Thionins are a family of low molecular weight cysteine-rich antimicrobial peptides. We isolated a cDNA encoding thionin gene from a flower bud cDNA library of Chinese cabbage (CFT). The gene contains 611 by nucleotides with 60 bp, and 150 by untranslated regions at its N- and C-terminal, respectively. The deduced amino acid sequence encoded 133 amino acids containing precursor polypeptide. The protein reveals that the precursor has a tripartite structure: a putative signal sequence at the N-terminus, followed by a mature thionin peptide, and a C-terminal acidic domain, which facilitates transport of the mature thionin through membrane. Genomic Southern blot analysis suggests that the CFT gene may be present as a single or two copy gene in the Chinese cabbage genome. Northern blot analysis shows that the gene is specifically expressed in flowers, but not in leaves, stems, or roots. When we analyzed the antifungal activity of the recombinant CFT protein, which was expressed in E. coli using the truncated cDNA region corresponding to the mature protein part, it was not active on fungal growth inhibition.
Alkaliphilic Bacillus sp. S-1 secretes a large amount (approximately 80% of total pullulanase activity) of an extracellular pullulanase (PUL-E). The pullulanase exists in two forms: a precursor form (PUL-I: $M_r$ 180,000), and a processed form (PUL-E: $M_r$ 140,000). Two forms were purified to homogeneity and their properties were compared. PUL-I was different in molecular weight, isoelectric point, $NH_2$-terminal amino acid sequence, and stabilities over pH and temperature ranges. The catalytic activities of PUL-I were also distinguishable in the $K_m$ and $V_{max}$ values for various substrates, and in the specific activity for pullulan hydrolysis. PUL-E showed 10-fold higher specific activities than PUL-I. However. PUL-I is immunologically identical to PUL-E, suggesting that PUL-I is initially synthesized and proteolytically processed to the mature form of PUL-E. Processing was inhibited by PMSF, but not by pepstatin, suggesting that some intracellular serine proteases could be responsible for processing of the PUL-I. PUL-I has a different conformational structure for antibody recognition from that of PUL-E. It is also postulated that the translocation of alkaline pullulanase(AP) in the bacterium possibly requires processing of the $NH_2$-terminal region of the AP protein. Processing of the precursor involves a conformational shift. resulting in a mature form. Therefore. precursor processing not only cleaves the signal peptide, but also induces conformational shift. allowing development of active form of the enzyme.
A stockperson has a significant influence on the productivity and welfare of his animals depending on his stockmanship competence. In this study, stockmanship competence (SC) is defined as the capacity of the stockperson to ensure the welfare of his animals by providing his animals' needs. The study was conducted to evaluate the SC of backyard goat raisers and examine its relationship to goat productivity and economic profitability. There were 101 respondents for this study who have all undertaken farmer livestock school on integrated goat management (FLS-IGM). Interview was conducted in Region I, Philippines on September 3 to 30, 2012 and March 4 to 17, 2013. Data on SC, goat productivity and farmer's income were gathered. Questions regarding SC were formulated based on the Philippine Recommendations for Goat Production and from other scientific literature. Housing, feeding, breeding and health and husbandry management were the indicators used in computing stockmanship competence index score (SCIS). Pearson correlation using Statistical Package for Social Science (SPSS) was carried out to analyse the relationship between SCIS, productivity and income. Based from the results of the study, a majority of the respondents raised native and upgraded goats. The computed mean SCIS before and after undergoing FLS-IGM were 38.52% and 75.81% respectively, a percentage difference of 65.23%. Both index scores resulted in significant differences in productivity and income. The median mature weight and mortality rate of goats before FLS-IGM was 14 kg and 30% respectively. After FLS-IGM, median mature weight was 19 kg and mortality rate decreased from 30% to 11.11%. Likewise, fewer goat diseases were observed by farmers who were able to undergo FLS-IGM. With regard to income, there was a 127.34% difference on the median net income derived by farmers. Result implies that improved SC could lead not only to increased productivity and income of backyard goat raisers but also to better animal welfare.
Breast milk production and factors related to milk production were examined by test-weighting method in the part of Kangwon Province longitudinally from 0.5 to 5 months of postpartum. Milk production of total mothers averaged 639, 789, 871, 843, 848 and 851g/day at 0.5, 1, 2, 3, 4 and 5 months of postpartum respectively. Mean Milk production of multiparae appeared significantly higher than those of primiparac. The number of feed per day showed decreasing trend from 9.6 to 8.4 during the lactation. Mature milk production had a correlation with transitional milk production and no correlation with infants weight at birth, gestational period and weight gain during pregnancy of mothers. The energy, protein and lipid consumption of total mothers had a positive correlation with breast milk production.
This study describes the growth of broiler chickens to the two forms of Gompertz function for application in broiler production models. The first form is based on the estimated mature weight ($W_A$), while the second is based on the estimated hatch weight ($W_O$). Both equations gave identical estimation because they are mathematically identical. To fit the growth curve of commercial broilers that marketed at 35-42 days, it is unnecessary to keep broilers to near maturity (> day 140) to obtain growth data for deriving the Gompertz function. This date does not improve the curve fitting of the early growing period. Additionally, a high mortality and health problem occurred to this type of chicken after day 105.
Lipoxygenase and Kunitz trypsin inhibitor protein of mature soybean [Glycine max (L.) Merr.] seed are main anti-nutritional factors in soybean seed. A new soybean cultivar, "Gaechuck#1" with the traits of black seed coat, green cotyledon, lipoxygenase2,3 and Kunitz trypsin inhibitor protein free was developed. It was selected from the population derived the cross of "Gyeongsang#1" and C242. Plants of "Gaechuck#1" have a determinate growth habit with purple flowers, brown pubescence, black seed coat, black hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on dry weight basis have averaged 39.1% and 16.2%, respectively. It has shown resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. "Gaechuck#1" matured on 5-10 October with a plant height of 50 cm. The 100-seed weight of "Gaechuck#1" was 23.2g. Yield of "Gaechuck#1" was averaged 2.2 ton/ha from 2005 to 2007.
Lipoxygenase and Kunitz trypsin inhibitor protein are the main antinutritional factor in mature soybean seed. A new soybean cultivar, "Gaechuck#2" with yellow seed coat, lipoxygenase2,3-free and Kunitz trypsin inhibitor protein-free was developed. It was selected from the population derived from the cross between "Jinpumkong2ho" and C242. Plants of "Gaechuck#2" have determinate growth habit with purple flowers, tawny pubescence, yellow seed coat, yellow hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on a dry weight basis were 40.7% and 18.7%, respectively. It has shown a resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. Gaechuck#2 matured in 4 October with plant height of 54cm and a 100-seed weight of 24.4g. Average Yield of Gaechuck#2 was 230 - 250 kg/10a in 2005 - 2007.

This study was carried out to investigate the effect of stem-cutting (SC) length and portion on the major agronomic characters related with dry matter production and accumulation, and to examine their relationships in cassava. When cassava was planted with longer SCs or with older portion SCs, major canopy or source characters like leaf number and leaf area index developed excessively, while tuber yields could be reduced due to the decrease of root/shoot ratio, relative growth rate, root dry weight, and harvest index, particularly in the bitter varieties (high cyanide-level varieties). It was considered that the sweet varieties (low cyanide-level varieties) be earlybulked with higher tuber yield when they are planted with the 25-30 cm long SCs or with the SCs from young type to semi-mature portions of mother stems, while the bitter varieties with the 15-20 cm long SCs or with the SCs from semi-mature to hardwood portions of mother stems, respectively. However, a significant interaction between length and portion of SC was not observed in all agronomic characters.
So as to compare the differences of major agronomic traits according to fall plant types, two cultivars and eight lines which were derived from the same pedigree, were planted and analyzed. The heading and mature times, and culum length of erect plant types were earlier or longer, but the spike number per ㎡ and yield were smaller than the those of prostrate types. Also, 1000 grain weith, assortment rate and light penetration rate in erect types were greater than in prostrate types. The heading time was positively correlated with mature time and yield, but negatively correlated with culum length, 1000 grain weight, assortment rate and light penetration rate. Yield was negatively correlated with light penetration rate. If the lines with prostrate types in fall season, fast regrowth after wintering and rapid elongation of stem were bred and selected, it will be of great benefit in the area requiring sever winter hardiness.
The purpose of this study was to identify preteen girls and their mothers' apparel image preference and apparel purchase behavior. This study was particularly interested in verifying the influencing factors of plus size preteen girls' apparel choice by investigating the correlations between preteens' physical characteristic and their apparel related variables. Data were obtained from the questionnaire by 190 preteen girls and 120 mothers. The influential power of preteens on apparel purchase decision was greater than that of their mothers, and their preferred apparel images were comfortable/neat, cute/girlish, and mature/chic. Comparing to preteen girls, their mothers preferred comfortable/neat and cute/girlish images to mature/chic apparel image. Their physical maturity levels were significantly correlated with apparel image preference. Moreover, plus size preteen girls' interests in appearance and clothing was higher than that of normal weight preteen girls, this implied potentiality in apparel market for plus size preteen.
