharide before hydrolysis.

Studies on the Pectolytic Enzymes from Byssochlamys fulva II (Byssochlamys fulca가 생성하는 펙틴질 분해효소에 관한 연구 II)

  • Polygalacturonase of Byssochlamys fulva was purified and characterized. Specific activity increased from 2.21 units/mg protein to 10.47 units/mg protein through $(NH_4)_2SO_4$ treatment, SephadexG-100 gel filtration, and DEAE-Sephadex ion exchange chromatography. Divalent cations, such as $Ca^{++}\;and\;Cu^{++}$, increased polygalacturonase activity greatly. Added as $10^{-3}M$ concentration, $Ca^{++}$ ion enhanced enzyme activity 9.8folds. Optimum temperature was $50^{\circ}C$ and optimum pH was 5.0. Activation energy of reaction was 8.69 Kcal/mole. Michaelis-Menten $constant(K_M)\;and\;V_{max}$ of reaction were $6.27{\times}10^{-3}mole/l\;and\;2.85{$\mu}moles/min$. Polygalacturonase of Byssochlamys fulva preferred polygalacturonic acid to pectin as substrate and was presumed as endo-type on the basis of the relationship between viscosity reduction and substrate degradation. Molecular weight of polygalacturonase was estimated as 55,000.

  • $\beta $-Glucosidase (EC 3.2.1.21) was purified from Aspergillus niger SFN-416 by a sequential process of ammonium sulfate precipitation, Sepadex G-100 and DEAE-Sephacel column chromatography. Molecular weight of the enzyme was 46, 000 daltons. The K$_{m}$ and V$_{max}$ values for PNPG were 0.67 mM and 25 moles/ml $\cdot $min., respectively. The optimum pH and temperature of the enzyme activity were 3.5 and 58$\circ $C, respectively. The enzyme activity was decreased by addition of metal ions, and increased by addition of metanol, ethanol, isopropanol and 1-butanol at a concentration of 10% (v/v). Stability of the enzyme was increased by addition of isopropanol and 1-butanol at a concentration of 10% (v/v).

  • In this paper, after we make high speed gear requested high precision, we got data on the sound level is Max 60,000rpm for tooth modification. We make a decision about tooth modification after predicting exactly amount of tooth interference by FEM. The purpose of this research is that modified gear is to drive under stable sound level. To accomplish its, Beam elements are used to simulate the same condition as a real gear drive by FEM. From result of simulation and experiment, if it has holes to decrease whole weight of high speed gear, it is not good effect because it has much more deformation than not.

  • Die sinking electrical discharge machining(EDM) was conducted for ceramic composite of 33 weight percent TiC based on AI$_{2}$O$_{3}$ ceramic matrix according to the change of current and duty factor(DF). Material removal rate(MRR) was increased as the current and the duty factor increased, but better surface mor-pholoty was obtained in the region of lower current and duty factor. From the scanning electron microscopy(SEM) photographs and the energy dispersive X-ray spectroscopy(EDX) of the EDMed surface, EDM trace formed by one discharge spark was analyzed. Although the bending strength after EDM was highly decreased, reliability obtained by weibull analysis was increased twice. The bending strength was recovered or more by barrel polishing after EDM. From the FEM analysis of temperature for one spark, the possible melting region of AI$_{2}$O$_{3}$and TiC was obtained.

Inhibitory Effects of Complex of Mulberry Extract on Degenerative Arthritis (상심자추출물 등 복합물의 퇴행성관절염 억제효과)

  • Complex of mulberry extract (CME) is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). In this study, we investigated prevention effects of CME on degenerative arthritis. The $SC_{50}$ value of DPPH radical scavenging by CME was $158.49{\pm}11.35{\mu}/mL$. We found that CME significantly reduced the production of nitric oxide (NO) and protein expression of cyclooxygenase-2 (COX-2) in RAW 264.7 cells which were activated by LPS. Experiments using animal model of degenerative arthritis showed that CME (400 mg/kg body weight) inhibited the production of TNF-${\alpha}$ (77.5%) and IL-$1{\beta}$ (95.0%). Furthermore, it was observed that CME reduced to 85.9% of paw edema induced by carrageenan. These results suggest that CME could improve degenerative arthritis.

  • Poly(A)+ RNA was selected from Canavalia lineata root nodule RNA through oligo(dT) cellulose column and used for construction of a cDNA library using λgt10-EcoRI arms. The size of the library was 7.2$\times$105 pfu/mL. A full length leghemoglobin (Lb) cDNA clone, pCILb1(687 bp) isolated with soybean Lb probe, contained one open reading frame (ORF) of 447 bp with 54 bp plus 186 bp at 5' and 3' untranslated region, respectively. A consensus sequence of plant translation start region (AAAATGGG) was found at 5' untranslated region, and two polyadenylation-related sequence (AATAAA, AATAAG) and a conserved motif between them (gACTTGTT) were found upstream of poly(A)+ tail consisted of 13 (A)s at 3' untranslated region. The ORF encoded a polypeptide consisted of 149 amino acids with a molecular weight of 16.2 kD. Deduced amino acid sequences showed high degree of homology values with those of other Lbs ranging from 66% (Casuarina glauca) to 85% (Glycine max).

  • Recently more attention is paid to the development of high-speed permanent magnet (PM) synchronous generators driven by gas-turbine, since they are conductive to high efficiency, high power density, small size, low weight, simple mechanical construction, easy maintenance and good reliability. In this paper, the performance analysis of a high-speed PM synchronous generator for military power application considering the min-max operating speed is presented. The output current and power versus DC-link voltage loci can obtained by solving the PM machine's steady-state equations for variable resistive load.

  • This Paper deals with the headlamp lens surface temperature distribution of p/c headlamp using the three categories of H4 halogen bulbs. Glass is gradually replaced by P/C for the lens material off vehicle headlamp due to the weight reduction and stream lined body of a vehicle. With this trend, the newly established standards for a headlamp with a P/C lens in Europe requires that the heat generated by a bulb should not distort the lens surface. Also the requirements fur the bulb of a headlamp are being enforced in U.S.A & Europe. However, such requirements are not established yet in Korea. By using three kinds (60/55w, 100/90w, 130/90w) of H4 halogen bulbs in this experiment, the surface temperature distribution and Max. temperature on the lens were measured. The results of this study implies the necessity of requirement fur the bulb off headlamp.

Degradation of Polyvinyl Alcohol by Brevibacillus laterosporus: metabolic Pathway of Polyvinyl Alcohol to Acetate

  • Approximately 0.1 mg/ml of polyvinyl alcohol (PVA) was degraded by the growing cell, Brevibacillus laterospours, for 30 h, and 0.2 mg/ml of PVA was degraded by the cell-free extract that was isolated from Brevibacillus laterosporus. Approximately $0.29{\mu}g$/ml of acetic acid was produced from PVA by using the cell-free extract as a catalyst for 40 min. $V_{max}\;and\;K_m$ value of purified PAV-degradation enzyme was 3.75g/l and 2.75 g/l/min in reaction with EDTA and 3.99 g/l and 2.98 g/l/min in reaction without EDTA, respectively. Molecular weight of the purified enzyme determined by SDS-PAGE was 63,000 Da. Alcohol dehydrogenase and aldehyde dehydrogenase activities were qualitatively detected on a native acrylamide gel by an active staining method, indicating the existence of the metabolic pathway to use PVA as a substrate.
